Compositional and Morphological Analyses of Black Precipitate Formed on the Surface of Heating Appliance (수돗물 가열시 형성되는 흑색 침전물의 성분 분석)

  • The compositional and morphological analyses have been carried out by employing XRD, EPMA, and SEM for the black-colored precipitate which is formed on the surface of heating appliances by heating tap water. XRD analysis has showed that the precipitate was mainly composed of calcium carbonate with a crystal structure of calcite. Also, small amounts of precipitate by SEM showed that the microscopic shape of the precipitate was usually angular spherical.

The Nitrite-Scavenging Effects by Component of Oolong and Black Tea Extracts (오룡차 및 홍차 추출물의 아질산염 분해작용)

  • The present study was conducted to elucidate the functional property of tea extract obtained from tea extracts, semi-fermented tea(oolong tea) and fermented tea(black tea). Tea extracts exhibited remarkable nitrite-scavenging actions, and the action increased with the increased of the amount of tea extracts. The nitrite-scavenging actionof tea extracts showed pH dependent, highest at pH 1.2 and lowest at pH6.0. The nitrite-scavenging rate in tea extracts/amine/nitrite systems proved to be faster than that in amine/nitrite systems. To screen the nitrite-scavenging factors, tea extracts were fractionated into water-soluble, methanol-soluble, methanol-precipitate and crude catechin fraction. Among these fractions of tea extracts, the crude catechin fraction possessed greater nitrite-scavenging action than the other fractions. The nitrite-scavenging action of tea extracts increased with the contents of total phenols and an absorbance at 280nm, nitrite-scavenging factors were supposed to be and catechins in tea polyphenol compounds.

  • A surface renewable pH electrode was prepared by utilizing composite electrode technique. Iridium oxide micro-fine particles was prepared by hydrolysis of $(NH_4)_2IrCl_6$ at elevated temperature. The iridium oxide particles were mixed with well-dispersed carbon black and then filtered. The mixture was suspended in DMF containing PVC as a binder. The mixture was precipitated rapidly by adding large amount of water. The precipitate was ground and pressure-molded to iridium oxide composite electrode material. The electrode showed linear response between pH 1-13 with 50 to 60 mV/pH slope. The electrode maintained the pH response without appreciable slope drift for 170 days if stored in deionized water. The electrode surface can be renewed reproducibly by simple grinding process whenever contaminated or deactivated.

Isolation and Optimal Culture Conditions of Brevibacillus sp. KMU-391 against Black Root Pathogens Caused by Didymella bryoniae (덩굴마름병원균인 Didymella bryoniae의 생물학적 방제를 위한 길항세균의 분리 및 특성)

  • We isolated a bacterium which produces antifungal substances from the Sanktpeterburg soils at Russia. The iso-lated strain was identified as Brevibacillus sp. and shown a strong antifungal activity on plant pathogenic fungi. Brevibacillus sp. KMU-391 produced maximum level of antifungal substances under incubation aerobically at $30^{\circ}C$ for 48 hours in trypticase soybean broth containing 1.0% sucrose and 1.0% polypeptone at 180 rpm and initiated pH adjusted to 7.0. Precipitate of culture broth by $30{\sim}60%$ ammonium sulfate precipitation exhibited strong antifungal activity against Didymella bryoniae by dry cell weight. Butanol extract of cultured broth also shown fungal growth inhibitory activity against Botrytis cinerea KACC 40573, Botrytis fabae KACC 40962, Colletotrichum gloeosporioides KACC 40804, Colletotrichum orbiculare KACC 40808, Didymella bryoniae KACC 40669, Fusarium graminearum KACC 41040, Fusarium oxysporum KACC 40037, Fusarium oxysporum KACC 40052, Fusarium oxysporum f, sp. radicis-Iycopersici KACC 40537, Fusarium oxysporum KACC 40902, Monosporascus cannonballus KACC 40940, Phytophthora camvibora KACC 40160, Rhizoctonia solani AG-1(IA) KACC 40101, Rhizoctonia solani AG-4 KACC 40142, and Scleotinia scleotiorum KACC 41065 by agar diffusion method.

Petrological Characteristics and Deterioration State of Standing Buddha Statue in the Gwanchoksa Temple, Nonsan, Korea (논산 관촉사 석조미륵보살입상의 암석학적 특성과 풍화훼손도)

  • The Standing Buddha Statue in the Gwanchoksa temple consists of medium to coarse grained biotite granodiorite with dark grey color, and it has a week gneissosity along the pegmatite veins. The results of magnetic susceptibility and geochemical patterns of the host rock of Standing Buddha Statue and the basement rock suggest that both values are formed from the co-genetic magma with the same differentiation process. The CIAs of the basement rock and the Standing Buddha Statue are calculated to 51.43 and 50.86, and the WPIs are estimated 4.52 and 8.95, respectively. So the weathering potential from the host rock of Standing Buddha Statue and basement rock prove to be high. The Standing Buddha Statue is terribly damaged with physical weathering from deterioration and exfoliation, and are scattered with secondary pollutant and precipitate. Basement rock is also in danger of ground collapse because of irregularly developed discontinuity system. Most surface of Standing Buddha Statue is seriously discolored into yellowish brown and dark gray, or black precipitates are also formed. Moreover, it is heavily covered with crustose lichen, fungi and algae, or moss are also found. In order to control the influential factors with the complex deterioration of Standing Buddha Statue, it is needed to rearrange a site environments, and conservation scientific management is required to protect it from covering lichens, exfoliations and fractures.

Deterioration Assessment and Conservational Scientific Diagnosis of the Stone Pagoda in the Bunhwangsa temple, Gyeongju, Korea (경주 분황사석탑의 풍화훼손도 평가와 보존과학적 진단)

  • The stone pagoda of the Bunhwangsa temple made by piling small brick-shaped stones. The major rock forming stone bricks are andesites with variable genesis. Rock properties of the pagoda roof stone suffer partly including multiple peel-offs, exfoliation, decomposition like onion peels, cracks forming round lines and falling off stone pieces. The stylobates and tabernacles in all the four directions the pagoda are mostly composed of granitic rocks. Those rock properties are heavily contaminated by lichens and mosses with the often marks of inorganic contamination by secondary hydrates that are dark black or yellowish brown. Within the four tabernacles and northern pagoda body situated to relatively high humidity. There are even light gray precipitate looking like stalactites between the northern and western rocks of the body Their major minerals are calcite, gypsum and clays. The stone lion standing in the southeast and northeast side are alkali granite, while that in the southwest and northwest lithic tuff. Total rock properties of the pagoda are 9,708 pieces, among the all properties, fractured blocks are 11.0%, fall out blocks are 6.7% and covered blocks by precipitates are 7.0%, respectively. The pagoda has highly deteriorated the functions of the rock properties due to physical, chemical and biological weathering, therefore, we suggest that this pagoda has need to do long term monitoring and synthetic conservation researches.

The Mechanism to Oxidative Polymerization of N-Methyl-p-Aminophenol and Thermal Characteristics of the Polymer Formed (N-Methyl-p-Aminophenol의 酸化重合機構 및 生成重合體의 熱特性)

  • N-Methyl-p-aminophenol was polymerized by oxidative couplng in the aqueous iron chelate solution in the presence of oxygen, and black precipitate of oligo-(N-methyl-p-aminophenol) was formed quantitatively. In this oxidative polymerization reaction, methyl group attached to N in the monomer was partly eliminated, and it was clarified by the infrared spectra from the fact that the absorption of ${\delta}\;asym\;CH_3\;1460\;cm^{-1}$ and ${\delta}\;sym\;CH_3\;1380\;cm^{-1}$ in acetone insoluble fraction was much weaker than that in acetone soluble fraction. From Thermo-gravimetric analysis, oligo-(N-methyl-p-aminophenol) showed about 40% weight loss at $600^{\circ}C$ and it was less heat-resistant than oligo (p-aminophenol) that methyl group was not contained. In pyrolysis of oligo-(N-methyl-p-aminophenol) in He atmosphere, monomer N-methyl-p-aminophenol and water were formed, and in the pyrolytic gases, $H_2,\;CO,\;CO_2$ were detected by gas chromatography. From the above facts, to the structural change on oligo-(N-methyl-p-aminophenol) when it was heat-treated, it was considered that original linear structure was partly degraded, and the most of the oligomer was to go in with melt polycondensation to form polymer, and heat-resistant cyclic structure was formed at a time.

Effect of Silicon on the Corrosion Characteristics of Zirconium (Zr의 부식특성에 미치는 Si의 영향)

  • Zr-Si binary alloys containing 0.01 to O.lwt.%Si were prepared to investigate the effect of Si on the corrosion behavior of Zr. Corrosion test was performed in pure water at 36$0^{\circ}C$ under a pressure of 2660psi for 100days. The alloys containing 0.01 wt. % and 0.05wt. %Si had the black and uniform oxide film and didn't show the transition of corrosion rate. However. the alloys containing O.lwt.%Si had white oxide film and showed the trasition of corrosion rate at 70 days corrosion test. The weight gain increased with the increasing Si content from 0.01 to 0.1 wt.%. The variation of Si contents had no effect on changing the oxide structure but had significant effect on the electrical resistivity of oxide. The electrical resistivity decreased with increasing Si content. The fraction of precipitates in the Zr-Si binary alloys. identified as tetragonal $Zr_{3}$Si increased with increasing Si content. The increase of the volume fraction of precipitates is thought to be responsible for the increase of weight gain due to short circuit effect of precipitate.

In-situ Precipitation of Arsenic and Copper in Soil by Microbiological Sulfate Reduction (미생물학적 황산염 환원에 의한 토양 내 비소와 구리의 원위치 침전)

  • Microbiological sulfate reduction is the transformation of sulfate to sulfide catalyzed by the activity of sulfate-reducing bacteria using sulfate as an electron acceptor. Low solubility of metal sulfides leads to precipitation of the sulfides in solution. The effects of microbiological sulfate reduction on in-situ precipitation of arsenic and copper were investigated for the heavy metal-contaminated soil around the Songcheon Au-Ag mine site. Total concentrations of As, Cu, and Pb were 1,311 mg/kg, 146 mg/kg, and 294 mg/kg, respectively, after aqua regia digestion. In batch-type experiments, indigenous sulfate-reducing bacteria rapidly decreased sulfate concentration and redox potential and led to substantial removal of dissolved As and Cu from solution. Optimal concentrations of carbon source and sulfate for effective microbial sulfate reduction were 0.2~0.5% (w/v) and 100~200 mg/L, respectively. More than 98% of injected As and Cu were removed in the effluents from both microbial and chemical columns designed for metal sulfides to be precipitated. However, after the injection of oxygen-rich solution, the microbial column showed the enhanced long-term stability of in-situ precipitated metals when compared with the chemical column which showed immediate increase in dissolved As and Cu due to oxidative dissolution of the sulfides. Black precipitates formed in the microbial column during the experiments and were identified as iron sulfide and copper sulfide. Arsenic was observed to be adsorbed on surface of iron sulfide precipitate.

Studies on the analysis of phytin by the Chelatometric method (Chelate 법(法)에 의(依)한 Phytin 분석(分析)에 관(關)한 연구(硏究))

  • Phytin is a salt(mainly calcium and magnesium) of phytic acid and its purity and molecular formula can be determined by assaying the contents of phosporus, calcium and magnesium in phytin. In order to devise a new method for the quantitative analysis of the three elements in phytin, the chelatometric method was developed as follows: 1) As the pretreatment for phytin analysis, it was ashfied st $550{\sim}600^{\circ}C$ in the presence of concentrated nitric acid. This dry process is more accurate than the wet process. 2) Phosphorus, calcium and megnesium were analyzed by the conventional and the new method described here, for the phytin sample decomposed by the dry process. The ashfied phytin solution in hydrochloric acid was partitioned into cation and anion fractions by means of a ration exchange resin. A portion of the ration fraction was adjusted to pH 7.0, followed by readjustment to pH 10 and titrated with standard EDTA solution using the BT [Eriochrome black T] indicator to obtain the combined value of calcium and magnesium. Another portion of the ration fraction was made to pH 7.0, and a small volume of standard EDTA solution was added to it. pH was adjusted to $12{\sim}13$ with 8 N KOH and it was titrate by a standard EDTA solution in the presence of N-N[2-Hydroxy-1-(2-hydroxy-4-sulfo-1-naphytate)-3-naphthoic acid] diluted powder indicator in order to obtain the calcium content. Magnesium content was calculated from the difference between the two values. From the anion fraction the magnesium ammonium phosphate precipitate was obtained. The precipitate was dissolved in hydrochloric acid, and a standard EDTA solution was added to it. The solution was adjusted to pH 7.0 and then readjusted to pH 10.0 by a buffer solution and titrated with a standard magnesium sulfate solution in the presence of BT indicator to obtain the phosphorus content. The analytical data for phosphorus, calcium and magnesium were 98.9%, 97.1% and 99.1% respectively, in reference to the theoretical values for the formula $C_6H_6O_{24}P_6Mg_4CaNa_2{\cdot}5H_2O$. Statical analysis indicated a good coincidence of the theoretical and experimental values. On the other hand, the observed values for the three elements by the conventional method were 92.4%, 86.8% and 93.8%, respectively, revealing a remarkable difference from the theoretical. 3) When sodium phytate was admixed with starch and subjected to the analysis of phosphorus, calcium and magnesium by the chelatometric method, their recovery was almost 100% 4) In order to confirm the accuracy of this method, phytic acid was reacted with calcium chloride and magnesium chloride in the molar ratio of phytic: calcium chloride: magnesium chloride=1 : 5 : 20 to obtain sodium phytate containing one calcium atom and four magnesium atoms per molecule of sodium phytate. The analytical data for phosporus, calcium and magnesium were coincident with those as determine d by the aforementioned method. The new method employing the dry process, ion exchange resin and chelatometric assay of phosphorus, calcium and magnesium is considered accurate and rapid for the determination of phytin.
