The sensitivities of 270 pathogens (124 Streptococci, 118 Staphylococci, 10 Corynebacterium pyogenes and 18 Escherichia coli) isolated from clinical or subclinical cases of bovine mastitis during lactation to 11 antibiotics were determined by the agar plate dilution method. All cultures of Streptococci were inhibited at $1.56{\mu}g/ml$ of ampicillin and 1.56 units/ml of penicillin G. Most of the cultures were inhibited at $3.125{\mu}g/ml$ of leukomycin, but were resistant to kanamycin, streptomycin, chloramphenicol, erythromycin and colistin at concentration of $12.5{\mu}g/ml$. More than 93% of the Staphlococcal cultures were sensitive to kanamycin, leukomycin, ampicillin, tetracycline, ledermycin and minocycline at concentrations of $12.5{\mu}g/ml$ or less, and sensitive to penicillin at concentration of 3.125 units/ml, but for more than 71% of the cultures to chloramphenicol, erythromycin and colistin the concentrations required to inhibit growth were 100 ${\mu}g/ml$ or higher. All 10 cultures of Corynebacterium pyogenes were inhibited by leucomycin, ampicillin and minocycline at concentration of $1.56{\mu}g/ml$ and by penicillin G at concentration of 0.78 units/ml, but all the cultures required at least $400{\mu}g/ml$ or higher of streptomycin, erythromycin and colistin for inhibition. More than 83% of E. coli cultures were sensitive to erythromycin and minocycline at concentration of $12.5{\mu}g/ml$, but resistant to leucomycin and chloramphenicol at concentration of $100{\mu}g/ml$.

This study was conducted to analyze penicillin G (PEG), streptomycin (STR) and neomycin (NEO) residues in milk of healthy lactating cows. Milk samples were collected from all four quarters of 12 dairy cows 2−7 days after intramammary infusions of an ointment containing PEG, STR and NEO once (n = 4; group I) or twice (n = 4, group II) daily. Ultra-performance liquid chromatography coupled with electrospray tandem mass spectrometry was used to determine the antibiotic residues in the samples. The correlation coefficient ($r^2$) of the calibration curves for all antibiotics was > 0.999 and the limits of detection and quantification were $0.002-0.005{\mu}g/mL$ and $0.007-0.02{\mu}g/mL$, respectively. Recovery rates were ranged from 75.5 to 92.3%. In group I, PEG, STR and NEO residues were detected in milk at 2, 3 and 2 days post-treatment, respectively, which were below the maximum residue limit (MRL). In group II, PEG, STR and NEO residues were detected in milk at 2, 3 and 3 days post-treatment, respectively, which were bellow the MRL. These results suggest that a 3-day for milk withdrawal period after the ointment treatment might be sufficient for reduction of the antibiotic residues below the MRL.

Staphylococcus aureus is a representative pathogenic bacterium carefully controlled in the dairy industry because it causes bovine mastitis and thus, can enter the dairy chain. Furthermore, the emergence of multi-drug resistant S. aureus is a big problem. We previously isolated a Lactococcus lactis strain producing a bacteriocin that exhibited strong antimicrobial activity against S. aureus. In this study, we investigated the synergistic inhibition of S. aureus by the bacteriocin and a bacteriophage (SAP84) which is specific to the organism. The bacteriocin (12.5-100 AU/mL) inhibited the growth of S. aureus KCTC 3881 in a dose-dependent manner, as did the bacteriophage SAP84 (0.001-1 MOI; multiplicity of infection). Co-treatment with the bacteriocin (100 AU/mL) and the bacteriophage (0.1 MOI) significantly inhibited the growth of S. aureus compared to each treatment alone (bacteriocin or bacteriophage), indicating the two components showed synergistic inhibition of S. aureus. Therefore, the bacteriocin and bacteriophage combination can be used as a good strategy for controlling pathogenic bacteria.
In vitro antifungal susceptibility test was carried out on 53 strains of Candida spp isolated from milk of dairy cows with subclinical mastitis and teat cups of milking machines, Nystatin, clotrimazole, miconazole, econazole, 5-fluorocytosine, cycloheximide, haloprogin and griseofulvin were tested by the agar dilution method. The 84.8% to 98.2% of Candida strains were inhibited by clotrimazole, econazole and miconazole at $${\leq_-}25{\mu}g/ml$$, and clotrimazole was most active. Interspecies differences of antifungal susceptibility were recognized and these were as follows. C albicans was most sensitive to clotrimazole (GM-MIC, $5.49{\mu}g/ml$) followed by 5-fluorocytosine, econazole and miconazole. C pseudotropicalis and C guilliermondii were notably sensitive to haloprogin, clotrimazole, miconazole, cconazole, 5-fluorocytosine, and haloprogin (GM-MIC, $0.17{\sim}0.19{\mu}g/ml$) was most active. C krusei was most sensitive to cycloheximide (GM-MIC, $0.54{\mu}g/ml$) followed by clotrimazole, haloprogin, miconazole and econazole. C parapsilosis was somewhat sensitive to econazole, cycloheximide, clotrimazole, and econazole (GM-MIC, $7.26{\mu}g/ml$) was most active. C tropicalis showed very low sensitivity to all tested drugs (GM-MIC, $${\geq_-}20.32{\mu}g/ml$$).
The present study was carried out to investigate the antibacterial activity against Staphylococcus aureus with antibiotic-resistance in vitro and the skin irritation in rabbits with surfactin produced by Bacillus subtilus Complex BC1212. The antibacterial activities of selected antimicrobial agents (surfactin, amoxacillin, colistin, norfloxacin and streptomycin) were evaluated by using the broth microdilution method. As the results, the minimum inhibitory concentration (MIC) of the surfactin was less than 15.6 ㎍/ml. In the skin irritation test, two out of 4 rabbits showed very slight edema at 24 h after the administration of surfactin, and then recovered at 72 h. The change of body weight was normal during the skin irritation test. The primary irritation index in accordance with the Draize evaluation of topical reaction was calculated to be '0.125', which meant not irritating. Based on these results, it could be concluded that the test agent, surfactin, was a non-irritant. We could also think that the surfactin may be useful for the treatment of S. aureus infections such as bovine mastitis.
The prevalence of yeasts in mammary glands of dairy cows and teat cups of milking machines was studied in Chinju area. The rate of subclinical yeast infection in 330 quarters was 3.6%. Of 12 isolates from the milk, 4 Candida pseudotropicalis, 3 C tropicalis, 2 C krusei, 2 C albicans and 1 Rhodotorula spp were identified. The 91.7% of the isolates belonged to the genus Candida and C pseudotropicalis was the predominant species. From 20.5% of 200 teat cups tested, 51 strains of yeasts were isolated. These were 13 C pseudotropicalis, 9 C guilliermondii, 7 C tropicalis, 5 C krusei, 5 C parapsilosis, 3 C albicans, 2 Torulopsis glabrata, 2 Geotrichum candidum and 5 unidentified yeasts. C pseudotropicalis was most frequently encountcred.
Probiotics colonize the intestines and exert an antibacterial effect on pathogens. Therefore, probiotics could be used as a preventive agent against lethal infections. To isolate probiotic microorganisms, 116 bacterial strains were isolated from healthy cow's milk and were subjected to Gram-stain, morphology and biochemical analyses, Vitek analysis, and 16S rRNA analysis. One of the strains identified as Bacillus (B.) thuringiensis 87 was found to grow very well at pH 4.0~7.0 and to be resistant to high concentrations of bile salts (0.3~0.9% w/v). B. thuringiensis was susceptible to the antibiotics used in the treatment of bovine mastitis, yet it exhibited an antimicrobial effect against Staphylococcus (S.) aureus 305. Moreover, it protected mice from experimental lethal infections of E. coli O55, Salmonella typhimurium 01D, and S. aureus 305 through a significant induction of interferon-${\gamma}$, even at four-week post-administration of B. thuringiensis. Although oral administration of B. thuringiensis 87 did not provide significant protection against these lethal challenges, these results suggest that B. thuringiensis 87 could be a feasible candidate as a probiotic strain.
Oligomeric proanthocyanidins (OPCs), classified as condensed tannins, have significant antioxidation, anti-inflammation and anti-cancer effects. This study was performed to investigate the anti-inflammatory effects of OPCs and the mechanism underlying these effects in lipopolysaccharide (LPS)-stimulated bovine mammary epithelial cells (MAC-T). Real-time PCR and ELISA assays indicated that OPC treatment at 1, 3 and 5 ㎍/ml significantly reduced the mRNA and protein, respectively, of oxidant indicators cyclooxygenase-2 (COX-2) (p < 0.05) and inducible nitric oxide synthase (iNOS) (p < 0.01) as well as inflammation cytokines interleukin (IL)-6 (p < 0.01), IL-1β (p < 0.01) and tumor necrosis factor-α (TNF-α) (p < 0.05) in LPS-induced MAC-T cells. Moreover, OPCs downregulated LPS-induced phosphorylation of p65 and inhibitor of nuclear factor kappa B (NF-κB) (IκB) in the NF-κB signaling pathway (p < 0.01), and they inhibited p65 translocation from the cytoplasm to the nucleus as revealed by immunofluorescence test and western blot. Additionally, OPCs decreased phosphorylation of p38, extracellular signal regulated kinase and c-jun NH2-terminal kinase in the MAPK signaling pathway (p < 0.01). In conclusion, the anti-inflammatory and antioxidant activities of OPCs involve NF-κB and MAPK signaling pathways, thus inhibiting expression of pro-inflammatory factors and oxidation indicators. These findings provide novel experimental evidence for the further practical application of OPCs in prevention and treatment of bovine mastitis.
The recombinant bovine somatotropin (rbST) has been used for increasing milk production of dairy cows without adverse health effects. This study was conducted to compare effects of supplementation with $Boostin^{(R)}$-250 containing 250 mg of rbST on milk production with those of $Posilac^{(R)}$ and $Boostin^{(R)}$-S. And safety of rbST supplementation on target animals was also observed. Each twenty-five lactating dairy cows were assigned randomly to one of four groups. $Boostin^{(R)}$-250 and vehicle (control) were administered weekly. $Boostin^{(R)}$-S and $Posilac^{(R)}$ were administered two week intervals. Milk yield, milk components, milk somatic cell count, health status, and body condition score of cows were examined. Supplementation with $Posilac^{(R)}$, $Boostin^{(R)}$-S, and $Boostin^{(R)}$-250 induced more milk yield than control group by 2.9 kg/day (12.3%), 4.2 kg/day (17.9%), and 4.1 kg/day (17.4%), respectively. There was a significant difference in milk yield among three rbST treatment groups and control group (${\alpha}$ = 0.05). The rbST supplementation did not increase the incidence of clinical mastitis and milk somatic cell counts. Supplementation with rbST did not significantly affect milk components (milk fat, protein, and solid not fat). The rbST supplementation of the dairy cows after peak milk yield did not cause negative effect on BCS. However, some cows less than 100 days in milking had decreased BCSs after rbST supplementation. In conclusion, milk production in 250 mg of rbST administered cows every week was similar to that of 500 mg of rbST administered cows every 2 weeks. And supplementation of 250 mg of rbST every week could reduce metabolic stress in cows.
