CHEMOSENSITIVITY OF CANCER CELLS TO ANTICANCER DRUGS USING DYE EXCLUSION ASSAY, [3H] THYMIDINE INCORPORATION, AND CLONOGENIC ASSAY (두경부악성종양세포주의 항암제감수성 시험에 관한 실험적 연구)

  • The in vitro predictive tests in cancer chemotherapy of cancer cell lines to anticancer drugs were determined using novel dye exclusion assay [NDEA], [3H] thymidine incorporation, and clonogenic assay [CA>. Antitumor effect of Bleomycin, Cis-platin, Vinblastine, Methotrexate to HEp-2, B16 cell lines using rapid assays was compared with [CA> in this study. In dye exclusion assay of B l6 cell line, cancer cells were sensitive to Bleomycin at all concentrations, to Vinblastine at the level of peak plasma concentration [PPC], ${\times}1/10$ [PPC](P<0.05). And Bleomycin revealed relatively good cytotoxicity than that of CDDP and vinblastine at ${\times}10$[PPC], (P<0.05). HEp-2 cells were resistive to methotrexate at the level of ${\times}100$[PPC] (P<0.05) In [3H] thymidine incorporation assay, B 16 cells were sensitive to Bleomycin, CDDP, Vinblastine at the level of [PPC], ${\times}10$ [PPC](P<0.01). Dose-dependent drugs of bleomycin, CDDP were more sensitive than Vinblastine at high concentration (P<0.05). In clonogenic assay, HEp-2 cell line was sensitive to three drugs of all concentrations except ${\times}10$ [PPC] of CDDP. B 16 cell line was sensitive to all drugs(P<0,01). In comparison of chemosensitivity tests among three assays, the results were correlated(${\gamma}=0.99$, P<0.05).

Effects of Flavonol Derivatives on the Micronudei Formation by N-methyl-N'-nitro-N-nitrosoguanidine and the Enhancement of Bleomycin-induced Chromosome Aberration by N-methyl-N'-nitro-N-nitrosoguanidine

  • Flavonol derivatives were tested for their anticlastogenic effect against induction of micronuclei by n-methyl-n'-nitor-n-nitorsoguanidine(MNNG), and against induction of chromosome aberration by bleomycin or MNNG.belomycin. For micronudeus assay, each flavonol derivative (0, 0.001, 0.01, 0.1, 1, 10 and 100 mg/kg) was administered orally twice with 24 h interval, together with intraperitioneally administered MNNG(150 mg/kg). The result showed that msot flavonol derivatives tested were effective in suppresing the frequencies of micronude induced by MNNG. For chromosome aberration assy, each flavonol derivative (0, 0.1, 1, 10m and 100 mg/kg)was administered to mice orally in vivo, and then mice were sacrificed and spleen lymphocyte cultures were made. Bleomycin $(3\;\mu$g/ml) was treated to the mouse spleen hymphocyte cultures at 24 h after con A initiation. There wre nomarked decrease tendencies in chromosome aberration unless all doses of galangin and some doses of several flavonol derivatives tested. In the another experiment, we have evaluated the effect of flavonol derivatives on the enhancement of bleomycin-induced chromsome aberration by MNNG. Most of flavonol derivtives reduced the incidence of chromosome aberration induced by in vitro treatment of bleomycin followed by in vivo treatment of MNNG. Galangin particulary showed a dose-dependent decrease tendency. Other flavonol derivative showed slightly suggest that most of flavonol derivatives may be capable of protecting the inhibition of suggest that most of flavonol derivatives may be capable of protecting the inhibition of DNA-repair by MNNG. Our data indicate clearly that flavonol derivatives can suppress MNNG-induced genotoxicity such as an induction of MNPCEs. Therfore, our results could suggest that flavonol derivtives may be useful as a chemopreventive agent of MNNG.

Maltol, an Antioxidant Component of Korean Red Ginseng, Shows Little Prooxidant Activity

  • Some antioxidant phenolic compounds exhibit prooxidant activity mainly due to their abilities to reduce $Fe^{3+}\; to\; Fe^{2+}.$ Reducing ability and prooxidant activity of maltol, an antioxidant component of Korean red ginseng, were compared with those of pyrogallol. Maltol at 2 mM did not appreciably reduce$ Fe^{3+}\; to\; Fe^{2+}$ and also failed to reduce nitroblue tetrazolium. Stimulation of hydroxyl radical mediated-deoxyribose degradation by pyrogallol was maximal at 60 .mu.M. Maltol stimulated the deoxyribose degradation to a much less extent, and a similar stimulatory effect was observed at a concentration of more than 100-fold higher than that of pyrogallol. The stimulatory effect of maltol reached a plateau over 1 mM, suggesting the removal of hydroxyl radicals by excess maltol. In bleomycin-$Fe^{3+}$-DNA assay, maltol at 2 mM produced a 2.5-fold increase of the iron-bleomycin-dependent DNA degradation over the basal value, whereas pyrogallol at 10 .mu.M accelerated DNA degradation by ca. 10-fold. Furthermore, maltol inhibited $Fe^{2+}$-stimulated DNA degradation by bleomycin. These results strongly suggested that maltol is an antioxidant with little prooxidant activity.

  • 2,3,6-Tribromo-4,5-dihydroxybenzyl methyl ether (TDB) from the methanolic extract of the red alga Symphyocladia latiuscula exhibits major antioxidant activity. In this study, the activity of TDB against oxidative damage in deoxyribose and DNA was investigated in vitro for potential applications in preventing mutagenesis caused by DNA damage. TDB inhibited the oxidation of deoxyribose at concentrations of up to $1{\mu}g$/mL in the presence of $Fe^{+3}$-EDTA/$H_2O_2$. Furthermore, TDB showed no pro oxidant activity as determined by absence of the reduction of bleomycin-$Fe^{+3}$ to bleomycin-$Fe^{+2}$, which leads to DNA damage. Based on these results, TDB demonstrated considerable antioxidant activity without prooxidant properties.

The Protective Effects of Green Tea Catechin on The Bleomycin and Cyclophosphamide Induced Cytotoxicity

  • Green tea and tea polyphenols have been studied extensively as cancer chemopreventive agents in recent years. Epigallocatechin-3-gallate (EGCG) is widely recognized as a powerful antioxidant and a free radical scavenger. The purpose of this study was to evaluate the protective effects of green tea catechins (GTC) on the Bleomycin- and Cyclophosphamide-induced cytotoxicity. Cell viability was measured by MTT assay. In the protective effect of GTC, the cell viability was significantly increased by the treatment of GTC. Furthermore, GTC showed the higher protective effect than EGCG and vitamin E. These results suggest that GTC has the protective effect which is related to the prevention of cancer. Our studies show that the continuous presence of EGCG can reduce radical-induced DNA damage in Chinese hamster lung fibroblast cells (CHL cells).

A STUDY ON THE RADIOSENSITIVITY AND CHEMOSENSITIVITY OF A-253 CELL LINE IN VITRO (시험관내 A-253 세포주의 방사선 및 항암제 감수성에 관한 연구)

  • The purpose of this study was to aid in the prediction of tumor cell tolerance to radiotherapy and/or chemotherapy. For this study, cell surviving curves were obtained for human epidermoid carcinoma A-253 cell line using semiautomated MTT assay. 2,4,6,8,10 Gy were irradiated at a dose rate of 210 cGy/min using /sup 60/Co Irradiator ALDORADO 8. After irradiation, A-253 cell lines(2×10⁴cells/mil were exposed to bleomycin or cisplatin for 1 hour. The viable cells were determined for each radiation dose with/without 2 /lg/mi of drug at the 3rd day. And they were compared to control values. The results were obtained as follows : 1. The surviving curve with gentle slope was obtained after irradiation of 2, 4, 6, 8, 10 Gy on A-253 cell line. 2. The cytotoxicity of bleomycin or cisplatin at the concentration of 2㎍/ml was great on A-253 cell line. But, there was no significant difference between the cytotoxicity of bleomycin and that of cisplatin. 3. There were significant differences of surviving fractions after irradiation with 2㎍/mi of bleomycin compared with irradiation only on A-253 cell line. 4. There were significant differences of surviving fractions after irradiation with 2㎍/ml of cisplatin compared with irradiation only on A-253 cell line. 5. There were no significant differences of surviving fractions between the groups of irradiation with bleomycin and the groups of irradiation with cisplatin on A-253 cell line.

After irradiation, YAC-1 cell lines(3×10⁴cells/ml) were exposed to bleomycin or cisplatin for 1 hour. The viable cells were determined for each radiation dose and/or each concentration of drug at the 4th day. And they were compared to control values. The obtained results were as follows : 1. The surviving curve with gentle slope was obtained after irradiation of 2, 4, 6, 8, 10 Gy on YAC-1 cell line. 2. The cytotoxicity of bleomycin or cisplatin was increased significantly at all concentration of 0.2㎍/ml, 2㎍/ml and 20㎍/ml on YAC-1 cell line (P<0.01). And the cytotoxicity of cisplatin was greater than that of bleomycin at all concentration on YAC-1 cell line (P<0.01). 3. There were no significant differences of surviving fractions among 4Gy, 6Gy and 8Gy after irradiation of each radiation dose with 2㎍/ml of bleomycin compared with irradiation only on YAC-1 cell line. 4. There was significant difference of surviving fraction between 2Gy and 10Gy after irradiation of each radiation dose with 2㎍/ml of cisplatin compared with irradiation only on YAC-1 cell line(P<0.05). 5. There were significant differences of surviving fractions between the groups of irradiation only and the groups of irradiation with 2㎍/ml of bleomycin or cisplatin at all doses of 2, 4, 6, 8 and 10Gy on YAC-1 cell line(P<0.05).

  • Objectives: The purpose of this study was to aid in the prediction of tumor cell tolerance to radiotherapy and/or chemotherapy. Material and Methods: Human epidermoid carcinoma A-431 cell lines were irradiated by 2, 4, 6, 8, 10Gy at a dose rate of 210cGy/min using /sup 60/Co Irradiator ALDORADO 8 and then were exposed to bleomycin or cisplatin at concentration of 2㎍/㎖ for 1 hour. The viable cells were determined for each radiation dose and/or each drug at the 4th day and cell surviving curves were obtained using semiautomated MTT assay. Results: The surviving fraction after irradiation of 2Gy was 0.99, and there was not significant difference of surviving fraction in comparison with the control group on A-431 cell line(P>0.05). But there were significant differences of surviving fractions at doses of 4, 6, 8, 10Gy in comparison with the control group(P<0.05). The cytotoxicity of bleomycin or cisplatin was significantly different in comparison with the control group on A-43l cell line (P<0.05). And the cytotoxicity of cisplatin was greater than that of bleomycin on A-431 cell line (P<0.05). There were significant differences of surviving fractions after irradiation of 2, 4, 6, 8, 10Gy with bleomycin or cisplatin in comparison with each group of irradiation only on A-431 cellline(P<0.05). There were significant differences of surviving fractions between the groups of irradiation with bleomycin and cisplatin at doses of 2, 4Gy(P<0.05), but there were not significant differences of surviving fractions at doses of 6, 8, 10Gy on A-431 cell line (P>0.05).

After irradiation, MG-63 cell lines(3×10⁴ cells/ml) were exposed to bleomycin and cisplatin at concentration of 0.2, 2, 20㎍/㎖ for 1 hour respectively. The viable cells were determined for each radiation dose and/or each concentration of drug. And they were compared to control values. The obtained results were as follows : 1. There was significant difference of surviving fraction at 4, 6, 8, 10Gy on MG-63 cell line(p<0.05). 2. There was significant difference of cytotoxicity of bleomycin or cisplatin at all concentration of 0.2, 2, 20㎍/㎖ (p<0.05) on MG-63 cell line. The cytotoxicity of cisplatin was more effective than bleomycin at concentration of 20㎍/㎖ on MG-63 cell line. 3. There was significant difference of cytotoxicity of bleomycin or cisplatin at all concentration after irradiation of 2Gy on MG-63 cell line. 4. There was significant difference of cytotoxicity of bloemycin or cisplatin at concentration of 20㎍/㎖ after irradiation than that of irradiation alone(p<0.01). But there was no significant difference of cytotoxicity of bleomycin at concentration of 20㎍/㎖ after irradiation of l0Gy than that of irradiation alone.
