Effect of Crab Shell Supplementation in Feeds on Taurine Content of Cow s Milk (게껍질의 급여가 우유내 타우린함량에 미치는 영향)

  • 본 논문에서는 게껍질분말 급여에 의한 천연 타우린 강화우유의 생산 가능성을 평가하고자 25두의 젖소를 5군으로 나누고, 각기 대조사료, 1%(1%CS), 3%(3%CS) 및 5% 게껍질분말사료(5%CS), 그리고 formaldehyde로 처리되 3% 게껍질분말사료 (3%FCS)를 30일간 급여하였다. 게껍질분말 단백질의 아미노산조성은 phenylalanine이 가장 높았고, glutamate, aspartate, glycine, serine histidine, arginine의 순으로 나타났으며, 다른 동물조직의 단백질과는 달리 branched-chain 아미노산의 함량이 비교적 낮았다. 게껍질에 함유된 유리아미노산 중 타우린은 가장 고농도로 존재하였으며, 특히 집게다리껍질에 509$\mu$mol/100 g이 함유되어 등뚜껑(319$\mu$mol/100g) 및 게껍질분말(296 $\mu$mol/100 g)에 비해 월등히 그 함량이 높았다. 게껍질분말의 급여가 우유의 타우린농도에 미치는 영향을 평가한 결과 p<0.01 수준에서 유의성이 관찰되었다. 1%CS군의 원유에는 5.84$\pm$0.73 $\mu$mole/100mL의 타우린이 함유되어 대조구 (4.84$\pm$0.29 $\mu$mol/100mL)에 비해 20%정도 더 높았으며, 3%CS군의 원유에는 7.21$\pm$0.77$\mu$mole/100 mL의 타우린이 함유되어 대조구에 비해 49%정도 유의적으로 증가하였다.(p<0.01). 한편, 3%FCS군의 경우 원유의 타우린농도가 3%CS군에 비해 유?거으로 감소하였고, 따라서 게껍질의 formaldehyde 처리는 반추위내에서 타우린을 보호하는데 효과적이지 못한 것으로 나타났다. 본 논문은 게껍질 급여에 의해 우유의 타우린농도가 증가하였음을 밝히는 최초의 보고이며, 본 연구의 결과는 앞으로 기타 축산제품의 타우린함량을 증가시키는데 폭넓게 응용되어질 수 있을 것이다.

Effect of Intraruminal Sucrose Infusion on Volatile Fatty Acid Production and Microbial Protein Synthesis in Sheep

  • Effects of sucrose supplement on the pattern of VFA production and microbial protein synthesis in the rumen were examined in sheep consuming basal diet of grass silage (2.5 kg fresh wt/d) that was provided in 24 equal meals each day by an automatic feeder. Four mature wethers were allocated to four experimental treatments in a 4${\times}$4 Latin square design with periods lasting 14 days. The treatments were (1) the basal diet, (2) supplemented with 150 g sucrose and 7.0 g urea, (3) 300 g sucrose and 13 g urea, and (4) 450 g sucrose and 20 g urea given as a continuous intraruminal infusion for 24 h. All infusions were given in 2 litres of aqueous solution per day using a peristaltic pump. The effect of sucrose level on rumen mean pH was significantly linear (p<0.01). There were not significant differences in the concentration of ammonia-N, total VFA and the molar proportions of acetate, propionate and butyrate with the level of sucrose infusion. The molar proportions of isobutyric acid (p<0.05) and isovaleric acid (p<0.001) were significantly reduced when the infused amount of sucrose was increased. The flow of microbial N was linearly (p<0.001) increased with sucrose and urea level. High levels of readily fermentable carbohydrate in a ration reduced the efficiency of microbial protein synthesis in the rumen. It was demonstrated that of the individual fatty acids, only the molar proportion of isovalerate showed a significant negative correlation (R2=$0.3501^{**}$) with the amount of microbial N produced and a significant positive correlation (R2=$0.2735^{**}$) with the efficiency of microbial growth.

Compositions of Fatty Acids, Inorganic Components and Volatile Organic Acids in Korean Valerian Roots (한국산 쥐오줌풀의 지방산, 무기성분 및 유기산 조성)

  • This study was carried out to investigate the compositions of fatty acids, inorganic components and volatile organic acids from Korean valerian roots, Valeriana fauriei var. dasycarpa Hara and Valeriana officinalis var. latifolia Miq. The contents of total lipids ranged from 3.7 % to 4.5% and the major fatty acids were linoleic, linolenic and palmitic acid. Ash contents ranged from 4.3% to 6.3%. and the contents of Na, Fe, Zn and Cu showed some quantitative differences depending upon grown region or species. Fourty components were identified from acidic fraction of dichloromethane extract of V. fauriei var. dasycarpa Hara, of which the major components were 3-methyl butanoic, dimethoxy-2-propenoic. 3, 4-dimethoxy benzoic and 3-methyl pentanoic acid.

Novel Naphthalene Based Lariat-Type Crown Ethers Using Direct Single Electron Transfer Photochemical Strategy

  • This study explored a direct SET-photochemical strategy to construct a new family of thioene conjugated-naphthalamide fluorophore based lariat-crown ethers which show strong binding properties towards heavy metal ions. Irradiations of designed nitrogen branched (trimethylsilyl)methylthio-terminated polyethylenoxy-tethered naphthalimides in acidic methanol solutions have led to highly efficient photocyclization reactions to generate naphthalamide based lariat type thiadiazacrown ethers directly in chemo- and regio-selective manners which undergo very facile secondary dehydration reactions during separation processes to produce their corresponding amidoenethio ether cyclic products tethered with electron donating diethyleneoxy- and diethyenethio-side arm chains. Fluorescence and metal cation binding properties of the lariat type enamidothio products were examined. The photocyclized amidoenethio products, thioene conjugated naphthalamide fluorophore containing lariat-thiadiazacrowns exhibited strong fluorescence emissions in region of 330-450 nm along with intramolecular exciplex emissions in region of 450-560 nm with their maxima at 508 nm. Divalent cation $Hg^{2+}$ and $Pb^{2+}$ showed strong binding to sulfur atom(s) in side arm chain and atoms in enethiadiazacrown ether rings which led to significant enhancement of fluorescence from its chromophore singlet excited state and concomitant quenching of exciplex emission. The dual fluorescence emission responses towards divalent cations might provide a new guide for design and development of fluorescence sensors for detecting those metals.

Expression in Escherichia coli of a Putative Human Acetohydroxyacid Synthase

  • A human gene has been reported that may encode the enzyme acetohydroxyacid synthase. Previously this enzyme was thought to be absent from animals although it is present in plants and many microorganisms. In plants, this enzyme is the target of a number of commercial herbicides and the use of these compounds may need to be reassessed if the human enzyme exists and proves to be susceptible to inhibition. Here we report the construction of several plasmid vectors containing the cDNA sequence for this protein, and their expression in Escherichia coli. High levels of expression were observed, but most of the protein proved to be insoluble. The small amounts of soluble protein contained little or no acetohydroxyacid synthase activity. Attempts to refold the insoluble protein were successful insofar as the protein became soluble. However, the refolded protein did not gain any acetohydroxyacid synthase activity. In vivo complementation tests of an E. coli mutant produced no evidence that the protein is active. Incorrect folding, or the lack of another subunit, may explain the data but we favor the interpretation that this gene does not encode an acetohydroxyacid synthase.

Characterization of KI-24, a Novel Murine Monoclonal Antibody with Specific Reactivity for the Human Immunodeficiency Virus-1 p24 Protein

  • The HIV-1 p24(202-221) sequence ETINNEEEWDRVHPV HAGP contains a B-cell epitope with the earliest immune response and the highest antibody titer against anti-mouse sera obtained by immunization with p24 antigens. A novel mouse monoclonal antibody (mAb) was generated against the immunodominant B-cell epitope of the HIV-1 p24 capsid protein, p24(202-221). BALB/c mice were immunized with the four branched multiple antigenic peptide (MAP) containing the HIV-1p24(202-221) sequence, and antibody-secreting hybridoma were produced by fusion of mouse splenocytes with P3X63Ag8.653, mouse myeloma cells. One clone which produced the antigen-specific mAb named KI-24 (Isotype IgG1, light chain: ${\kappa}$) was identified. mAb KI-24 was highly specific for both the p24(202-221) and p24 proteins when analyzed by ELISA and Western blotting. Since p24(202-221) also contains a cytotoxic T-lymphocyte epitope, this specfic peptide epitope and the monoclonal antibody with specific reactivity against the p24 protein and p24(202-221) can be used in peptide vaccine development and p24 antigen detection from HIV patients.

Influence of Isobutyric Acid Supplementation on Nutrient Intake, Its Utilization, Blood Metabolites and Growth Performance of Crossbred Calves Fed Wheat Straw Based Low Protein Diets

  • The effects of dietary supplementation with the sodium salt of isobutyric acid in a low protein (10% CP) wheat straw based diet on nutrient utilization, blood metabolites and growth performance were studied with male crossbred calves. The calves were divided into two equal groups of 6 each. The animals of the control group were fed a basal diet consisting of wheat straw, concentrate mixture and green oat fodder in 40:40:20 proportion whereas BCFA supplemented group received the basal diet+isobutyric acid at 0.75 percent of basal diet. The duration of study was 120 days. The feed intake between experimental groups did not differ significantly and the average total DMI (% BW) was 1.99 and $1.95kg\;day^{-1}$ in control and BCFA supplemented diets. The dietary supplementation of BCFA improved (p<0.01) the DM, OM, CP (p<0.05), NDF and cellulose digestibilities by 8.50, 9.01, 5.39, 17.78 and 18.44 per cent over those fed control diet. The total N retention on BCFA supplementation was improved (p<0.01) due to the decreased (p<0.05) faecal N excretion. The BCFA supplementation did not alter the blood circulatory levels of glucose, total protein, albumin, urea N and amino acids. However after 120 days of experimental feeding a significant (p<0.05) increase in the concentrations of non-esterified fatty acid was observed in control group. The DCP intake and the DCP content of experimental diets was similar in both groups. However, the TDN content of BCFA supplemented diet was significantly (p<0.01) higher (64.35%) than that of control (59.60%). The total live weight gain in BCFA supplemented diet increased by 15.94% over control. The average daily gain and efficiency of feed conversion were also improved in BCFA fed calves by 13.38 and 26.71% respectively, compared to control. It is concluded that dietary supplementation with isobutyric acid improved the digestibility of nutrients and growth performance of calves.

Chemical Composition, Nitrogen Fractions and Amino Acids Profile of Milk from Different Animal Species

  • Milk composition is an imperative aspect which influences the quality of dairy products. The objective of study was to compare the chemical composition, nitrogen fractions and amino acids profile of milk from buffalo, cow, sheep, goat, and camel. Sheep milk was found to be highest in fat ($6.82%{\pm}0.04%$), solid-not-fat ($11.24%{\pm}0.02%$), total solids ($18.05%{\pm}0.05%$), protein ($5.15%{\pm}0.06%$) and casein ($3.87%{\pm}0.04%$) contents followed by buffalo milk. Maximum whey proteins were observed in camel milk ($0.80%{\pm}0.03%$), buffalo ($0.68%{\pm}0.02%$) and sheep ($0.66%{\pm}0.02%$) milk. The non-protein-nitrogen contents varied from 0.33% to 0.62% among different milk species. The highest r-values were recorded for correlations between crude protein and casein in buffalo (r = 0.82), cow (r = 0.88), sheep (r = 0.86) and goat milk (r = 0.98). The caseins and whey proteins were also positively correlated with true proteins in all milk species. A favorable balance of branched-chain amino acids; leucine, isoleucine, and valine were found both in casein and whey proteins. Leucine content was highest in cow ($108{\pm}2.3mg/g$), camel ($96{\pm}2.2mg/g$) and buffalo ($90{\pm}2.4mg/g$) milk caseins. Maximum concentrations of isoleucine, phenylalanine, and histidine were noticed in goat milk caseins. Glutamic acid and proline were dominant among non-essential amino acids. Conclusively, current exploration is important for milk processors to design nutritious and consistent quality end products.

Multisystemic Aspergillosis with Granulomas in Layer Chickens (산란계에서의 전신 다발성 육아종성 Aspergillosis 발생)

  • A case of aspergillosis in 39-day-old layer chickens having a history of gradual emaciation and subsequently death with nervous signs such as torticollis and lack of equilibrium was documented. Based on the results from serology and polymerase chain reaction (PCR) test, this flock was not affected with known viral or bacterial diseases. On postmortem examination of the affected birds, multiple white to yellow nodules measuring 1~5 mm in diameter were observed in the lungs, cerebrum, liver and kidney. Microscopically, these nodules were identified as granulomatous lesions characterized by mixed population of multinucleated giant cells and lymphocytes. By periodic acid-schiff staining and nucleotide sequencing analysis, Aspergillus flavus with characteristic septate and branched hyphae were identified in the granuloma of lung and cerebrum. This case was a chronic and multisystemic aspergillosis specialized to central nervous system caused by Aspergillus flavus infection in the layer flocks.

Chemotaxonomic and Phylogenetic Study on the Oligotrophic Bacteria Isolated from Forest Soil

  • Oligotrophic bacteria isolated from forest soil showed a specific community consisting of various taxonomic groups compared with those in other soil or aquatic habitats. Based on the cell shape, the isolates were divided into four groups: regular rod, curved/spiral rod, irregular rod, and prosthecate bacteria. The cellular fatty acids 60 oligotrophic isolates were analyzed. The 30 fatty acids which were identified or characterized are classified. At the dendrogram based on cellular fatty acid composition, four clusters(I-IV) were separated at a euclidian distance of about 50. Cluster 3 and 4-a strains were containing Q-8, these strains are accommodated in the Proteobacteria gamma and beta subdivision. The chemotaxonomic profiles of the cluster 4-a strains showed good agreement with those of the genus Burkholderia. Cluster 3 was characterized by the presence of branched-chain fatty acids, iso-C15:0, iso-C17:1, and iso-C17:0 as the major components. These chemotaxonomy suggested the close relationship of the isolates with Xathomonas/Sterotrophomonas group. Based on the 16S rDNA sequence analysis, the two representative strains(MH256 and MA828) of cluster 3 showed the close relation to genera, Xathomonas/Sterotrophomonas, but were not included in these genera. These strains were even further away from core Xanthomonas, and clearly were seen to branch outside the cluster formed by the Sterotrophomonas maltophilia. MH256 and MA828 16S rDNA sequence was different enough to put new genus on a separate branch. The isolates with Q-10 were also studied. They are corresponded to the two large groups in Proteobacteria alpha subdivision. One was incorporated in the genus Bradyrhizobium cluster, which also includes Agromonas, a genus for oligotrophic bacteria. The strains of the other group showed high similarity to the genus Agrobacterium.
