YH439의 알콜성 지방간생성 억제작용 (Decreased Induction of Alcoholic Fatty Liver by YH430 in Rats)

  • A single large dose of ethanol as well as chronic ethanol consumption produces alcoholic fatty liver in human and experimental animals. We examined the effects of YH439, a potential hepatoprotective agent, on alcoholic fatty liver generation in adult female rats. In rats treated with YH439 (250 mg/kg, po) 4 hr prior to a single dose of ethanol (6 g/kg, po), a significant decrease in hepatic triglyceride accumulation was observed. YH439 also has an inhibitory effect on hepatic triglyceride and cholesterol accumulation induced by repeated ethanol treatments for one week. Because it has been known that induction of alcoholic fatty liver is associated with lipid peroxidation and/or hepatic glutathione depression, the effect of YH439 on these parameters was determined in the livers of rats treated with ethanol. Coadministration with YH439 inhibited MDA formation and gIutathione depression induced by acute or repeated ethanol administration. In order to determine the effect of YH439 on ethanol metabolism in vivo, disappearance of ethanol from blood was measured. In rats treated with a single dose of ethanol (6 g/kg, po), the ethanol concentration in blood reached a peak approximately 120 min following the treatment which declined linearly for 18 hrs. YH439 had no effect on the decline of blood ethanol concentration regardless of the dose of ethanol given to rats. These results in this study suggest that YH439 has an inhibitory effect on fatty liver generation induced by acute or repeated ethanol consumption through a mechanism not directly related to the rate of ethanol metabolism in vivo.

소토사자환이 ob/ob mouse의 혈당, 고지혈증, Polyol Pathway 및 항산화작용에 미치는 영향 (Effects of Sotosajahwan on Blood Glucose, Hyperlipidemia, Polyol Pathway and Antioxidative Mechanism in ob/ob Mouse)

  • Effects of Sotosaja hwan on Blood Glucose, Hyperlipidemia, Polyol Pathway and Antioxidative Mechanism in ob/ob Mouse Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Sotosaja-hwan has been known to be effective for the antiaging and composed of four crude herbs. In male ob/ob mouse in severe obesity, hyperinsulinemia and hyperlipidemia, which are features of NIDDM, the hyperglycemic activites and mechanisms of Sotosaja-hwan were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 30 and 90 mg Sotosaja-hwan per 1 kg of body weight for 14 days. The effects of Sotosaja-hwan extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of superoxide anion radical $({\cdot}O_2)$, MDA+HAE, GSH/GSSG ratio, and also the enzyme activities involved in polyol pathway. Sotosaja-hwan lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Sotosaja-hwan treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Sotosaja-hwan-administered groups. Sotosaja-hwan inhibited the generation of ${\cdot}O_2$ in the kidney. Finally, MDA+HAE levels was increased and GSH/GSSG ratio was decreased in the ob/ob mice, whereas those were improved in the Sotosaja-hwan-administered groups. Sotosaja-hwan showed the antidiabetic and anti hyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging reactive oxygen species and reducing the MDA+HAE levels in the ob/ob mice.

Bleeding Efficiency and Meat Oxidative Stability and Microbiological Quality of New Zealand White Rabbits Subjected to Halal Slaughter without Stunning and Gas Stun-killing

  • A study was conducted to compare the effect of halal slaughter without stunning and gas stun killing followed by bleeding on residual blood content and storage stability of rabbit meat. Eighty male New Zealand white rabbits were divided into two groups of 40 animals each and subjected to either halal slaughter without stunning (HS) or gas stun-kill (GK). The volume of blood lost during exsanguination was measured. Residual blood was further quantified by determination of haemoglobin content in Longissimus lumborum (LL) muscle. Storage stability of the meat was evaluated by microbiological analysis and measuring lipid oxidation in terms of thiobarbituric acid reactive substances (TBARS). HS resulted in significantly higher blood loss than GK. HS had significantly lower residual haemoglobin in LL muscle compared to GK. Slaughter method had no effect on rabbit meat lipid oxidation at 0, 1, and 3 d postmortem. However, at 5 and 8 days of storage at $4^{\circ}C$, significant differences (p<0.05) were found, with meat from the GK group exhibiting significantly higher levels of MDA than that from HS. At day 3, greater growth of Pseudomonas aeroginosa and E. coli were observed in the GK group (p<0.05) with B. thermosphacta and total aerobic counts remained unaffected by slaughter method. At days 5 and 7 postmortem, bacterial counts for all tested microbes were affected by slaughter method, with GK exhibiting significantly higher growth than HS. It can be concluded that slaughter method can affect keeping quality of rabbit meat, and HS may be a favourable option compared to GK due to high bleed out.

패 추출물이 제2형 당뇨병 환자의 혈당 및 항산화 체계에 미치는 영향 (Effects of Ishige okamurae Extract Supplement on Blood Glucose and Antioxidant Systems in Type 2 Diabetic Patients)

  • 본 연구는 패추출물 섭취를 통해 제 2형 당뇨병 환자의 혈당, 혈중지질 및 항산화 체계 개선에 있어서 패추출물의 효능을 규명하고자 하였다. 동결건조 한 패추출물을 200 mg 캡슐로 만들어 제 2형 당뇨환자에게 아침과 저녁 식전에 4캡슐씩 하루 8캡슐(1600 mg)을 10주간 섭취하도록 하였다. 위약군은 동량의 corn starch를 실험군과 동일하게 제조한 후 같은 방법으로 섭취하게 하였고, 이중맹검법을 사용하였다. 실험대상자는 총 46명이며 실험군 24명, 위약군 22명으로 구성되었으며, 두 군의 일반적 특성 및 생활습관은 유의적 차이가 없었다. 패추출물 섭취군에서 공복혈당은 10주간 섭취 후 $136.50{\pm}31.96$ mg/dL에서 $130.90{\pm}29.88$ mg/dL로 감소하였으나 유의적인 차이를 보이지 않았다. 당화혈색소는 $7.34{\pm}0.78%$에서 $6.82{\pm}0.98%$로 유의적(p<0.01)으로 감소하였다. 특히 당화혈색소가 8% 미만인 저당화혈색소 군에서 유의적으로(p<0.05) 감소하는 것으로 나타났다. 인슐린저 항성은 패추출물군의 경우 $2.77{\pm}0.49$ mmol/L에서 $2.19{\pm}0.62$ mmol/L로 감소하였으나 유의성이 없었으며 위약군도 유의한 변화가 없었다. 패추출물의 섭취가 10주간 이루어지면서 혈청지질의 변화는 유의하지 않았으나 HDL-콜레스테롤 경우 위약군에서 감소한 반면 패추출물군에서는 $41.14{\pm}7.86$ mg/dL에서 $46.82{\pm}5.69$ mg/dL로 증가하였다. LDL-콜레스테롤은 $115.43{\pm}27.47$ mg/dL에서 $108.73{\pm}22.36$ mg/dL로 감소하였으나 유의한 변화는 없었다. 지질과산화물 농도는 패추출물 섭취군에서 $6.83{\pm}1.05$ nmol/MDA에서 $5.72{\pm}1.28$ nmol/MDA로 그 함량이 감소되었고, superoxide dismutase, glutathione peroxidase, catalase와 같은 항산화 효소계의 활성은 위약군에 비하여 패추출물 섭취군에서 증가하였으나 유의적인 차이를 보이지 않았다. 간기능 지표인 AST, ALT, ${\gamma}$-GTP와 신장 기능 지표인 BUN, creatinine에서 유의적인 차이 없이 정상범위를 나타내었다. 이상의 연구 결과 한국인 제2형 당뇨병 환자에서 패추출물의 섭취는 지난 2~3개월간의 평균 혈당관리 상태를 파악할 수 있는 지표인 당화혈색소 감소에 효과적인 것으로 나타났으며, 패추출물 복용 후에도 간 및 신장기능이 정상으로 확인되었으므로 패추출물은 당뇨병 환자들의 평균 혈당관리에 도움을 줄 수 있을 것으로 사료된다.

커피가루 첨가식이가 당뇨 쥐의 혈당 및 간 조직 항산화효소 활성에 미치는 영향 (Effects of coffee powder supplementation on the blood glucose and antioxidative enzyme activity of liver tissue in STZ-induced diabetic rats)

  • 커피첨가식이를 섭취한 당뇨 쥐에서 혈당, 혈중 지질농도, 간지질함량 및 간조직의 항산화효소인 SOD, GPx, CAT에 미치는 영향을 분석한 결과를 요약하면 다음과 같다. 실험시작 시 체중은 실험군 간에 유의적인 차이가 없었고 실험종료 시 체중은 당뇨군내에서 커피군이 대조군보다 유의적으로 높았으며, 실험기간동안 체중증가량은 당뇨군이 비당뇨군보다 유의적으로 낮았으나 커피첨가식이 섭취에 따른 유의적인 차이는 없었다. 평균 식이섭취량은 당뇨군이 비당뇨군보다 유의적으로 높았고, 당뇨군내에서는 커피군이 대조군보다 유의적으로 높았으며, 식이효율은 당뇨군이 비당뇨군보다 유의적으로 낮았으나 커피섭취에 따른 유의적인 차이는 없었다. 당뇨군과 비당뇨군 모두 혈중 HDL-콜레스테롤 농도는 커피군이 대조군보다 유의적으로 높았고, LDL-콜레스테롤 농도와 atherogenic index는 커피군이 대조군보다 유의적으로 낮았다. 간의 중성지방 함량은 당뇨군과 비당뇨군 모두 커피군이 유의적으로 낮았고, 총콜레스테롤 함량은 커피섭취에 따른 유의적인 차이가 없었다. 간 조직의 MDA 함량은 당뇨군과 비당뇨군 모두 커피군이 대조군보다 유의적으로 낮았다. 간 조직의 항산화효소인 SOD, GPx, CAT 활성은 당뇨군과 비당뇨군 모두 커피섭취에 따른 유의적인 차이는 없었다. 혈당은 당뇨군과 비당뇨군 모두 커피군이 대조군보다 유의적으로 낮았다. 결론적으로 당뇨 쥐에서 0.5% 커피첨가식이가 간의 항산화효소 활성에는 유의적인 영향이 없었으나, 당뇨 쥐의 혈당을 낮추었고, 혈중 HDL-콜레스테롤 농도는 높였으며, 혈중 LDL-콜레스테롤 농도, atherogenic index, 간 중성지방 함량 및 과산화지질 MDA는 낮추어 혈당과 혈중 및 간 지질 개선에 도움이 되는 것으로 나타났다.

체외순환 시 백혈구 제거필터 사용의 임상효과 (The Clinical Effects of Leukocyte-Depleting Filter on Cardiopulmonary Bypass)

  • 배경: 개심술 시 체외순환회로 및 산화기의 합성표면에 대한 혈액의 접촉은 백혈구를 활성화시켜 전신 염증 반응을 일으키며, 활성화된 백혈구에 의해 생성된 산소유리기는 수술 후 심장 및 폐기능 손상의 주요 원인이 된다. 저자들은 체외순환동안 백혈구 제거필터 사용에 따른 임상효과를 평가하기 위해 본 연구를 실시하였다. 대상 및 방법: 성인 심장질환 환자 30명을 대상으로 백혈구 제거필터를 체외순환 회로에 포함시켜 수술한 필터군(n=15)과 필터를 사용하지 않은 대조군(n=15)으로 분류한 뒤, 테외순환 동안 말초혈액 내 백혈구 및 혈소판 수의 변화, 체외순환 전과 후의 관상정맥동혈액 내 malondialdehyde(MDA), troponin-T(TnT), 5'-nucleotidase(5'-NT의 농도변화, 대동맥 교차차단 해제 후 심박동 자연회복률, 수술 전과 후의 심장지수 및 폐혈관 저항의 변화, 수술 후 출혈량, 혈액제제 사용량과 창상 합병증 발생률 등을 양 군간에 비교하였다. 결과: 체외순환 종료 시 총 백혈구 수는 필터군이 대조군에 비해 유의하게 낮았으나(9,567$\pm$842/㎣ vs 13,573$\pm$1,167/㎣, p<0.01), 혈수판 수는 양 군간에 차이가 없었다(138,133$\pm$10,519/㎣ va 156,733$\pm$10,735/㎣, p=0.22). 수술 직후 관상정맥동 혈액내 MDA(3.78$\pm$0.31$\mu$mo1/L vs 5.86$\pm$0.65$\mu$mo1/L, p<0.01), TnT(0.40$\pm$0.04 ng/mL vs 0.59$\pm$0.08 ng/mL, p<0.05), 그리고 5'-NT(3.88$\pm$0.61 U/L vs 5.08 $\pm$0.90 UL, p<0.05) 등의 활성도는 필터군이 대조군 보다 더 낮았다(p<0.05). 수술후 심장지수는 필터군이 대조군보다 높았고(3.26$\pm$0.18 L/$m^2$/min vs 2.75$\pm$0.17 L/$m^2$/min, p=0.05), 폐혈관 저항은 필터군이 대조군 보다 낮았다(65.87$\pm$7.59 dyne/sec/ $cm^{5}$ vs 110.80$\pm$12.22 dyne/sec/$cm^{5}$ , p.0.01). 수술 후 심박동 자연회복률은 필터군이 대조군 보다 높았고(12명 vs 8명, p<0.05), 수술 후 창상합병증 발생률은 필터군이 대조군보다 낮았다(1명 vs 4명, p<0.05). 수술 후 첫 24시간 출혈량은 필터군이 대조군보다 많았다.(614$\pm$ 107 mL vs 380$\pm$71 mL, p=0.05).

허혈/재관류 심장의 산화손상에서 미토콘드리아의 역할 (Role of Mitochondria in Oxidative Damage of Post-Ischemic Reperfused Hearts)

  • Restoration of the blood flow after a period of ischemia is accompanied by generation of toxic oxygen radicals. This phenomenon may account for the occurrence of reperfusion-mediated tissue injury in ischemic hearts. In in vitro studies, although oxygen radicals can be generated from a variety of sources, including xanthine oxidase system, activated leucocytes, mitochondria and others, the most important source and mechanism of oxygen radical production in the post-ischemic reperfused hearts is unclear. In the present study, we tested the hypothesis that the respiratory chain of mitochondria might be an important source of oxygen radicals which are responsible for the development of the reperfusion injury of ischemic hearts. Langendorff-perfused, isolated rat hearts were subjected to 30 min of global ischemia at $37^{\circ}C$, followed by reperfusion. Amytal, a reversible inhibitor of mitochondrial respiration, was employed to assess the mitochondrial contributions to the development of the reperfusion injury. Intact mitochonria were isolated from the control and the post-ischemic reperfused hearts. Mitochondrial oxygen radical generation was measured by chemiluminescence method and the oxidative tissue damage was estimated by measuring a lipid peroxidation product, malondialdehyde(MDA). To evaluate the extent of the reperfusion injury, post-ischemic functional recovery and lactate dehydrogenase(LDH) release were assessed and compared in Amytal-treated and -untreated hearts. Upon reperfusion of the ischemic hearts, MDA release into the coronary effluent was markedly increased. MDA content of mitochondria isolated from the post-ischemic reperfused hearts was increased to 152% of preischemic value, whereas minimal change was observed in extramitochondrial fraction. The generation of superoxide anion was increased about twice in mitochondria from the reperfused hearts than in those from the control hearts. Amytal inhibited the mitochondrial superoxide generation significantly and also suppressed MDA production in the reperfused hearts. Additionally, Amytal prevented the contractile dysfunction and the increased release of LDH observed in the reperfused hearts. In conclusion, these results indicate that the respiratory chain of mitochondria may be an important source of oxygen radical formation in post-ischemic reperfused hearts, and that oxygen radicals originating from the mitochondria may contribute to the development of myocardial reperfusion injury.

구기자 추출물과 생과즙의 간독성 보호효과 (Hepatoprotective Effects of Lycium chinense Mill Fruit Extracts and Fresh Fruit Juice)

  • 국내에서 선별 육종된 불로 구기자의 처리방법에 따른 간기능 보호효과를 측정하기 위하여 사염화탄소$(CCl_4)$ 투여로 급성 간독성을 유발시킨 흰쥐를 대상으로 연구하였다. 건조구기자 추출물(DFE), 볶은 구기자 추출물 (RFE), 그리고 생과즙(FFE)을 경구 투여한 결과 군간의 체중증가량은 차이를 나타내지 않았다. 한편, 체중 100 g 당 간 무게는 처리조건에 관계없이 구기자 추출물 및 생과즙 투여군이 유의적으로 낮은 간 무게를 나타내었다(p<0.01). 이것은 구기자 추출물과 생과즙 섭취가 $CCl_4$ 투여에 의한 지방간 형성 또는 간 독성에 의한 간 무게증가를 다소 완화하여준 효과를 나타내는 것으로 판단된다. 사염화탄소$(CCl_4)$ 투여에 의하여 흰쥐의 혈청 GPT 및 GOT 활성과 LDH 활성은 유의적으로 증가하였으나 볶은 구기자 추출물 투여군(RFEC)과 생과즙 투여군(FFJC) 모두 GPT, GOT 활성이 유의적으로 감소되었다. 혈중 총 콜레스테롤 함량은 구기자 추출물과 생과즙 투여군 모두 대조군(CON)과 비교하여 유의적인 증가를 나타내지 않았고 중성 지방함량은 대조군에 비해 유의적인 증가를 나타내었다(p<0.05). 간 조직의 MDA 함량과 GST, cytochrome P-450 활성 결과로 보아 구기자 추출물과 생과즙을 섭취 시 사염화탄소에 의한 독성을 완화시키는 것으로 나타났다. 따라서 이러한 모든 결과를 종합해 볼 때, 구기자 추출물과 생과즙은 $CCl_4$ 투여로 인한 지질 과산화, 지방변성, 간 세포 괴사 등을 억제 시키는 간 독성 보호 효과가 있는 것으로 생각된다.

Cinnamaldehyde Attenuates Cataractogenesis via Restoration of Hypertension and Oxidative Stress in Fructose-Fed Hypertensive rats

  • Objectives: Several studies have revealed that systemic hypertension is strongly associated with cataractogenesis. However, the pathophysiology and treatment is often unclear. In this study, we evaluated the anti-cataractogenic effect of cinnamaldehyde (CA), a natural organic compound, in rats with fructose-induced hypertension. Methods: The rats were divided into six groups. For six weeks, the normal group received a suspension of 0.5% carboxy methyl cellulose (10 mL/kg/day, p.o.) while five other groups received a 10% (w/v) fructose solution in their drinking water to induce hypertension. By the end of the third week hypertension had been induced in all the animals receiving fructose. From the beginning of the fourth week to the end of the sixth week, one of those five groups (control) continued to receive only 10% (w/v) fructose solution, one group (standard) received ramipril (1 mg/kg/day, p.o.) plus 10% (w/v) fructose solution, and three groups (experimental) received CA at doses of 20, 30, and 40 mg/kg/day p.o., plus 10% (w/v) fructose solution. Blood pressure was measured weekly using a non-invasive blood pressure apparatus. After six weeks, the animals were sacrificed, and the anti-cataractogenic effects on the eye lenses were evaluated. Results: Administration of fructose elevated both the systolic and the diastolic blood pressures, which were significantly reduced by CA at all dose levels. In the control group, a significant increase in the malonaldehyde (MDA) level and decreases in the total protein, $Ca^{2+}$adenosine triphosphate (ATP)ase activity, glutathione peroxidase, catalase, superoxide dismutase and glutathione levels, as compared to the normal group, were observed. Administration of CA at all doses significantly restored the enzymatic, non-enzymatic, antioxidants, total protein, and $Ca^{2+}$ATPase levels, but decreased the MDA level, as compared to the control group. Conclusion: The present study revealed that CA modulated the antioxidant parameters of the serum and lens homogenates in hypertension-induced cataractogenic animals.

RhGLP-1 (7-36) protects diabetic rats against cerebral ischemia-reperfusion injury via up-regulating expression of Nrf2/HO-1 and increasing the activities of SOD

  • The present study aimed to explore the neuroprotective effect and possible mechanisms of rhGLP-1 (7-36) against transient ischemia/reperfusion injuries induced by middle cerebral artery occlusion (MCAO) in type 2 diabetic rats. First, diabetic rats were established by a combination of a high-fat diet and low-dose streptozotocin (STZ) (30 mg/kg, intraperitoneally). Second, they were subjected to MCAO for 2 h, then treated with rhGLP-1 (7-36) (10, 20, $40{\mu}g/kg$ i.p.) at the same time of reperfusion. In the following 3 days, they were injected with rhGLP-1 (7-36) at the same dose and route for three times each day. After 72 h, hypoglycemic effects were assessed by blood glucose changes, and neuroprotective effects were evaluated by neurological deficits, infarct volume and histomorphology. Mechanisms were investigated by detecting the distribution and expression of the nuclear factor erythroid-derived factor 2 related factor 2 (Nrf2) in ischemic brain tissue, the levels of phospho-PI3 kinase (PI3K)/PI3K ratio and heme-oxygenase-1 (HO-l), as well as the activities of superoxide dismutase (SOD) and the contents of malondialdehyde (MDA). Our results showed that rhGLP-1 (7-36) significantly reduced blood glucose and infarction volume, alleviated neurological deficits, enhanced the density of surviving neurons and vascular proliferation. The nuclear positive cells ratio and expression of Nrf2, the levels of P-PI3K/PI3K ratio and HO-l increased, the activities of SOD increased and the contents of MDA decreased. The current results indicated the protective effect of rhGLP-1 (7-36) in diabetic rats following MCAO/R that may be concerned with reducing blood glucose, up-regulating expression of Nrf2/HO-1 and increasing the activities of SOD.
