  • 한국에 자생하는 초롱꽃식물은 분화 및 조경용으로써의 이용가치가 높은 식물이다. 본 연구는 애기초롱꽃(C. punctata Lam.)을 모본으로, 자주초롱꽃(C. punctata Lam. var. rubriflora Mak.)을 부본으로 하여 표현형이 왜화된 $F_1$ 식물 '직녀'를 육 성하였다. 2001년 6월에 교배 시작하여 10월에 채종하였다. 이후 $5^{\circ}C$ 저온저장 후 2002년 3월에 파종하였다. $F_1$ 품종 특성으로는 화색은 자주색이며 초장, 엽장, 엽폭, 화경 등에서 각각의 모본에 비해 중간형질을 보였다. 개화특성은 각 소화경의 개화기간은 평균 14일이며, 식물체 전체 개화기간은 4개월이었고, 재배 시 추대를 위한 저온감응을 꼭 필요로 하는 식물로써 관수 시 과습은 피하는 것이 좋다.

Genetic Parameter Estimation in Seedstock Swine Population for Growth Performances

  • The objective of this study was to estimate genetic parameters that are to be used for across-herd genetic evaluations of seed stock pigs at GGP level. Performance data with pedigree information collected from swine breeder farms in Korea were provided by Korea Animal Improvement Association (AIAK). Performance data were composed of final body weights at test days and ultrasound measures of back fat thickness (BF), rib eye area (EMA) and retail cut percentage (RCP). Breeds of swine tested were Landrace, Yorkshire and Duroc. Days to 90 kg body weight (DAYS90) were estimated with linear function of age and ADG calculated from body weights at test days. Ultrasound measures were taken with A-mode ultrasound scanners by trained technicians. Number of performance records after censoring outliers and keeping records pigs only born from year 2000 were of 78,068 Duroc pigs, 101,821 Landrace pigs and 281,421 Yorkshire pigs. Models included contemporary groups defined by the same herd and the same seasons of births of the same year, which was regarded as fixed along with the effect of sex for all traits and body weight at test day as a linear covariate for ultrasound measures. REML estimation was processed with REMLF90 program. Heritability estimates were 0.40, 0.32, 0.21 0.39 for DAYS90, ADG, BF, EMA, RCP, respectively for Duroc population. Respective heritability estimates for Landrace population were 0.43, 0.41, 0.22, and 0.43 and for Yorkshire population were 0.36, 0.38, 0.22, and 0.42. Genetic correlation coefficients of DAYS90 with BF, EMA, or RCP were estimated to be 0.00 to 0.09, -0.15 to -0.25, 0.22 to 0.28, respectively for three breeds populations. Genetic correlation coefficients estimated between BF and EMA was -0.33 to -0.39. Genetic correlation coefficient estimated between BF and RCP was high and negative (-0.78 to -0.85) but the environmental correlation coefficients between these two traits was medium and negative (near -0.35), which describes a highly correlated genetic response to selection on one or the other of these traits. Genetic Trends of all three breeds tend to be towards bigger EMA or greater RCP and shorter DAYS90 especially from generations born after year 2000.

  • Quantitative trait loci, a genetic concept for explaining the inheritance of non-Mendelian traits in 1940s, have been realized as particular fragments of chromosome even unique genes in most crops in 21st century. However, only very a small portion of QTL has been screened out by geneticists comparing to a great number of genes underneath the quantitative traits. These identified QTL even have been seldom used into breeding program because crop breeders may not find the QTL in their breeding populations in their field station. Several key points will be proposed to meet the challenges of QTL analysis today: a fine mapping population and the related reference genetic map, QTL evaluation in multiple environments, recognizing real QTL with small genetic effect, map integration.

식물유전 및 육종학 연구에서의 분자생물학적 마커기술의 이용 (Utilization of Molecular Markers in Plant Genetics and Breeding)

  • The understanding on the plant genome is accelerated with the fast advance of molecular biological techniques. The molecular dissecting of the plant genome has made possible the precise genotyping the plants, which can be utilized for molecular breeding program. As well, the molecular cloning of genes interested can facilitate the process of gene transfer between intra-and inter-generic taxa. Moreover, the manipulation of the agronomically important QTL genes, which can be rarely performed by the conventional genetic methods, is also possible by the utilization of molecular markers. In addition to these genetical applications, molecular markers are useful in the areas of plant taxonomy and management of germplasm by fingerprinting analysis. This paper describes the theoretical aspects marker technologies and practical applications of each marker technique.

버어리종 연초 곁순발생의 품종간 차이 (Varietal Difference of Sucker producing Characteristics in Burley Tobacco(Nicotiana tabacum L. var. burley))

  • This study was carried out to obtain the basic information about the sucker producing characteristics of certain burley tobacco varieties(Nicotiana tabacum L. var. burley) at Suwon and Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 1999. The number and weight of suckers for 12 varieties or breeding lines were investigated three times, 45 days after transplanting for ground sucker, 10 days after topping, and 10 days after maleic hydrazide(MH) applying for upstalk sucker. KB 103 and KB 9416-1 produced less ground suckers and upstalk suckers than other entries did. KB 9210-8 produced less ground suckers and (Male sterile TC 612 x KB 108)F$_1$ also produced less upstalk suckers than other entries did. There was no differences among entries in sucker production at 10 days after MH treatment. Between the weight of total sucker and days to flower revealed the negative correlation, but not significant. The low sucker producing entries described above could be used as a parental line in the breeding program for the low sucker producing variety.

한국 환경농업의 성공적 정착을 위한 정책적 및 기술적 접근과제 (Technical and Political Approaches for Successful Settling of the Environmental Agriculture System in Korea)

  • This study aims to point out the basic strategy of agrotechnical and -political approaches toward successful settling for the environmental agriculture system in Korea. Although environmental agriculture & forestry strategy toward the 21st century and it's action program were released in 1996, some scientist from environmental agriculture and organic agriculture still argue on the errors of it's strategy and action program in terms of environmental sound functions. Especially it's strategy and action program of government did not focus the reduction of fertilizer and pesticide as the matters of weight and importance to reduction of fertilizer and pesticide as the matters of weight and importance to realize an environmental sound agriculture system. And korean organic agriculture gets a point in dispute, because 1) they do not practice of Basic Standard of IFOAM and FAO/WHO Codex draft, and 2) organic farming is defined quite differently from internationally recognized core aspects of organic agriculture. Organic farming, in Korea, is taken to mean the replacement of chemical fertilizer by organic manure and avoidance of agricultural chemicals. In the paper it is discussed the necessity, purpose, effect and consequence of basic standard for korean organic agriculture. The reformation of the certification system and t he direct payment system in environmental agriculture & forestry strategy toward the 21st century and it's action program by government, and on the need of the teaching program and curriculum in high education institution, the breeding program of resistant cultivars, the soil testing for optimum fertilization strategy, the nutrient management program, the declaration for 2020 environemntal agriculture are also discussed.

Inbreeding Levels and Pedigree Structure of Landrace, Yorkshire and Duroc Populations of Major Swine Breeding Farms in Republic of Korea

  • The registration data of 15 populations from nine major swine breeding farms were investigated to check levels of inbreeding and the current status of pedigree structures of breeding stocks. The average rate of inbreeding per generation was 0.208%, 0.209%, 0.098%, 0.307% and 0.071% for farms D, S, K, H, and Y in Duroc, 0.071%, 0.188%, 0.685%, 0.336%, and 0.449% for farms S, H, C, J, and W in Landrace, and 0.243%, 0.123%, 0.103%, 0.165%, and 0.286% for farms D, S, G, H, and J in Yorkshire, respectively. The average inbreeding rate was highest for Landrace, intermediate for Yorkshire, and lowest for Duroc farms. In Landrace and Yorkshire populations there were few immigrant animals per generation. In Duroc, however, there were quite large numbers of immigrant animals per generation compared to other breeds. The effective population sizes calculated from the average rate of inbreeding were distributed between 73.0 and 708.7. Specific values were 204.8, 239.7, 508.8, 163.0 and 708.2 for farms D, S, K, H, and Y in Duroc, 708.7, 266.5, 73.0, 148.9, and 111.3 for farms S, H, C, J, and W in Landrace, and 205.5, 406.0, 486.9, 302.6 and 175.0 for farms D, S, G, H, and J in Yorkshire, respectively. The values were acceptable for natural selection for fitness and inbreeding depression. The results showed that there was no cause for concern over the current inbreeding level of major swine breeding farm populations and the inbreeding level was within an acceptable range.

Proteome Analysis of Disease Resistance against Ralstonia solanacearum in Potato Cultivar CT206-10

  • Potato is one of the most important crops worldwide. Its commercial cultivars are highly susceptible to many fungal and bacterial diseases. Among these, bacterial wilt caused by Ralstonia solanacearum causes significant yield loss. In the present study, integrated proteomics and genomics approaches were used in order to identify bacterial wilt resistant genes from Rs resistance potato cultivar CT-206-10. 2-DE and MALDI-TOF/TOF-MS analysis identified eight differentially abundant proteins including glycine-rich RNA binding protein (GRP), tomato stress induced-1 (TSI-1) protein, pathogenesis-related (STH-2) protein and pentatricopeptide repeat containing (PPR) protein in response to Rs infection. Further, semi-quantitative RT-PCR identified up-regulation in transcript levels of all these genes upon Rs infection. Taken together, our results showed the involvement of the identified proteins in the Rs stress tolerance in potato. In the future, it would be interesting to raise the transgenic plants to further validate their involvement in resistance against Rs in potato.

Genetic diversity analysis of high yielding rice (Oryza sativa) varieties cultivated in Bangladesh

  • Investigation of genetic diversity and molecular characterization in high yielding rice varieties is important for their identification. The experiment was conducted during 2016 - 2017 to analyse the genetic diversity of fifteen high yielding rice varieties in Bangladesh by using random amplification of polymorphic DNA (RAPD) markers. Polymorphism was revealed in 12 RAPD primers out of 30, whereas no other reaction was detected on the remaining 18 primers. The 40 out of 45 bands (88.89%) polymorphics were produced by the primers and ranged from 50 to 100%. The maximum number of polymorphic bands was produced by the primer OPB-18 whereas the lowest number of polymorphic bands belonged to OPC-12. The genetic similarity coefficients were determined with the RAPD data, which ranged from 0.47 to 0.94. The unweighted paired group of arithmetic means (UPGMA) dendrogram presented the studied rice varieties into two major clusters. Moreover, the value of Nei's genetic diversity is 0.26 and the Shanon information index is 0.41. The study produced distinct positions, suggesting that the genotypes were different from each other. The results indicated that these markers could be efficient for comparing the genetic relationships, patterns of variation, and measurement of genetic distance among rice varieties. Considering all of these results, RAPD analysis is found to be an effective tool for estimating the genetic diversity of different rice varieties. The outcomes of this research may contribute to the germplasm data of rice accessions and a future breeding program of rice genotypes.

Pathogenicity Determinants of Sclerotinia sclerotiorum and Their Association to Its Aggressiveness on Brassica juncea

  • White rot or stem rot caused by Sclerotinia sclerotiorum is one of the most destructive fungal diseases that have become a serious threat to the successful cultivation of oilseed Brassicas. The study was designed with an aim to investigate the association between the pathogenic aggressiveness and pathogenicity determinants of this pathogen specifically in Brassica for the first time. For this, a total of 58 isolates of S. sclerotiorum from different geographical regions were collected and purified. These isolates were inoculated on a Brassica juncea cv. RL-1359 and they exhibited high level of variation in their disease progression. The isolates were grouped and then 24 isolates were selected for the biochemical analysis of pathogenicity determinants. The isolates varied significantly with respect to their total organic acids, oxalic acid production and pectin methyl esterase and polygalacturonase activity. The oxalic acid production corresponded to the disease progression of the isolates; the isolates with higher oxalic acid production were the more aggressive ones and vice-versa. This is, in our knowledge, the first study to establish a correlation between oxalic acid production and pathogenic aggressiveness of S. sclerotiorum on B. juncea. However, the pectinases' enzyme activity did not follow the trend as of disease progression. These suggest an indispensable role of oxalic acid in pathogenicity of the fungus and the potential to be used as biochemical marker for preliminary assessment of pathogenic aggressiveness of various isolates before incorporating them in a breeding program.
