Nephroprotective effects of a crude drug-preparation (Ekongsan) were determined from cisplatin induced renal injury in vivo and in vitro. Ekongsan decreased cisplatin induced the cytotoxicity on rabbit kidney proximal tubule and human renal cortical cells by MTT assays and sustained glucose consumption on cisplatin-induced human renal cortical tissue. Levels of creatinine and blood urea nitrogen (BUN) in serum after administration of cisplatin (0.75 mg/kg, i.p.) to Ekongsan (0.75 g/kg/d, p.o.) pretreated rats were markedly lower compared to those of cisplatin-treated rats. Moreover, the administration of Ekongsan significantly inhibited the loss of body weight of cisplatin-injected rats. These findings suggest that Ekongsan is an active prescription in protection against nephrotoxicity of cisplatin.
Objectives : This study was carried out to research the protective effects of Sagunja-Tang(SA) through in vivo experiments, and tried to investigate the relation between oxidation of liver tissues and deficiency of Qi. Methods : Acute liver injury which initiated from free radical induced by $CCl_4$, were applied to mice and metabolic data were obtained. In order to measure the degree of liver injury, serum level of alanine aminotransferase(AST), aspartate aminotransferase(ALT), creatinine, blood urea nitrogen(BUN), total protein(TP) and glucose were measured. Lipid peroxidation of liver slice was examined by measuring malondialdehyde(MDA), a product of lipid peroxidation. Results : SA had protective effects on $CCl_4$ induced acute liver injury by decreasing serum level of ALT. Kidney injury was induced by injection of $CCl_4$ too, and SA protected kidney injury by decreasing serum level of creatinine and BUN. Conclusions : Through this study, we found that SA have healing effects on liver and kidney injury of $CCl_4$ induced oxidative stress that is similar to deficiency of Qi. And further studies have to be followed to certify the mechanisms.
The pharmacokinetics of atenolol (25 mg/kg, i.v.) in the folate-induced renal failure rabbits was studied. Renal failure was induced by the i.v. injection of folate (50, 100, and 200 mg/kg). At folate dose of 100 and 200 mg/kg, the serum creatinine concentration (Scr) and blood urea nitrogen (BUN) increased significantly compared with control rabbits. Plasma concentrations and AUC of atenolol increased significantly at folate dose of 100 and 200 mg/kg. The elimination rate constant $(K_{el})$ and total body clearance $(CL_t)$ of atenolol decreased significantly, and half-life ($t_{1/2}$) and mean residence time (MRT) of atenolol increased significantly at folate dose of 100 and 200 mg/kg. The serum creatinine concentration $(S_{cr})$ correlated well (p<0.05) with half-life $(t_{1/2})$ and elimination rate constant $(K_{el})$ of atenolol, as well as BUN with AUC and total body clearance $(CL_t)$ of atenolol.

In this study, we investigated the effect of silkworm, and its by-products on exercise endurance, muscle mass, and fatigue recovery using ICR mice model. Powders of silkworm, pupae, dongchunghacho, and silk powder were suspended in water and feed to mice for 2 weeks. The forced swimming time was increased by 4.3, 4.1 and 2.8 seconds after silkworm, pupae, and dongchughacho administration compared to DW fed group. Increases in exercise ability were achieved by difference mechanism according to feeding materials. Pupae increased muscle mass by 129% compared with the control group which may effect on elongation of swimming time. Dongchunghacho improved the plasma concentrations of fatigue markers such as creatinine, blood urea nitrogen (BUN), and D-lactate. Silkworm administration showed dual effect, the muscle mass induction (114% vs. control) and anti-fatigue (plasma creatinine, BUN, and D-lactate were 63, 75, and 78% vs. con) effect which contributed most elongated swimming time. In conclusion, silkworm and its by-products including pupae and dongchunghacho with the predominant protein and bioactive components improved muscle mass and showed anti-fatigue effect which could promote exercise performance ability.

This study was conducted to evaluate changes in the lysine to digestible energy (DE) ratio on performance, apparent ileal and fecal nutrient digestibilities as well as blood urea nitrogen (BUN), and to estimate optimal lysine:DE ratios for growing pigs of different sexes. A total of 150 pigs ($(Landrace{\times}Yorkshire){\times}Duroc$, 16.78 kg average body weight, 75 barrows and 75 gilts) was randomly allotted into a $2{\times}3$ (sex by diet) factorial design. Three diets were formulated to contain a crude protein level of 19%, a DE level of 3.5 Mcal/kg with three lysine:DE ratios of 3.2 (low), 3.5 (middle) and 3.8 (high) g lysine/Mcal DE per kg diet for both barrows and gilts throughout the study. With increasing dietary lysine:DE ratio, the average daily gain (ADG) of barrows decreased but there was no significant difference among treatments (p>0.05). However, ADG was significantly higher in gilts fed the diet containing the high lysine:DE ratio (p<0.05), followed by the middle and low lysine:DE ratio dietary groups. No significant effects of lysine:DE ratios on feed intake (ADFI) and feed conversion (F/G) were observed for barrows and gilts during overall period (p>0.05), while the optimal F/G was found in barrows fed diets of low and in gilts fed high lysine:DE ratio. Blood urea nitrogen had a positive relationship with growth rate. The results showed that the optimal lysine:DE ratios were 3.2 and 3.8 g lysine/Mcal DE per kg diet for barrows and gilts of 16 to 57 kg body weight, respectively.
Acetaminophen (APAP) and gentamicin are widely used for many patients, but little in-formation is available regarding the combined effects of APAP and gentamicin. This study was aimed to investigate the potent nephrotoxicity following combined-treatment with APAP and gentamicin. Serum biochemical parameters and histopathological changes in the kidney were observed in female SD rats after continuous daily treatment with either 600 mg/kg/day APAP, and/or 300 mg/kg/day gentamicin for 3 days, and compared with saline sham-treated control animals. APAP and gentamicin combination-treated rats exhibited inconsistent increasing tendency in blood urea nitrogen (BUN) by 96 hours after the last treatment, compared to control or the animals treated with each drug. The relative kidney weights were also increased. Histopathological findings of kidneys revealed that necrosis of proximal convoluted tubules were higher in rats treated with APAP and gentamicin combination than the rats treated with each drug alone. These results suggest that combination use of both drugs have more severe nephrotoxicity than treating each drug alone.
An aqueous extract of oriental herbal composition named Onchengyeum and curcumin, an antioxidant isolated from turmeric (Curcuma Zonga L.) reduced hepatotoxicity induced by carbon tetrachloride ($CCI_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), blood urea nitrogen (BUN), creatinine (CRE), total cholesterol (T-CHO), triglyceride (TG), low density lipoprotein cholesterol (LDL-CHO), high density lipoprotein cholesterol (HDL-CHO), total protein (TP), albumin (ALB) and total bilirubin (BIL) in serum. Hepatic parameters monitored were levels of cholesterol (CHO), triglyceride (TG), and malondialdehyde (MDA) and activities of cytochrome P450 (CYP), NADPH-CYP reductase, superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), glutathione S-transferase (GST), glutathione reductase (GR), and glutathione peroxidase (GPx). The histopathological examination showed that the treatment of Onchengyeum and curcumin relieved the ballooning degeneration of hepatocytes which had been generated by $CCI_4$. The results suggested that hepatoprotective effects of Onchengyeum and curcumin possibly are due to their promising antioxidative activity.
The present investigation evaluated the safety of the methanol extract from the fruit body of Phellinus pini Ames (PPA) by determining its potential toxicity after a subacute administration in rats. The extract was orally administered in doses of 1 g/kg, 2 g/kg, and 4 g/kg daily for 14 days to rats. Body weight, biochemical, and hematological parameters were determined at the end of 14 days of daily administration. The no-observed adverse effect levels (NOAEL) of the extract were 4 g/kg, when given by gavage routes. Daily oral administration of PPA extract for up to 14 days did not result in the death of significant changes in the body weight, hematological, and mainly biological parameters. In biological analysis, some significant changes occurred, including triglyceride and blood urea nitrogen (BUN), indicating that the PPA extract has liver and kidney-modulating activity. The PPA extract was found to be low or non-toxic in rats.
$\beta$-Immunan was proteoglycan obtained from mycelium of Ganoderma lucidum IY009. In this study, the protective effects of $\beta$-Immunan, against the CDDP induced in vitro cytotoxicity and in vivo renal toxicity, was measured. Concentration dependent cytotoxicities of CDDP in normal kidney cells (Vero, TCMK-l) were reduced by $\beta$-Immunan treatment. Increased renal toxicity factors, such as elevation of blood urea nitrogen (BUN) and serum creatinine, reduction of kidney weight and malonidialdehyde (MDA), by intraperitoneal administration of CDDP in rats was improved. These results indicated that $\beta$-Immunan have a protective effects against the CDDP induced renal toxicity, however, it needed to confirm the detailed mechanism for therapeutic effects.
Water extracts of 21 traditional Korean herbal drugs were prepared, and a dose range of 100 mg/kg to 400 mg/kg was administered orally into mice once a day for five days. Changes of serum enzyme activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), blood urea nitrogen (BUN), alkaline phosphatase, body weight changes and histo pathological examination of various organs were investigated. Water extract of Ephedra Herba caused severe body weight loss at a dose of 100 mg/kg and death from a dose level of 200 mg/kg by oral administration. Angelica koreanae Radix and Anthrisci Radix showed a slight body weight loss and damages to liver and kidney.
