Several animal species including pigs are directly involved in the zoonotic transmission of the hepatitis E virus (HEV) to humans. This study was conducted to detect HEV in bovine and porcine raw livers by nested reverse transcription-polymerase chain reaction. Zoonotic HEV strains were identified in 1.0 and 3.0% of the tested bovine and porcine livers, respectively. HEV-4 was detected in the bovine livers, but both HEV-3 and HEV-4 were identified in the porcine livers. These results indicate that zoonotic transmission of HEV may occur via consumption of raw or undercooked livers of pigs and cattle.
A case of disseminated lymphoblastic lymphosarcoma is reported in an aborted bovine fetus 7 month in gestation. Grossly, numerous tan firm nodules, 2 to 5 mm in diameter were scattered throughout the myocardium and skeletal muscle. The corticomedullary junction of the kidney was discolored to whitish tinct. Histologically, compact sheet of monomorphic lymphoblastic lymphoid cell infiltration was noted in the myocardium and skeletal muscle. Neoplastic cell infiltration was also noted in the corticomeullary junction and deep cortical regions of the kidney, lung and the liver. This is believed to be the first reported case of lymphosarcoma in an aborted bovine fetus in Korea.
The surface of albumin microspheres was modified with methotrexate(MTX) by using 1,3-dicyclohexylcarbodiimide (DCC). Surface-modified albumin microspheres entrapping no MTX (SAMS), free MTX (SAMSF) and MTX-bovine serum albumin(BSA) conjugates(SAMSC) were prepared. The organ-targeting ability of free $[^3H]MTX,\;[^3H]MTX-BSA$ conjugate and the above microspheres was evaluated after i.v. administration of the preparations, equivalent to 150 nCi via the tail vein of mice. The total radioactivity in the lung increased immediately in a few minutes after i.v. injection of the microspheres, and then declined for the period of 3-4 weeks. However, the radioactivity in the liver, spleen and kidney increased slowly during the rapid decrease in radioactivity in the lung. This suggested that the microspheres could be entrapped rapidly in the lung through mechanical filtration because of their large size and slowly redistributed to the liver, spleen and kidney due to either the microspheres being degraded enough for the size to allow passage through the capillary beds of the lung and/or the release of $[^3H]MTX\;or\;[^3H]MTX-BSA$ conjugates from the microspheres. The amount of $60{sim}70%$ of the dose was targeted to the liver after the i.v. injection of SAMS, SAMSF and SAMSC, and the values of $(R_e\;^*\;_{e)liver}$ from the microspheres were $5{\sim}7$ compared to free MTX. This suggested that the liver-targeting ability from surface-modified albumin microspheres could be $5{\sim}7$ times as that of free MTX. The liver-targeted drug was accumulated in the Kupffer cells at the initial stage, thereafter the drug in the Kupffer cell was slowly transferred into the hepatocytes. The value of AUQ for liver from SAMS was higher than that from SAMSF, but much lower than that from SAMSC. This suggest that MTX bound to their surface could be eliminated slower than the entrapped free MTX, and faster than the entrapped MTX-BSA conjugates. This is consistent with the in vitro release rates order in the presence of a proteolytic enzyme. Also, surface-modified MTX was scarcely released in the absence of a proteolytic enzyme. Therefore, the surface-modified MTX nay be released (or eliminated) rapidly from SAMSC at the target site, and thereafter MTX may be released (or eliminated) slowly from the entrapped MTX-BSA conjugates in SAMSC for a long period.

In attempt to develop a drug delivery system using serum albumin microspheres, bovine serum albumin microspheres containing antitumar agent. Cytarabine, were prepared. The shape, surface characteristics, size distribution, behavior of in vivo distribution, drug release behavior, and degradation of albumin microsphers in animal liver issue homogenate and proteolytic enzyme were investigated. The shape of albumin microspheres was spherical and the surface was smooth and compact. The size distribution of the albumin microspheres was effected by dispertion forces during emulsification and albumin concentration. Distribution of albumin microspheres after imtravenous administration in rabbit was achieved immediately. In vitro, albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin concentration ratio and size distribution. This study was designed to evaluate the efficacy of antioxidants and amino acid with buffalo rat liver cell(BRLC), bovine oviductal epithelial cell(BOEC) and STOC monolayers in supporting the development of in vitro matured(IVM) and in vitro fertilized(IVF) bovine oocytes. Bovine embryos developed to the 2~8 cell stage after in vitro fertilization were cultured for 5 to 6 days at 39$^{\circ}C$ in CR1aa containing antioxidants and amino acids with various somatic cells. Embryo development was examined and cell numbers of blastocysts were counted by fluorescence staining method. In experiment 1, the proportion of embryos that reached the blastocyst stage in control, catalase(250U), SOD(600U), glutathione(100$\mu$M) and taurine(2.5mM) with BRLC were 11.4, 8, 0, 16.7 and 43.4 respectively. Taurine(2.5mM) with BRLC group was significantly the highest among treatments(P<0.05). In experiment 2, in vitro development rate into blastocyst in control, catalase(250U), SOD(600U), glutathione(100$\mu$M) and taurine(2.5mM) with BOEC were 15.8, 23.5, 22.8, 28.6 and 56.9 respectively. In experiment 3, embryonic development in all treatments as control, catalase(250U), SOD(600U), glutathione(100$\mu$M) and taurine(2.5mM) added to CR1aa with STO cells were 23.5, 24.5, 17.0, 28.8 and 50.0 blastocysts. These results show that antioxidants and amino acids with somatic cells can provide a significant benefit for coculture of early bovine embryos derived from IVM and IVF.
This study was performed for assessing carcinogenicity of several synthetic hormones; Diethylstilbesterol (DES), 17-ethinylestradiol ($EE_2$) and Bovine somatotrophin (BST). Six weeks old F344 rats were divided into five groups and given an intraperitoneally injection of 200 mg of diethylnitrosamine (DENA). At two week after beginnig of experiment, DES, $EE_2$, BST. Phenobarbital were administered to group 1, 2, 3 and 4 respectively, group 4 is positive control and group 5 is negative control. At the same time, all groups received a single i.p. injection of D-galactosamine at a dose of 300 mg/kg and underwent 2/3 partial hepatectomy at week 5. All rats were sacrificed at the end of week 8 for assessment of liver lesion development. The liver was processed for immunohistochmical staining for GST-P and quantitatively analyzed by image analyzer. It was concluded that two synthetic estrogen hormones (DES, $EE_2$) was different significantly (p < 0.01) but BST was not different as compared with control group. Therefore, we thought that DES, $EE_2$ was promoting effects and BST was not in rat hepatocarcinogenesis.
A new method for the simultaneous determination of six glucocorticoids (betamethasone, dexamethasone, prednisone, prednisolone, methylprednisolone, and flumethasone) in meats (bovine muscle, bovine liver) were established. Samples were effectively extracted using C18 cartridge with ethylacetate. Chromatographic separation was achieved using C18 column and negative electrospray ionization mass spectrometry was performed in the Multiple Reaction Monitoring mode for the effective quantitation and qualification of glucocorticoids. Acetonitrile and water (0.1% formic acid) were used as mobile phase and additive for effective electrospray ionization, and gave good chromatographic separation and mass spectrometric sensitivity. The limit of detection (LODs) and the limit of quantitation (LOQs) in spiked blank samples depending on types of matrix and pharmaceuticals were ranged from 0.2 to $1.0\;{\mu}g$/kg and 0.8 to $3.4\;{\mu}g$/kg, respectively. And the recoveries were between 89.5 to 119.6%. The established method showed good recoveries, accuracies, precisions and fast sample preparation and it will be applied to assay of glucocorticoids residues in meat.
The purpose of this experiment was to determine the effects of thiol compounds, $\beta$-mercaptoethanol($\beta$-ME) and cystearrone with buffalo rat liver cell(BRLC) co-culture on the development and intracellular glutathione(GSH) concentrations of bovine embryos produced by in vitro inaturation(IVM) and in vitro fertilization(IVF). Bovine IVM /IVF embryos developed to 2~8 cell stage were co-cultured with BRLC in GRlaa with or without thiol compounds. The developmental rate beyond morulae stage in CRlaa containing 0, 10,25 and 50$\pi$M $\beta$-ME with BRLG were 63.0, 74.0, 72.3 and 77.1%, respectively. And the developmental rate with 0, 25, 50 and 75$\pi$M cystearnine with BRLC were 69.6, 77.6, 81.0 and 76.8%, respectively. The developmental rate beyond morulae stage of GRlaa containing thiol compound with BRLG group was higher than that of control group. The intracellular GSH concentrations of blastocysts cultured for 5 days in GRlaa containing 0 and 50$\pi$M $\beta$-ME or cysteamine with BRLG were 81.2 and 86.4, 83.2 and 84.2pM, respectively. The intracellular GSH concentrations of blastocysts in GRlaa containing thiol compounds with BRLG was slightly higher than that of control group The cell numbers of blastocysts were not difference in all experimental groups. These results indicate that thiol compounds with BRLG co-culture was increased the percentage of developed into morulae and blastocysts, and intracellular GSII concentrations of blastocysts embryos.

Simultaneous multiresidue analysis using liquid chromatography determination for five benzimidazole anthelmintics(thiabendazole, oxibendazole, albendazole, mebendazole and fenbendazole) in bovine muscle, liver and omasum has been described. Blank or benzimidazole-fortified samples(0.5g) were blended with bulk $C_{18}$($40{\mu}m$, 18% load, endcapped, 2g). A column made from the resultant $C_{18}$/animal tissue matrix was first washed with hexane($8m{\ell}$), following which the benzimidazoles were eluted with acetonitrile($8m{\ell}$). Analytes of extracted sample were determined by liquid chromatography with UV detector at 290nm. Correlation coefficients of standard curves for individual benzimidazole isolated from fortified samples, using internal standardization, were linear($0.991{\pm}0.007$ to $0.996{\pm}0.005$) with average relative percentage recoveries from $62.1{\pm}3.8(%)$ to $92.3{\pm}7.5(%)$ for the concentration range($0.2{\sim}6.4{\mu}g/g$), respectively. Recoveries rates of TBZ, MBZ in liver, OBZ, MBZ in muscle and TBZ, MBZ in omasium from fortified benzimidazole were 92.%, 87.3%, 74.5%, 82.7%, 75.2% and 83.5% at condition II, respectively. Condition II showed higher recoveries rates than condition I. These results indicated that the matrix solid phase dispersion(MSPD) methodology is acceptable for the determination of 5 benzimidazole anthelmintics and may also suitable for other matrixes of food animal origin.
