Background: The intestinal microbiota is an important regulator of bone health. In previous studies we have shown that intestinal microbiota dysbiosis, induced by treatment with broad spectrum antibiotics (ABX) followed by natural repopulation, results in gut barrier dysfunction and bone loss. We have also shown that treatment with probiotics or a gut barrier enhancer can inhibit dysbiosis-induced bone loss. The overall goal of this project was to test the effect of Korean Red Ginseng (KRG) extract on bone and gut health using antibiotics (ABX) dysbiosis-induced bone loss model in mice. Methods: Adult male mice (Balb/C, 12-week old) were administered broad spectrum antibiotics (ampicillin and neomycin) for 2 weeks followed by 4 weeks of natural repopulation. During this 4-week period, mice were treated with vehicle (water) or KRG extract. Other controls included mice that did not receive either antibiotics or KRG extract and mice that received only KRG extract. At the end of the experiments, we assessed various parameters to assess bone, microbiota and in vivo intestinal permeability. Results: Consistent with our previous results, post-ABX- dysbiosis led to significant bone loss. Importantly, this was associated with a decrease in gut microbiota alpha diversity and an increase in intestinal permeability. All these effects including bone loss were prevented by KRG extract treatment. Furthermore, our studies identified multiple genera including Lactobacillus and rc4-4 as well as Alistipes finegoldii to be potentially linked to the effect of KRG extract on gut-bone axis. Conclusion: Together, our results demonstrate that KRG extract regulates the gut-bone axis and is effective at preventing dysbiosis-induced bone loss in mice.
Chronic kidney disease-mineral bone disorder (CKD-MBD) is a systemic disorder of mineral and bone metabolism caused by CKD. Patients with early-stage CKD who present with disordered regulation of bone and mineral metabolism may be asymptomatic. However, if untreated, the condition can be a significant barrier in achieving optimal bone strength, linear growth, and cardiovascular health in pediatric patients with CKD. Thus, the current study evaluated the definition, pathogenesis, diagnosis, and management of pediatric CKD-MBD.
Purpose: This study was to investigate lifestyles and identify factors influencing the bone mineral density (BMD) among employees in a community. Methods: Data were collected from 199 employees. Their lifestyles and BMIs were measured with a self-report questionnaire. As for their BMDs, their calcanei were measured with Quantitative Ultrasound. Results: Assessing their BMDs with reference to the WHO standards, 52.8% of the subjects' calcaneus BMDs were normal and 44.2% showed osteopenis, 3.0% showed osteoporosis. Age (odds ratio=1.05, p=.029), exercise (odds ratio=.31, p=.006) and the number of eating meals per day (odds ratio=1.97, p=.046) were significant factors influencing the subjects' BMDs. Conclusion: Strategies to emphasize exercise and dietary habits in employees are important for improving their bone mineral density. Programs for improving bone mineral density based on life cycle need to be developed.
This study investigated anthropometric and biochemical characteristics, nutrient intakes, and bone density of Korean elderly men (over 65 yrs). Data on bone density and anthropometric (height, weight, waist circumference, body mass index and blood pressure) and biochemical (total cholesterol, high-density lipoprotein (HDL)-cholesterol, low-density lipoprotein (LDL)-cholesterol, triglyceride and hemoglobin) characteristics, nutrient intakes, and nutrient density were obtained from the Fifth Korea National Health and Nutrition Examination Survey (KNHANES, 2008~2011). Subjects were categorized into smoking and non-smoking groups, and smoking groups were divided into three groups by smoking period (under 20 yrs, 21~40 yrs and over 41 yrs). Serum triglyceride concentrations of the smoking group aged 21~40 yrs were higher than those of other groups (P<0.01), whereas other biochemical factors were not different. Intake ratios of energy, protein, phosphorus and sodium in subjects were over 100% of Dietary Reference Intake for Koreans (KDRI). Nutrient densities according to intakes of thiamin, riboflavin, and niacin per 1,000 kcal were significantly different among the groups (P<0.05). Bone density of subjects decreased according to smoking period (T-score of total femur in non-smoking group -0.3108, and -0.2918, -0.4941, -0.6847 in smoking group, respectively). Ratio of osteoporosis was 38.1% in the non-smoking group and 44.4%, 51.1%, and 64.0% in the smoking group, respectively. The findings of the present study show that smoking may be associated with bone health, higher ratio of osteoporosis, and low nutrient density in elderly men. Therefore, practical and systematic non-smoking programs are required to improve the bone density of elderly men as well as maintain healthy bone levels and desirable lifestyle.
This study investigated changes of mechanical characteristics in the trabecular and cortical bone of the lumbar vertebrae of the ovariectomised(OVX) rat. In previous researches, there were many studies for morphology of osteoporotic bones based on Micro-CT (Micro - Computed Tomography). However, there were few studies for detecting and tracking changes of mechanical characteristics in the trabecular bone and cortical bone of the lumbar vertebrae for the OVX rat. For this study, one female Sprague-Dawley rat was used: an OVX rat. The 4th lumbar of the OVX rat was utilized as a specimen. Detecting and tracking changes of mechanical characteristics could be investigated in the trabecular and cortical bone of the lumbar vertebrae for an OVX rat using In-vivo Micro-CT. An OVX rat was scanned at week 0 (just before surgery), at week 4, and week 8 after surgery. Micro finite element(${\mu}\;FE$) analysis was used to investigate mechanical characteristics in the trabecular and cortical bone of the lumbar vertebrae for an OVX rat.
국민건강영양조사 제 5기 1 2차년도에 참여한 남성을 대상으로 연령에 따른 부위별 골밀도 변화, 골 소실률에 대해서 알아보고자 시도 하였다. 연령에 따른 골밀도 변화를 가장 잘 보여 주기 위해 Cubic regression을 사용하였고, 척추, 대퇴경부, 대퇴골 전자부, Ward 부위 골밀도 차이는 분산분석을 이용하여 분석하였다. 연구결과, 요추, 대퇴골 전체, 대퇴경부, 대퇴골 전자부, Ward 부위의 최대 골밀도는 20-24세였으며, 75-79세에서 골 소실률이 가장 높았다. 따라서, 남성에서는 75세 이상에서 골다공증의 집중적인 관리가 필요할 것이며, 남성에서의 골다공증 진단기준은 국제 임상 골밀도 학회(The International Society for Clinical Densitometry; ISCD)의 지침을 따르는 방안을 고려해 봐야 할 것이다.
The objective of this study is to propose a finite element based design of the dental implant replacing unction and shape of natural teeth. For this, geometric actors were varied to investigate stress distribution of the alveolar bone around dental implant. In this study, the results were obtained based on the theory of linear elastic, with geometrically axisymmetric assumption. Geometric actors determining implant shape are ranged as 0.2mm-0.6mm, 0.04mm-0.1mm, 0.46mm-0.84mm or height of thread, radius of curvature of thread, and pitch, respectively. The stresses in the alveolar bone caused by biting force playa major role in determining implant stability. Especially, the stress concentration in the cortical bone causes bone resorption and finally makes the implant unstable. Therefore, the stress distributions were investigated on the side of the alveolar bone focusing on the area of cortical bone. The maximum von Mises stress was found to increase up to 6% as the height of thread increases, while its value was to decrease to 19% when the radius of curvature increase within the assigned ranges. For the variation of pitch, the larger size of pitch results in greater maximum von Mises stress when the length of the implant under consideration is fixed. The existence of the neck below the shoulder did not affect the stress distribution in the region of alveolar bone. However, the stresses on the side of the implant near the neck were found to be different by 20% approximately. Therefore, the neck can provide the stability of the implant against continuing biting movement. As a conclusion, the finite element based study shows a potential in designing the dental implant systematically.
The purpose of this study is to evaluate the effect of loading at three different occlusal surface position of the gold alloy crown on the stress distributions in surrounding bone, utilizing 3-dimensional finite element method. A three dimensional finite element model of an implant with simplified gold alloy crown and supporting bone was developed for this study. A oblique or vertical load of 100 N was applied at the following position at each FE model : 1) center of occlusal surface, 2) a point on the buccal side away from center of occlusal surface (COS) by 2.8mm, 3) a point on the lingual side away from COS by 2.8mm. In the results, Minimum von Mises stresses under vertical load or oblique load of 100N were about 6MPa at the center of occlusal surface and about 40MPa at the point on the buccal side, respectively. From the results we could come to the conclusion that occlusive loading position could be an important factor for establishment of structural safety of supporting bone.
Background: Bone mineral density (BMD) is a lifetime marker of estrogen in a woman's body and has been associated with increased breast cancer risk. Nonetheless the actual association is still debatable. Furthermore, estrogen is very crucial in maintaining human bone density and gradually decreases over age. A systematic search was conducted to assess any association of BMD with breast cancer risk factors among premenopausal and postmenopausal women. Materials and Methods: Review identification was performed through databases searching on MEDLINE, CINAHL and SCOPUS and 19 qualified studies were elected. The keywords used were "bone mineral density", "breast cancer", and "breast density". Results: A total of 19 articles showed variation with the majority of the studies focused on postmenopausal and a few focused on premenopausal women. Overall there was no concensus on effects. Conclusions: An enormous effort is being undertaken by researchers to prove that BMD might be one of the significant risk factors for breast cancer.
Calcium density in human bones decreases as people are getting older due to the interior or exterior environmental factors. Bone aging forms osteoporosis. And this can bring out various spine fractures which develops a complications. Thus the prediction of seniliy is one of the important factors in spine diseases. Once spine aged, diverse fractures occur such as compression fracture and micro fracture. Side images of the spine by the digital radiography (DR) were prepared, and pixel arbitrary unit with Image J was measured from one spot in the lumbar bone part. By calculating pixel arbitrary unit of the simple contrast, it was obtained that the value of pixel arbitrary unit decreased as seniliy of bones increased. By simply applying Image J to the seniliy of patient's spine, the seniliy of bones predicts the level of danger with only digital radiography(2D) image. consequently we show that Image J value of pixel arbitrary unit index for predicts the level of precaution of osteoporosis patient.
