A survey on ornamental diseases in 2000 and 2001 showed frequent occurrence of blight symptoms on leaves and stems of columbine and bleeding heart in botanical gardens of Gangwon and Chungnam provinces in Korea. A total of 65 isolates of Streptobotrys sp. were obtained from the infected leaves and stems of the two plants. All isolates were identified as Streptobotrys caulophylli based on their morphological and cultural characteristics. Blight symptoms were induced on leaves of the two plants by artificial inoculation with the isolates. This is the first report that S. caulophylli causes leaf and stem blight on columbine and bleeding heart in the world.
The gesneriaceous perennial plant Titanotrichum oldhamii has beautiful foliage and attractive bright yellow flowers. However, breeding of T. oldhamii by conventional sexual hybridization may be difficult because sexual reproduction of this species is very rare. In the present study, plant regeneration systems via both direct and indirect formation of adventitious shoots from leaf explants were established as the first step toward breeding T. oldhamii by using biotechnological techniques. Adventitious shoots were formed efficiently on medium containing $0.1mg\;l^{-1}$ benzyladenine. Histological observation showed that shoot formation on this medium occurred directly from leaf epidermal cells without callus formation. On the other hand, leaf explants formed calluses on medium containing $0.1mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid. The calluses could be maintained by monthly subculturing to fresh medium of the same composition. When the calluses were transferred to plant growth regulator-free medium, they formed adventitious shoots. Directly and indirectly formed shoots rooted well on medium containing $0.1mg\;l^{-1}$ indole-3-butyric acid. Plantlets thus obtained were successfully acclimatized and grew vigorously in the greenhouse. Flow cytometry analysis indicated that no variation in the ploidy level was observed in plants regenerated via direct shoot formation, whereas chromosome doubling occurred in several plants regenerated via indirect shoot formation. Regenerated plants with the same ploidy level as the mother plants showed almost the same phenotype as the mother plants, whereas chromosome-doubled plants showed apparent morphological alterations: they had small and crispate flowers, and round and deep green leaves.

As part of a study to understand the current status of Korean plant species propagated in foreign countries, we conducted extensive surveys on Korean living collections in the Arnold Arboretum of Harvard University, the oldest public arboretum in North America. Specifically, we collected and identified the voucher specimens of the living collections and further examined the collection databases to understand the history of how Korean plants were introduced to the Arboretum. We found that a total of 154 Korean taxa (belonging to 49 families and 81 genera) have been planted in the arboretum. Most plants originated from seeds that were directly collected in Korea or obtained via seed exchanges with botanical gardens in Asia, Europe, and U.S.A. Of the 154 taxa, 16 are Korean endemic species, and three are listed as endangered species. In addition, 62 taxa are as floristic regional indicator species of Korea used by the Ministry of Environment.
The purpose of this study was to investigate satisfaction of student, adult and forest interpreters about facilities and program of children's garden, which was constructed as edutainment space within Korea National Arboretum on May 2011. The results are as follows. First, an information about construction of children's garden should be provided actively, because 70.8% student and 68.6% adult of respondent did not know existence of children's garden. Second, it has been analysed that a fundamental conception of GREEN-I be reflected in construction children's garden, especially, reflecting conception of Enjoy and Edutainment was comparatively high. Third, It was found that ecological experiencing activities program of children's garden was positively to develop environmental literacy for student. Fourth, It was found that satisfaction of uncommon planting area was very low among planting area and satisfaction of forest interpreters was very low about place of ecological experience learning using five senses. Finally, for effective operation of children's garden, children's garden should be provided planting for environmental education connected with the school curriculum and ecological experiencing activities program to enjoy time with the family. Therefore, these results will be applied to improve children's garden and could be applied to construct other children's garden in botanical gardens.
Inspecting herbaria collections of Laurencia rigida highlighted frequent misidentifications between L. rigida and L. heteroclada f. decussata, two poorly studied taxa from Australia. Recent collections of DNA material, including from topotype material, allowed for re-examination of these two taxa using molecular techniques. Detailed morphological and molecular analyses based on two markers (rbcL and COI-5P) strongly supported these two taxa as being distinct from each other and requiring nomenclatural changes. Comprehensive morphological analyses highlighted features useful for accurate identifications. Interestingly, L. rigida was found to belong to the genus Palisada with evidence from both the morphology and molecular data. Therefore, this study proposed recognizing L. rigida as Palisada rigida comb. nov. Molecular data for L. heteroclada f. decussata on the other hand supported its separation from L. heteroclada, with too great a molecular distance to be considered a variety. Morphological characters that best separated P. rigida from L. decussata included seven characters; number of pericentral cells per vegetative axial segment, the presence of secondary pit connections, the presence of lenticular thickenings, tetrasporangia alignment, the presence of corps en cerise, holdfast morphology, and overall plant shape. Morphologically, L. heteroclada f. decussata was also separated from L. heteroclada, particularly by the following characteristics; ultimate branchlets morphologies, lower order branch lengths, primary axis and holdfast morphologies. Therefore, it was proposed that L. heteroclada f. decussata is recognized at a species level as L. decussata comb. et stat. nov.

We conducted phylogenetic analyses using multiplexed inter-simple sequence repeat genotyping by sequencing and compared chloroplast DNA sequences among Ardisia japonica, A. pusilla, and morphologically intermediate plants found on Jeju Island, Korea. Our network analysis demonstrated that the intermediate plants were genetically positioned between A. japonica and A. pusilla. Our comparison of the intergenic spacer between the psbA and trnH genes in chloroplast DNA indicated that four nucleotide substitutions separate A. japonica and A. pusilla, whereas the intermediate plants exhibited the A. japonica haplotype. Our results suggest that the intermediate plants on Jeju Island represent a natural hybrid of A. japonica, as the maternal species, and A. pusilla, and that they are attributable to Ardisia×walkeri. This record constitutes the first documented occurrence of the hybrid taxon in Korea.

With a history spanning 9,000 years, yeast has played a central role in the traditional production of fermented foods, particularly alcohol and bakery. Recent research has highlighted the versatility of yeast in various industries, including the production of bioactive compounds, bioethanol, pigments, and enzymes, and as a host for molecular biology studies. In Korea, yeast isolation has traditionally focused on traditional fermented foods such as soybean paste. However, there is a growing trend in the isolation and characterization of yeasts from natural environments such as flowers and fruits for industrial applications. In this study, we isolated and identified 12 yeasts from various natural environments in Korea, including botanical gardens and parks that are not listed on the National Species List of Korea (NSLK). These newly discovered species included Sakaguchia oryzae, Cystobasidium raffinophilum, Meira argovae, Kazachstania humilis, Meyerozyma smithsonii, Anthracocystis trispicatae, Naganishia brisbanensis, Tremella yokohamensis, Kwoniella shandongensis, Kwoniella newhampshirensis, Aureobasidium proteae, and Rhodotorula dairenensis.
일제강점기 조선의 궁궐은 많은 부분 훼손되었다. 창경궁은 특히 동물원과 식물원, 박물관 등이 건립되면서 가장 많이 훼철된 궁궐이다. 본 연구는 창경궁 내전 구역에서 가장 중요한 공간으로 여겨졌던 자경전 터의 바로 옆에 자리 잡은 장서각(이왕가박물관)의 건립 과정을 사료와 현장 조사를 통해 고찰하였다. 1911년에 건립된 장서각은 창경궁 전체를 내려다보는 위치이자 멀리 남산의 경성신사를 바라볼 수 있는 곳에 자리 잡았다. 장서각의 건립 과정에서 주변 공간의 변화는 다음과 같이 요약할 수 있다. 첫째, 1910년대 초에는 장서각 터를 조성하기 위해 자경전 후면부 동산의 지형과 장고(醬庫) 일부가 훼손되었다. 장서각 전면부에 앞마당을 조성하면서 장대석 석축을 시공하였으며, 남쪽으로 계단을 설치하였다. 이 과정에서 양화당 후면부의 석계의 원형은 파괴되었고, 정일재나 기타 건물 등은 철거되었을 것으로 추정된다. 둘째, 1920년대에는 많은 전각이 헐리고 평탄화 작업을 거쳐 동식물원과 박물관이 완공되었다. 장서각이 완공된 후에는 내전과 주변의 관람동선도 변경되었다. 장서각 앞마당과 계단 주변의 화계에는 벚나무와 모란을 심고, 양화당과 집복헌 사이에는 일본식 정원을 조성하였다. 셋째, 1930년대에는 장서각 주변의 동선이 현재의 모습으로 완성되었으며, 박물관인 장서각과 동식물원, 그리고 벚나무 동산이 된 창경궁은 일본식 문화공원으로 탈바꿈되었다. 그 이후 철거될 때 까지 주변 공간은 크게 변하지 않았다. 현재 궁궐의 복원은 문화재청의 장기적이고 국가적인 사업이다. 본 연구의 결과는 차후 창경궁 내전권역의 복원 계획에 중요한 자료를 제공할 것이며, 향후 관련 연구자들에게 또 다른 정보를 제공할 것으로 기대한다.
1484년 9월 성종조에 조성된 창경궁은 순종 1년(1907) 창경원으로 격하되기 전까지 조선시대 궁원으로서의 모습을 보여준 전통공간이다. 일제강점기에 창경궁 내에 대온실, 박물관, 동 식물원이 개설되며 전통건축물과 궁궐의 정원이 소실되거나 변형되었다. 1984년 "창경궁 정비계획"에 의해 일제강점기 설치된 각종 시설과 관람로 등이 정비되었고, 전통적인 궁궐로 복원하고자 1986년 중건 공사를 통해 현재의 모습으로 복원한 이후 30여 년간 1980년대 중건 계획의 모습대로 유지 관리되고 있다. 창경궁 궁원(宮苑)의 모습은 19세기 초반 창경궁을 그린 "동궐도"에 그 모습이 잘 나타나고 있으며, "동궐도" 분석을 통해 조선시대 궁원의 원형을 확인할 수 있다. 1984년부터 시작된 창경궁 식생조사는 2005년, 2010년, 2016년에 걸쳐 4회 전수조사 되었고 식생조사 결과를 토대로 창경궁 내 식생 현황을 확인할 수 있었다. 30여 년간의 식생조사 결과와 "동궐도" 식생 분석을 통해 파악된 창경궁 내 식생 현황을 비교 분석해 봄으로써 창경궁 식생 현황이 조선시대 궁원의 모습으로 접근해 가는지 확인할 수 있으며 전통 궁원의 모습을 보전하기 위한 식생보전방향을 제시하였다.
우리나라에서 앞으로 적용가능한 식물의 서식처 및 종의 선정에 대한 보전평가기준 특히 식물적 가치에 대한 기준을 마련하기 위하여 1969년부터 1986년 사이에 발표된 야생동물의 보전가치의 평가에 관련된 논문에 발표된 기준을 재정리하였다. 희귀성, 다양성, 크기 및 정도 및 자연성 등은 대부분의 논문에서 사용되고 있는 기준이었다. 위와 같은 내용을 우리나라의 상황과 비교하여 서식처의 크기를 고려하여 16개의 요인을 정리하였다. 인간의 간섭, 생태적 분쇄성 및 희귀성 등은 서식처의 규모가 비교적 좁은 경우에 고려되어야 할 요인인 반면, 지역, 생태적 및 지리적 단위내에서의 다양성 위치 및 자연성 등은 입지의 규모가 큰 지역에 고려되어야 할 요인으로 판단되었다. 유일성, 전형성, 연구 및 교육적 가치 등은 규모의 크기 여하를 불문하고 고려되어야 할 요인으로 판단되었다. 본 기준이 우리나라에서 실제로 적용되어 앞으로 적용가능한 평가기준이 될 수 있도록 해야 하는 과제가 남아 있기는 하지만, 장차 우리나라에서 마련되어야 할 보호지역 및 특정식물의 평가기준을 위한 지침이 될 수 있으리라 판단된다.
