The effects of vitamin $D_{3}$ supplementation on bone mineral density and bone mineral content in ovariectmized rats were investigated. Forty female Sprague-Dawley rats (body weight 180 g) were divided into two group, ovariectomy and Sham groups, which were each randomly divided into two subgroups that were fed casein and casein supplemented with vitamin $D_{3}$ diets. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXlmus (GE Lunar Co, Wisconsin, USA) in spine and femur on 6 weeks after feeding. Osteocalcin and urinary DPD crosslinks value were measured as markers of bone formation and resorption. The body weight gain and food efficiency ratio (FER) were higher in OVX groups than in Sham groups regardless of diets. Serum Ca concentration and urinary Ca excretion were higher in vitamin $D_{3}$ supplemented group than in casein group in Sham and OVX. Crosslinks values were higher in OVX groups than in Sham groups. Spine BMD and femur BMD of ovariectomy group were significantly lower than Sham groups, however vitamin $D_{3}$ supplemented groups were significantly higher than control groups in Sham and OVX. In conclusion, dietary vitamin $D_{3}$ supplementation on ovariectimized rats were significantly increased bone mineral density and bone mineral content in spine and femur.
To investigate the effect of dietary Ca levels on metabolic changes of Ca and skeleton in postmenopausal women, 10-month-old ovariectomized female rats were compared with 2 month old rats. The rats were fed either 0.2% or 1.2% Ca diets for 16 weeks. Food intake and weight gain as higher in rats fed high Ca diets and in ovariectomized rats. Apparent Ca absorption as higher, and Ca balance was lower in the low Ca groups. Vertebrae density was higher in old rats or those fed a high Ca diets. The old rats and ovariectomized rats showed decreased bone formation, increased bone resorption and kidney function deterioration resulting in increased urinary Ca excretion. Contradictory to the above observation, old rats and ovariectomized rats still showed higher bone mass and bone ash content. Therefore aging was not fully onging in 10-month-old rats. Bone weights, mineral contents, and mineral/wt ratio were lower in ovariectomized rats. Dietary Ca level did not affect urinary Ca excretion, urinary protein excretion, GFR, serum alkaline phosphatase, or urinary hydroxyporline excretion. This means that dietary Ca level did not influence kidney function or bone turnover. However Ca content and the ash content of femur, 4th vertebra, and scapula were increased in high Ca groups. Therefore, it is considered that decreased bone formation and accelerated bone resorption may account for the increased osteoporotic risk in women in menopause after middle age. However, Ca metabolism can be improved and bone components can be maintained if Ca is supplemented.
Modification of the diet during childhood and adolescence may be an effective strategy for maximizing the peak bone mass. Many supplementation studies have suggested a positive effect of the increased vitamin D intake on the bone mineral status in the elderly. However to date all studies have been conducted on old men and postmenopausal women. The aim of this study was to examine the effects of vitamin D supplementation on the bone mineral density and bone mineral content in growing rats. Twenty Sprague-Dawley female rats were divided into two groups; Control, and vitamin D supplementation. The bone mineral density(BMD) and bone mineral content(BMC) were measured using PIXImus in the spine and femur. Vitamin D supplementation did not affect the level of weight gain, mean food intake and food efficiency ratio. In addition, vitamin D supplementation had no added effect on the spine and femur BMD, and BMC. There were no significant differences in the spine BMD/weight and BMC/weight between the groups, but the spine BMD/weight and BMC/weight was 11 % higher in the vitamin D supplementation group. The femur BMD/weight and femur BMC/weight were significantly higher in the vitamin D supplementation group 9 weeks after the experiment. These results provide evidence of the beneficial effects of vitamin D supplementation on the BMD during the growth period.
Peri-acetabular bone ingrowth plays a crucial role in long-term stability of press-fit acetabular cups. A poor bone ingrowth often results in increased cup migration, leading to aseptic loosening of the implant. The rate of peri-prosthetic bone formation is also affected by the polar gap that may be introduced during implantation. Applying a mechano-regulatory tissue differentiation algorithm on a two-dimensional plane strain microscale model, representing implant-bone interface, the objectives of the study are to gain an insight into the process of peri-prosthetic tissue differentiation and to investigate its relationship with implant-bone relative displacement and size of the polar gap. Implant-bone relative displacement was found to have a considerable influence on bone healing and peri-acetabular bone ingrowth. An increase in implant-bone relative displacement from $20{\mu}m$ to $100{\mu}m$ resulted in an increase in fibrous tissue formation from 22% to 60% and reduction in bone formation from 70% to 38% within the polar gap. The increase in fibrous tissue formation and subsequent decrease in bone formation leads to weakening of the implant-bone interface strength. In comparison, the effect of polar gap on bone healing and peri-acetabular bone ingrowth was less pronounced. Polar gap up to 5 mm was found to be progressively filled with bone under favourable implant-bone relative displacements of $20{\mu}m$ along tangential and $20{\mu}m$ along normal directions. However, the average Young's modulus of the newly formed tissue layer reduced from 2200 MPa to 1200 MPa with an increase in polar gap from 0.5 mm to 5 mm, suggesting the formation of a low strength tissue for increased polar gap. Based on this study, it may be concluded that a polar gap less than 0.5 mm seems favourable for an increase in strength of the implant-bone interface.
An experiment was conducted with male broiler chicks to determine the effect of different dietary phosphorus sources and evaluate the biological availability of phosphorus sources. The biological availability of phosphorus from dicalcium phosphate (DCP) was used as a reference standard (100%) compared to defluorinate phosphate (DFP). DCP and DFP was supplemented to a corn-soy basal diet at levels of 0.05, 0.15, 0.25, 0.35%. Each of 24 pens of 10 male broiler chicks with three replications was used for three weeks. The results indicated that weight gain, feed in-take, feed efficiency, nonphytic phosphorus (NPP) and tibia ash were significantly different among treatments. Dietary supplemental phosphorus of DFP improved weight gain, NPP intake and feed efficiency consistently, whereas supplements of DCP did not show consistent increase. Regression equations was used for the availability of DPP compared with DCP when percent bone ash was a function of total phosphorus in the diet. The percent (%) bone ash of DFP groups compared to that of DCP groups showed a value of 59.98% as a slope ratio. DFP indicated lower biological availability compared to DCP, but it's dietary supplementation tended to increase bone ash and maximize the growth of young broiler chicks.
This study examined the effect of dietary lipids and ovariectomy(OVX) on bone mass in rats. The experimental animals were divided into sham operation(S) or OVX(O) groups and then each group was divided into soybean oil(SS, OS), beef tallow(SB, OB), and fish oil(SF, OF) diet group. Experimental diets were fed for 16 weeks. Weight gain tended to be higher in OVX groups than in sham groups. Weigth gain was the highest in beef tallow group (SB) and the lowest in fish oil group(SF). There were no difference in weight, length, breaking force, strength and circumference of femur mong groups. However, volume of femur tended to be higher in OVX groups than in sham groups. Ash content of femur was significantly lower in OVX than sham groups but there were no difference between SB and OB group. Ca content in the soybean oil and beef tallow group were not influenced by OVX but Ca content in fish oil group was significantly higher in OF than SF group. The SB group showed the hyighest Ca content and the SF group showed the lowest Ca content. P content of femur was significantly lower in OVX groups than in sham groups but there were no difference between SB and OB group. The SS group showed the highest P content and the OS and OF group showed lower P content compare to other groups. From this result, it suggested that beef tallow positively influenced maintenance of bone mass by means of accumulation of body fat in ovariectomy.
An important related question is whether arginine has influence bone metabolism. The effect of arginine supplements on bone markers and related hormones were studied in young female Sprague-Dawley rats fed either an arginine supplemented diet or control diet. Twenty four rats (body weight 83${\pm}$5 g) were randomly assigned to one of two groups, consuming casein or casein with supplemented arginine diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, estrogen, insulin-like growth factor-1 (IGF-1), parathyroid hormone (PTH) and calcitonin were analyzed using radioimmunoassay kits. The weight gain and mean food intake were not affected regardless of diets. The rats fed arginine-supplemented diet had not significantly different in ALP, osteocalcin, crosslinks value, PTH, estradiol, and IGF-1 compared to those fed casein diet group. The arginine-supplemented group had significantly higher growth hormone and calcitonin than casein group. This study suggests that arginine is beneficial for bone formation in growing female rats. Therefore exposure to diet which rich in arginine early in life may have benefits for bone formation and osteoporosis prevention.
The need for refining dietary nutrient levels and limited information regarding Holstein steer and phosphorus supplementation led to the objective of examining the response of removing supplemental dietary phosphorus from a corn-based finishing diet offered to yearling Holstein steers. Two groups of yearling Holstein steers were utilized to study responses of increasing dietary phosphorus level during the finishing period. In Exp. 1, 96 Holstein steers (419 kg) were blocked into four weight groups. Dietary treatments included no additional phosphorus (NDC) or the inclusion of dicalcium phosphate (DCP) to achieve 0.30% phosphorus (P) in the complete diet. Daily gain, DMI and carcass traits were not different (p>0.05). Overall gain efficiency was slightly lower for NDC (p<0.05). Exp. 2 consisted of 78 Holstein steers (491 kg) blocked into two weight groups. Steers were harvested on d 84 and 112 d on test with carcass data collected following a 48-h chill. No differences (p>0.05) were detected for DMI, ADG, or gain efficiency. No differences (p>0.05) were detected in carcass characteristics in this experiment. Percent bone ash, calcium, P, and bending moment also did not differ among treatments (p>0.05). Removing supplemental phosphorus in the feedlot diet did not produce adverse effects on steer performance, carcass, or rib bone characteristics for yearling Holstein steers.

The objective of this experiment was to assess the use of different vitamin D metabolites in the feed of broiler chickens and the effects of the metabolites on performance, bone parameters and meat quality. A total of 952 one-day-old male broiler chicks were distributed in a completely randomised design, with four treatments, seven replicates and 34 birds per experimental unit. The treatments consisted of four different sources of vitamin D included in the diet, $D_3$, $25(OH)D_3$, $1,25(OH)_2D_3$, and $1{\alpha}(OH)D_3$, providing 2000 and 1600 IU of vitamin D in the starter (1 to 21 d) and growth phases (22 to 42 d), respectively. Mean weight, feed:gain and weight gain throughout the rearing period were less in animals fed $1{\alpha}(OH)D_3$ when compared with the other treatments (p<0.05). No significant differences were noted among the treatments (p>0.05) for various bone parameters. Meat colour differed among the treatments (p>0.05). All of the metabolites used in the diets, with the exception of $1{\alpha}(OH)D_3$, can be used for broiler chickens without problems for performance and bone quality, however, some aspects of meat quality were affected.
This study is to find out effects of hot water soluble extract from green tea, one of the Korean favorites, on the calcium metabolism and bone strength in body. To do so, calcium, phosphate, creatinine concentration and ALP activity in blood and the content of calcium and ash in the organ, the length, weight, strength in bone were measured. In addition, to find the calcium metabolism, the level of calcium intake, excretion, retention were measured. Twenty male Sprague-Dawley rats were divided into two groups and isoloated soy protein was provided as the source of protein and CaCO₃ was provided as the source of calcium. 0.5% hot water soluble extract from green tea was provided to the green tea groups and for the control group deionized water was provided. The results are as follows ; 1. There is no difference between the experimental groups in diet intake, weight gain, and the feed intake. 2. Feed efficiency ratio was low in the group which hot water soluble extract from green tea was provided. 3. There is no difference between groups the level of calcium, phosphorus, creatinine and ALP activity in serum. 4. There is no difference between groups weight, contents of ash and calcium in kidney and liver. 5. There is no difference between groups in calcium intake, absorption, excretion, and retention. 6. There is no difference between groups weight, length and strength in bone. In summary, when hot water soluble extract from green tea was provided with the amount of 150-200mg, which is taken when people generally drink as favorite tea, weight gain was reduced due to the decrease of feed efficiency ratio. However, it did not affect the availability of calcium in body at all. Thus, even if a big quantity of green tea powder or solid of hot green tea extract is not provided, the quantity obtained when people drink green tea lowers the feed efficiency ratio without reducing availability of calcium in body.
