Improvement of reliability of an ISFET pH-meter by employing multiple sensors

  • The ISFET(ion sensitive field effect transistor), a semiconductor ion sensor, has many advantages over conventional ion sensors. Various single-sensor type ISFET pH-meters have been developed. However, they could not be applied in fields because their performances are directly affected by the sensor condition. With only one sensor, the system could be easily damaged from environmental factors, and reliability of it is decreased. Therefore, a 4-channel PH-meter system is proposed to improve the reliability of ISFET pH-meter. It has 4 ISFETS as ion sensor, and a software which contains a new calibration and measurement algorithm appropriate to the system. The reliability of the system was proved by measuring hydrogen ion concentration in the pH standard solutions and buffer solutions.

  • In order to maximize the growth of Bifidobacterium lactis BL 740 and soy isoflavone agycones production, we investigated the optimization of a culture medium containing soy hypocotyls, which are the byproducts of the soy manufacturing process, and soy proteins. The ingredients of the medium containing soy materials (S-medium) were selected by fractional factorial design (FFD) and central composite design (CCD) within a desirable range. The FFD was applied by six factors: glucose, cellobiose, fructooligosaccharide, soy peptone, soy protein, and soy hypocotyl. Soy protein, soy peptone, and soy hypocotyl were found to be significant factors from the result of FFD for both the growth of B. lactis BL 740 and aglycone production. The CCD was then applied with three variables found from FFD at five levels each and the optimum values were determined for the three variables: soy peptone, soy protein, and soy hypocotyl. In the case of the growth of B. lactics BL740, the proposed optimal media contained 12.73 g/L of soy protein, 29.55 g/L of soy peptone, and 130.67 g/L of soy hypocotyl. To produce isoflavone aglycones, optimized media was composed of 2.06 g/L, soy protein, 1.25 g/L of soy peptone, and 60.02 g/L of soy hypocotyl.

Effect of Boron on Growth and Yields of Perilla(Perilla frutescens var. japonica HARA), Melon(Cucumis melo L.) and Cucumber(Cucumis sativus L.) (붕소 시용이 들깨, 참외 및 오이의 생육과 수량에 미치는 영향)

  • A pot experiment was carried out to investigate the effect of boron on the growth, symptom, yield, and content of boron in Perilla(Perilla frutescens var. japonica HARA), Melon(Cucumis melo L.) and Cucumber(Cucumis sativus L.). The results obtained are as follows. Perilla. melon and cucumber uptake a lot of boron from the soil, and toxic effects of boron result in leaf tip yellowing by progressive necrosis. Yield of perilla, melon and cucumber are decreased at the $3{\sim}5mg\;kg^{-1}$, $5{\sim}10mg\;kg^{-1}$ of boron applicated in soil, respectively. Boron content in the leaf is the highest among various parts plant, and the tip and margin of the leaf contained more boron than central part of the leaf. Boron applicated was moved and accumulated to the plant and soil.

Irrigation Water Quality of the Kyoungan Stream. (경안천수계의 농업용수로서 수질검토)

  • The water quality in Kyoungan stream was surveyed at 6 sites for 7 months from March to September in 1994, 1995 and 1997. The overall results are summarized as follows: Yearly COD concentration of Kyuongan stream was decreased from 26.3 mg/l in 1994 to 17.8 mg/l in 1997, but those of $NH_4-N$ and $NO_3-N$ were not changed. The water quality of Kyoungan stream in July and August was better than any other months due to dilution with rainwater. The water quality along Kyoungan stream was deteriorated with inflow of tributaries contaminated with livestock wastewater and sewage water in the mid-stream, but thereafter it was recovered with inflow of Gonjiam steam and Beon stream.

A Study on Quantity and Quality of Collected Rainwater by Collected Materials (우수 이용을 위한 포집재료별 포집수량과 수질에 관한 연구)

  • In this study, quantity and quality of collected rainwater by sand, gravel, soil, lawn and concrete surface, as collection materials were investigated and Rainwater Collection Prediction Model was developed to predict the amount of collected rainwater. The quantity of collected rainwater in concrete surface, gravel, sand, soil and lawn collection system was 1,067L(93.2%), 1,006L(87.8%), 902L(78.8%), 800L(69.9%), 788.5L(68.8%) for 8 months period, respectively. The average turbidity of collected rainwater in concrete surface, gravel, sand, soil and lawn collection system was 3.2NTU, 2.2NTU, 1.9NTU, 1.7NTU, 1.5NTU for 8 months period, respectively. For sand collection material, predicted amount by the Model and actual collected amount were 931.5L and 902L, which were very closed. For gravel collection material, predicted amount by Model and actual collected amount were 1,028.21. and 1,006L, which were very closed. To simulate the optimal rainwater storage volume, the rainfall and evaporation data in Dae-jeon city were used. For sand collection system with 30m2 area, the maximum storage volume was $17m^3$ and 62% of the year was secured for use of 240L/day.

Two Unrecorded Species of the Snapper (Perciformes: Lutjanidae) Collected from Jeju Island, Korea

  • Two unrecorded species of the snapper, Lutjanus malabaricus (296.0 mm standard length [SL]) and L. stellatus (350.0 mm SL) belonging to the family Lutjanidae, were first collected from the western coastal waters of Jeju Island, Korea. L. malabaricus is characterized by having a dark marking on the upper half of the caudal peduncle, a band of vomerine teeth that does not protrude posteriorly at the middle, and nine anal soft rays. Compared to its Korean relative, L. malabaricus is distinguishable by having rows of scales that run obliquely in the dorsal-posterior direction above the lateral line (vs. parallel to the lateral line in L. argentimaculatus). L. stellatus can be identified by the absence of wavy blue lines on the head (vs. many blue lines in L. rivulatus) and presence of a white spot above the lateral line (vs. a black blotch on the lateral line in L. rivulatus). We propose new Korean names, "Jin-hong-tung-dom" and "Huin-jeom-tung-dom," for L. malabaricus and L. stellatus, respectively.

Suspension Culture of an Antibacterial Peptide Producing Cell Line from Bombina orientalis

  • The suspension culture of an anchorage-dependent cell line (Bok-l) from Bombina orientalis was successful in respects of cost and efficiency. The amount of cells obtained from the suspension culture was almost equivalent to that from the anchorage-dependent culture. This result shows the possibility of suspension culture for scale-up. The cells in suspension produced an antibacterial peptide as much as anchorage-dependent cells did. The cell growth ($6.0\times10^6cells/m\ell$) and viability (>80%) at 10 rpm were higher than that at 0 rpm ($1.9\times10^6cells/m\ell$, 65~80%) and 30 rpm ($1.8\times10^6cells/m\ell$ 40~76%). The size of cells became smaller at the agitation rate of 30 rpm. The antibacterial activities of cell extracts from suspension cultured cells were confirmed against gram-negative and gram-positive bacteria by the inhibition zone assay and the liquid growth inhibition assay.
