Effects of d-${\alpha}$-tocopherol supplements on lipid metabolism in a high-fat diet-fed animal model

  • High-fat diet up-regulates either insulin resistance or triglycerides, which is assumed to be related to the expression of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ and PPAR-${\gamma}$. The beneficial effects of vitamin E on insulin resistance are well known; however, it is not clear if vitamin E with a high-fat diet alters the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$. We investigated the effects of d-${\alpha}$-tocopherol supplementation on insulin sensitivity, blood lipid profiles, lipid peroxidation, and the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat (HF) diet-fed male C57BL/6J model of insulin resistance. The animals were given a regular diet (CON; 10% fat), a HF diet containing 45% fat, or a HF diet plus d-${\alpha}$-tocopherol (HF-E) for a period of 20 weeks. The results showed that the HF diet induced insulin resistance and altered the lipid profile, specifically the triglyceride (TG) and total cholesterol (TC) levels (P < 0.05). In this animal model, supplementation with d-${\alpha}$-tocopherol improved insulin resistance as well as the serum levels of TG and very-low-density lipoprotein-cholesterol (VLDL-C) (P < 0.05). Moreover, the treatment decreased the levels of malondialdehyde (MDA) in the serum and liver while increasing hepatic PPAR-${\alpha}$ expression and decreasing PPAR-${\gamma}$ expression. In conclusion, the oral administration of d-${\alpha}$-tocopherol with a high-fat diet had positive effects on insulin resistance, lipid profiles, and oxidative stress through the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat diet-fed male mice.

제주지역 성인의 Apolipoprotein E Phenotype 분포와 식생활 및 혈청지질 농도의 관련인자 연구 (The Effects of Dietary Patterns and Apolipoprotein E Phenotype on the Blood Lipid Profiles of Individuals from Cheju Area)

  • The purpose of this study was to determine the relation between serum lipid profiles, apolipoprotein E phenotype, and dietary patterns in a cross-section of healthy individuals from Cheju-Do. Age, gender, anthropometric measurements, blood pressure, dietary consumption, drinking / smoking habits and menopausal status were surveyed. Total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, fasting blood glucose, and insulin levels were measured from overnight fasting blood. The study involved a total of 286 individuals(147 men and 139 women) between the ages of 20 and 60 years old. All of the subjects were recruited from a population of healthy individuals living in Cheju-Do. The results of the study are as follows : 1) Among the males, those in their 20's had the maximum food intake, while those in their 40's had the minimum food intake. For the females, food intake was the highest for those in their 30's. Energy and nutrient intakes were directly proportional to the amount of food intake. Men in their 30's were heavier than other men and women in their 40's were heavier than other women. The activity index for men in their 20's and 30's appeared to be lower than that of men above 40. The activity index of women in their 20's appeared to be lowest among all aged groups, and the index appeared to increase from the age of 30 onwards. 2) In terms of changes In serum constituents with age, men in their 40's appeared to have the highest levels of serum constituents such as lipids, glucose, and insulin. Men in their 50's showed the highest levels of serum LDL-cholesterol and glucose. Men in their 30's showed peak levels of serum triglycerides. On the other hand, women in their 50's appeared to have peak levels of serum total cholesterol, LDL-cholesterol, and triglycerides. There was no ch:ange with age in HDL-cholesterol and insulin levels for men and women. The percentage of the subjects had the following apo E phenotypes : E3/3, 91.3% ; E3/2, 5.4% ; E4/3, 2.5% ; E4/2, 0.7%. Lee's reserch with Korean female college students showed that the percentage of ApoE3/3, E3/2, E 4/2, E4/3, and E4/4 were 84.8%, 6.7%, 6.7%, 0.9%, 0.9%, respectively. The number of samples with ApoE mutation was so small that there was no statistical significance in the relation between apolipoprotein E phenotype and se겨m lipids. 3) To investigate the relati onship between weight and serum constituents, the subjects of this study were divided into three groups by BMI underweight, normal weight, and overweight. The serum constituents of men and women below the age 40 in the overweight groups belonged to the normal domain. On the other hand, serum cholesterol levels of both men and women above the age 40 in the overweight groups remained in the borderline-high region(above 200mg/dl), and the mean value of LDL-cholesterol(above 130mg/dl) and triglycerides of men were above normal. Fasting blood glucose levels also remained in the borderline-high region. Total cholesterol levels of women above the age 40 in the overweight group was in the borderline-high region. 건강진단 후 추구 관리의 현황 및 의의 (Significance of Follow-up Blood Test after Health Examination)

  • After a health examination, abnormal patients were classified into two groups. One needs clinical treatment immediately, while the other one needs check-up in 2~3 month after for blood tests and preventive medical study. To discover the status and importance of the follow-up study in a health program, blood test upon health examination was compared to the test from the follow-up. We examined patients of ages from 20's to 70's. Among these patients, most of them were between 40's and 50's, and in male patients the follow-up test were more freguent than female. We examined blood obtained from January 2007 to December 2008. We analyzed and compared initial and the follow up blood test. Lipid profile, liver test, thyroid function tests (TFTs), glucose fasting, and HbA1c were examined from the blood tests. We also surveyed their distribution and status. The value of the follow up blood tests was significantly decreased with the most striking differences in lipid profiles and liver test. Follow up study is able to improve life style of patients through doctor's advice in a health program. Therefore, we can conclude that the follow up blood test is also quite critical.

이소플라본 섭취 수준이 주폐경기와 폐경 후 여성의 혈청 내 지질 패턴과 Total Antioxidant Status에 미치는 영향 (Dose Respone Relationship of Isoflavone Supplementation on Plasma Lipid Profiles and Total Antioxidant Status in Perimenopausal and postmenopausal Women)

  • There is growing evidence that soy isoflavones play beneficial role in the prevention of cardiovascular disease. However, current data are not sufficient to determine the effective dose of isoflavone on improving plasma lipid profiles and total antioxidant capacity in Korean menopausal women. In the present study, 24 perimenopausal and 23 postmenopausal women were divided into 3 groups and were given soy isoflavone supplements of 100, 150, 200mg/day for 12weeks. Blood samples were taken before and after 6, 12weeks of soy isoflavone supplementation for analysis of total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride and total antioxidant status(TAS). In the perimenopausal women, serum total cholesterol level decreased significantly in all three groups after 12weeks of isoflavone supplementation(p< 0.05). However, serum LDL cholesterol level significantly lowered only in 150 and 200mg groups, and serum triglyceride level decreased significantly in 200mg group(p<0.05). In the postmenopausal women, the 150mg isoflavone supplementation group demonstrated significant effects on lowering serum total cholesterol, LDL cholesterol levels and raising HDL cholesterol level after 12weeks of isoflavone supplementation(p<0.05). Two hundred mg isoflavone supplementation group had a significantly higher TAS than other groups in both perimenopausal and postmenopausal women. (Korean J Nutrition 34(3) : 322∼329, 2001)

The Effect of Nutrition Education Using MyPlate on Lipid Profiles, Glycemic Indices, and Inflammatory Markers in Diabetic Patients

  • As a result of a nutrition transition, chronic diseases, including diabetes, have increased in Iran. Nutrition education is a cost-effective method for modifying diet and controlling diabetes. This study aimed to examine the effect of nutrition education using MyPlate recommendations on glycemic and lipid profiles and inflammatory markers in Iranian adults diagnosed with type 2 diabetes. A 12-week randomized clinical trial was conducted on 44 adults aged 30-50 years from Ahvaz, Iran. The participants were divided into education and control groups. The education participants were taught the MyPlate recommendations. Serum levels of fasting blood sugar (FBS), hemoglobin A1c (HbA1c), lipid profiles, and inflammatory markers, including high sensitivity C-reactive protein (hs-CRP), tumor necrosis factor-α, and adiponectin, were measured at the baseline and the end of the study. The results showed that serum levels of FBS (p = 0.014) and HbA1c (p < 0.001) decreased significantly in the education group at the end of the study. The serum level of low-density lipoprotein in the education group declined significantly at the end of the study (p = 0.043). Furthermore, the serum level of hs-CRP (p = 0.005) declined significantly while the level of adiponectin (p = 0.035) increased in the education group at the end of the study. The evidence of this study showed that nutrition education using MyPlate recommendations is an effective method for controlling diabetes complications. A longitudinal analysis with a larger sample size is recommended to confirm the evidence of this study.

비만하지 않은 다낭성난소증후군 환자에서 메트포민 효용성의 예비 연구 (The Effect of Metformin in Non-Obese Women with Polycystic Ovary Syndrome; Pilot Study)

  • 목 적: 본 예비 연구는 비만하지 않은 다낭성난소증후군 환자들에게서 인슐린 저항성, 혈중 호르몬 농도 및 혈중 지질 농도에 관한 metformin의 효과를 알아보기 위해 수행하였다. 연구방법: 본 연구는 2006년 6월부터 2007년 9월까지 본원에서 다낭성난소증후군으로 진단받은 16명의 비만하지 않은 환자를 대상으로 하였다. 혈액 검사는 metformin 치료 전과 치료 후 6개월이 경과한 후에 실시하였으며 공복 혈당과 공복 인슐린 치, 75 g의 액상 포도당 섭취 후 2시간째의 혈당과 혈중 인슐린 농도를 측정 하였다. 혈중 호르몬 및 혈중 지질 농도는 FSH, LH, estradiol, testosterone, free testosterone, serum lipid profile을 포함하였다. 인슐린 저항성을 평가하기 위해 공복 시 혈당/인슐린 비율 (fasting glucose/insulin ratio; FGIR)과 액상 포도당 섭취 후 2시간째의 혈당/인슐린 비율을 산출하여 추정하였다. 또한 HOMA beta cell function 및 HOMA IR을 산출함으로써 인슐린 저항성 및 췌장 베타세포 기능을 조사하였다. 결 과: Metformin으로 치료한 후, 75 g 경구 당 부하 후 2시간째 측정한 glucose/insulin 비율은 증가되었으며 (p=0.04), HOMA IR은 치료 후 유의하게 감소가 있었다 (p=0.000). 그러나 혈중 지질 농도는 의미 있는 변화를 보이지 않았다. 혈중 free testosterone 농도는 치료 후 감소하였고 (p=0.001), LH 농도 및 LH/FSH 비도 감소하였다 (p=0.000, p=0.034). 결 론: 본 예비 연구는 metformin이 비만하지 않은 다낭성난소증후군 환자들에게서 인슐린 감수성을 개선하고, 고안드로겐혈증을 완화시키는 효과가 있을 수 있다는 것을 보여준다. Metformin의 역할을 결정하기 위하여 향후 보다 다수의 환자 군을 대상으로 하여 장기적인 추적 관찰을 수반한 심도 있는 연구가 이루어져야 할 것이다.

승마운동의 유형에 따른 노인의 혈중 지질의 차이에 관한 연구 (Differences in the Serum Lipid Profile by Types of Horseback Riding Exercises in Senior Citizens)

  • Purpose : We investigated the effects of horseback riding and horseback riding machine exercise on the serum lipid profile in elderly individuals. Methods : Our study included 30 healthy elderly individuals who were randomly assigned to 2 groups: the horseback riding and the horseback riding machine exercise group, with each group comprising 15 individuals. The riding exercise program was performed 25 times a week over 12 weeks. The repeated measures analysis of variance was used to determine changes in serum lipid profiles before and after exercise 6 and 12 weeks after exercise. Results : An intergroup comparison of blood tests showed that triglyceride, total cholesterol and low-density lipoprotein-C levels were decreased in both groups after 12 weeks of the riding exercise program. However, both groups showed an increase in high-density lipoprotein-C levels, although this difference was not statistically significant. Conclusion : Horseback riding and horseback riding machine exercise showed a positive effect on the serum lipid profile in elderly individuals.
