Analysis of Korean Patent Current Status Related to Food using Edible Insects (식용곤충을 이용한 식품 관련 국내 특허 현황 분석)

  • The purpose of this paper was to analyze domestic patents for food using edible insects. From January 1, 1980 to December 31, 2021, patents filed with the Korean Intellectual Property Office were searched, and a total of 242 valid patents were selected. The trend of Korean patent applications for edible insect food has increased since 2015, with 57 cases (the highest number) in 2017. As for the edible insects used in food, Bombyx mori L. were the most common with 127 cases, followed by Tenebrio molitor L. with 118 cases. By type of applicant, individuals accounted for the most, with 132 cases. As a result of grouping patents by food classification, 67 cases of edible insects were used in snacks, breads, and rice cakes. As a result of patent analysis, an effort was made to supplement insufficient nutrients by adding nutritionally excellent edible insects to existing foods, and efforts were made to improve the sensory properties of insect foods. It is expected that this study will contribute to establishing future R&D directions and patent application strategies related to edible insect food.

In vitro screening of anti-skin aging and antioxidant properties of aqueous/solvent extracts from distinctive stages of silkworm (Bombyx mori L.) pupae

  • Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.

Development and Validation of a Unique HPLC-ELSD Method for Analysis of 1-Deoxynojirimycin Derived from Silkworms (누에에 함유된 1-Deoxynojirimycin의 분석을 위한 HPLC-ELSD 분석법 밸리데이션)

  • A simple and accurate assay was developed for the quantitative analysis of 1-deoxynojirimycin (1-DNJ) derived from the silkworm (Bombyx mori). Normal-phase high-performance liquid chromatography coupled with an evaporative light scattering detector (HPLC-ELSD) and a hydrophilic interaction liquid chromatography column was used. Various parameters were applied to optimize the analysis method. The limits of detection and quantification of 1-DNJ were 2.97 × 10-3 and 9.00 × 10-3 mg/mL, respectively. The calibration curve showed good linearity results. The concentration range and the r2 value were 0.0625-1.0 mg/mL and 0.9997, respectively. The accuracy test demonstrated a significantly high recovery rate (89.95-103.22%). The relative standard deviation was ≤ 1.00%. Thus, a method for the accurate identification and quantitative analysis of 1-DNJ in silkworms was developed. Moreover, in this procedure, the process of derivatization of 1-DNJ, which was required in previous experiments, could be eliminated. This technique may be actively utilized for the development of pharmaceuticals and health functional foods using 1-DNJ.

Antijuvenoid Action of Terpenoid Imidazole Compound on Larval - Pupal - Adult Development of Silkworm, Bombyx mori L.

  • Precocious metamorphosis was induced in two popular bivoltine breeds i.e. YB and $NB_4D_2$ by an imidazole compound having anti-juvenile hormone activity. The chemical was administered by feeding treatment with mulberry leaf to freshly moulted (0-6 hrs) IV instar larvae. The dose of the chemical was found to be breed specific being 650 ppm for YB and 500 ppm for $NB_4D_2$. The chemical caused complete skipping up of the fifth instar larvae which is most susceptible to diseases. But IV instar was somewhat prolonged. As a result the total larval period was reduced by 4-5 days. However, some of the rearing and reeling parameters like cocoon yield, cocoon weight, shell weight, shell %, filament length and fecundity were reduced for that particular generation only. The effective rate of rearing (ERR %) was significantly increased in trimoulters during the most unfavourable August - September seed crop season. Number of cocoons / kg and number of male moth were significantly higher in trimoulters. These may be useful for preparation of $multi{\times}bi$ hybrid seed. Fine denier was also found in trimoulter cocoon which is the cause of getting fine silk filament from trimoulter cocoons. Normal mating behaviour and emergence pattern was recorded in trimoulters. Bivoltine trimoulter males also showed competence for mating with multivoltine females. These results suggest the possibility of getting trimoulter males during hot and humid seasons when rearing of bivoltine is almost impossible particularly in Eastern and North Eastern India.

Reproductive Performance of Breeds and Hybrid of Silkworm, Bombyx mori L. with Special Reference to Egg Laying Rhythmicity

  • The reproductive performance of multivoltine breed 'Nistari' and bivoltine breeds as NB18, P5 and a bivoltine hybrid (NB18 ${\times}$ P5) of B. mori were studied in different seasons in terms of total number of eggs laid, unlaid and hatching efficiency of the laid eggs to critically assess the breeds / hybrid potentiality. The bivoltine hybrid showed better performance in all the seasons as compared to the breeds investigated, as expected due to possibility of hybrid vigour. Among the breeds, bivoltine P5 had a better egg laid performance in S1 (February - March), S2 (May - June) and S3 (September - October) seasons but with an increase in the number of unlaid eggs. The hatching percentage, although didn't show any definitive trend, still suggested a marginal better performance in the hybrid. Most importantly, egg laying rhythmicity was studied by mating the female moth at 6.00 a.m. for $3{\frac{1}{2}}$ hours, and subsequently allowing the laying of eggs both under BOD condition ($25{\pm}1^{\circ}C$ and $85{\pm}5%$ RH) and at ambient temperature ($23-28^{\circ}C$ and $74.7{\pm}5%$) separately. Maximum (90%) egg laid occurred from 3.30 to 7.30 pm (i.e. 4 - 6 h after decoupling). Similar laying patterns were observed when females where mated at 12 noon and decoupled at 3.30 pm, revealing that late photoperiod and early scotoperiod were favourable for egg laying in case of multivoltine breed and bivoltine prefers late photoperiod for egg laying.

Effects and Mechanisms of Silkworm Powder as a Blood Glucose-Lowerinly Agent

  • Cocoon production, which is a representative of traditional sericulture shifted into silkworm powder production in the spring of 1995. This, infect, signifies the change from the dress-centered textile business to the bio-industry and the functional resource industry. One of the most outstanding shifting is utilization of silkworm larvae for anti-diabetic agent. In Asian countries including Korea, silkworm powder derived from the domestic silkworm (Bombyx mori L.) has long been favored for anti-diabetic agent, but its efficacy was not tested until last decade by modern scientific methods. In this article, we reviewed the major researches on the silkworm powder as a blood glucose-lowering substance. After the beginning test of the efficacy of silkworm powder by a cooperative research between Department of Sericulture and Entomlogy, NIAST, RDA and Kyung Hee University, substantial data have been accumulated so far, In a serial experiment to select best condition, the fifth instar larvae prepared by freeze dry method turned out to have the best blood glucose-lowering effect. In the pharmacological experiment to understand the mechanism of silkworm powder in small intestine, the silkworm powder turned out to inhibit the activity of ${\alpha}$-glucosidase, by competitively binding to $\alpha$-type disaccharides. The animal experiment showed that the extract of silkworm powder prevents a rapid increase of blood glucose level after meal and prevents hunger and law blood glucose level during empty stomach. In the experiment to isolate the major component of silkworm powder, which exerts blood glucose-lowering effect, 1-deoxynojirimy-cin (DNJ) was eventually mass-purified, and it turned out that DNJ isolated from silkworm powder was excellent in its blood glucose-lowering effect. In the experiment to understand the personal difference of the efficacy of the silkworm powder, clinical candidates were divided on the basis of the criterion of traditional Chinese medicine: Tae-Yang, Tae-Um, So-yang, and So-Um. The result showed that silkworm powder has a tendency to reduce blood glucose level at fasting and at 2 hours after meal, and this trend was somewhat obvious in the Tae-Um body type. In summary, we reviewed scientific papers on the efficacy of silkworm powder and its purified DNJ as a blood glucose-lowering agent. These suggest that silkworm powder truly possesses blood glucose-lowering effect as documented in the traditional Chinese medicine, although further researches will be required to develop them as "medical" resource instead of functional food.

Study of Miscibility of Natural Silk by Molecular Dynamics Calculation of Solubility Parameter (용해도 파라미터의 분자동역학 계산을 통한 천연 실크 소재의 혼화성 연구)

  • In recent years, polymer membranes, which are actively used in various industrial fields, have the advantage of being able to impart unique properties through the control of chemical structures and physical properties in the film-fabrication process, as well as through fabricating blend membranes mixed with various materials. In this study, the solubility parameter, which can be used as an index of miscibility with other materials, was calculated using molecular dynamics using a silkworm (Bombyx mori) silk polymer which has a wide potential to be used as an eco-friendly natural material. When the solubility parameter of polyvinylalcohol (PVA), which is also environmentally friendly and biocompatible, was calculated by molecular dynamics and compared with each other, it was confirmed that the two polymer materials had similar solubility parameter values. In conclusion, it was theoretically proved that the two polymers could blend well with each other, which was confirmed through experiments.

Effects of Silkworm Hemolymph on Cell Viability and hCTLA4Ig Production in Transgenic Rice Cell Suspension Cultures

  • Silkworm hemolymph (SH), prepared from fifth-instar larvae of Bombyx mori and heat-treated at $60^{\circ}C$ for 30 min, was used to improve cell viability and the production of human cytotoxic T-lymphocyte antigen 4-immunoglobulin (hCTLA4Ig) in transgenic Oryza sativa L. cell suspension cultures. Even though SH could not elevate cell viability at the concentrations up to 3% (v/v), addition of 0.3% (v/v) SH to a culture medium enhanced the production of hCTLA4Ig by 36.8% over an SH-free medium. Moreover, the production period of hCTLA4Ig could be shortened in a 0.3% (v/v) SH-added medium compared with that in an SH-free culture. As a result, addition of 0.3% (v/v) SH improved the productivity of hCTLA4Ig significantly in transgenic rice cell cultures.

  • Ten promising polyvoltine mulberry silkworm strains(SDMGl, SDMG2, SDMG3, SDMG4, SDMWl, SDMW2, RMWl, RMW2, RMW3 and RMW4) that are superior in quantitative and qualitative traits have been synthesized in the polyvoltine breeding laboratory of Andhra Pradesh State Sericulture Research & Development Institute, Hindupur through systematic hybridization and appropriate selection methods. After the genotypes were found homozygous for the desired traits, they have been crossed with 3 bivoltine testers $(APS8, APS4 and {NB_2}{D_4})$ and thirty new hybrid combinations were developed for the assessment of their hybrid performance. Phenotypic expressions of economically important quantitative and qualitative traits of fist filial generation were measured and subjected for statistical analysis. Evaluation Index and Subordinate Function methods were employed for the assessment of hybrid performance since they are widely used in silkworm hybrid evaluation. Total of seven poly${\times}$bivoltine combinations, which ranked high in both the methods, were selected as potential combinations for further field test. These combinations also ranked significantly higher than the control hybrid (APMl${\times}$APS8).
