노르플록사신의 체내동태에 대한 신장해의 영향 (Effect of Renal Failure on Pharmacokinetics of Norfloxacin in Rabbits)

  • The pharmacokinetics of norfloxacin (100 mg/kg, oral) in renal failure rabbits was studied. Renal failure rabbits were induced by the i.v. injection of folate (50,100 and 150 mg/kg). These produced significant increases of serum creatinine concentration $(S_{cr})$ and blood urea nitrogen (BUN). Plasma concentration and AUC of norfloxacin significantly increased. Elimination rate constant $(K_{el})$ of norfloxacin significantly decreased, and half-life $(t_{1/2})$ of norfloxacin significantly increased. Correlation between serum creatinine concentration $(S_{cr})$ and half-life $(t_{1/2})$ of norfloxacin, and correlation between BUN and AUC of norfloxacin have linear relationship respectively. These results suggest that adjustment or the dosage regimen of norfloxacin is desirable, and serum creatinine concentration $(S_{cr})$ as well as BUN can be used an index for adjusting the dosage regimen of norfloxacin in renal failure.

Anti-fatigue effects of Elaeagnus multiflora fruit extracts in mice

  • The fruit, leaves, and roots of Elaeagnus multiflora Thunb. have been used in traditional Chinese medicine to treat cough, diarrhea, and itching. However, the anti-fatigue effects of E. multiflora fruit (EMF) extract have not been studied in detail. The aim of this study was to examine the effect of EMF on fatigue and exercise performance in BALB/c mice. EMF was orally administered to mice at four doses (10, 50, 100, and 200 mg/kg/day) for 2 weeks. The anti-fatigue activity was evaluated by determining the exhaustive swimming time. Blood lactate and glucose levels and serum lactate levels after a 10 min swimming time, as well as ammonia, creatine kinase (CK), blood urea nitrogen (BUN), lactate dehydrogenase (LDH), and glycogen contents after exhaustive swimming time were measured. The exhaustive swimming time of the EMF 200 group was significantly increased (p <0.01). The EMF groups showed significantly low levels of CK, BUN, LDH, and lactate compared with the control group (p <0.05). Increased liver glycogen was observed in the EMF 200 group (p <0.05). These results suggest that EMF can be utilized as an efficacious natural resource for its anti-fatigue effects.

  • 단세포 조류 생물인 Chlorella vulgaris(클로렐라)는 다양한 생리 활성을 가진 기능성 소재로 이용되고 있다. 본 연구에서는 강제 수영 부하 실험과 혈액 생화학적 지표에 대한 클로렐라의 효과에 대해 연구하였다. 혈액 생화학적 지표로는 Blood urea nitrogen(BUN), creatine kinase(CK), lactic dehydrogenase(LDH), glucose(Glc), total protein(TP), albumin을 혈액 생화학적 지표로 측정하였다. 매일 0.05, 0.1, 0.15 g/kg 농도의 클로렐라를 각각 실험 군별로 마우스에 구강 투여했다. 클로렐라를 투여한지 3일, 7일째 되는 날 강제 수영 부하 실험을 시행한 결과 0.15 g/kg 클로렐라를 투여한 그룹에서 유의적으로 부동시간을 감소시켰다. 또한 혈청 중 BUN 수치를 낮췄으며, CK, LDH 수치는 감소하는 경향을 나타냈다. 클로렐라 투여 시 혈 중 Glc 수치는 높아졌으나, TP 와 albumin 수치는 변화가 없었다. 이상의 결과들은 클로렐라가 육체적 지구력 향상 효과가 있는 것을 시사하고 있다.

Effects of Yeonlyeonggobon-dan on the Blood of Arsenic-poisoned Rats

  • Sodium arsenate and Yeonlyeonggobon-dan (nianlinggubendan) extract (YGD), a herbal restorative were treated p.o. 20 mg/kg and 500 mg/kg, respectively, and concurrently to rats, and examined the biochemical parameters in blood. The values of white blood cell (WBC), red blood cell (RBC), hemoglobin (Hgb) and hematocrit (Hct) in each group did not show significant variance. The value of aspartate aminotrasferase (AST) of arsenic-treated group was increased for 2 weeks significantly while that of the group of concurrent administration with YGD became low significantly compared with arsenic-treated group and the value of alkaline phosphatase (ALP) of arsenic-treated group was decreased while that of the group of concurrent administration with YGD was increased significantly compared with arsenic-treated group. In arsenic-treated groups, the value of glucose (Glu), and those of lactic dehydrogenase (LDH), blood urea nitrogen (BUN) and triglyceride (TG) were decreased at first but increased later while the groups of concurrent administration with YGD showed significant recovery from the toxicity of arsenic.

Evaluation of FSP (Fermented Soy Protein) to Replace Soybean Meal in Weaned Pigs: Growth Performance, Blood Urea Nitrogen and Total Protein Concentrations in Serum and Nutrient Digestibility

  • A total of one hundred and forty four weaned pigs with an average BW of $8.09{\pm}0.05$ kg were used in a 28 day study to investigate the effects of fermented soy protein on growth performance, blood urea nitrogen and total protein concentrations in serum and nutrient digestibility in weaner pigs. Pigs were blocked by initial body weight and randomly allocated to one of four dietary treatments in a randomized complete block design. There were six replications per treatment. Dietary treatments included: SBM (corn-soybean meal basal diet), F 5, 10 and 15 (fermented soy product was used at 5, 10 and 15% to replace soybean meal in basal diet, respectively). ADG (average daily gain) and ADFI (average daily feed intake) were not affected (p>0.05) by dietary treatments during the entire 4-wk study period. There were linear increments in feed efficiency (p<0.01) as the dietary FSP level increased during the entire feeding period. No significant differences were observed for dry matter and nitrogen digestibility during the experimental period (p>0.05). Digestibilities of histidine, lysine and methionine were increased as the FSP level increased (linear effect, p<0.05, p<0.01). Among non-essential amino acids, alanine, glutamic acid, serine, tyrosine and total non essential amino acid digestibilities were increased linearly (p<0.05, p<0.01). There were quadratic effects in protein digestibility (p<0.05). Total amino acid digestibility of the F15 diet was improved compared with the F5 diet (p<0.05). There were no significant differences in fecal consistency score among the treatments (p>0.05). At the end of experiment, BUN (blood urea nitrogen) concentration was increased as the FSP level increased (linear effect, p<0.01) and total protein concentration was lowest (p<0.05) for pigs fed the SBM diet among treatments. In conclusion, the feeding of 10 or 15% FSP to nursery pigs improved feed efficiency, amino acid digestibility and blood urea nitrogen and total protein concentrations in blood.

Improving Productive and Reproductive Performance of Holstein Dairy Cows through Dry Period Management

  • To determine the effects of dry period (DP) length on milk yield, milk composition, some blood metabolites, complete blood count (CBC), body weight and score and follicular status, twenty five primiparous and multiparous Holstein cows were assigned to a completely randomized design with DP-60 (n = 13) and DP-20 (n = 12) dry period lengths. Cows in the DP-60 produced more milk, protein, SNF, serum non-esterified fatty acids (NEFA) and beta hydroxyl butyrate acid (BHBA) compared with cows in DP-20 ($p{\leq}0.05$). Serum glucose, blood urea nitrogen (BUN), urea, and glutamic oxaloacetic transaminase (GOT) were all similar among the treatments. Body Condition Score (BCS), body weight (BW), complete blood count (CBC) and health problems were similar between the treatments. Diameter of the first dominant follicle and diameter of the dominant follicle on d 14 were different among the treatments. Thus, results of this study showed that reducing the dry period length to DP-20 had a negative effect on milk production, milk composition and reproductive performance in Holstein dairy cows.

Hematology, Serum Biochemistry, and Acute Phase Proteins in Hanwoo (Bos taurus coreanae) Calves with Diarrhea

  • This study was conducted to identify the useful blood variables in diagnosing calf diarrhea in Hanwoo calves and good indicators for calf diarrhea. In 530 Hanwoo calves, fecal scores were recorded on a scale of 0 to 3, and blood samples were collected and analyzed for hematology, serum biochemistry, and acute phase proteins. Among the blood variables, 16 blood variables showed significant differences (p < 0.01) according to fecal scores. After reference intervals of these 16 blood variables were calculated, the distributions of calves by calculated reference intervals showed a significant difference (p < 0.001) and linear associations (p < 0.001) in blood urea nitrogen (BUN), glucose (GLU), blood sodium concentration (Na), blood potassium concentration (K), fibrinogen (Fib), and haptoglobin (Hp). Of 6 blood variables, the optimal cut-off values were calculated for BUN, K, Fib, and Hp, and the area under the curve was 0.5 or more: BUN (9.5 mg/dL, AUC: 0.623), K (5.8 mmol/L, AUC: 0.599), Fib (650.0 mg/dL, AUC: 0.706), and Hp (12.5 mg/dL, AUC: 0.847). These findings could be useful in evaluating calves with diarrhea and making decision of further treatment of calf diarrhea in Hanwoo calves.

Determination of Optimal Dietary Sulfur Amino Acids Ratio Relative to Lysine for Growing Barrows and Gilts

  • This experiment was conducted to investigate the effects of dietary SAA (sulfur-containing amino acids) on growth performance, nutrient digestibility and blood urea nitrogen (BUN) content, and to determine the optimal SAA:lysine ratio for growing barrows and gilts. A total of 150 pigs (75 barrows and 75 gilts, Landrace${\times}$Yorkshire${\times}$Duroc) were assigned to 6 treatments with 5 replicates of 5 pigs per pen. All pigs were fed diets containing either 1.12 (for barrows) or 1.33% (for gilts) dietary lysine with increasing SAA levels (50, 55 and 60% of dietary lysine) in a $2{\times}3$ factorial design. Throughout the whole experimental period (15 to 54 kg body weight), there was no interaction between sexes and SAA:lysine ratios on ADG, ADFI and FCR. However, increasing the SAA:lysine ratio from 50 to 60% in a diet showed a trend to increase ADG and ADFI of barrows. None of differences in nutrient digestibilities except for calcium and phosphorus were observed and gilts showed higher digestibility of calcium and phosphorus (p<0.05). Among dietary SAA:lysine ratios, there were no differences in apparent nutrient digestibility. Mean values of the essential amino acids (EAA), non-essential amino acids (NEAA) and total amino acids (TAA) digestibilities were higher in gilts than barrows (p<0.01). However, no differences in mean value of EAA, NEAA and TAA digestibilities were observed among dietary SAA:lysine ratios. Between sexes and among SAA:lysine ratios, no significant difference in BUN concentration was observed. This study demonstrated that the optimal inclusion ratio of SAA:lysine was 55% and below 50% in barrows and gilts, respectively.

Therapeutic Plasma Exchange in Pediatric Kidney Disease: 23-year Experience at the Severance Children's Hospital in Korea

  • Purpose: The American Society for Apheresis provides clinical guidelines for therapeutic apheresis in adults, but there are no guidelines for children. This study aimed to analyze the effect of therapeutic plasma exchange (TPE) in pediatric patients with various kidney diseases in Korea. Methods: We retrospectively reviewed the data of 16 children (up to 18 years of age) who were admitted to Severance Children's Hospital with refractory kidney disease. All patients received TPE between 1994 and 2016. Clinical and laboratory characteristics such as age, weight, sex, change in blood urea nitrogen (BUN), and creatinine level before and after TPE, and complications after TPE were analyzed. Results: The mean age and weight of the 16 patients at the time of TPE was $11.3{\pm}4.0$ years and $34.6{\pm}17.5$ kg, respectively. The BUN level was 35.4 mg/dL before TPE and significantly decreased to 21.5 mg/dL (P=0.025) at 1 week and 20.5 mg/dL (P= 0.01) at 1 month after TPE. The creatinine level significantly decreased from 1.20 mg/dL before TPE to 0.90 mg/dL (P=0.02) at 1 week after TPE. Four complications (hypovolemia, anemia, hypocalcemia, and thrombocytopenia) were reported, but were not fatal. Conclusion: Our findings suggest that TPE is an effective therapeutic modality in children with refractory kidney disease and can be indicated for the treatment of various kidney diseases.

시험스트레스가 대학생의 질소대사에 미치는 영향 (Effect of Examination-stress on Nitrogen Metabolism of College Students)

  • This study was performed to investigate effects of examination-stress and protein supplementation on nitrogen metabolism and blood protein levels of Korean college students. Experiment was conducted at the beginning of a academic term and during midterm examination. During midterm examination, subjects were classified into two groups randomly : protein supplemental group(male n=6, female n=10) and placebo group(male n=4, female n=9). Protein capsules(2g/day) above 10% of indispensible amino acids requirement estimates were given to supplemental group for 10 days. At the begining of the term, male students(n=12) ingested 223.15mgN/kg/d, excreted 20.7mgN/kg/d in feces, and excreted 94.31mgN/kg/d in urine. Their apparent protein protein digestibility was 90.72%, true N balance was +100.11mgN/kg/d, and the mean maintenance N requirement of mixed Korena diet calculated was 112.13mgN/kg/d. Female students(n=19) ingested 171.44mgN/kg/d, excreted 22.13mgN/kg/d in feces, and excreted 122.92mgN/kg/d in urine. Their apparent protein digestibility was 86.76%, true N blance was + 18.39mgN/kg/d, and the mean maintenance N requirement calculated was 135.31mgN/kg/d. Blood levels of serum total protein, albumin, and BUN were within normal range. During midterm examination, fecal and urinary N excretions of female subjects(n=19) were increased, especially urea N markedly, and urea N/creatinine N ratio was augumented significantly. Apparent protein digestibility of male subjects(n=10) was decreased. Examination-stress showed 8.05mgN/kg/d (7.2%) increase of mean maintenance N requirement in male and 8.55mgN/kg/d(6.3%) increase in female students in comparison with that of the beginning of the term. Serum total protein and albumin levels showed no significant change, but serum transferrin level of female were decreased significantly. During midterm examination, females supplemented with protein capsules(2g/d)had no significant increase in fecal and urinary N excretions.
