Inhibition of melanogenesis by sodium 2-mercaptoethanesulfonate

  • Sodium 2-mercaptoethanesulfonate (mesna) is a protective agent that is widely used in medicine because of its antioxidant effects. Recently, reactive oxygen species (ROS) were shown to increase pigmentation. Thus, ROS scavengers and inhibitors of ROS production may suppress melanogenesis. Forkhead box-O3a (FoxO3a) is an antimelanogenic factor that mediates ROS-induced skin pigmentation. In this study, we aimed to investigate the whitening effect of mesna and the signaling mechanism mediating this effect. Human melanoma (MNT-1) cells were used in this study. mRNA and protein expression were measured by real-time quantitative PCR and Western blotting analysis to track changes in FoxO3a-related signals induced by mesna. An immunofluorescence assay was performed to determine the nuclear translocation of FoxO3a. When MNT-1 melanoma cells were treated with mesna, melanin production and secretion decreased. These effects were accompanied by increases in FoxO3a activation and nuclear translocation, resulting in downregulation of four master genes of melanogenesis: MITF, TYR, TRP1, and TRP2. We found that mesna, an antioxidant and radical scavenger, suppresses melanin production and may therefore be a useful agent for the clinical treatment of hyperpigmentation disorders.

An Experimental Study on the Development of a Cabin Noise Reduction System for Improving Ship Habitability (선박 거주성 향상을 위한 선실 소음 저감 시스템 개발에 관한 실험적 연구)

  • Ship noise is one of the important factors for the living and health of seafarers, and ef orts to reduce ship noise are actively underway. There are two methods of noise reduction: passive noise Control (PNC) and active noise control (ANC). Unlike cars and airplanes, ANC is not widely used for noise reduction on ships. This study aimed to reduce the noise generated in the engine room by using soundproof panels and high-frequency vibration generators, as well as active noise control (ANC). For this purpose, an experimental model was made using an acrylic box, and the noise reduction effect was measured under four conditions. The experimental results are as follows: First, the soundproof panel had a noise reduction effect in all ranges from 55 dB to 85 dB. In the case of using a high-frequency vibration generator, there was no ef ect in the low noise range such as 55 dB(A), but there was a noise reduction effect in the high noise range such as 70.8 dB(A) and 85 dB(A).Second, when the soundproof panel and the high-frequency vibration generator were used simultaneously, the noise reduction ef ect was up to -2.2 dB(A). The results of this experiment were obtained from an experimental model made of acrylic, and they may be different from actual ships made of steel plate. In future studies, we plan to experiment using iron plate (considering the material, thickness, and structure) used in actual ships. We hope that this study will help to improve the living environment and health of seafarers on ships.

An Experimental Study on the High Strength Lightweight Self-Compacting Concrete (고강도경량 자기충전콘크리트에 관한 실험적 연구)

  • This paper was to evaluate the high strength lightweight self-compacting concrete(HLSCC) manufactured by Nan-Su, which main factor, Packing Factor(PF) for mixing design, has been modified and improved. We have examined HLSCC performance at its fresh condition as well as its mechanical properties at the hardened condition. The evaluation of HLSCC fluidity has been conducted per the standard of second class rating of JSCE, by three categories of flowability(slump-flow), segregation resistance ability(time required to reach 500mm of slump-flow and time required to flow through V-funnel) and filling ability(U-box test) of fresh concrete. The compressive strength of HLSSC at 28 days has come out to more than 30MPa in all mixes. The relationship between the compressive strength-splitting tensile strength and compressive strength-modulus of elasticity of HLSSC were similar those of typical lightweight concrete. Compressive strength and dry density of HLSCC at 28 days from the multiple regression analysis resulted as $f_c=-0.16LC-0.008LS+50.05(R=0.83)\;and\;f_d=-3.598LC-2.244LS+2,310(R=0.99)$, respectively.

A Research for Computation of Bearing Capacity and Settlement of Foundation Considering Scale Effect in Weathered-granite Layer (화강풍화토에서 Scale Effect를 고려한 기초의 지지력 및 침하량 산정에 관한 연구)

  • When calculating bearing capacity and settlement of actual foundation from plate test result fur design and construction of shallow foundation, scale effect should be considered. But, adequate guide and test result of scale effect were not prepared yet in Korea. So, to analyze the relations of bearing capacity and settlement as the difference of loading plate sizes, model test and field loading test were performed with different loading plate on weathered-granite layer. Model tests were conducted with water content, compaction number, saturated unit weight and plate size(Dl5, 25cm) in soil-box$(2,000\times 2,000\times 1,000mm)$ formed soil layer. Field loading tests were carried out with diameters of loading plate$(D15, 25, 30, 40, 75\times 75, 140\times 210cm)$ on the same soil condition. Finally, we presented the prediction formula of bearing and settlement for computating scale offset in design of shallow foundation through result analysis of load test and numerical simulation on weathered soil and rock.

Small Bowel Sparing Effect of Small Bowel Displacement System in 3D-CRT and IMRT for Cervix Cancer (자궁경부암의 3D-CRT와 IMRT시 소장전위장치의 소장 선량에 대한 영향)

  • Purpose : In radiotherapy for cervix cancer, both 3-dimensioal radiotherapy (3D-CRT) and intensity-modulated radiation therapy (IMRT) could reduce the dose to the small bowel (SB), while the small bowel displacement system (SBDS) could reduce the SB volume in the pelvic cavity. To evaluate the effect of the SBDS on the dose to the SB in 3D-CRT and IMRT plans, 3D-CRT and IMRT plans, with or without SBDS, were compared. Materials and Methods : Ten consecutive uterine cervix cancer patients, receiving curative radiotherapy, were accrued. Ten pairs of computerized tomography (CT) scans were obtained in the prone position, with or without SBDS, which consisted of a Styrofoam compression device and an individualized custom-made abdominal immobilization device. Both 3D-CRT, using the 4-field box technique, and IMRT plans, with 7 portals of 15 MV X-ray, were generated for each CT image, and proscribed 50 Gy (25 fractions) to the isocenter. For the SB, the volume change due to the SBDS and the DVHs of the four different plans were analyzed using palled t-tests. Results : The SBDS significantly reduced the mean SB volume from 522 to 262 cm$^{3}$ (49.8$\%$ reduction). The SB volumes that received a dose of 10$\~$50 Gy were significantly reduced in 3D-CRT (65$\~$80$\%$ reduction) and IMRT plans (54$\~$67$\%$ reduction) using the SBDS. When the SB volumes that received 20$\~$50 Gy were compared between the 3D-CRT and IMRT plans, those of the IMRT without the SBDS were significantly less, by 6$\~$7$\%$, than those for the 3D-CRT without the SBDS, but the volume difference was less than 1$\%$ when using the SBDS. Conclusion : The SBDS reduced the radiation dose to the SB in both the 3D-CRT and IMRT plans, so could reduce the radiation injury of the SB.

Implication of High Mobility Group Box 1 (HMGB1) in Multicellular Tumor Spheroid (MTS) Culture-induced Epithelial-mesenchymal Transition (Multicellular tumor spheroid (MTS) 배양에 의한 EMT에서 HMGB1의 역할)

  • As tumors develop, they encounter microenvironmental stress, such as hypoxia and glucose depletion, due to poor vascular function, thereby leading to necrosis, which is observed in solid tumors. Necrotic cells are known to release cellular cytoplasmic contents, such as high mobility group box 1 (HMGB1), into the extracellular space. The release of HMGB1, a proinflammatory and tumor-promoting cytokine, plays an important role in promoting inflammation and metabolism during tumor development. Recently, HMGB1 was shown to induce the epithelial-mesenchymal transition (EMT) and metastasis. However, the underlying mechanism of the HMGB1-induced EMT, invasion, and metastasis is unclear. In this study, we showed that noninvasive breast cancer cells MCF-7 formed tightly packed, rounded spheroids and that the cells in the inner regions of a multicellular tumor spheroid (MTS), an in vitro model of a solid tumor, led to necrosis due to an insufficient supply of O2 and glucose. In addition, after 7 d of MTS culture, the EMT was induced via the transcription factor Snail. We also showed that HMGB1 receptors, including RAGE, TLR2, and TLR4, were induced by MTS culture. RAGE, TLR2, and TLR4 shRNA inhibited MTS growth, supporting the idea that RAGE/TLR2/TLR4 play critical roles in MTS growth. They also prevented MTS culture-induced Snail expression, pointing to RAGE/TLR2/TLR4-dependent Snail expression. RAGE, TLR2, and TLR4 shRNA suppressed the MTS-induced EMT. In human cancer tissues, high levels of RAGE, TLR2, and TLR4 were detected. These findings demonstrated that the HMGB-RAGE/TLR2/TLR4-Snail axis played a crucial role in the growth of the MTS and MTS culture-induced EMT.

Implementation of Fingerprint Cognition System Based on Embedded LINUX (임베디드 리눅스 기반의 지문 인식 시스템 구현)

  • In this paper, we have designed a fingerprint cognition system based on the embedded Linux. The proposed algorithm in this paper use the wavelet transform to derive the special feature vector from the captured fingerprint and a probabilistic neural network is used to compare the feature vectors for the fingerprints. The system consists of server PC based on the Linux and the client based on the embedded Linux. The client is a Tynux box-x board using the PXA-255 CPU. For the acquisition of the fingerprint image, we use d the AS-S2 semiconductor sensor. The system is likely to be used to develop a police inspection system.

Recognition of small-obstacles using a camera and program for a mobile (이동로봇을 위한 카메라를 이용한 소형 장해물 인식)

  • This paper describes an image processing algorithm for recognition of small-obstacles using a camera and program for a mobile robot in indoor environment. Mobile robot could meet small-obstacles such as a small plastic bottle of about 1l in quantity, a small box of 7$\times$7$\times$7 cm$^3$ in volume, and so on in its designated path, and could be disturbed by them in the locomotion of a mobile robot. So, it is necessary to research on the image processing algorithm for recognition of small-obstacles using a camera and program. In this paper, 2-D the image processing algorithm for recognition of small-obstacles using a camera and program for a mobile robot in indoor environment was developed. The characteristic test of the developed program to confirm the recognition of small-obstacles was performed. It is shown that the developed program could recognize small-obstacles accurately.

A Study on the Characteristics of Coupling Loss Factor associated with Fluid Loading (접수구조물의 연성손실계수 변화에 관한 연구)

  • 본 연구에서는 연성손실계수가 구조물의 진동에 미치는 영향을 살펴보았다. 세부시스템의 두께가 동일할 경우 평판으로 이루어진 구조물이 접수되면 연성손실계수가 감소하게 된다. 특히, 저주파수 대역에서 연성손실계수가 크게 낮아졌다. 그 주된 원인은 접수로 인해 평판에서 전파되는 굽힘파의 그룹속도가 줄어들기 때문이며, 접수로 인한 파워투과계수의 변화는 크지 않았다. 내부에 격벽을 가진 steel box에 공기중 연성손실계수와 접수시 연성손실계수를 적용하여 세부시스템의 속도를 구한 결과, 공기중 연성손실계수를 이용하여 구한 속도가 접수시 연성손실계수를 이용하여 구한 속도보다 약 2-3 dB정도 높게 나타났다. 선박과 같은 복잡한 구조물인 경우 연성손실계수에 의한 속도 차이는 더욱 커질 것으로 추정된다. 따라서, SEA를 이용한 접수 구조물의 진동해석에서 해석오차를 줄이기 위해서는 연성손실계수에 접수의 영향을 고려해 주어야 함을 확인하였다. 특히, 접수효과가 고려된 연성손실계수를 사용하면 SEA가 큰 오차를 보이는 중, 저주파수 대역에서 해석 결과에 대한 신뢰성을 높일 것으로 판단된다.

Implementation of Small Sized Designs for Economic Estimation of Second-Order Models (2차 모형의 경제적 추정을 위한 소형실험계획의 활용)

  • Response surface methodology (RSM) is a useful collection of experimentation techniques for developing, improving, and optimizing products and processes. When we are to estimate second-order regression model and optimize quality characteristic by RSM, central composite designs and Box-Behnken designs are widely in use. However, in developing cutting-edge products, it is very crucial to reduce the time of experimentation as much as possible. In this paper small-sized second-order designs are introduced and their estimation abilities are compared in terms of D-optimality, A-optimality, and variance of regression coefficients, ease of experimentation, number of experiments. Then we present a guideline of using specific designs for specific experimentation circumstances. The result of this study will be beneficial to experimenters who face experiments which are expensive, difficult, or time-consuming.
