Acute beetroot juice supplementation does not attenuate knee extensor exercise muscle fatigue in a healthy young population

  • [Purpose] The effect of acute nitrate supplementation on muscle fatigue is largely unknown. This study aimed to evaluate the effect of acute nitrate supplementation on muscle fatigue. [Methods] Thirty-five recreationally active subjects consumed 140 ml of beetroot (BR) juice (nitrate: 8 mmol·d-1) or placebo (PL) 12 and 2.5 hours before two exercise sessions. Peak torque was measured during 50 repetitions, at maximal effort, and during concentric knee extensions at 90°·s-1. Blood pressure (BP) was recorded pre- and post-exercise. [Results] Peak torque, maximum work, rate of fatigue, and rate of work fatigue were similar between the BR and PL conditions. Post-exercise diastolic BP (BR: 67.2 ± 9.8 vs. PL: 64.5 ± 7.9 mmHg, p < 0.05) and mean arterial pressure (BR: 91.6 ± 9.3 vs. PL: 88.8 ± 8.2 mmHg, p < 0.05) were higher with BR supplementation. [Conclusion] These findings suggest that the acute intake of BR juice had no effect on knee extensor muscle strength or fatigue but increased BP in a healthy recreationally active population.

Shewanella algae BrY를 이용한 영가철 칼럼의 TCE 처리 수명연장 (Durability Extension of Fe(0) Column with Shewanella Algae BrY on TCE Treatment)

  • 영가철은 trichloroethylene(TCE)과 같은 염소계 유기오염물질 제거에 탁월한 효과를 가지고 있어서, 반응벽체를 이용한 오염된 토양 및 지하수의 현장처리에 반응매질로 자주 사용되고 있다. 하지만 영가철의 빠른 반응성으로 인하여 반응벽체의 수명이 다하고, 탈염소화 과정 중 생성된 산화철이 영가철 표면에 침적 되어 반응표면적을 줄임으로써 반응성이 떨어지게 된다. 이러한 영가철 반응벽체의 단점을 보완하기 위한 방법이 연구되어 왔고, 그 중 철환원균을 이용한 연구가 본 연구에서 시도되었다. 실험에 사용한 Shewanella algae BrY는 철환원균의 일종으로서 Dissimilatory Iron Reducing Bacteria(DIRB)로 분류된다. 본 연구에서는 이전의 배치실험 연구결과를 바탕으로 칼럼실험을 통해 TCE의 농도를 30mg/L과 67.5mg/L의 두 가지로 비교하고, 유량을 8mL/hr, 16mL/hr의 두 가지로 비교하여 세 개의 칼럼 실험을 실시하여 영가철 반응벽체의 TCE의 제거 및 산화된 철의 철환원균을 이용한 환원과정을 칼럼실험을 통해 측정했다. 그 결과 철환원균에 의한 산화철의 환원은 오염물질의 농도가 높을 경우 처리량의 증가로 인하여 반응벽체의 수명을 단축시키는 원인이 되었고, 유량의 증가 역시 유입되는 오염물질의 증가로 인하여 수명의 단축을 가져왔다. 그러나 유량증가의 경우 유속의 증가를 가져와서 Shewanella algae BrY가 매질표면에 작용하여 산화철을 환원시킬 수 있는 시간을 감소시키고 착상되는 것을 방해하기 때문에 오염물질의 농도보다 큰 영향을 미치는 결과를 보여준다. 칼럼실험결과 Shewanella algae BrY에 의한 산화철의 환원은 TCE의 탈염소화 후 생성된 산화철의 침적에 의하여 발생되는 반응벽체의 수명감소를 줄일 수 있다는 결과를 얻었다.

Channeling of Intermediates Derived from Medium-Chain Fatty Acids and De novo-SYnthesized Fatty Acids to Polyhydroxyalkanoic Acid by 2-Bromooctanoic Acid in Pseudomonas fluorescens BM07

  • 2-Bromooctanoic acid (2-BrOA) is known to block the formation of polyhydroxyalkanoic acid (PHA) in Pseudomonasfluorescens BM07 without any influence on the cell growth when grown on fructose, but it inhibits the cell growth when grown on octanoate (OA) (Lee et al., Appl. Environ. Microbiol. 67: 4963- 4974, 2001). We investigated the role of 2-BrOA in the PHA synthesis of the bacterium grown with mixtures of fructose and fatty acids. OA, 11­phenoxyundecanoic acid (1 1-POU), and 5-phenylvaleric acid (5-PV) were selected as model substrates. When supplemented with 50 mM fructose, all these carboxylic acids suppressed the formation of PHA from fructose, however, the ~-oxidation coenzyme A monomers derived from the carboxylic acids were efficiently polymerized, but the conversion yield [(mol of carboxylate substrate converted into PHA)/(mol of carboxylate substrate in the feed)] was low (e.g., maximally $\~53\%$ for 5 mM 11-POU). Addition of 2-BrOA (up to 5 mM) to the mixed carbon sources raised the conversion yield sensitively and effectively only at low levels of the acid substrates (e.g., 2 mM 1 1-POU or 5 mM OA): For instance, $100\%$ of 2 mM ll-POU were converted into PHA in the presence of 5 mM 2-BrOA, whereas only $\~10\%$ of the 1 1-POU were converted in the absence of 2-BrOA. However, at highly saturated suppressing levels (e.g., 5 mM ll-POU), 2-BrOA inhibitor showed no significant additional effect on the conversion ($60- 70\%$ conversion irrespective of 2-BrOA level). The existence of competitive and compensative relationship between 2­BrOA and all the carboxylic acid substrates used may indicate 'Present address: Section on Brain Physiology and Metabolism, Bldg. 10, Rm. 6N202, National Institute on Agmg, National Institute of Health, Bethesda, MD 20892, U.S.A. that all the acid substrate-derived inhibiting species bind to the same site as the 2-BrOA inhibiting species does. We, therefore, suggest that 2-BrOA can be used for efficiently increasing the yield of conversion of expensive substituted fatty acids into PHA and then substituted 3-hydroxyacids by hydrolyzing it.

다양한 휘도에서 호흡 알코올 농도에 따른 대비감도 변화 연구 (A Study on the Change of Contrast Sensitivity with Breath Alcohol Concentration in Various Luminance)

  • 목적: 호흡 알코올 농도를 제한한 상태에서 알코올 섭취로 인한 호흡 알코올 농도(breath alcohol concentration, BrAC) 증가가 대비감도에 미치는 영향을 알아보았다. 방법: 20대의 남성 23명(평균연령 $21.17{\pm}2.19$세, body mass index(BMI) $22.09{\pm}2.16$)을 대상으로 Watson 공식을 기반으로 개발된 BAC(blood alcohol concentration) Dosing Software를 이용하여 호흡 알코올 농도 0.05%와 0.08%에 도달하기 위한 알코올 양을 산정하여 투여한 후 다양한 휘도(명소시, 박명시, 박명시 눈부심) 상태에서 대비감도를 측정하여 비교 분석하였다. 결과: 호흡 알코올 농도가 증가할수록 모든 공간주파수에서 대비감도는 감소하였다. 정점대비감도는 명소시 상태에서 6 cycle per degree(cpd), 박명시 상태에서 3 cpd로서 호흡 알코올 농도가 증가하여도 정점이동이 나타나지 않았지만, 박명시 눈부심 상태에서는 0%와 0.05%에서는 6 cpd, 0.08%에서는 3 cpd로서 호흡 알코올 농도 증가에 따른 정점이동이 나타났다. 결론: 음주에 의한 호흡 알코올 농도 증가는 모든 공간주파수에서 대비감도의 감소와 정점대비감도의 이동을 유발함으로써 다양한 시작업에 영향을 미치고 안전사고의 원인이 될 수 있으므로 주의하여야 한다.

도시하천도로의 EAP수립을 위한 침수특성분석 - 중랑천 동부간선도로를 중심으로 - (Analysis of Inundation Characteristics for EAP of Highway in Urban Stream - Dongbu Highway in Jungrang Stream -)

  • 본 연구에서는 침수위험이 큰 도시하천내에 건설된 도로의 침수로 인한 피해를 미연에 방지하고 적절한 교통통제계획, EAP 등을 수립하기 위한 수문, 수리학적 분석과정을 제시하였다. 연구대상지역으로 우리나라의 대표적 도시하천인 중랑천 유역의 좌우안에 위치한 동부간선도로를 선정하고 비상대처계획 수립을 위한 기초자료를 작성하였다. HEC-HMS에 의하여 유출 해석을 실시하였으며, UNET을 이용하여 주요 지점 및 구간별 침수특성을 분석하였다. 이문철교부근(하구에서 약 9.5 km)과 월릉교부근(하구에서 약 11.5 km)에서 침수위험이 가장 높아 이문철교부근은 10년 빈도시에, 월릉교부근의 좌안도로는 20년 빈도시에 각각 침수가 되는 것으로 나타났다. 누가강우량과 지속시간을 고려한 침수특성 분석결과 강우 지속시간 7시간 이하에서 누가강우량이 250 mm이상일 경우에는 월계1교지점의 위험홍수위(EL.17.84 m)를 초과하는 것으로 분석되었다. 한강의 배수위를 고려하지 않은 경우에는 C2(월계1교-중랑교, 좌안), C1(월계1교-중랑교, 우안), D(중랑교-군자교)구간순으로 침수위험이 높은 것으로 나타났으나 배수위를 고려한 경우에는 D2(중랑교-군자교, 좌안), E(군자교-용비교)구간의 침수위험이 오히려 높은 것으로 분석되었다.

  • This research was performed in order to investigate the anti-inflammatory effects of Bupleuri Radix(BR) on the Immune response in vitro. Cellular proliferation and cytokine production were measured in mast cells or mouse B cells or CD4 Th cells. BR water extract inhibited the secretions of TNF-$\alpha$ and IL-6 in PMA/A23187 stimulated HMC-1 cells. It increased proliferation but did not affect the expressions of CD69 or CD23 in rIL-4/anti-CD40 activated S cells. BR reduced surface IgE expression and secreted IgE but increased the production of IL-4, IFN-$\gamma$ and IgG1 in the same cells. BR caused an increase in proliferation in anti-CD3/anti-CD28 stimulated CD4 Th cells but it did not affect the differentiation of Th1 or Th2 cells. However, IL-2 was increased in BR treated Th2 cells. Considering the above-mentioned results, BR can be applied to a broad range of anti-inflammatory reactions, but our data suggest that it will not be likely to exert any effects on type 1 allergic response.

Ileal Amino Acid Digestibility of Broken Rice Fed to Postweaned Piglets with or without Multicarbohydrase and Phytase Supplementation

  • Most of amino acid (AA) digestibility values for feed ingredients are obtained using pigs cannulated in the distal ileum. The ileal-cannulated pig model uses pigs older than six weeks due to difficulties related to implanting the T-cannula in distal ileum of younger pigs and complications during the post-surgical recovery. However, to properly formulate the diet of weaned pigs, the nutritive value of feed ingredients should be determined with younger pigs. Thus, 25 weaned pigs were used to determine the apparent total tract digestibility (ATTD) of nutrients, energy, and apparent ileal digestibility (AID) and standardized ileal digestibility (SID) ileal AA digestibility of broken rice (BR), with or without multicarbohydrase (MC) and phytase (Phy) supplementation. Piglets were weaned at 23 d of age and individually housed in digestibility cages until 45 d of age. The trial consisted of 7 d of adaptation to the experimental diets and 3 d of excreta (feces and urine) collection. Ileal digesta was collected at slaughter (about 6 weeks of age). A completely randomized experimental design was used to determine the effects of MC and Phy. Reference diets (RD, 5% casein) was replaced by 30% of BR with or without MC, Phy, or MC+Phy. The RD was used to quantify endogenous AA losses. BR with Phy supplied had increased the ATTD of dry matter (p<0.05) and SID of histidine (p = 0.05), arginine, leucine, lysine, valine, alanine, and proline (p<0.05). BR with MC had been increased digestible energy and protein and SID for histidine (p<0.05). There was no interaction between Phy and MC on the BR nutrient digestibilities. Standardized amino acid digestibilities of BR, without enzymes, were lower than those values reported in the literature. The MC and Phy improved the digestibility of some nutrients and energy of BR in post-weaned piglet diets.

Genetic mapping and sequence analysis of Phi class Glutathione S-transferases (BrGSTFs) candidates from Brassica rapa

  • Glutathione S-transferases (GSTs) are multifunctional proteins encoded by a large gene family divided into Phi, Tau, Theta, Zeta, Lambda and DHAR classes on the basis of sequence identity. The Phi(F) and Tau(U) classes are plant-specific and ubiquitous. Their roles have been defined as herbicide detoxification and responses to biotic and abiotic stresses. Fifty-two members of the GST super-family were identified in the Arabidopsis thaliana genome, 13 members of which belong to the Phi class of GSTs (AtGSTFs). Based on the sequence similarities of AtGSTFs, 11 BAC clones were identified from Brassica rapa. Seven unique sequences of ORFs designated the Phi class candidates of GST derived from B. rapa (BrGSTFs) were detected from these 11 BAC clones by blast search and sequence alignment. Some of BrGSTFs were present in the same BAC clones indicating that BrGSTFs could also be clustered as usual in plant. They were mapped on B. rapa linkage group 2, 3, 9 and 10 and their nucleotide and amino acid sequences were highly similar to those of AtGSTFs. In addition, in silico analysis of BrGSTFs using Korea Brassica Genome Project 24K oligochip and microarray database for cold, salt and drought stresses revealed 15 unigenes to be highly similar to AtGSTFs and six of these were identical to one of BrGSTFs identified in the BAC clones indicating their expression. The sequences of BrGSTFs and unigenes identified in this study will facilitate further studies to apply GST genes to medical and agriculture purposes.

속성배추를 이용한 식물 탐구 수업이 초등학생의 식물 개념 변화에 미치는 효과 (Effects of the Plant Inquiry Instruction using Rapid-cycling Brassica rapa on the Change of Plant Concept of the Elementary School Students)

  • This study is intended to develop plant inquiry instruction using Rapid-cycling Brassica rapa (RcBr) whose life cycle is relatively short, and to apply it to the elementary science instruction of the 4th grader and examine their plant concept, science inquiry skills and science-related attitudes. The materials were consisted of laboratory manuals for the students as well as teachers' guide. By observing the life cycle of RcBr, students can experience the conceptual learning of the plant's life cycle. In addition, this study investigated the cause of change in science inquiry skills and science-related attitudes by interviewing 12 students. It has shown that plant inquiry instruction using RcBr has meaningful effects on students' understanding of the plant concept, improving students' science inquiry skills, and changing students' science-related attitudes. Students who showed improvement in science inquiry skills were able to answer questions regarding science knowledge correctly. And students whose science related attitudes were improved had a positive attitude on cultivating RcBr. Students told that RcBr was an interesting and good material to inquire plant. Because of its small size and its relatively short life cycle of RcBR, it should be a desirable plant material for the inquiry instruction which can give rise to useful and meaningful results for the elementary school students.

유산견(流産犬) 태아(胎兒)로부터 Brucella suis의 분리(分離) (Isolation of Brucella Suis from Aborted Fetus of a Dog)

  • 우리나라 가축중(家畜中) 우(牛) 및 돈(豚)의 brucellosis는 알려져 있으나 견(犬)에서의 brucella 분리(分離)에 관(關)한 보고(報告)는 볼 수 없다. 저자(著者)등은 1971년(年) 8월(月) German shepherd의 유산(流産)한 55일(日) 견태(犬胎)에서 brucella라고 생각(生覺)되는 균(菌)을 분리(分離)하여 그 성상(性狀)을 추구(追究)하여 Brucella suis로 동정(同定)하였다. 이로서 우리나라에도 brucella에 의(依)한 견(犬)의 유산(流産)이 있음을 알수 있다. 유산견(流産犬)의 혈청항체가(血淸抗體價)는 Br. suis에 대(對)하여 320배(倍)였다. 대구시(大邱市) 도축장(屠畜場)에서 얻은 86례(例)의 견(犬)의 혈청(血淸)의 brucella에 대(對)한 항체가(抗體價)를 검사(檢査)하였던바 Br. abortus에는 1례(例)만이 80배양성(倍陽性)이었고, Br. suis에는 3례(例)가 160배(倍), 8례(例)가 80배양성(倍陽性)였다. 분리균(分離菌)에 대(對)한 항체가(抗體價)는 대체(大體)로 Br. suis에 대(對)한 항체가(抗體價)와 평행(平行)하였다.
