  • A laser speckle is a random pattern that has a granular appearance produced by reflected light when a coherent laser illuminates an irregular course surface. Laser speckle system has many advantages. It can detect some animals functional parts. Moreover, it relatively consists of simple and in-expensive system. It is very important that detecting micro-vessels through image processed image. Current study is to improve image quality through variable image processing method. But this paper made laser speckle endoscope for miniaturization and commercialization laser speckle system. We had endoscope test through goldfish's tail. We will compare the processed speckle image and halogen image.

Serum neuron specific enolase is increased in pediatric acute encephalitis syndrome

  • Purpose: This study aimed to investigate whether serum neuron-specific enolase (NSE) was expressed in acute encephalitis syndrome (AES) that causes neuronal damage in children. Methods: This prospective observational study was conducted in the pediatric neurology ward of Soetomo Hospital. Cases of AES with ages ranging from 1 month to 12 years were included. Cases that were categorized as simple and complex febrile seizures constituted the non-AES group. Blood was collected for the measurement of NSE within 24 hours of hemodynamic stabilization. The median NSE values of both groups were compared by using the Mann-Whitney U test. All statistical analyses were performed with SPSS version 12 for Windows. Results: In the study period, 30 patients were enrolled. Glasgow Coma Scale mostly decreased in the AES group by about 40% in the level ${\leq}8$. All patients in the AES group suffered from status epilepticus and 46.67% of them had body temperature >$40^{\circ}C$. Most of the cases in the AES group had longer duration of stay in the hospital. The median serum NSE level in the AES group was 157.86 ng/mL, and this value was significantly higher than that of the non-AES group (10.96 ng/mL; P<0.05). Conclusion: AES cases showed higher levels of serum NSE. These results indicate that serum NSE is a good indicator of neuronal brain injury.

  • Although rheumatoid arthritis (RA) is the most common chronic inflammatory autoimmune disease, diagnosis of RA is currently based on clinical manifestations, and there is no simple, practical assessment tool in the clinical field to assess disease activity and severity. Recently, there has been increasing interest in the discovery of new diagnostic RA biomarkers that can assist in evaluating disease activity, severity, and treatment response. Proteomics, the large-scale study of the proteome, has emerged as a powerful technique for protein identification and characterization. For the past 10 years, proteomic techniques have been applied to different biological samples (synovial tissue/fluid, blood, and urine) from RA patients and experimental animal models. In this review, we summarize the current state of the application of proteomics in RA and its importance in identifying biomarkers and treatment targets.

Micro Cell Counter Using a Fixed Control Volume Between Double Electrical Sensing Zones (다수의 계수구역간의 검사체적을 이용한 소형 세포농도센서)

  • We present a novel flow-rate independent cell counter using a fixed control volume between double electrical sensing zones. The previous device based on the single electrical cell sensing in a given flow-rate requires an accurate fluid volume measurement or precision flow rate control. The present cell counter, however, offers the flow-rate independent method for the cell concentration measurement with counting cells in a fixed control volume of $22.9{\pm}0.98{\mu}{\ell}$. In the experimental study, using the RBC (Red Blood Cell), we have compared the measured RBC concentrations from the fabricated devices with those from Hemacytometer. The previous and present devices show the maximum errors of $20.3\%\;and\;16.1\%$, which are in the measurement error range of Hemacytometer (about $20\%$). The present device also shows the flow-rate independent performance at the constant flow-rates ($5{\mu}{\ell}/min$ and $10{\mu}{\ell}/min$) and the varying flow-rate (4, 2, and $4{\mu}{\ell}/min$). Therefore, we demonstrate that the present cell counter is a simple and automated method for the cell concentration measurement without requiring an accurate fluid measurement and precision flow-rate control.

A Comparative Study of Antegrade Cardioplegia Versus Retrograde Cardioplegia for Myocardial Protection during the Open Heart Surgery (순행성 관관류법과 역행성 관관류법의 임상적 비교연구)

  • During aortic valve surgery, cardioplegic solution is delivered through direct cannulation of both coronary ostia. Since this approach may cause an intimal injury leading to acute dissection or late ostial stenosis, this study was undertaken to evaluate myocardial protective effect of retrograde perfusion of cardioplegia [RCSP <% RRAP] in 18 clinical cases, which were compared with antegrade perfusion of cardioplegia in 27 clinical cases. This study were investigated 1] cease and return of electromechanical activity after cardioplegia infusion 2] the myocardial temperature during operation 3] the aortic cross clamping time and total bypass time 4] frequency of DC shock for defibrillation 5] need for inotropic drugs after operation 6] electrocardiographic evidence of myocardial infarction or ventricular arrhythmia after operation 7] the enzymes activity during preoperative and postoperative period as an evaluation of myocardial ischemic injury and 8] operative mortality rate The combination of retrograde cardioplegia and topical cooling with ice slush yielded promptly hypothermia of myocardium and shorter aortic cross-clamping time compared with antegrade cardioplegia [P < 0.05]. The temperature of the interventricular septum was maintained below 20oC by continuous perfusion or intermittent perfusion of cold blood cardioplegia and other results were no statistically significant difference between the two methods [P >0.05]. This technique provides clear operative field and avoids some serious complications which are caused by coronary ostial cannulation. These results suggested that the retrograde perfusion of cardioplegia is a simple, safe, and effective means of myocardial protection during open heart surgery.

  • The effect of prior cumulative mixing was evaluated by means of a simple regression model which used as a dependent variable percentage(%) change of each repeated observed value for the initial value. The red blood cell count (RBC), hematocrit (Hct), and mean corpuscular volume(MCV) showed a significant decrease (p<.01), but % of basophils had a significant increase (p<.05). The ratios of rate of change (regression coefficient/SD) were .0012 for RBC, .0020 for Hct, .0024 for MCV, and .0011 for % of basophils. Of the 19 parameters evaluated, 21% changed.

  • The purpose of this study was to determine the effects of laser Irradiation on Human plasma $\beta-endorphin$ levels, by treating with low level helium-neon (He-Ne) and Infrared(lR) laser. The Laser was fixed frequency of 2400Hz by continuous scanning and irradiating time was 8 minutes each point. Blood samples were taken at before, after, after 15min's treatment and Plasma $\beta-endorphin$ was measured by radioimmunoassay. The samples for this study were 6 normal subjects(3male, 3female). The data were analyzed by paired t-test, one-way ANOVA and simple regression. The results of this study were as follows : 1. The human plasma $\beta-endorphin$ levels were noted as significant increase in after-treatment $(22.84{\pm}10.63pg/ml)$ as compared with before-treatment $(16.96{\pm}9.23pg/ml)$ and significant increase in after 15min's $(27.27{\pm}8.81pg/ml)$ as compared with after-treatment (p<0.05). 2. There were no significant changes in plasma g-endorphin levels between male and female. 3. The human plasma $\beta-endorphin$ levels were high associated in between session reliability (p<0.05).

Direct Detection of Shigella flexneri and Salmonella typhimurium in Human Feces by Real-Time PCR

  • We have established a SYBR Green-based realtime PCR method using AnyDirect solution, which enhances PCR from whole blood, for direct amplification of the virA gene of Shigella flexneri and the invA gene of Salmonella typhimurium from human feces without prior DNA purification. When we compared the efficiency of conventional or realtime PCR amplification of the virA and invA genes from the supernatant of boiled feces supplemented with S. flexneri and S. typhimurium in the presence or absence of AnyDirect solution, amplification products were detected only in reactions to which AnyDirect solution had been added. The detection limit of real-time PCR was $1{\times}10^4\;CFU/g$ feces for S. flexneri and $2{\times}10^4\;CFU/g$ feces for S. typhimurium; this sensitivity level was comparable to other studies. Our real-time PCR assay with AnyDirect solution is simple, rapid, sensitive, and specific, and allows simultaneous detection of S. flexneri and S. typhimurium directly from fecal samples without prior DNA purification.

Real-Time Monitoring of Catheter-Related Biofilm Infection in Mice

  • This study was done to establish a mouse model for catheter-related biofilm infection suitable to bioluminescence imaging (BLI). Biofilm formation of Pseudomonas aeruginosa (P. aeruginosa) Xen5 grown on catheter disks in vitro and in an implanted mouse model was real-time monitored during a 7-day study period using BLI. The numbers of integrated brightness (IB) and viable bacterial count (VBC) in the biofilm disks in vitro were highest at 24 h after inoculation; the IB of biofilm in vivo was increased until 24 h after implantation. A statistical correlation was observed between IB and VBC in vitro by linear regression analysis. The actual VBC value in vivo can be estimated accurately by IB without sacrifice. In addition, we monitored the change in white blood cells (WBCs) during infection. The number of WBCs on day 7 was significantly higher in the infection group than in the control group. This study indicates that BLI is a simple, fast, and sensitive method to measure catheter biofilm infection in mice.

Experimental Studies on the Acupuncture Prescription for Electroacupuncture Analgesia of the Cattle (소의 전침마취에 적용하는 경혈배합에 관한 실험적연구)

  • In order to develop the effective methods of electroacupuncture anesthesia to bovine species. eight animals of Korean native cow, which are healthy and have ordinary perception response. were examined to study on the acupuncture prescription. In the present study, six kinds of acupuncture prescription methods were applied with four kinds of meridian points which consist of Sam Yang Rack, Keuk Moon, Boo Yang and Sam Eum Kyo. The results are summarized as follows. 1. All kinds of methods applied here were simple in needling technique but considerably effective on analgesia. It was also confirmed that the methods could be applied to the all clinical signs, because these were not depended to vary on the position of operation area and the body restrain. 2. The results, which were obtained by applying to all meridian points at both legs selected by diagonal lines(e. g., right foreleg and left hind leg, and vice versa) and to all meridian points at the all legs, were revealed the same analgesia effect. However, the result applied to meridian points at one side of body did not show the good effect for analgesia. 3. When the acupuncture prescription was applied to Sam Yang Rack and Keuk Moon at both forelegs, and Boo Yang and Sam Eum Kyo at both hind-legs, the good effects of regional analgesia appeared at the anterior body and both forelegs, and the posterior body and both hind-legs, respectivery. 4. There was no sognificant difference in changing biochemical pictures of blood and serum during the experiment.
