4-tert-octylphenol (OP) is a surfactant additive widely used in the manufacture of a variety of detergents and plastic products. Also, OP is known to have estrogenic activity by interacting with development and functions of endocrine system. This study was carried out to obtain toxicokinetic parameters of OP in male Sprague-Dawley rats. Male rats were administered OP, by either single oral (gavage) applications of 50, 100 or 200 mg/kg body weight. or a single intravenous injections of 1, 5 or 10 mg/kg body weight. Blood samples taken at several time intervals after administration were obtained from the femoral artery. Analysis of blood samples for OP was performed by gas chromatography mass spectrometry (GC/MS). The detection limit of OP was 1.9 ng/$m\ell$ at SIM (selected ion monitoring) mode of GC/MS. Calibration curve for analysis of the concentrations of OP in plasma was (OP/butylphenol peak area ratio) = 0.0294 $\times$ (plasma cone.) + 0.028 ($r^2$= 0.9991). The OP plasma concentration was 3921 ng/$m\ell$ immediately after single intravenous application, decreased rapidly within 45 min, and was detectable at low concentration up to 6 hr after application. When administered orally in rats (50, 100 and 200 mg/kg), OP was detected in the blood early after gavage administration, indicating the rapid initial uptake from gastrointestinal tract, with Tmax obtained from 0.67~0.83 hr. Using the AUC (area under the curve) of plasma concentration vs. time, low oral bioavailabilities of 1.2, 5.0 and 5.3% were calculated for the 50, 100 and 200 mg/kg groups, respectively.

The purpose of this study was to investigate the effects of lotus leaf on hyperglycemia and dyslipidemia in animal model of diabetes. Inhibitory activity of ethanol extract of lotus leaf against yeast ${\alpha}$-glucosidase was measured in vitro. The effect of lotus leaf on the postprandial increase in blood glucose levels was assessed in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without lotus leaf extract (500 mg/kg) was administered to the rats after an overnight fast, and postprandial plasma glucose levels were monitored. Four-week-old db/db mice were fed a basal diet or a diet containing 1% lotus leaf extract for 7 weeks after 1 week of acclimation to study the chronic effect of lotus leaf. After sacrifice, plasma glucose, insulin, triglycerides (TG), total cholesterol (CHOL), high-density lipoprotein (HDL)-CHOL, and blood glycated hemoglobin levels were measured. Lotus leaf extract inhibited ${\alpha}$-glucosidase activity by 37.9%, which was 1.3 times stronger than inhibition by acarbose at a concentration of 0.5 mg/mL in vitro. Oral administration of lotus leaf extract significantly decreased the area under the glucose response curve by 35.1% compared with that in the control group (P < 0.01). Chronic feeding of lotus leaf extract significantly lowered plasma glucose and blood glycated hemoglobin compared with those in the control group. Lotus leaf extract significantly reduced plasma TG and total CHOL and elevated HDL-CHOL levels compared with those in the control group. Therefore, we conclude that lotus leaf is effective for controlling hyperglycemia and dyslipidemia in an animal model of diabetes mellitus.
The present study was conducted to investigate te effect of sea tangle and hypoglycemic agent(Metformin) on lipid peroxidation and antioxidants levels in normal and diabetic rats. Male Sprague-Dawley rats were fed AIN-76 based experimental diets containing 5% (w/w) cellulose or 15%(w/w) sea tangle for 3 weeks, and then rats of diabetic groups were rendered diabetic by intramuscular injection of streptozotocin(STZ, 45mg/kg BW). After induction of diabetes Metformin(350mg/kg BW) was given once a day using a feeding tube for 5 days. Blood glucose level in diabetic rats fed sea tangle was significantly lower than that of diabetic rats fed cellulose. Metformin feeding resulted in further lowering blood glucose. Plasma and liver microsomal levels of lipid peroxides were increased significantly in diabetic rats as compared to normal rats, and the plasma level tended to be decreased by sea tangle feeding. Plasma level of retinol was reduced by STZ treatment, but it was increased by Metformin feeding in diabetic rats fed sea tangle. The liver contents of retinyl plamitate were reduced in diabetic rats but recovered by sea tangle feeding to some extent. Liver contents of total vitamin A were increased significantly by sea tangle in diabetic rats. Although difference in either plasma or liver level of $\alpha$-tocopherol by diet and STZ treatment was not significant, $\alpha$-tocopherol levels were the highest in the group with simultaneous feeding of sea tangle and metformin. Liver contents of zinc and copper were not influenced by either STZ treatment or sea tangle feeding. The present study indicates that the lowering blood glucose level could be attained by simultaneous trial of sea tangle diet and hypoglycemic agent and the increased oxidative stress caused by STZz treatment could be relieved by sea tangle feeding.

The aim of this study was to investigate the effects of three herb supplementations on blood metabolites, hormones, antioxidant activity, immunoglobulin (Ig) G concentration, and ruminal fermentation in steers. Four Holstein steers in a $4{\times}4$ Latin square design received four herb treatments. The treatments consisted of the steers' regular diets with addition of: 1) nothing (control), 2) peppermint, 3) clove, and 4) lemongrass at 5% of the diet (DM basis). Clove supplementation increased the plasma concentration of cholesterol by about 10% (from 79 to 87 mg/dl). Peppermint and lemongrass feeding resulted in an increase in the concentrations of plasma urea nitrogen (from 5.9 to 6.9 and 6.4 mg/dl, respectively). The three herb treatments had no effect on other metabolites and hormones. Steers receiving clove supplementation showed a higher plasma antioxidant activity. The three herb treatments caused lower concentrations of IgG in the blood. Peppermint and lemongrass feedings increased, and clove feeding decreased ruminal concentrations of ammonia. There were no significant differences in VFA concentrations among herbal treatments, except for the decrease in propionate concentration in steers receiving clove treatment. This study suggested that clove feeding changed cholesterol metabolism and increased antioxidant activity in plasma, and feeding of three herbs affected immunity system and ruminal fermentation in steers.

The effects of green tea (Camellia sinensis) waste silage and supplemental polyethylene glycol (PEG) on rumen fermentation and blood components were studied in cattle. Six Holstein steers were fed three diets in a 3${\times}$3 Latin square design, replicated twice. One diet was a control with no added silage, and the other two diets were supplemented (20% of the dry matter) with green tea waste silage either with (PEG) or without PEG (tea). Most of the fermentation parameters including major volatile fatty acids (VFA) were not affected by the diet treatments. The concentrations of high density lipoprotein cholesterol in the PEG group and urea nitrogen in the tea and PEG groups were greater than those in the control before morning feeding. The plasma 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid equivalent concentration was not different before morning feeding, but 3 h after morning feeding, its concentrations in both the tea and PEG groups were higher than in the control. Although the concentration of plasma vitamin A in the animals was not affected by feeding green tea waste silage, the concentrations of plasma vitamin E were significantly higher in the tea and PEG groups than in the control, both before and 3 h after morning feeding. The results from the present study suggest that feeding diets containing 20% of the dietary dry matter as green tea waste silage to Holstein steers has no negative impact on their ruminal fermentation, and increases their plasma antioxidative activity and concentration of vitamin E.
Palm kernel cake (PKC), a by-product of oil palm seeds after extraction of their oil. The aim of this study was to investigate the effects of different levels of PKC on growth performance and blood lipids in rats. A total of 64 Sprague-Dawley (8 weeks of age) male rats were assigned individually to four treatments with different levels of PKC in the diet: 0, 15, 20 and 25%. No differences (p<0.05) were found in daily feed intake (6-8 g/day), body weight, growth rate and epididymal fat weight for all the dietary groups. Plasma protein and very low density lipoprotein (VLDL) triacylglycerol (TG) were higher (p<0.05) for 20% PKC fed rats than the control rats. Conversely, the plasma cholesterol and TG and VLDL-phospholipid (PL) concentrations of the control rats were higher (p<0.05) than those of PKC fed rats. The VLDL-protein, total cholesterol, free cholesterol (FC) and cholesteryl ester (CE) were not significantly different (p>0.05) among the treatment groups. Rats fed PKC had greater (p<0.05) ratios of total surface to core lipid components [(FC+PL)/(CE+TG)] than control rats. The results reflect dissimilarities of VLDL particle size between PKC treatment and control rats, where the plasma of the PKC treated rats contained more lipid rich VLDL. In conclusion, there was no adverse effect on growth performance when inclusion of PKC up to 25%. However, fibre content may affect the plasma lipid concentrations.
Background: We investigated the tolerability and pharmacokinetic properties of various ginsenosides, including Rb1, Rb2, Rc, Rd, and compound K, after single or multiple administrations of red ginseng extract in human beings. Methods: Red ginseng extract (dried ginseng > 60%) was administered once and repeatedly for 15 days to 15 healthy Korean people. After single and repeated administration of red ginsengextract, blood sample collection, measurement of blood pressure and body temperature, and routine laboratory test were conducted over 48-h test periods. Results: Repeated administration of high-dose red ginseng for 15 days was well tolerated and did not produce significant changes in body temperature or blood pressure. The plasma concentrations of Rb1, Rb2, and Rc were stable and showed similar area under the plasma concentration-time curve (AUC) values after 15 days of repeated administration. Their AUC values after repeated administration of red ginseng extract for 15 days accumulated 4.5- to 6.7-fold compared with single-dose AUC. However, the plasma concentrations of Rd and compound K showed large interindividual variations but correlated well between AUC of Rd and compound K. Compound K did not accumulate after 15 days of repeated administration of red ginseng extract. Conclusion: A good correlation between the AUC values of Rd and compound K might be the result of intestinal biotransformation of Rb1, Rb2, and Rc to Rd and subsequently to compound K, rather than the intestinal permeability of these ginsenosides. A strategy to increase biotransformation or reduce metabolic intersubject variability may increase the plasma concentrations of Rd and compound K.
Objectives : Acupuncture has been used as treatment of disease in the korean medicine. In this study, it was investigated that effects of reduction of acupuncture techniques of five evolutive phase for appling excess in the heart, kidney on cardiovascular system as blood pressure and renin and atrial natriuretic peptide (ANP) in plasma, cardiac hypertrophy. Materials and methods : The experiments were performed on Sprague Dawley rats, 2K1C hypertension model was prepared by constricting the left renal artery with a sliver clip. Animals were then divided into seven groups, 2K1C induced and no treated group (Control), acupuncture on SP3 HT7(AC-1), LR1 KI1 (AC-2), SP3 HT7 LR1 KI1 (AC-3). The treatments were performed once a day for 10 days in rats. Results : The results are that the blood pressure was significantly decreased at 15days in AC-1 group. The cardiac hypertrophy was significantly decrease in AC-3 group. The activity of plasma ANP was increased in all groups without AC-1 group and the that of plasma Rein was decrease in AC-1, AC-2 groups than control group. Conclusions : These results suggest that acupuncture at SP3 HT7 and SP3 HT7 LR1 KI1 can be used as a therapy for controlling renal hypertension induced by 2K1C in the rats.
Quinones have been reported to undergo nonenzymatic reaction with thiols to generate reactive oxygens. It is therefore possible that the nonenzymatic reaction of quinones with thiols in plasma could lead to potentJared cellular toxicity or disease. When 1 mM menadione was added in plasma under pH 11.2, 7.4 and 5.0, the increase in oxygen consumption rate was the order of pH 11.2 > pH 7.4 > pH 5.0. In addition, oxygen consumption rates under plasma anticoagulated with trisodium citrate solution (pH 7.85) was significantly higher than those with acid-citrate-dextrose solution (pH 6.87). SOD and catalase reduced the rate of oxygen consumption induced by menadione in plasma. Taken together, these results suggest that the menadione-induced increased oxygen consumption was due to nonenzymatic reaction of menadione with thiols in the plasma. The presence of plasma has an additive effect on the increased oxygen consumption rates induced by the menadione treatments on our model tissue, platelets, as compared between washed platelet (WP) and platelet rich plasma (PRP). Cytotoxicity, as determined by LDH release, are well correlated with the oxygen consumption rates observed in each system and strongly suggest that menadione-induced cytotoxicity can be increased with the presence of blood plasma.
This study was carried out to investigate the effects of garlic vinegar supplementation on changes in body weight, blood glucose and serum triglyceride and cholesterol. Rats were fed a cholesterol-diet with or without garlic vinegar (20% garlic juice )supplement for 28days. Body weights in rats fed a diet containing garlic vinegar were significantly lower than those of control rats. The concentration of plasma glucose, total cholesterol . LDL-cholesterol , and HDL-cholesterol were not significantly different between the two groups for a period of up to four weeks. However, the concentration of plasma triglyceride was slightly decreased in the garlic vinegar-supplemented group. Results suggest that the supplementation of garlic vinegar is beneficial for weight reduction in high-cholesterol fed rats.
