300 MHz proton NMR spectra of human blood plasma were analyzed by deconvolution of spectrum, and we compared its results with Fossel's test in normal (15 cases), liver cancer (14 cases) , and other cancer (14 cases) groups. This analysis had enabled us to obtain dynamic characteristics of each individual lipoprotein. As a result of deconvolution method, the VLDL and chylomicron intensity level were found to be elevated in the patients with liver cancer. Moment ratio values of $CH_2$ resonance in the raw spectrum were found to be higher than the normal group for patients with, malignant tumors other than liver cancer. These differences between the three groups could not be found in the conventional Fossel's test. We could simulate plasma spectra by addition of spectra of individual lipoproteins through deconvolution method. Further clinical trials in larger populations and additional biochemical method may shed new light on many of clinical and biochemical interests for knowing characteristics about lipoprotein not separated from blood and the background of Fossel test.

Proteolytic enzymes secreted by Aspergillus, as pathogenicity factors, affect blood coagulation and fibrinolysis, and therefore the target proteins of their action in the bloodstream are of significant interest. In the present study, the action of the isolated protease of A. terreus 2 on different human plasma proteins was shown. The protease of A. terreus 2 exhibited the highest proteolytic activity against hemoglobin, which was 2.5 times higher than the albuminolytic activity shown in both of the protein substrates used. In addition, the protease has significant ability to hydrolyze both fibrin and fibrinogen. However, the inability of the A. terreus 2 protease to coagulate rabbit blood plasma and coagulate human and bovine fibrinogen indicates the severity of the enzyme's action on human blood coagulation factors. It should be considered as a potential indicator of this isolated protease's participation in fungal pathogenesis. The protease shows no hemolytic activity. Furthermore, its activity is insignificantly inhibited by thrombin inhibitors, and is not inhibited by plasmin inhibitors.
Blood glucose and immunoreactive insulin (IRI) were measured during oral glucose tolerance test in 15 thyrotoxic patients and 8 normal controls, to study the glucose metabolism in thyrotoxicosis. Following were the results; 1. In thyrotoxicosis, there is noticed late rise and late fall of plasma IRI durnig oral glucose tolerance test, like as phenomenon of mild diabetes mellitus. 2. When the thyrotoxic patients were divided into normal and abnormal responsive groups after the level of blood glucose by Wilkerson Criteria, no significant differences in plasma IRI levels were noticed between two groups. 3. This result may be interpreted as relative deficiency of insulin secretion from panceas and suggest genetically related defects.
Atherosclerotic vascular disease is a major cause of the increased morbidity and mortality assciated with diabetes mellitus. The prominent role of nutrition in hypercholesteolemia and atherosclerosis is generally accepted. Diet is a key element in the management of diabetes (type I-IDDM), yet the appropriate diet for patient with diabetes mellitus is not well known. Dietary protein has been shown to have a significant effect on plasma cholesterol levels in both experimental animals and humans. The present experiment was designed to determine the effect of the dietary protein level(20% vs 60%) on plasma glucose concentration, lipids profile, insulin and glucagon levels from non-diabetic and streptozotocin-induced diabetic rats. Results showed that a high protein diet decreased triglyceride concentration in diabetic rats. Also diabetic rats fed a high protein diet were hypocholesterolemic than rats fed a control diet. There were no effects by level of protein on fasting blood glucose concentration and insulin/glucagon ratio. Results from the present study suggest that a high protein diet may be beneficial to control pasma lipids in chemically-induced diabetic rats.
Objective : This study was performed to investigate the effects of aqua-acupuncture of Jibaikjihwangtang in two-kidney one clip Goldblatt hypertensive rats and spontaneously hypertensive rats. Methods : we injected aqua-acupuncture solution into Shin-Soo ($BL_{23}$) which corresponds to human acupuncture point in two-kidney one clip Goldblatt hypertensive rats and spontaneously hypertensive rats. Systolic blood pressure, renin activity, aldosterone and atrial natriuretic peptide (ANP) plasma levels were tested. Results : Systolic blood pressure decreased significantly after aqua-acupuncture of jibaikjihwangtang. Acupuncture group in two-kidney one clip Goldblatt hyper-tensive rats had deference with control group. In plasma levels of atrial natriuretic peptide, acupuncture group of spontaneously hypertensive rats increased meaningfully but to two-kidney one clip Goldblatt hypertensive rats it was decreased meaningfully. In Serum Aldosterone density, the acupuncture group of spontaneously hypertensive rats had significant alteration than control group, but the acupuncture group of two-kidney one clip Goldblatt hypertensive rats had decreased alteration than control group. Conclusion : According to these results, after Aqua-Acupuncture of Jibaikjihwangtang blood pressure decreased significantly and data suggest that blood pressure reduction activity connected with renin activity reduction in renal hypertensive rat.
In order to investigate the relationship between plasma renin activity and metabolism of Ca and Na in blood pressure, the habitual dietary intakes of Na and Ca urinary excretion of Ca, Na and K, and plasma level of renin activity, aldosterone, and indices of Ca metabolism were measured in 27 untreated hypertensive women and 30 age-matched normal women on a free diet. Hypertensive subjects were classified into high renin hypertensive (HH), medium renin hypertensive(HM) and low renin showed no significant difference among normotensive, LH, MH and HM groups. It appeared that 25-(OH) Vit D3 level of HH group was significantly higher than LH group(p<0.05). There was significant difference in habitual intake of Ca between normotensive and LH groups. However, habitual intake of Na showed no significant difference among normotensive, LH, MH and HH group. Positive correlation of systolic and diastolic blood pressure with PTH(r=0.324, r=0.375) and urinary Ca(r=0.496, r=0.278) and a negative correlation of systolic blood pressure with habitual Ca intake(r=-0.371) existed(p<0.05). A relative magnitude of factors affecting hypertension was analyzed by multiple regression analysis. Overall results about relative influence of independent variables to dependent variable (systolic blood pressure) indicated that urinary Ca was the higher correlation in all subjects(p<0.0001), followed by age and aldosterone. PTH showed a significant correlation for relative influence on diastolic blood pressure in all subjects. The above results indicated that renin-aldosterone system and Ca regulating hormone had a mutual relationship in hypertension.
In oriental medicine, Gamibangpungtongsungsan (KBTS) has been used as a therapeutic agent for the treatments of acute stage of cerebrovascular diseases and hypertension. In the present study, underlying mechanism on KBTS effects was investigated using spontaneously hypertensive rats (SHR) by determining related parameters such as blood pressure, heart-beat rates, and hormones and plasma constituents. The major finding are summarized as follows. 1. KBTS treatment at concentrations lower than $125\;{\mu}g/m{\ell}$ did not show any cytotoxicity on cultured human fibroblast cells. 2. KBTS treatment in SHR significantly decreased blood pressure and heart-beat rate compared with untreated control. 3. KBTS treatment in SHR decreased aldosterone levels in the blood compared with untreated control, but the difference was not statistically significant. 4. KBTS treatment in SHR significantly decreased dopamine, norepinephrine and epinephrine levels in the blood compared with untreated control. 5. KBTS treatment in SHR decreased plasma ion concentrations such as Na+, K+, Ca2+, Cl- compared with untreated control; decreases in Na+ and Cl- were statistically significant. 6. KBTS treatment in SHR significantly decreased TNF-$\alpha$, IL-6, and IL-10 levels in the blood compared with untreated control. Thus, the present data show evidence on anti-hypertension activity of KBTS in an experimental animal system, which can provide further insights into the development of anti-hypertension therapeutic agents.
Comparative blood analysis was carried out to investigate the change of blood constituent in TAE-EUM-IN group and control group. RBC, WBC, hemoglobin, hematocrit, total protein, triglyceride, phospholipid, total cholesterol, LDL-cholesterol, HDL-cholesterol, BUN, creatinine, ACTH, cortisol and prostaglandin E and $F_2{\alpha}$ were measured. And the following results were obtained: 1. In the change of blood cell, the value of hematocrit showed significant difference, while that of RBC, WBC and himoglobin didn't. 2. In the change of protein, the value of total protein showed significant difference, but that of albumin didn't. 3. In the change of lipid and cholesterol in serum, the value of triglyceride, phospholipid, total cholesterol and LDL-cholesterol indicated significant difference, while that of HDL-cholesterol indicated significant difference, while that of HDL-cholesterol didn't. 4. The value of BUN in serum represented significant difference, but that of creatinine didn't. 5. The value of cortisol in plasma showed significant difference, but that of ACTH didn't. 6. The value of prostaglandin E and $F_2{\alpha}$ in plasma showed significant difference. These results suggest that investigation on hormones and blood constituents enables objectification in differentiation of four types of physical constitution.
In the present study, we determined the degree of hydrolysis (DH) of porcine blood plasma proteins, albumin, and globulin hydrolyzed by six proteases (alcalase, neutrase, flavourzyme, protamex, trypsin, and papain) for various reaction times. Moreover, antimicrobial activities of hydrolysates against five pathogenic microorganisms (Bacillus cereus, Staphylococcus aureus, Salmonella Typhimurium, Escherichia coli, and Shigella flexneri) were investigated. Alcalase, trypsin, and papain hydrolysates of the three porcine blood proteins showed higher DH values than hydrolysates produced by the other three proteases. DH of the three porcine blood proteins hydrolyzed by the six proteases failed to increase after 2 h of hydrolysis. In antimicrobial tests, hydrolysates (hydrolysis time of 2 h) showed antibacterial activity only against B. cereus. Albumin hydrolysates showed higher antimicrobial activity than globulin and plasma hydrolysates. Albumin hydrolysates obtained with flavourzyme, protamex, and trypsin showed higher antimicrobial activity than those obtained with the other three proteases.
This study was performed to investigate the effects of different dietary magnesium levels on systolic blood pressure and mineral distribution in normotensive and spontaneously hypertensive rats. In experiment 1, Normotensive rats(NTR ; Sprague Dawley, Female) were given diets containing regular magnesium (0.05% Mg ; rMg), marginal magnesium (0.01% Mg ; mMg) or marginal magnesium with stress(0.01% Mg + stress ; mMg + Str). In experiment 2, spontaneously hypertensive rats (SHR ; Kyoto Wistar, Femal) were fed diets containing regular magnesium(0.05% Mg ; rMg) and high magnesium (0.2% Mg ; hMG). The following were found ; 1) NTR treated with marginal magnesium with stress showed significant increase in systolic blood pressure (SBP). Marginal magnesium diet without stress resulted in nonsignificant increase in SBP. Significant in crease of blood pressure showed in NTR treated with marginal magnesium and stress was associated with decreased magnesium and increased calcium content in femur, reticulocyte and plasma. 2) In experiment 2, magnesium supplementation to SHR showed significant attenuation of their systolic blood pressure with increasing age. The attenuation of SBP showed in SHR was associated with increased magnesium, lowered calcium content in cardiac muscle and reticulocyte and decreased plasma sodium and aldosterone level.
