Effect of Temperature and Salinity on Production of Resting Egg in Korean Rotifer, Brachionus plicatilis (L and S-type)

  • Production of resting egg from the Korean rotifer, Brachiunus plicatilis (L and S-type) was investigated at different temperatures (L-type : 20, 24, $28^{\circ}C$, S-type : 28 32, $36^{\circ}C$) and salinities (10, 20,30 ppt). The rotifer was cultured in 25 ml test tube and fed on Nannochloris oculata. With regard to mixis rate, L-type rotifer showed higher rate at lower temperature, and the highest rate was observed at 20 ppt of salinity at each temperature of the experiment. However, for S-type rotifer, the optimum temperature and salinity were $28\~32^{\circ}C$ and 20 ppt, respectively. The highest number of resting egg was 173 eggs/ml in 16 days at $24^{\circ}C$, 10 ppt for L-type rotifer and 410 eggs/ml in 14 days at $28^{\circ}C$, 10 ppt for S-type rotifer. The maximum number of resting egg produced per 10,000 rotifers was 8,122 eggs at $20^{\circ}C$, 20 ppt for L-type rotifer and 8,700 eggs at $28^{\circ}C$, 20 ppt for S-type rotifer. The maximum number of resting egg produced $10^8$ cells of N. oculata was 50.7 eggs for L-type rotifer ($24^{\circ}C$, 20 ppt) and 79.6 eggs for S-type rotifer ($32^{\circ}C$, 10 ppt). The number of resting egg produced per day was $1\~11$ eggs/ml for L-type rotifers and $21\~35$ eggs/ml for S-type rotifer in 9 combination experiments. In this study, S-type rotifer is better than L-type rotifer in resting egg production, and the optimum temperature and salinity for resting egg production were $20^{\circ}C$, 20 ppt for L-type rotifer and $28^{\circ}C$, 20 ppt for S-type rotifer. This result shows the difference of Korean rotifer in the optimum condition for resting egg production from other rotifers reported earlier.

Growth Performance of the Rotifer Brachionus plicatilus and the Larvae of Two Bivalves Fed on the Cryptophyte Teleaulax amphioxeia (은편모류 Teleaulax amphioxeia의 윤충류 Brachionus plicatilus 및 이매패류 유생 2종에 대한 먹이 기능성 평가)

  • The aim of this study was to verify the performance of Teleaulax amphioxeia as prey for Brachionus plicatilis and for the larvae of the bivalves (Pacific oyster Crassostrea gigas and Manila clam Ruditapes philippinarum) to select the best of five T. amphioxeia strains. The five strains of T. amphioxeia were collected from the coasts of Korea, purecultured and then fed to the rotifer and bivalve larvae. The density and fecundity rate of the rotifer fed on strain 01 were the highest. The instantaneous growth rate of the rotifer fed strain 08-2 was the highest. The maximum density and fecundity rate of the rotifer fed strain 08-2 were significantly higher than those fed Chlorella ellipsoidea, while instantaneous growth rate showed the opposite pattern. Survival rates of the Manila clam larvae fed each strain of Teleaulax showed no significant difference between strains. Survival rates of the Pacific oyster fed Teleaulax were significantly higher than those fed Chaetoceros sp., but lower than those fed I. galbana. This study indicates that strain 08-2 showed better growth for B. plicatilis and better survival for the bivalve larvae than did the other strains, and that T. amphioxeia can also be used as a prey organism for rotifers or bivalve larvae.

Ecotoxic Evaluations of BDE-47 and BDE-209 using Rotifer (Brachionus plicatilis) (해산로티퍼 (Brachionus plicatilis)를 이용한 브롬화난연제 (BDE-47, BDE-209)의 생태독성평가)

  • The toxic assessment of the PBDEs (BDE-47, BDE-209) has been comprehensively investigated by using the rates of survival and population growth in the marine rotifer, Brachionus plicatilis. Chiefly, the survival rate was determined after a measurement of 24 hours of exposure to the BDE-47 (2,2'4,4'-Tetrabromodiphenyl ether) and the BDE-209 (2,2',4,4'-Decabromodiphenyl ether) was performed. The BDE-47 reduced survival rate in dose-dependent manner and a significant reduction were noted to have occurred at a concentration of greater than $3.9mg\;L^{-1}$, but the BDE-209 had no effect which was subsequently observed in this study. The population growth rate (r) was determined after 72 hours of exposure to toxicants in the study. It was observed that the r value in the controls (absence PBDEs) was greater than 0.5, and that it decreased as the dose-dependent manner as recorded. The survival rate when exposed to BDE-47 and BDE-209, $EC_{50}$ value was $13mg\;L^{-1}$ and $>1,000mg\;L^{-1}$, and population growth rate was $3.67mg\;L^{-1}$ and $862.75mg\;L^{-1}$, respectively. Therefore, the BDE-47 is considered to be 76-235 times more harmful than the BDE-209 as noted. In this study, the ecotoxicological bioassay using a noted survival rate and population growth rate of B. plicatilis can be used as a baseline data for the continued establishment of the environmental quality standard of the incidences of the BDE-47 and BDE-209 in a marine environment.

Production and Hatching Rate of Resing Egg of Korean Rotifer, Brachionus plicatilis (S-type) with Different Diets (먹이에 따른 한국산 S-tyoe Rotifer, Brachionus plicatilis의 내구란 생산과 부화율)

  • Dietary values of phytoplanktons, concentrated Chlorella and commercial yeasts were investigated for the resting egg production of the Korean rotifer, B. plioatilis S-type. The hatching rate of the resting egg of the rotifer fed on different diets was also studied. The highest production of the resting egg was 3,760 eggs/20 ml with N. oculata. However, the hatching rate was not significant among 4 phytoplankton species. The highest production of resting eggs among 9 diet groups ($\omega$-yeast, baker's yeast, 2 refrigerated Marine Chlorella, frozen Marine Chlorella, frozen Chlorella ($70\%$) + baker's yeast ($30\%$), frozen Chlorella ($30\%$) + baker's yeast ($70\%$), 2 refrigerated freshwater Chiorella) was 283 eggs/ml in the frozen Chlorella ($30\%$) +baker's yeast ($70\%$) and the refrigerated freshwater Chlorella. The highest number of resting egg from 10,000 rotifers and 1 mg dry wieght diet was 5,566 eggs and 2,131 eggs in the frozen Chlorella ($30\%$) + baker's yeast ($70\%$). However, the highest hatching rate of the resting eggs was $67.4\%$ in $\omega$-yeast. In this study, the results suggest that the baker's yeast with a small amount of frozen Chlorella seems to be good feeding regime for the economical production of resting eggs.

Establishment of Standard Methods for Marine Ecotoxicological Test (해양생태독성평가를 위한 표준시험방법 개발에 관한 연구)

  • Six standard methods for marine ecotoxicological tests were established(or applicated) using marine decomposer, primary producers and consumers. Development processes referred to the standard methods established by USEPA(United States Environmental Protection Agency), international organizations and European methods. However, the standard test species were selected among the domestic species generally found in the Korean waters and sediments. The test methods provide the culture/maintenance of test species, test methods, reproducibility and quality acceptance criteria etc. A total of nine test species were designated including bioluminescent bacteria(Vibrio fischeri), diatom(Skeletonema costatum), seaweed(Ulva pertusa), rotifer(Brachionus plicatilis), benthic copepod(Tigriopus japonicus), benthic amphipods(Mandibulophoxus mai, Monocorophium acherusicum), and fishes(Oryzias latipes, Paralichthys olivaceus). These test species represent the decomposer, primary producer and consumers in marine trophic system in Korean coastal ecosystems, and we recommend the "battery test" including at least one species from the each trophic level for marine ecotoxicological test.

  • Growth rate, first reproductive age, life span and total offspring numbers were measured to study the effects of water temperature and salinity on the propagation of rotifer, Brachionus plicatilis. Three types of rotifer, Large (L), Small (S) and Thailand Small (TS)-types, were cultured in the $4\times4$ factorial culture conditions with four different salinities (5, 15, 25 and $35\%o$) at each of four different water temperatures (15, 20, 25 and $29^{\circ}C$). The results are as follows; Under the 4 different salinity gradients %$(5\%o,\;15\%o,\;25\%o,\;and\;35\%o)\;at\;29^{\circ}C$, the mean growth rates of L, Sand TS-type were 0.60, 0.84 and 0.96, respectively. The first reproductive age of three types rotifer appeared to be early at high water temperature. The total offspring numbers of the three types were higher at $25^{\circ}C\;and\;29^{\circ}C$, with the maximum value of 28.3 on the average at $29^{\circ}C$ for TS-type, and the minimum value at $15^{\circ}C$ for S-type. Life span decreased with high water temperature and increased with low water temperature. L-type and TS-type rotifer showed the longest life span of 13.5 days on the average at $15^{\circ}C$, and S-type showed shortest 6.2 days on the average at $29^{\circ}C$.

Production of extracellular polysaccharide by Monilinia fructigena for aquaculture

  • Production of extracellular polysaccharide by Monilinia fructigena in B-I medium containing cereals was higher than that in glucose medium. Productivities in B-I medium and glucose medium were 0.7g/l nd 0.2-03g/l respectively. The maximum content of polysaccharide occurred at the rising point from the lowest pH of culture. As the apparent viscosity of the polysaccharide solution increased, the flow Index (m) decreased, and the consistency Index (Kc) also increased. The polysaccharide solution was a typical pseudoplastic fluid. The mycelium was separated from the culture solution by $300\mu m$ mesh-filter and the polysaccharide was precipitated by adding 50% of ethanol (v/v). The amount of the polysaccharide removed from the filtrated solution was 0.45 g/l and the amount adhered to the mycelium was 0.25g/l. In experiments for investigating growth enhancement of rotifer (Brachionus plicatilis) by the polysaccharide, the dose of the polysaccharide was 1mg per 10,000 organisms of rotifer. Maximum specific growth rate of rotifer with feed consisting of sea Chlorella sp. and the polysaccharide was 1.095/day in the batch culture for 10 days. A semi-continuous culture was done for 30 days, the biomass of rotifer could be harvested twice. Maximum specific growth rate with sea Chlorella sp. and the polysaccharide was 0.734/day before the first harvest, and 1.685/day before the second harvest. Productivity was 38 $cells/ml\; \cdot\; day$ with sea Chlorella sp. and the polysaccharide.

Ecotoxicological Evaluation of Sewage Sludge Using Bioluminescent Marine Bacteria and Rotifer

  • Bioassay using the marine bacteria, Vibrio fischeri and rotifer, Brachionus plicatilis, and chemical analyses were conducted to assess the toxicity of the various sewage sludges, one of the major ocean dumped materials in the Yellow Sea of Korea. Sludge elutriates extracted by filtered seawater were used to estimate the ecotoxicity of the sludge. Chemical characterization included the analyses of organic contents, heavy metals, and persistent organic pollutants in sludge. Bacterial bioluminescent inhibition (15 min), rotifer mortality (24 hr) and rotifer population growth inhibition (48 hr) assay were conducted to estimate the sludge toxicity. EC50 15 min (inhibition concentration of bioluminescence after 15 minutes exposed) values by Microtox(R) bioassay clearly revealed different toxicity levels depending on the sludge sources. Highest toxicity for the bacteria was found with the sludge extract from dyeing waste and followed by industrial waste, livestock waste, and leather processing waste. Clear toxic effects on the bacteria were not found in the sludge extract from filtration bed sludge and rural sewage sludge. Consistent with Microtox(R) results, rotifer neonate mortality and population growth inhibition test also showed highest toxicity in dyeing waste and low in filtration bed and rural sewage sludge. High concentrations of persistent organic pollutants (POPs) and heavy metals were measured in the samples from the industrial wastes, leather processing plant waste sludge, and urban sewage sludge. However, there was no significant correlation between pollutant concentration levels and the toxicity values of the sludge. This suggests that the ecotoxicity in addition to the chemical analyses of various sludge samples must be estimated before release of potential harmful waste in the natural environment as part of an ecological risk assessment.

Possibility to Develope the Multistage Culture System for Larvae Cultivation by Computer Simulation (컴퓨터 시뮬레이션을 통한 치어 사육용 다단계 배양시스템의 개발 가능성 탐색)

  • The possibility for developing multistage culture system to cultivate larvae by computer simulation with basic experiments was investigated. This culture system was composed of 3 stages. At the 1st stage, Chlorella sp. were cultivated and at 2nd stage Chlorella sp. were supplied to rotifer (Brachionus plicatilis), and rotifer were supplied to larvae at the 3rd stage. In this study, Chlorella sp. were cultivated by batch culture to search for the possibility of continuous feeding to rotifer at 2 nd stage. The maximum specific growth rate ($\mu$$_{max}$) of Chlorella sp. at the logarithmic phase was 0.56 [1/day]. Rotifer was cultivated by fed-batch culture at the feeding rate of Chlorella sp., $10^3$, $10^4$, $10^{5}$ and $10^{6}$ [cells/rotiferㆍh] to search for the relation between the feeding rate of Chlorella sp. and the growth rate of rotifer. As the results, the minimum feeding rate of Chlorella sp. was 2.8 ${\times}$ $10^4$ [cells/rotiferㆍday] in the multistage culture system, then the change of rotifer concentration at 2nd stage was simulated by computer. The required amount of rotifer for the growth of larvae was also increased as the growth of larvae. On the 9th day of the culture, the rotifer uptake rate of larvae was 250 [cells/rotiferㆍday]. Based on these basic experiments and results, It was suggested that the possibility of multistage culture system to cultivate larvae with continuous feeding of Chlorella sp. and rotifer.r.
