Protective Effects of Boyanghwanoh-tang on Serum and Glucose Deprivation-induced Apoptosis of PC12 Cells (보양환오탕이 영양혈청결핍에 의한 PC12 세포의 고사에 미치는 영향)

  • Objectives : Boyanghwanoh-tang (Buyanhaiwu-tang) has been used as a prescription for stroke, senile and vascular dementia, ischemic brain and heart damage in Oriental traditional medicine. However, there is little known about the mechanism by which the water extracts of Boyanghwanoh-tang (Buyanhaiwu-tang) rescue cells fromthese damages, and little is known about the protective mechanisms of Boyanghwanoh-tang (Buyanhaiwu-tang) on oxidative stress in neuronal cells. Therefore, we have investigated the role of Boyanghwanoh-tang (Buyanhaiwu-tang) on serum and glucose deprived apoptosis in PC12 cells. Methods : PC12 Cells have been used extensively as a model for studying the cellular and molecular effects of neuronal cells. The viability of cells was measured by MIT assay. We used DNA fragmentation and caspase 1, 2, 3, 6, 9-likeproteases activation assay. Transcriptional activation of NF-kB was assessed by using electrophoretic mobility shift assay. Results : Boyanghwanoh-tang (Buyanhaiwu-tang) rescued PC12 cells from apoptotic death by serum and glucose deprivation in a dose-dependent manner. The nuclear staining of PC12 cells clearly showed that Boyanghwanoh-tang (Buyanhaiwu-tang) attenuated nuclear condensation and fragmentation, which represent typical neuronal apoptotic characteristics. Boyanghwanoh-tang (Buyanhaiwu-tang) also prevents fragmentation of genomic DNA and activation of caspase 3-like protease in serum and glucose deprived PC12 cells. Furthermore, Boyanghwanoh-tang (Buyanhaiwu-tang) reduced the activation of NF-kB by serum and glucose-deprived apoptosis. Conclusions : These findings suggest that serum and glucose deprivation induces reduced glutathione (GSH) depletion, and consequently, apoptosis through endogenously produced reactive oxygen species in PC12 cells. Also, our data indicated that Boyanghwanoh-tang (Buyanhaiwu-tang) has protective effects against the serum and glucose deprived deaths of PC12 cells, which are mediated by the generation of GSH that, in turn, can reduce oxidative stress caused by reactive oxygen species (ROS) such as hydrogen peroxide.

Protective Effects of Boyanghwanoh-tang on Zinc-mediated Cytotoxicity in H9c2 Cardiomyoblast Cells (산화적 손상에 의해 유발된 심근세포 독성에 대한 보양환오탕(補陽還五湯)의 방어효과)

  • The water extract of Boyanghwanoh-tang has been used for treatment of ischemic vascular disease in oriental medicine. However, little is known about the mechanism by which the water extract of Boyanghwanoh-tang rescues cells from these damages. Therefore, this study was designed to evaluate the protective effects of Boyanghwanoh-tang on zinc-mediated cytotoxicity in H9c2 cardiomyoblast cells. This study demonstrates that, after treatment of H9c2 cells with zinc, there was a decrease in cell viability in a dose dependent manner, and there was a chromatin condensation. Zinc induced the change of cell morphology. In addition, zinc induced mitochondrial dysfunction. Zinc-induced H9c2 cell death was remarkably prevented by the pretreatment of Boyanghwanoh-tang consistently with increase of the peroxoredoxin 1, 2, 3, 5, and 6 expression. Taken together, the results suggest that zinc induced severe cell death in H9c2 cardiomyoblast cells, and that protective effects of Boyanghwanoh-tang against oxidative injuries are achieved through regulation of peroxiredoin expression.

Experimental study on the effects of Boyanghwanoh-tang(補陽還五湯) on blood pressure and hyperlipidemia (고혈압(高血壓) 및 고지혈증(高脂血症)에 대(對)한 보양환오탕(補陽還五湯)의 실험적(實驗的) 연구(硏究))

  • In order to study the effects of Boyanghwanoh-tang(補陽還五湯) on blood pressure and hyperlipidemia, experimental study was performed on Hypertension in normal rats, SHR, and on Hyperlipidemia induced by Triton WR-1339 and 2% cholesterol Fed-diet in normal rats. Also the level of Total cholesterol, Triglyceride, LDL-cholesterol, HDL-cholesterol, Phospholipid were measured. The results are summarized as follows : 1. After Boyanghwanoh-tang(補陽還五湯) was given to normal rats and SHR, the results showed that inhibitory effects on blood pressure were not significant but exitory effects on heart beat were significant. 2. In the model of hyperlipidemia Rats induced by 2% Cholesterol Fed-Diet, Boyanghwanoh-tang(補陽還五湯) had significantly decreasing effects on Total cholesterol, Triglyceride, LDL-cholesterol, Phospholipid level in serum but no significantly increasing effects on HDL-cholesterol level in serum. 3. In the model of hyperlipidemia Rats induced by Triton WR-1339, Boyanghwanoh-tang(補陽還五湯) had significantly decreasing effects on LDL-cholesterol, Phospholipid level in serum and significantly increasing effects on HDL-cholesterol level in serum but there were not significantly decreasing effects on Total cholesterol, Triglyceride. These results show that Boyanghwanoh-tang(補陽還五湯) has significantly inhibitory effects on hyperlipidemia and thal it could be clinically applied for hyperlipidemia.

A Case Report on Right Hemiparesis after Left Thalamus Infarction Treated with Boyanghwanoh-tang-gagam and Complex Korean Medicine Treatment (보양환오탕가감방 및 복합한의치료로 호전된 시상 부위 뇌경색 후 우측 편마비 환자 치험 1례)

  • Objectives: This study reports the case of a patient with right hemiparesis after a left thalamic infarction, in which the patient experienced improved clinical symptoms (hemiparesis, constipation) after treatment with a complex Korean medicine treatment. Methods: A 81-year-old male patient diagnosed with acute left thalamic infarction was treated with daily acupuncture and herbal medicine (Boyanghwanoh-tang-gagam) three times a day. Jowiseunggi-tang extract granules were administered to treat constipation by adjusting the dosage according to the reported symptoms. The patient's clinical course was assessed using changes in Modified Barthel Index (MBI) score, Manual Muscle Testing (MMT) grade, and defecation frequency . Results: The patient's MBI score improved from 35 to 69 and was discharged with the ability to walk under supervision. The MMT grade also improved from 3/4 to 5/5 in the upper/lower extremities. Other clinical symptoms, including constipation, also improved, leading to the discontinuation of magnesium medication. Conclusion: These findings suggest that complex Korean medicine treatments, including herbal medicine and acupuncture, may be an effective treatment for post-stroke hemiplegic patients.

A Case Report on Hemiplegia in a Patient with an Intracerebral Hemorrhage of the Basal Ganglia of the Corona Radiata Treated with Traditional Korean and Western Medicine (기저핵-대뇌부챗살 뇌출혈로 인한 반신부전마비에 대한 한·양방 병행 치험 1례)

  • Objective: This study examined the case of a 63-year-old man with a history of hypertension and gout who had developed an intracerebral hemorrhage (ICH) of the right basal ganglia of the corona radiata. Methods: The patient was treated with herbal medicine (Boyanghwanoh-tang and powdered Oryeong-san extract), Western medicine (Anticonvulsants, antihypertensive drugs, psychotropic agents, and others), acupuncture, moxibustion, cupping, and rehabilitative therapy. Their effects were evaluated using the Manual Muscle Test (MMT), Korean version of the modified Barthel Index (K-MBI), National Institute of Health's Stroke Scale (NIHSS), and Modified Rankin Scale (mRS). Results: After treatment, the MMT grade improved from 3/3+ to 4+/4+. Additionally, the K-MBI score improved from 62 to 77. Conclusion: The results suggest that combined traditional Korean and Western medicine can effectively treat patients with ICH.

  • Alzheimer's disease (AD) is progressive neurodegenerative disease, which is pathologically characterized by neuritic plaques and neurofibrillary tangles associated with the acetylcholinesterase, apolipoprotein E and butylcholinesterase, and by mutations in the presenilin genes PS1 and PS2, and amyloid precursor proteins (APPs) overexpression. The present research is to examine the inhibition effect of KBT on PS1, PS2 and APPs overexpression by detected to Western blotting. To verify the Effects of KBT on cognitive deficits further, we tested it on the scopolamine (1 mg/kg)-induced amnesia model of the mice using the Morris water maze tests, and there was ameliorative effects of memory impairment as a protection to scopolamine. KBT only partially blocked the increase in blood serum level of acetylcholinesterase and Uric acid induced by scopolamine, whereas blood glucose level was shown to attenuate the amnesia induced by scopolamine and inreased extracellular serum level compared with only scopolamine injection. In conclusion, studies of KBT that has been know as anti-choline and inhibition ablilities of APPs overexpression, this could also be used further as a important research data for a preventive and promising symptomatic treatment for Alzheimer's disease.
