제주 미천동굴 고사릿과 자생식물의 생태 고찰 (I) (Ecological Contemplation on Native Plant of Bracken at Micheon Cave in Je-ju Island)

  • There are many beautiful volcano caves in Je-ju island which is biggest volcanic island in Korea. The typical volcano caves are ManJang-gul, HyupJe-gul and MiCheon-gul, etc. Among them the MiCheon-gul name is originated from the beautiful things of cave which were more than thousands in there. This MiCheon-gul cave has another feature in special. It is about a native plant bracken living among the various live creatures and plants in MiCheon-gul cave, however, it is growing contrary under the vertex of the cave grown at retro-gravity, as one of the famous things of the proud cave. Therefore, it was contemplated and investigated on the habitat of a mode of life and ecological adaptation on the native plant of bracken include the geomorphic and geographic features of the MiCheon-gul cave.

Bracken-fern Extracts Induce Cell Cycle Arrest and Apoptosis in Certain Cancer Cell Lines

  • Bracken fern [Pteridium aquilinem (L.) kuhn (Dennstaedtiaceae)] is one of the most common species on the planet. It has been consumed by humans and animals for centuries. Use by some human groups is because they believe bracken fern is good for health as plant medicine. However, it is also one of the few known plants that can cause tumors in farm animals. Many interested groups have focused their attention on bracken fern because of these interesting features. In order to evaluate the biological effects of exposure to this plant in cellular level, human cancer cell lines were treated with the fern dichloromethane extracts and the genotoxic and cytotoxic effects were studied. Anti-proliferative/cytotoxic effects were evaluated by cell count, MTT assay and flow cytometry methods with three different cancer cell lines, TCC, NTERA2, and MCF-7, and two normal cells, HDF1 and HFF3. Pro-apoptotic effects of the extracts were determined by DAPI staining and comet assay, on TCC cancer cells compared to the normal control cell lines. Cellular morphology was examined by light microscopy. Our present study showed that the extract caused DNA damage and apoptosis at high concentrations ($200{\mu}g/mL$) and also it may induce cell cycle arrest (G2/M phase) at mild concentrations (50 and $30{\mu}g/mL$) depending on the cell type and tumor origin. These results indicate that bracken fern extract is a potent source of anticancer compounds that could be utilized pharmaceutically.

남해산 고사리의 이화학적 특성 및 항산화 활성 (Physicochemical Characteristics and Antioxidant Activity of Bracken (Pteridium aquilinum Kuhn) in Namhae)

  • In this study, we investigate the physicochemical characteristics of dried bracken (Pteridium aquilinum Kuhn) harvested in Namhae. The moisture, ash, crude protein and crude lipid content was $10.79{\pm}0.31%$, $6.16{\pm}0.04%$, $33.20{\pm}0.40%$ and $2.45{\pm}0.27%$, respectively. The total mineral (Ca, Fe, K, Mg, Na, Mn) content was $36720.1{\pm}495.7mg/kg$, with K being the highest at $31890.0{\pm}503.8mg/kg$. The total free amino acid content was 704.41 mg/100 g, the amount of methionine, citrulline and sarcosine being higher than the others. The water and 50% ethanol extracts from the dried bracken were evaluated for the total poly phenolic compound content, antioxidant activity and xanthine oxidase and ${\alpha}$-glucosidase inhibitory activity. The total polyphenol content in the 50% ethanol extract ($1574.86{\pm}18.31mg/100g$) was higher than in the water extract ($1240.24{\pm}16.32mg/100g$). The DPPH and ABT radical scavenging activity was increased according to the concentration of bracken extract, and the activity in the 50% ethanol extract was higher than in the water extract. In addition, the xanthine oxidase and ${\alpha}$-glucosidase inhibitory activity was also higher in the 50% ethanol extract than in the water extract. In conclusion, bracken has great antioxidative and antidiabetic effects and can be used as a preventive agent for oxidation and diabetes.

남서해안 고사리 생육지의 토양화학적 특성과 고사리식물체의 무기성분 함량 (Nutrient Contents of Bracken (Pteridium aquilinum L.) and Soil Chemical Properties of Its Habitat in the Coastal Area)

  • 전남 신안군 내 고사리 재배농가들의 기술지원을 위하여 고사리 재배시험을 수행하면서 고사리 생육지의 토양특성을 조사하였고, 나물용 고사리와 성숙고사리의 식물체를 채취하여 이들의 무기성분 함량들을 분석하였으며 또한 고사리를 재배하고 있는 농가들의 포장토양과 고사리 시료들을 채취하여 분석 검토한 결과는 다음과 같다. 고사리 자생지의 토양은 pH가 5.2, 유기물 19 g $kg^{-1}$, 유효인산 20 mg $kg^{-1}$ 이었고, 치환성 칼륨, 칼슘 및 마그네슘의 함량은 각각 0.32, 2.0, 및 1.3 $cmol_c\;kg^{-1}$이 었으며, 재배지토양의 화학성은 pH가 5.7, 유기물 13 g$kg^{-1}$, 유효인산 367 mg $kg^{-1}$ 이었고, 치환성 칼륨, 칼슘 및 마그네슘의 함량은 각각 0.81, 4.0, 및 1.4 $cmol_c\;kg^{-1}$로 고사리 자생지의 토양 pH, 유효인산 및 치환성 칼슘의 함량이 매우 낮았으며, 유기물함량은 다소 높았다. 자생나물용 고사리의 무기성분 함량은 질소 4.40, 인 0.55, 칼륨 3.40, 칼슘 0.22, 마그네슘 0.32, 나트륨 0.09% 이었고, 철은 126, 망간 210, 구리 23, 아연 75, 붕소 11 mg $kg^{-1}$ 이었으며, 재배고사리의 무기성분 함량 중 질소는 5.50, 인 0.73, 칼륨 3.55, 칼슘 0.17, 마그네슘 0.28, 나트륨 0.07% 이었고, 철은 120, 망간 252, 구리 19, 아연 72, 붕소 20 mg $kg^{-1}$ 이었다. 나물용 고사리와 성숙고사리의 무기성분 함량을 비교해 보면 3요소 성분을 비롯한 대부분의 다량성분과 미량성분 모두 나물용고사리에서 높았고, 고사리가 성숙되어 갈수록 낮아지는 경향을 보였으며, 칼슘과 망간의 함량만은 성숙되어 갈수록 높아지는 경향을 나타내었다. 자생고사리와 재배고사리 간의 무기성분 함량을 비교한 결과 질소, 인, 칼륨 등 3요소 성분은 재배고사리에서 높은 경향이었고, 마그네슘 함량은 자생고사리에서 다소 높은 경향을 보였으며 그 외의 성분들은 일정한 경향을 보이지 않았다. 농가의 고사리 재배에서 재배 1년차와 2년차로 구분하여 토양의 화학성과 고사리 생육을 조사한 결과 토양화학성은 pH가 5.5~5.7, 유기물 함량이 13 g $kg^{-1}$, 유효인산이 222~367 mg $kg^{-1}$, 치환성양이온 중 칼륨 0.40~0.81, 칼슘 3.4~4.0, 마그네슘 1.4~1.6 $cmol_c\;kg^{-1}$이었고, 고사리의 생육은 1년차의 포장은 간장 (지표에서 잎가지가 있는 부분까지)이 25 cm, 입모 수는 13개 $m^{-2}$ 이었으며, 2년차 포장에서는 간장이 89 cm, 입모수가 25개로 조사 되었다.

Didymella sp.에 의한 고사리 신규 마름병 발생 보고 (First Report of a Bracken Blight Disease Caused by Didymella sp.)

  • 최근 4년간 경남 남해군 창선면 일대 고사리 재배 포장에서 마름병 증상을 보이는 새로운 병해가 발생하였고 이 병해로 인하여 해당 지역 고사리 생산량이 해마다 크게 감소하고 있다. 2018년 5월, 7월, 10월에 이병 시료를 채취하여 그로부터 총 92개의 곰팡이 균주를 분리하였다. 이들 균주를 건전한 고사리 잎에 접종하여 총 22개 곰팡이 균주가 포장에서 발생된 병징과 동일한 병징을 일으킴이 확인되었다. 이 중 병원성이 가장 높은 2균주를 선발하여 ribosomal RNA, RNA polymerase ${\beta}$-subunit, ${\beta}$-tubulin 유전자 부위, internal transcribed region을 증폭하였다. 해당 증폭 부위의 염기서열을 이용한 계통수 작성을 통해 병원균 균주는 Didymella속의 clade에 속하며, 기존 균주 중 D. rumicicola 또는 D. acetosellae와 비슷하나 기존 종 수준의 subclade에는 포함되지 않았다. 따라서 고사리 마름병을 일으키는 병원균의 정확한 종 동정을 위해 추가적인 분석이 필요하다. 본 결과는 Didymella sp.에 의한 고사리 마름병 발생 첫 보고이다.

고사리 산업화를 위한 인공종자 개발 및 생산 (Synthetic Seed Development and Production for Industrialization of Eastern Bracken)

  • Ferns have been consumed as food in many countries for centuries. As rich sources of protein, fiber, minerals, vitamins, essential amino acids, and fatty acids, ferns provide important nutrients to humans. Eastern bracken (Pteridium aquilinum var. latiusculum (Desv.) Underw. ex A. Heller) is the most popular edible fern in South Korea where, additionally, it has long been used as an edible wild leaf vegetable. Recently, the production of eastern brackens in South Korea (2018) has reached 14,032 tons, for an annual revenue of 83.5 billion won, and even more eastern brackens are marketed if imports are taken into account as well. Most of the common ferns can be propagated using spores. However, fern farmers cultivate seedlings through traditional propagation methods, such as root pruning or rhizome division. These propagation methods exhibit limitations in forming roots and growing-points and are labor intensive. Quality seedlings of eastern bracken can be obtained through spore propagation, but the spores are fine and difficult to handle in the field. In addition, it would require appropriate environmental control. The production of synthetic seeds using encapsulation technology is easy to establish and it can be used to achieve high productivity at low cost. Synthetic seeds contain explants embedded into a seed foam, and they overcome the limitations of micropropagation and offer the possibility of using plug seedlings. Synthetic seed matrix, such as sodium alginate, has the advantages of low cost, low toxicity, and gel stability. The present study aimed to develop and produce synthetic seeds for the commercial exploitation of eastern bracken. Furthermore, we verified spore germination and the extent of gametophyte and sporophyte development achieved with our new synthetic seeds, whose production was intended to solve current problems with the handling, storage, and transportation of eastern bracken.

수입식품의 품질조사연구원(1) -중국산을 중심으로- (A Study on the Quality Evaluation of Imported Processed Foods(1)- With Special Reference to Chinese Products-)

  • The purpose of present paper is to compare and analyze the quality of imported Chinese products with the Korean local products in terms of proximate composition, character of external appearance , ingredient composition of dried bracken , dried radish cubers, meju , dried pollack, dried mussel , and dried brown seaweed(Undaria puinnatidifida) which were imported between Oct., and Dec. 1992 and distributed in the market. Moisture recovery of dried products resulted in changed shape and incomplete moisture recovery due to an excessive drying in consideration of hygroscopic property in the course of distribution. The amounts of water soluble brown pigment were two times greater in Korean dried pollack and bracken than those of Chinese products. The content of volatile basic nitrogen (VBN) were twice in Chinese dried pollack, and this may be due to foregin odor from a long transport time and distribution process. The contents of aflatoxin in Korean products were not detectable and these of Chinese products were 5 $\mu\textrm{g}$(%) in dried pollack, bracken and radish cuber. the remaining agricultural chemicals were not detectable in all products. Because of poor technique and facilities, the external appearance of Chinese products especially dried mussels and dried pollack were not uniformed in size and color. Discoloration and foregin substance such as hair, dust were checked , Korean products were mostly packed in small size in PP or bag, and Chinese products were packed in bulk size and distributed as a bulk.

식이섬유류(食餌纖維類)의 종류(種類)와 수준(水準)이 흰쥐의 분내 지방대사(脂肪代謝)에 미치는 영향(影響) (Effects of Dietary Fiber Sources and Levels on Lipid Metabolism in Rats)

  • The present study was designed to investigate the effects of dietary fiber sources and levels on serum lipids in rats fed a hypercholesterol diet containing 0.5% cholesterol. Male Sprague-Dawley rats weighing 52.7 ${\pm}$ 7.27g were devided into sevel groups by completely randomized block design and fed various dietary fibers which were prepared from Rice bran, Siraegi (The dried green parts of a radish) and Bracken Fern at a level of 5% and 10% respectively. The results obtained in this study are summarized as follows: 1. Body weight gains were not significantly different among the groups and food intake was significantly higher in rats fed a diet of Siraegi 10% than those fed the control diet. 2. Fecal wet weights were significantly increased in all experimental groups. 3. Levels of serum total cholesterol in all experimental groups were significantly lower than that of the control group except the level in the R 10(Rice bran 10%) group. 4. Free Cholesterol levels in the serum were increased in all experimental groups, especially the levels in the R 10 (Rice bran 10%) and S 10 (Siraegi 10%) groups were significantly higher than that of the control group. 5. Ester Cholesterol levels in the serum were significantly lower in rats fed the experimental diets than those fed the control diet. 6. There was a tendency of higher serum HDL-Cholesterol levels in R5 (Rice bran 5%), R10 (Rice bran 10%) and B5(Bracken 5%) groups compared to the control group however no significant differences were found between the control group and all the experimental groups except in the case of the S5 (Siraegi 5%) group. 7. LDL, VLDL-Cholesterol levels in the serum sere significantly reduced in the S5 (Siraegi 5%), B5 (Bracken 5%) and B10 (Bracken 10%) groups. 8. The HDL-C./Total-C. Ratio was increased in all experimental group especially the Ratios in the S5 and B5 groups were significantly higher than that of the control group.

고사리(Pteridium aquilinum) 뿌리 전분의 이화학적 특성에 관한 연구 -제2보 : 전분의 물리적 특성- (Physico-chemical Properties of Bracken (Pteridium aquilinum) Root Starch - II. Physical Properties-)

  • 고사리뿌리 전분의 이화학적인 특성연구의 일환으로 실시된 물리적 특성시험 결과는 다음과 같다. 전분입자의 미셀구조의 결합력을 보기 위한 흡수도, 팽윤력, 용해도 시험결과 감자전분이나 타피오카전분 보다 강한 결합을 하고 있으며 곡류전분보다는 약하며 팽윤력이 single-stage pattern을 이루고 있었다. 가열에 의한 전분입자의 복굴절성 상실, ${\alpha}$-아밀라아제에 의한 소화성 및 X-선 회절등으로 전분의 호화과정을 살펴본 결과 $55{\sim}60^{\circ}C$에서 호화가 시작되고 $60{\sim}70^{\circ}C$에서는 95%이상 호화되었다. 또한$3{\sim}6%$전분용액의 가열에 따른 점도변화률 amylograph를 사용하여 실험한 결과 pasting temperature $62{\sim}68^{\circ}C$, 최고정도 $80{\sim}840\;BU$, $50^{\circ}C$로 냉각후의 점도는 $110{\sim}555\;BU$였다. 따라서 다른 전분에 비해서 점도가 낮으나 setback이 현저히 느렸다. 수분함량 50%의 전분겔의 경도는 팥 전분보다는 약하고 감자나 밀전분에 비해서는 현저히 높았다. 전분겔의 저장중 노화속도는 밀 전분보다는 느리고 타피오카 전분보다는 빨랐다. 고사리뿌리 전분은 팥 전분겔특성에 큰 변화를 주지않지만 노화를 억제하였고 밀 전분겔에 첨가시에는 겔의 경도가 저하되어 부드러워지는 경향을 보였다.

한국산 고사리의 독성조사에 관한 연구 (A Toxicological Study of Young Fronds of Bracken Fern (Pteridium aquilinum var latiusculum) Collected in Kwang Ju Area)

  • 고사리의 ethanol extract를 독성실험한 결과는 mouse에 시료 10g/kg까지는 급성독$(LD_{50})$을 나타내지 않았다. Rat에 매일 시료40mg/kg, 400mg/kg, 1200mg/kg을 경구투여한 처음 7일간에는 체중, 해부소견, 혈액의 생화학적 검사에서 별이상이 없었다. 14일째 전시료투여군의 체중증가율은 대조군보다 평균 약 7% 감소를 보이고 혈청의 생화학적 측정치가 대부분 약간 증가하였다. 21일째 400mg/kg군과 1200mg/kg군의 체중증가율이 대조군보다 $6.7{\sim}5.7%$ 감소되고 1200mg/kg군 1필이 황색간증세를 보였다. 모든 시험군 GPT 및 GOT는 각각 $88{\sim}102u$$164{\sim}200u$로 증가하고 alkaline phosphatase도 상승하여 중독의 징후를 보였다. 28일째 400mg/kg군과 1200mg/kg군의 체중증가율이 대조군보다 감소되고 이들군에서 각각 $1{\sim}2$필이 황색간을 보였다. 1200mg/kg군 2필이 소장염을 보였다. 모든 시험군은 GPT $92{\sim}116u$와 GOT $56{\sim}215u$ 및 alkaline phosphatase $57{\sim}70u$로 증가 되었다.
