Changes in Poly ADP Ribose Polymerase Immune Response Cells of Cerebral Ischaemia Induced Rat by Transcranial Magnetic Stimulation of Alternating Current Approach

  • This study examined effect of a transcranial magnetic stimulation device with a commercial-frequency approach on the neuronal cell death caused ischemia. For a simple transcranial magnetic stimulation device, the experiment was conducted on an ischemia induced rat by transcranial magnetic stimulation of a commercial-frequency approach, controlling the firing angle using a Triac power device. The transcranial magnetic stimulation device was controlled at a voltage of 220 V 60 Hz and the trigger of the Triac gate was varied from $45^{\circ}$ up to $135^{\circ}$. Cerebral ischemia was caused by ligating the common carotid artery of male SD rats and reperfusion was performed again to blood after 5 minutes. Protein Expression was examined by Western blotting and the immune response cells reacting to the antibodies of Poly ADP ribose polymerase in the cerebral nerve cells. As a result, for the immune response cells of Poly ADP ribose polymerase related to necrosis, the transcranial magnetic stimulation device suppressed necrosis and had a protective effect on nerve cells. The effect was greatest within 12 hours after ischemia. Therefore, it is believed that in the case of brain damage caused by ischemia, the function of brain cells can be restored and the impairment can be improved by the application of transcranial magnetic stimulation.

Pain Relieving Effect of Sucrose Coating Pacifier in Neonates (신생아 통증완화를 위한 자당 코팅 노리개 젖꼭지의 적용 효과)

  • Purpose: This study is to assess and compare the analgegic effects of $25\%$ sucrose coating pacifier and pacifier. Method: The participants are 75 healthy infants of neonatal age 1-7 days and randomized to receive heel prick before 2 minutes to blood sampling and physiological test in Nursery of A Medical Center from 24. January to 28, February, 2003. The experimental group assigned to one of three treatment groups: no treatment, a pacifier, $25\%$ sucrose coating pacifier. Collected data were analyzed with the SPSS 11.0 program using $x^2$-test, one-way ANOVA and Scheffe, repeated mesured ANOVA. Results: The pain score of $25\%$ sucrose coating pacifier is lower than no treatment group and pacifier group. In heart rate, there were statistical significant differences between three groups. In repiratory rate, there were no statistical significant differences between three groups. In $SaO_2$, there were statistical significant difference between three groups. Conclusion: The $25\%$ sucrose coating pacifier showed pain relief effect in behavior responses and heart rate and $SaO_2$. Accordingly, the sucrose coating pacifier should be applied nursing intervention for simple pain management as heel prick.

  • Natural mineral water is generally quite different from ordinary drinking water due to its original nature and various properties. The complexity of natural mineral water requires, therefore, not only to identify its nature and proper characteristics, but also to classify them by a reasonable scientific basis of comparison. The study was concentrated on a possible classification technique to natural mineral waters by their constitutions and physico-ehemical properties. The classification was carried out by the computation of such numerical parameters as ionic equivalent percentage, electrolytic conductance or mobility, ionic molecular weight, molecular concentration, equivalent conductivity and degree of ionization in consideration of the determinative criteria as follows -particular single element or molecule -major components of natural waters as bicarbonate, sulphate, chloride,caloride, calcium, magnesium, and sodium -moleculat concentration related to blood osmotic pressure -water temperature at emergence from spring -contents of free carbon dioxide (CO2) -pH value of water -total dissolved solids or salts (NaCl) The results obtained proved out to be clearly distinguhhable from ordinary drinking water as far as concern natural mineral water as an example on the subject -simple water -bicarbonate-predominating water -cold spring -carbonated-non gaseous water -weak alkaline water -non saline water Putting these various results together, the sample turned out to be a kind of natural mineral water that can be used as a drinking water if microbiologically safe.

Evaluation of DNA Extraction Methods from Low Copy Number (LCN) DNA Samples for Forensic DNA Typing

  • DNA isolation for PCR-based short tandem repeat (STR) analysis is essential to recover high yields of amplifiable DNA from low copy number (LCN) DNA samples. There are different methods developed for DNA extraction from the small bloodstain and gloves, commonly found at crime scenes. In order to obtain STR profiles from LCN DNA samples, DNA extraction protocols, namely the automated $iPrep^{TM}$ $ChargeSwitch^{(R)}$ method, the automated $QIAcube^{TM}$ method, the automated $Maxwell^{(R)}$ 16 DNA $IQ^{TM}$ Resin method, and the manual $QIAamp^{(R)}$ DNA Micro Kit method, were evaluated. Extracted DNA was quantified by the $Quantifiler^{TM}$ Human DNA Quantification Kit and DNA profiled by $AmpFISTR^{(R)}$ $Identifiler^{(R)}$ Kit. Results were compared based on the amount of DNA obtained and the completeness of the STR profiles produced. The automated $iPrep^{TM}$ $ChargeSwitch^{(R)}$ and $QIAcube^{TM}$ methoas produced reproducible DNA of sufficient quantity and quality trom the dried blood spot. This two automated methods showed a quantity and quality comparable to those of the forensic manual standard protocols normally used in our laboratory. In our hands, the automated DNA extraction method is another obvious choice when the forensic case sample available is bloodstain. The findings of this study indicate that the manual simple modified $QIAamp^{(R)}$ DNA Micro Kit method is best method to recover high yields of amplifiable DNA from the numerous potential sources of LCN DNA samples.

Gastric Hemangioma Treated with Argon Plasma Coagulation in a Newborn Infant

  • Gastric hemangioma in the neonatal period is a very rare cause of upper gastrointestinal bleeding. We present a case of hemangioma limited to the gastric cavity in a 10-day-old infant. A huge, erythematous mass with bleeding was observed on the lesser curvature side of the upper part of the stomach. Surgical resection was ruled out because the location of the lesion was too close to the gastroesophageal junction. Medical treatment with intravenous $H_2$ blockers, octreotide, packed red blood cell infusions, local epinephrine injection at the lesion site, application of hemoclip, and gel-form embolization of the left gastric artery did not significantly alter the transfusion requirement. Hemostasis was achieved with endoscopic argon plasma coagulation (APC). After two sessions of APC, complete removal of the lesion was achieved. APC was a simple, safe and effective tool for hemostasis and the ablation of gastric hemangioma without significant complications.

Lateral Supramalleolar Flap for Reconstruction of Soft Tissue Defect around the Ankle Joint

  • Purpose: Soft tissue defect on foot and ankle is vulnerable and requires a thin flap for improvement of aesthetic and functional results. Lateral supramalleolar flap is a simple and fast procedure, which can preserve and supply reliable constant blood flow, and causes fewer donor site complications. The authors reviewed our cases and report the clinical results. Materials and Methods: Ten cases of soft tissue defects on the lower leg, around the ankle were treated with lateral supramalleolar flap. There were seven males and three females with a mean age of 54.8 years. The mean size of flaps was $5.9{\times}6.3$ cm and the mean follow-up period was 23 months. Flap survival and postoperative complications were evaluated. Results: Nine flaps survived completely without loss of flap. There was one case of partial wound dehiscence requiring debridement and repair, and another case of necrotic flap change requiring partial bone resection and closure. All patients were capable of weight bearing ambulation at the last follow up. Conclusion: The authors suggest that the lateral supramalleolar flap could be a useful option for treatment of soft tissue defect around the ankle joint.

Induction of Resveratrol Biosynthesis in Grape Skins and Leaves by Ultrasonication Treatment

  • Grapes (Vitis vinifera) are one of the most important fruits worldwide and are eaten raw or after conversion to jelly, jam, juice and wine. Grape skins are a major source of resveratrol (3,5,4'-trihydroxystilbene), which has the ability to reduce blood sugar as well as anticancer, anti-inflammatory, and other beneficial cardiovascular effects. In this study, we investigated the increased accumulation of resveratrol in grape skin and leaves following ultrasonication treatment, which has been shown to induce resveratrol accumulation in several plants. Various ultrasonication treatment times and incubation periods were employed to identify the optimum conditions for the maximum accumulation of resveratrol. Treatment and further incubation led to increased resveratrol in both grape skins and leaves, with the highest increases of 7.7-fold and 1.9-fold occurring in response to 5 min ultrasonication treatment followed by 6 hour incubation and 15 min ultrasonication treatment followed by 3 hour incubation, respectively. The underlying mechanism for the increased amounts of resveratrol were studied by employing a semi-quantitative RT-PCR to monitor the expression levels of the resveratrol synthase (RS) gene in response to ultrasonication treatment. The RS gene increased the expression in response to ultrasonication treatment, suggesting that up-regulation of the RS gene by ultrasonication treatment triggers increased amounts of resveratrol. Taken together, these data indicate that this simple ultrasonication treatment of grapes can be an efficient post-harvest technology for increasing resveratrol in grape skins in addition to cleaning the fruits.

Cytological diagnosis of adenocarcinoma using the mammary gland secretion from a dog (개의 유선분비물을 이용한 유선암종의 세포학적 진단)

  • A nine-year-old female Yorkshire terrier dog showing mammary secretion mixed with blood and pus for 5 month duration was presented. Cytologically mammary secretion consisted of many pleomorphic tumor cells, RBC, and neutrophil. Tumor cells were characterized by polyhedral pleomorphic nuclei with smudged chromatin and basophilic cytoplasm with many secretory vacuoles. N/C ratio was very high. Therefore it was diagnosed as mammary gland adenocarcinoma. Histological examination confirmed cytological diagnosis. From these results cytology isfor the diagnosis of secreting mammary gland adenocarcinoma is very simple and accurate for the diagnosis of secreting mammary gland adenocarcinoma.

Trido Mitral Valve Replacement with Dacron Collar Prosthetic Valve due to Paravalvular Leak (판막주위 누출에 대한 다크론 확장 인공판막을 이용한 판막치환술 치험)

  • The paravalvular leak after redo mitral prosthetic valve replacement is rare but serious complication when it does happen. This condition should be corrected surgically to increase life span and improve symptoms. But simple closure or patch closure of paravalvular leak are not effective in cases of weak annulus or broad defect. We report 3 cases of trido mitral valve replacement using mechanical valve with its sewing ring expanded by a collar of Dacron sheet. The prosthetic sewing ring is anchored on the weak mitral annulus with multiple interrupted sutures, while the Dacron collar is contineuously sutured to the left atrial wall for blood tight sealing. All of the three cases showed event free postoperative course.

  • Pulmonary Aspergillomas usually arise from proliferation of Aspergillus in preexisting parenchymal cavities.202 college students (99 men, 103 women) aged 18 to 26 years. Fasting blood samples were. The most common symptom in this disorder is hemoptysis, which may be minimal in amount or it may be massive & life threatening. The optimum therapy for pulmonary aspergilloma is controversial. The major options available include surgical resection of the lesion, a number of medical therapies, or simple observation of the patient for a time. Surgery is the most effective treatment but it is limited to some patient because most patients have underlying pulmonary disease. Thus the various form of medical therapy was available with successful result. The authors present a case of percutaneous intracavitary amphoterician B injection for the treatment of pulmonary aspergilloma & its successful effect for the repetitive hemoptysis.
