Measurement of red blood cell aggregation by analysis of light transmission in a pressure-driven slit flow system

  • The aggregation characteristics of red blood cells (RBCs) were measured using a newly developed light-transmission slit rheometer. Conventional methods of RBC disaggregation such as the rotational Couette system were replaced with a pressure-driven slit flow system with a vibrational mechanism. Using a vibration generator, one can disaggregate the RBC aggregates stored in the slit. While shear stress decreases exponentially, instantaneous pressure and the transmitted light intensity were measured over time. Applying an abrupt shearing flow after disaggregation caused a rapid elongation of the RBCs followed by loss of elongation with the decreasing shear stress. While the shear stress is further decreasing, the RBCs start to re-aggregate and the corresponding transmitted intensity increases with time, from which the aggregation indices can be obtained using a curve-fitting program.

  • In this study, we attempted to quantify the relative importance of assumptions regarding blood rheology. Three patient-specific carotid bifurcation geometries and time-varying flow rates were obtained using magnetic resonance imaging. For each subject, CFD simulations were carried out assuming two different non-Newtonian rheology models Carreau and Ballyk models) and rescaled Newtonian viscosities based on characteristic shear rates to account for the shear-thinning property of blood. The sensitivity of WSS and oscillatory shear index (OSI) were contextualized with respect to the reproducibility of the reconstructed geometry and to assumptions regarding the inlet boundary conditions. We conclude that the assumption of Newtonian fluid is reasonable for studies aimed at quantifying the distribution of WSS-based extrema in an image-based CFD model of carotid bifurcation.

Numerical study of the effects of periodic body acceleration (PGZ) and bifurcation angle in the stenosed artery bifurcation

  • This article describes the numerical investigation of blood flow in the stenosed artery bifurcation with acceleration of the human body. Using the commercial software FLUENT, three-dimensional analyses were performed for six simulation cases with different body accelerations and bifurcation angles. The blood flow was considered to be pulsation flow, and the blood was considered to be a non-Newtonian fluid based on the Carreau viscosity model. In order to consider periodic body acceleration, a modified, time-dependent, gravitational-force term was used in the momentum equation. As a result, flow variables, such as flow rate and wall shear stress, increase with body acceleration and decrease with bifurcation angle. High values of body acceleration generate back flow during the diastolic period, which increases flow fluctuation and the oscillatory shear index at the stenosis.

A noble RBC aggregometer with vibration-induced disaggregation mechanism

  • The aggregation of red blood cells (RBCs) is a major determinant of blood flow resistance passing through various veins. Available techniques for measuring RBC aggregation often require pretreating and washing after each measurement, which is not optimal for day-to-day clinical use. A laser reflection technique has been combined with a vibration-aided disaggregation mechanism, which shows significant advances in aggregometer design, operation and data analysis. The essential features of this design are in its simplicity and a disposable element that is in contact with the blood sample. Using extremely small quantities of blood, the RBCs subjected to vibrations can be quickly and completely disaggregated. This is followed by measuring the backscattered light intensity. The measurements with the present sensor were compared with those of a commercial aggregometer and a strong correlation was found between them. The newly-developed optical aggregometer can measure the RBC aggregability difference between young and old cell suspension with ease and accuracy.

Dynamic response of heat and mass transfer in blood flow through stenosed bifurcated arteries

  • The present study deals with a mathematical model describing the dynamic response of heat and mass transfer in blood flow through bifurcated arteries under stenotic condition. The geometry of the bifurcated arterial segment possessing constrictions in both the parent and the daughter arterial lumen frequently appearing in the diseased arteries causing malfunction of the cardiovascular system, is formulated mathematically with the introduction of the suitable curvatures at the lateral junction and the flow divider. The blood flowing through the artery is treated to be Newtonian. The nonlinear unsteady flow phenomena is governed by the Navier-Stokes equations while those of heat and mass transfer are controlled by the heat conduction and the convection-diffusion equations respectively. All these equations together with the appropriate boundary conditions describing the present biomechanical problem following the radial coordinate transformation are solved numerically by adopting finite difference technique. The respective profiles of the flow field, the temperature and the concentration and their distributions as well are obtained. The influences of the stenosis, the arterial wall motion and the unsteady behaviour of the system in terms of the heat and mass transfer on the blood stream in the entire arterial segment are high­lighted through several plots presented at the end of the paper in order to illustrate the applicability of the present model under study.

A Comparative Study on Constitutive Equations of Human Blood (인체혈액의 구성방정식에 관한 비교연구)

  • 인체 동맥혈관내 혈액의 유동현상을 수치적으로 해석하기 위해서는 혈액의 유변학 적 성질을 구성방정식으로 나타내어야한다. 본 연구에서는 혈액의 점성계수를 표현하기 위 하여 비뉴턴유체의 점성을 나타내는 식으로서 Carreau 모델, 수정 Cross 모델, 수정 Powell-Eyring 모델과 수정멱법칙모델을 사용하였고 원형관내 혈액의 정상유동을 수치모사 하기 위하여 겉보기점성계수를 이용하는 구성방정식을 운동량방정식에 적용하였다. Carreau 모델과 수정멱법칙모델을 적용할 때 레이놀즈수의 변화가 중심선상의 속도와 길이방향의 압 력변화에 미치는 영향을 고찰하였다. 전단율이 높은영역에서 혈액의 겉보기점성계수를 효과 적으로 나타낼수 있는 수정멱법칙모델을 제시하였다.

Steady Flow Analyses of Blood and the Blood Analogue Fluids in the Stenosed Circular and bifurcated Tubes (협착이 발생된 원관과 분기관내 혈액과 혈액대용유체의 정상유동해석)

  • 본 논문의 목적은 협착이 발생괸 원관과 분기관내 혈액과 혈액대용유체의 유동문제 에 수치해석방법을 적용하여 유동특성을 파악하는데 있다. 혈액대용유체로서는 Separan AP-273 500wppm 수용액과 Carpobol 934 1.0w% 수용액을 사용하였다. Carbopol 수용액의 유변학적 성질은 수정멱법칙모델, 그리고 혈액과 Separan 수용액의 유변학적 성질은 Carreau 모델로 나타내었다. 협착관유동에서 Carbopol 수용액의 재부착거리는 혈액이나 Separan 수용액의 경우보다 길고 협착으로 인한 압력강하는 Carbopol 수용액, 혈액, Separan 수용액의 순으로 작게 나타난다. 분기관유동에서 Separan 수용액의 압력손실은 혈 액과 Carbopol 수용액보다 작게 나탄나고 협착이 발생괸 부기관내에서 혈액과 Separan 수 용액의 압력손실은 협착이 없는 분기관의 압력손실보다 크게 증가한다.

Microchip-based cell aggregometer using stirring-disaggregation mechanism

  • A new microchip-based aggregometer that uses a stirring-aided disaggregation mechanism in a microchip was developed to measure red blood cell (RBC) aggregation in blood and RBC suspensions. Conventional methods of RBC disaggregation, such as the rotational Couette system, were replaced with a newly designed stirring-induced disaggregation mechanism. Using a stirrer in a microchip, the aggregated RBCs stored in a microchip can be easily disaggregated. With an abrupt halt of the stirring, the backscattered light intensity can be measured in a microchip with respect to time. The time recording of the backscattered light intensity (syllectogram) shows an exponential decreasing curve representing the RBC aggregation. By analyzing the syllectogram, aggregation indices such as AI and M were determined. The results showed excellent agreement with LORCA. The essential feature of this design is the incorporation of a disposable microchip and the stirring-induced disaggregation mechanism.
