Tea polysaccharide had high antioxidant activity and it could be used to cure diabetes. Antioxidant activity of tea poly-saccharide (TPS) from three kinds of tea (green tea, oolong tea and black tea) were compared, the result indicated that oolong tea polysaccharide (OTPS) had the highest antioxidant activity. In order to explicate the mechanism of antioxidant and hypoglycemic activity, the streptozotocin (STZ)-induced diabetes mice model (DM) was established. The influence of OTPS on blood-glucose, content of MDA and NO, and activities of GSH-PX, SOD, NOS in serum, kidney and liver were investigated. The result showed that after four weeks injection of OTPS to DM mice, the blood-glucose of three treatment group reduced by 14.5%,21.5% and 33.3%, respectively, comparing to the model control. The reduction effect of OTPS increased with the rise of dose. The activity of SOD and GSH-PX elevated significantly, while the activity of NOS decreased. The content of MDA and NO reduced significantly. The above results imply that antioxidant activity was enhanced. Comparing to XKW treatment, the effect of a dose of 300mg/(kg. bw) OTPS was much better. The research showed that the OTPS had a significant effect on reducing blood glucose, and could enhance the antioxidant activity of DM mice.
There are growing evidences suggesting a pivotal role of oxidative stress in the pathophysiology of preeclampsia. We investigated oxidative stress in the rat model of preeclampsia, and in clinical cases. Pregnant female rats were injected intraperitoneally with deoxycorticosterone acetate (DOCA) and given 0.9% saline as drinking water during their pregnancy. We assessed plasma $F_2-isoprostane(8-iso-PGF_{2{\alpha})$ and malondialdehyde (MDA) in a rat model, and the same markers in the plasma of maternal blood and fetal cord blood in pregnant women with preclampsia. Blood samples from the umbilical arteries and veins were collected separately. The concentrations of MDA were increased in the preeclampsia groups of animal and humans, compared with the control group; it was significantly increased in the umbilical artery and vein of the preeclampsia group. The concentrations of $F_2-isoprostane$ were elevated in the preeclampsia groups of animal and humans, compared with the control group, and the increase in $F_2-isoprostane$ concentration was prominent in the umbilical vein than umbilical artery of the preeclampsia group. Therefore, it appears that the placenta has an important role in the pathophysiology of preeclampsia, and the $F_2-isoprostane$ of the umbilical vein may serve as a relatively reliable marker for ischemic/hypoxic injury to the fetus during the perinatal period.
The protective effects of Phaleria macrocarpa (PM) against oxidative stress in diabetic rats were investigated. Diabetes was induced in male Sprague Dawley rats using alloxan (150 mg/kg i.p). After the administration of PM fractions for two weeks the diabetic symptoms, nephropathy and renal antioxidant enzymes were evaluated. The results showed that the oral PM treatments reduced blood glucose levels in diabetic rats. The PM fractions decreased kidney hypertrophy and diminished blood urea nitrogen (BUN) in diabetic rats. Malondialdehyde (MDA), a lipid peroxidation marker, was increased in diabetic animals, but was suppressed by the PM treatments. In addition, the superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activities, and glutathione (GSH) level in the alloxan-induced diabetic rats were significantly decreased compared with those in the normal rats, but were restored by PM treatments. The PM fractions also suppressed the level of MDA in the kidney. In conclusion, the anti hyperglycemic and anti-nephropathy of P. macrocarpa may be correlated to the increased renal antioxidant enzyme activity in the kidney.
Effects of oral taurine supplementation (6g/day)for 2-4 weeks on activities of red blood cell(RBC)total superoxide dismutase (SOD) and plasma glutathione peroxidase(GSH-Px) and the level of malondialdehyde(MDA) were evaluated in healthy female adults (23.6$\pm$0.3 years old). Compared to the value for 0 week plasma GSH-Px activity of the subjects was significantly lower after 2 weeks of taurine supplementation(p<0.05) and recovered to the value similar to 0 week after 4 weeks of taurine supplementation. RBC total SOD activity tended to be decreased after 2 weeks of taurine supplmentation compared to the values for 0 week although the difference between the means of the two group was not statistically significant. Plasma MDA level was not significantly decreased by taurine supplementation most probably due to the fact that the subjects participated in the present study were healthy and their antioxidant defense system had been in the 'normal' range. Plasma MDA concentration was negatively correlated with plasma taurine concentration(r=-0.2003m p<0.05) but tended to be positively correlated with plasma cholesterol concentration(r=0.2465, p=0.0645) as expected Plasma GSH-Px activity was positively associated with the percentage of 22:0 (r=0.2892, p<0.05) or 20:4w6(r=0.2939, p<0.05). On the other hand plasma MDA concentration was positively correlated with the percentage of 20:5w3 in plasma total lipids(r=0.2635 p<0.05) and negatively correlated with $\Delta$5 desaturation index of w6 fatty acids(20:3w6⇒20:4w6) in plamsa total lipids(r=-0.2714, p<0.05) as well as in phospholipids(r=-0.2864, p<0.05). From these results protective effect of taurine supplementation against lipid peroxidation and antioxidant defense system in humans appears to be minimal when the subjects are in a relatively healthy state. Further studies concerning the antioxidant efficacy of taurine should be conducted in human subjects under various disease states related to oxidative stress such as diabetes and artheroxclerosis.
Objectives : Alcohol-induced oxidative stress has been known to injure various tissues or organs. This stress is related with free radicals which are produced as the result of long-term alcohol consumption. Malonyldialdehyde(MDA) is produced by the interaction of free radicals and cell membrane lipids, and indicates the degree of lipid peroxidation indirectly. The purpose of this study was to investigate the relationship between red blood cell(RBC) membrane lipid peroxidation by free radicals, and associated hepatic injuries and hematologic changes. Methods : Thirty-three subjects diagnosed as alcohol dependence according to DSM-IV diagnostic criteria were evaluated within 72 hours after discontinuing alcohol drinking. Clinical characteristics were evaluated by CAGE questionnaire and Korean Michigan Alcoholism Screening Test(MAST). RBC membrane MDA level was measured as the marker of RBC membrane lipid peroxidation. Aspartate aminotransferase(AST), alanine aminotransferase(ALT) and gamma-glutamyltransferase(GGT) were used as the biochemical markers of liver damage due to alcohol ingestion. The alcohol-induced hematologic change was assessed by mean corpuscular volume(MCV). Results : The results were as follows. Clinical characteristics were not different between two groups having normal and abnormal levels of AST, ALT, GGT or MCV. The levels of MDA were not correlated with the clinical characteristics and serum levels of AST, ALT and GGT. However, there was a significant correlation between the levels of MDA and the value of MCV(p=0.017). Conclusions : These findings suggest that oxidative stress in alcohol dependence may not be reflected in liver enzyme markers such as AST, ALT and GGT, but may be reflected in MCV.
The antioxidant enzymes, lipid peroxidation and free radicals assessment were made of the effects of selenium, copper and magnesium on bovine endemic fluorosis under high fluoride, low selenium and low copper productive conditions. Thirty-two beef cattle were selected from high fluoride area, and randomly divided into four groups with eight cattle each as follows: (1) high fluoride control group (HFC); (2) supplemented group with 0.25 mg/kg selenium (HFSe); (3) supplemented group with 15 mg/kg copper (HFCu) and (4) supplemented group with 0.25 mg/kg selenium+15 mg/kg copper+1 mg/kg magnesium (HFSeCuMg) per day for 83 days. Moreover, eight beef cattle were selected from non-high fluoride area as normal control group. Blood samples were collected from cattle on 0 d, 30 d and 83 d respectively, to analyze the enzyme activities and concentration of GSH-px, CAT, SOD, MDA and free radicals. The results showed that the contents of free radicals and MDA in HFC group were significantly higher, and the whole blood GSH-px, CAT, erythrocyte SOD activities were lower than the normal control group. Free radicals, metabolic imbalance and antioxidant disorder therefore, play an important role in fluorosis. However, GSH-px, CAT and SOD activities in HFSe group and HFSeCuMg group at 30 d and 83 d were markedly higher than the same groups at the 0 d and the HFC group at the same time. Likewise, there was a corresponding reduction in the contents of free radicals and MDA. These findings indicated that supplementation with selenium, copper and magnesium elevated high fluoride bovine antioxidant enzymes, and decreased MDA and free radicals contents. But, the activities of supplementation selenium group did not increase until day 83. These results demonstrated that fluorosis was associated with lower serum Se and Cu levels than in the control, and it was therefore concluded that fluorosis is associated with decreased serum levels of these minerals. Long-term high fluoride intake under productive condition enhances oxidative stress in the blood, thereby disturbing the antioxidant defense of cattle. Increased oxidative stress could be one of the mediating factors in the pathogenesis of toxic manifestations of fluoride. It is benefical for high fluoride cattle supplemented with proper selenium, copper and magnesium to increase fluoride excretion and obtain the protective impact of the activity of oxidative enzymes, and to decrease lipid peroxidation and free radicals contents.

The purpose of this study is to help a recovery of fatigue through the effect of cold immersion treatment after rowing ergometer on blood fatigue substance, MDA and SOD in rowing athletes. For this, 10 subjects participated in this study and we divided them into cold water immersion group (CWI, n=10), non-cold immersion group (NCI, n=10). The exercise program was performed to 2000 m rowing ergometer for maintaining high intensity. The data was collected with regard to the interaction effect of the group and time among the CWI and NCI, ANOVA was used. As the post-hoc test, Bonferroni test was used. The significance was set at .05 and the following conclusions were deduced. For lactic acid, there were the main effect of time (p<.001) and significant difference in the both group (p<.001). Also, LDH were significant difference in the each group (p<.05). For MDA, there were the interaction between group and time (p<.05) and the main effect of group (p<.05) and time (p<.001). SOD were indicated main effect of group and time (p<.05), there was significant difference between each group in the after 30 min recovery (p<.05). Collectively, The results of this study suggest that positive effect on blood fatigue substances, reactive oxygen species and anti-oxidation enzyme through cold water immersion intervention. Therefore, we strongly recommend that performing the cold immersion intervention would be beneficial after high intervention exercise.
According to the epidemiological studies in chromium workers, hexavalent chromium is associated with the risk of lung cancer. Cellular oxidative damages by reactive oxygen species produced by hexavalent chromium exposure may play an important role in the carcinogenesis process. We investigated the availabilities of malondialdehyde measurement for the assessments of oxidative damages from chromium exposure with an experimental inhalation study in vivo. Lipid peroxidation, one kind of cellular oxidative damage, was measured in blood plasma of the rats which inhaled the hexavalent chromium mist for 1, 2 and 3 weeks. The concentrations of malondialdehyde (MDA), the metabolite of lipid peroxidation, in all exposed groups were higher than those of controls with dose-dependant manner. The levels of MDA were also correlated with urine chromium levels of the rats. Therefore, MDA as an indicator of lipid peroxidation could be proper biologic marker for the assessment of the oxidative damage from chromium exposure, which might be involved in carcinogenesis.
The purpose of this study was to investigate the effect of endurance training and Rooibos-tea treatment during 12 week on lipid peroxidation(MDA), oxidative DNA damage(8-hydroxyguanine), and antioxidant enzymes(SOD, GPX) in human. The subjects were divided into three groups; Train+Rooi, Train, and Rooi groups. The Train+Rooi and Rooi group took 3 g of Rooibos-tea for 12 weeks. Blood samples were taken from antecubital vein at before training, after 6week, and after 12 week training. Data were analyzed by two-way ANOVA with repeated measures using the SPSS/PC+. The results are summarized as follows: MDA and 8-hydroxyguanine concentration were no significantly differences between group(p>.05). SOD and GPX concentration were significantly increased in Train+Rooi, Rooi group than Train group(p<.05). This results suggested that effects of Rooibos-tea treatment has associated with improve scavenger of antioxidant.

The object of this study was to evaluate the effects of dietary supplementation of mulberry leaves and silkworm excreta ethanol extracts on weight performances, blood characteristics, cecal microflorae of chickens. Two hundred forty male broiler chicks(Ross) were fed diets for five weeks containing 0.1%(MLA) and 1%(MLB) of mulberry leaves ethanol extracts, and 0.1%(SEA) and 1%(SEB) of silkworm excreta ethanol extracts. Weight performance did show no significant difference in all test groups which were fed with supplementation of mulberry leaves and silkworm excreta ethanol extracts. They showed better weight gain and feed conversion than the negative control group which was fed only with forage without antibiotics. ABTS(2'-azine-bis[3-ethylbenzothiazoline-6-sulfonic acid]) test was conducted to investigate free radical scavenging activity of blood in tested groups. ABTS scavenging activities of tested groups were higher than control groups in significant level, though there was no significant difference(P = 0.396). Specifically, MLB group showed the highest scavenging activity. Blood-level concentration of MDA, which is an indicator of lipid peroxidation, was also decreased in tested groups and the lowest level was observed in SEA(P = 0.001). As storage time increased at $4^{\circ}C$, muscle-level MDA concentrations of all tested groups were generally increased and significant difference was obsereved between tested groups and controls in total increase of MDA concentration($P=4.417{\times}10^{-3}$). In cecal microflorae, SEA and SEB showed decreased total microbe population compared to NC($P=6.462{\times}10^{-5}$) and even to PC. Supplementation of mulberry leave and silkworm excreta ethanol extract did show a similar inhibition effect against Salmonella sp., furthermore, MLB did enhanced the growth of Lactobacillus sp.($P=3.636{\times}10^{-7}$). In summary, ethanol extract of silkworm excreta may be a potential alternative of antibiotics for chicks. In addition, both of ethanol extracts supplementation to broiler chicks would be very useful not only to improve antioxidant effect of blood but also to suppress lipid peroxidation without any loss of weight performance in poultry farming.
