Effects of Acai Berry Ethanolic Extracts on Production of Nitric Oxide and Activity of Angiotensin Converting Enzyme Related to Blood Circulation (혈액순환과 관련하여 nitric oxide 생성과 angiotensin converting enzyme 활성에 미치는 Acai berry 에탄올 추출물의 영향)

  • The aim of this study is to develop a supplementary therapeutic agent capable of promoting vascular circulation. The effects of Acai berry ethanolic extracts (ABEE) on activity of angiotensin converting enzyme from rabbit lung, production of nitro oxide in both murine macrophage cells and vascular endothelial cells as well as antioxidant effects were investigated in this study. First of all, it was observed the direct effects of ABEE on reducing power and antioxidant effect lipid peroxidation. In addition, ABEE showed a protective effect on DNA oxidation induced by hydroxyl radical. Furthermore, ABEE at 0.01% exerted approximately 50% inhibition on activity of angiotensin converting enzyme. ABEE increased the production of nitric oxide in endothelial cells, but decreased the induction of nitric oxide stimulated by lipopolysaccharide in microphage. The expression levels of superoxide dismutase (SOD)-2 and -3 were enhanced by ABEE treatment, however, the expression level of SOD-1 remained constant. Moreover, the expression level of nitric oxide synthases-1 (NOS-1), a constitutive enzyme, was increased by ABEE, but that of NOS-2, a inducible enzyme, was constant. It was also found that the level of Nrf-2, a transcription factor of SOD, was increased by ABEE. Therefore, these results demonstrate that ABEE could promote blood circulation via above actions, suggesting that may be helpful for health of blood vessel.

In vivo Biological Function of a Fibrinolytic Enzyme after Oral Adminstration (혈전용해효소의 경구투여에 의한 생체 내 작용)

  • A fibrinolytic enzyme gene (BCF-1) was subcloned to the pEB vector which is high expression vector in the Bacillus host. The enzyme was purified by using FPLC after ammonium sulfate precipitation. The enzyme was oral-administrated to the rat and checked the bleeding time, blood clotting time and fibrinolytic effect of the serum. In the bleeding time retardation test, it was longer about 1.7 fold in the feeding rat than without feeding. The serum of rat feeded with the enzyme had the fibrinolytic activity from 1 hour to 3 hours after oral-administration. After 3 hours from feeding, the fibrinolytic activity was decreased gradually. Also blood clotting time after bleeding was longer than that of control rat. The enzyme could be detected at band of 30,000 Da in the blood by western blotting. The enzyme was not harmful to the all internal organs of the rats. Taken together, the enzyme originated from B. subtilis BB-1 can be a candidate to develop the drug for thrombosis, arteriosclerosis and myocardial infarction.

Synergistic effect of exogenous multi-enzyme and phytase on growth performance, nutrients digestibility, blood metabolites, intestinal microflora and morphology in broilers fed corn-wheat-soybean meal diets

  • Objective: This study was conducted to investigate the synergistic effect of exogenous multienzyme and phytase on growth performance, nutrients digestibility, blood metabolites, intestinal microflora, and morphology in broilers fed corn-wheat-soybean meal diets. Methods: A 2×2 factorial design was used in this study. Four dietary treatments consisted of i) basal diets (corn-wheat-soybean meal based diets without multi-enzyme and phytase), ii) basal diets with phytase (0.05%), iii) basal diets with exogenous multi-enzyme (0.05%), and iv) basal diets with exogenous multi-enzyme including phytase (0.05%). A total of 480 broiler chickens (Ross 308 - one day old) were weighed and allotted to thirty-two cages (15 birds per cage), and chicks were randomly allocated to four dietary treatments. Results: The body weight gain and feed conversion rate were improved by supplementation of exogenous multi-enzyme containing phytase during the finisher period (p<0.05). The birds fed diets with exogenous multi-enzyme containing phytase had a significantly greater digestibility of dry matter, gross energy, crude protein, calcium, and phosphorus compared with birds fed non-supplemented diets (p<0.05). The chickens fed diets with exogenous multi-enzyme containing phytase showed a higher concentration of Ca and P in the serum (p<0.05). The population of Lactobacillus spp., Escherichia coli, and Clostridium were not affected in the ileum and cecum of chickens fed enzyme-supplemented diets. The dietary supplemental exogenous multi-enzyme containing phytase showed a significant improvement in villus height, crypt depth, and villus height and crypt depth ratio, compared to basal diets or dietary supplemental phytase (p<0.05). Conclusion: The supplementation of the exogenous multi-enzyme containing phytase synergistically improved the growth performance, nutrients digestibility, and villus height of the small intestine of broiler chickens fed a corn-wheat-soybean meal based diets.

The Effects of Changing Duty Cycle With Electrical Stimulation on Blood Lactate and Plasma Enzyme (전기자극 시 활동주기 형태의 변화가 혈중젖산과 혈장효소에 미치는 영향)

  • By measuring changes in blood lactate and plasma enzyme (CPK, GOT, GPT) with electrical stimulation applied at two duty cycles, this study is intended to look into which type of duty cycle may have more effects on blood lactate and plasma enzyme constituents through animal experiment so as to determine any duty cycle appropriate for electrical treatment. In this study, electrical stimulation was applied to total 20 Korean house rabbits (weight: 3~3.5 kg) by means of an electrical therapeutic apparatus called TS6000 (made in Netherlands) at duty cycle of 50% and 20% respectively for 30 minutes. Here, 5 cc of blood was collected from their carotid artery before stimulation and in 30 minutes after stimulation respectively to carry out biochemical experiment and analysis. As determined through the above experiment, blood lactate rate was increased to 333.07% at 50% duty cycle after experiment and 185.71% at 20% duty cycle after experiment respectively. In both cases, blood lactate rate was significantly increased to higher level after electrical stimulation than before. Moreover, the rate of change in the average of blood lactate rate at both duty cycles also showed significant differences. CPK rate was boosted to 301.82% at 50% duty cycle after experiment and 321.35% at 20% duty cycle after experiment respectively. In both cases, CPK rate was remarkably boosted to higher level after stimulation than before (p<.05). However, there was not any significant difference in the rate of change in average CPK at both duty cycles (p<.05). GOT rate was significantly boosted up to 38.97% at 50% duty cycle after experiment (p<.05), while it was slightly increased to 1.68% at 20% duty cycle after experiment without any significant difference. Rather, GPT rate dropped slightly at both duty cycles after experiment, but there was not any significant difference. Although blood lactate and GOT were relatively less generated at 20% duty cycle after electrical stimulation than at 50% duty cycle, the change of duty cycle didn't have any significant influence on CPK rate. In this regard, this study failed to come any consistent conclusion about the association between change of duty cycle and muscle fatigue. Therefore, it is advisable that follow-up studies seek various ways to a little more effectively apply electrical stimulation to laboratory animals by avoiding their muscle fatigue. GOT rate was significantly boosted up to 38.97% at 50% duty cycle after experiment (p<.05), while it was slightly increased to 1.68% at 20% duty cycle after experiment without any significant difference. Rather, GPT rate dropped slightly at both duty cycles after experiment, but there was not any significant difference. Although blood lactate and GOT were relatively less generated at 20% duty cycle after electrical stimulation than at 50% duty cycle, the change of duty cycle didn't have any significant influence on CPK rate. Effects of soybean supplementation on blood glucose, plasma lipid levels, and erythrocyte antioxidant enzyme activity in type 2 diabetes mellitus patients

  • The purpose of this study was to investigate the effect of soybean on blood glucose and lipid concentrations, and antioxidant enzyme activity in type 2 diabetes mellitus(DM) patients. We divided patients into two groups and fed them, respectively, a basal diet(control group) and a basal diet with 69 g/d of soybean(soybean group) for 4 weeks. Pills with roasted soybean powder were provided to the soybean supplementation group three times a day. Macronutrients intake except dietary fiber was similar between the two groups. No significant differences were observed in dietary intakes or body weight before and after the supplementation. Energy composition ratio of C:F:P was 65:19:16 in the control group, 64:20:16 in the soybean group. The blood parameters of subjects before supplementation, such as fasting blood glucose, postprandial glucose level, total cholesterol, triglyceride, LDL-cholesterol and $HbA_1C$ were not different between the two groups. After supplementation, fasting blood glucose(p<0.001), postprandial glucose level(p<0.001) and serum triglyceride level(p<0.05) were significantly reduced in the soybean group in comparison with the control group. The total cholesterol level was not significantly different between the control and the supplemented group after 4 weeks of treatment. TBARS levels of the soybean group were not significantly different from those of the control group. The activities of catalase(p<0.01) and glutathione peroxidase(p<0.05) were significantly higher in the soybean group compared to the control group. The results of this study suggest that soybean supplementation would be helpful to control blood glucose and serum lipid in diabetic patients. Also, soybean showed an antioxidant activity that may contribute to enhance the effect of antioxidant defense. This activity contributes to protection against oxidative damage in type 2 DM patients. Soybean may have potential use in the disease management of patients with DM.

Detection of PSS Gene through Genomic DNA of Umbilical Cord Blood by PCR-RFLP in Piglets (자돈의 제대혈 Genomic DNA를 이용한 PSS 유전자 검색)

  • This study was carried out to find out PSS(Porcine Stress Syndrome) with the PSE(Pale, Soft, Exudative) in different piglets. These experiments were accomplished with the aid of PCR-RFLP(Polymerase Chain Reaction-Restriction Fragment Length Polymorphism). The samples were collected and examined from umbilical cord blood of piglets of Yorkshire, Landrace and Crossbred. And then, the PCR products were digested by restriction enzyme, Hha I. The results obtained were as follows; The PCR products of the blood genomic DNA of ryanodine receptor gene were length of 1 .8kb in umbilical cord blood. Normal type(NN), heterozygous type(Nn) and recessively homozygous type(nn, PSS) as a result of digestion of restriction enzyme, Hha 1, were 90.0%, 10.0% and 0.0% in Yorkshire piglets, 76.2%, 19.0% and 4.8% in Landrace, 69.1%, 23.8% and 7.1% in crossbred, respectively. As already showing the above results, the blood from piglets umbilical cord can be availably used for the determination of genotypes of PSS because of easiness of blood collection without stress in live piglets.

  • Angiotensin converting enzyme (ACE) inhibitors were isolated and purified from pig blood plasma. Pig blood plasma was obtained after removing blood cells by centrfugation, followed by the addition of anticoagulant to whole pig blood. To precipitate plasma proteins, pig blood plasma was treated with 4% trichloroacetic acid (TCA) as a final concentration. ACE inhibitors were isolated from plasma protein hydrolysates and TCA supernatant, using ultrafiltration, gel permeation chromatography, and reverse-phase high pressure liquid chromatography. ACE inhibitors isolated from plasma hydrolysates and TCA supernatant had $IC_{50}$ values of $23\;{\mu}M$ and $2\;{\mu}M$, pentapeptide, respectively.

Comparison of Blood Metabolites and Enzyme Activities at Different Slaughter Ages of Hanwoo Cattle

  • This study was conducted to investigate changes of blood metabolites and enzyme activities at different slaughter ages of Hanwoo cattle. Blood samples were taken from six hundred thirty two steers (24~33 months of age) and one hundred twenty eight bulls (17~24 months of age) by venipuncture from caudal vein immediately prior to slaughter. Glucose concentrations were linearly increased (P<0.001) with increases of the slaughter age in steers, and blood urea nitrogen (BUN) concentrations were linearly decreased (P<0.001) with increases of the slaughter age in steers. Likewise, glucose and albumin concentrations in bulls were linearly decreased (P=0.009 and P<0.001, respectively) with increases of the slaughter age. BUN, cholesterol, triglycerides, and total cholesterol concentrations were linearly increased (P<0.001, 0.018 and 0.002, respectively), and creatinine concentrations were quadratically increased (P=0.009) with increases of the slaughter age. Activity of gamma glutamyltransferase (${\gamma}$-GT) and aspartate aminotransferase (AST) was linearly increased (P<0.001) with increases of the slaughter age in both steers and bulls, whereas alanine aminotransferase (ALT) was not affected by increasing slaughter age. In addition, the concentrations of ${\gamma}$-GT and AST were higher (P<0.05) for bulls than those of steers at 24 months of age. Increased levels of ${\gamma}$-GT and AST indicate impaired liver function; it may be associated with increases of concentrate level which is a concomitant of the extended fattening periods in Hanwoo feeding. In conclusion, the results in the present study may be one of the useful information for diagnosis of the metabolic disorder in Hanwoo cattle.

Pregnancy Diagnosis in Sows by Using an On-Farm Blood Progesterone Test

  • To improve animal production, a simple and accurate pregnancy diagnosis plays a very important role. Therefore, the purpose of this study was to develop an on-farm blood progesterone enzyme immunoassay (EIA) system for monitoring the early pregnancy in sows. Star tubes coated with mouse monoclonal anti-progesterone antibody were used for this proposed EIA system which was tested in field trials. The results could be obtained within 30 minutes either by spectrophotometry or the naked eye. Heparinized fresh blood samples collected from the ear vein of sows 17-22 days after breeding (day 0) were tested qualitatively to diagnose sows as pregnant or non-pregnant with high ( > 3 ng/ml) or low ($${{\leq_-}}3ng/ml$$) progesterone in the blood. To provided a double check data, plasma progesterone levels were also measured quantitatively by the same EIA system with some modification. Total agreement of diagnosis by the on-farm EIA kit and by farrowing or abortion from 128 tested sows was found to be 92.2% accuracy (93.1% on pregnant diagnosis and 83.3% on non-pregnant diagnosis). It was concluded that the on-farm EIA blood progesterone test is a very useful method for monitoring the early pregnancy status of sows.

Effect of Supplementation of Dietary Sea Tangle on the Renal Oxidative Stress in Diabetic Rats (식이 다시마의 섭취가 당뇨 쥐 신장의 산화적 스트레스 및 당뇨성 병변에 미치는 영향)

  • Diabetic nephropathy has been increasing, although blood glucose and blood pressure can be controlled by angiotensin converting enzyme(ACE) or advanced glycosylation end products(AGE) inhibitors in the diabetic patients. We investigated the effect of dietary supplementation of sea tangle on the blood glucose, and pathological scoring of diabetic kidneys in the streptozotocin(STZ) induced diabetic rats. Male Sprague-Dawley rats were divided into normal rats fed control diet and diabetic rats fed control diet or control diet supplemented with powder or oater extract of sea tangle. Diabetes was induced by a single injection of STZ(60mg/kg, ip) in citrate buffer. The animals were fed the experimental diet and water for 13 weeks. Dietary supplementation of sea tangle decreased blood glucose in the diabetic rats. However, dietary supplementation of sea tangle did not affect the antioxidant enzyme activities, MDA content and pathology of diabetic kidneys. These results indicate that decreased blood glucose by sea tangle could not delay the progression of diabetic kidney disease.
