A Study on the Intake Patterns of Health Food of the Elderly Aged over 60 Years in the Chuneheon Area (춘천지역 60세 이상 노년층의 건강식품 섭취실태 조사)

  • This study was carried out to investigate the intake patterns of health food according to the kind of disease of the elderly aged over 60 years living in the Chuncheon area. This study was conducted by using a questionnaire from December 1999 to the March 2000. The data was analyzed using a SPSS program. The results were summarized as follows : The poorer the self- perceived health condition, the lower the level of activity of daily living(ADL) and instrumental activities of daily living(IADL) were the higher. the frequency score of health food intake. The incidence of disease, such as arthritis(44.7%), hypertension(18.8%), digestive diseases(17.1%), diabetes(10.5%) were found to be higher. than other chronic diseases among the elderly. The frequency score of health food intake was not significantly different according to self-perceived health status, disease status, or the level of ADL and IADl.. The must important source of information for health food intake was the recommendation of friends and relatives(68.7%). Twenty eight point two percent of the elderly considered health foods to be effective, however 8.3% of the elderly considered them to be ineffective. The elderly with chronic diseases had taken health floods in the descending order of neuralgia-arthritis, hypertension, digestive disease, diabetes. The elderly with digestive diseases, diabetes, renal disease, neuralgia- arthritis and respiratory disease were inclined to habitually take health floods once or twice per year. Intake of Korean traditional folk food was as high as intake of registered health food and Chinese tonic medicine. The most frequently taken health food was tonic medicine, which is fo11owed by pumpkin, ginseng products, herbal tea formula with black goat, deer antler and Kye-so-ju. Intakes of health food such as tonic medicine, blood of the deer, royal jelly, aloe were found to be higher among the elderly with chronic diseases such as diabetes, hypertension, and digestive disease. However, intakes of ginseng products, deer antler, pumpkin, herbal tea formula with black goat and Kye-so-ju were found to be higher among the healthy elderly. The elderly with neuralgia-arthritis had taken sixteen kinds of health foods. From these results, a wide consumer education program should be developed to convince people of the importance of well-balanced diet and to choose proper health foods according to the elderly's health conditions. Also, comprehensive and scientific research into Korean traditional folk foods are needed for the correct use.

Cloning of Farm Animals in Japan; The Present and the Future

  • 1. About fifty thousand of cattle embryos were transferred and 16000 ET-calves were born in 1999. Eighty percents of embryos were collected from Japanese Black beef donors and transferred to dairy Holstein heifers and cows. Since 1985, we have achieved in bovine in vitro fertilization using immature oocytes Collected from ovaries of slaughterhouse. Now over 8000 embryos fertilized by Japanese Black bull, as Kitaguni 7 -8 or Mitsufuku, famousbulls as high marbling score of progeny tests were sold to dairy farmers and transferred to their dairy cattle every year. 2. Embryo splitting for identical twins is demonstrated an useful tool to supply a bull for semen collection and a steer for beef performance test. According to the data of Dr.Hashiyada (2001), 296 pairs of split-half-embryos were transferred to recipients and 98 gave births of 112 calves (23 pairs of identical twins and 66 singletons). 3. A blastomere-nuclear-transferred cloned calf was born in 1990 by a joint research with Drs.Tsunoda, National Institute of Animal Industry (NIAI) and Ushijima, Chiba Prefectural Farm Animal Center. The fruits of this technology were applied to the production of a calf from a cell of long-term-cultured inner cell mass (1998, Itoh et al, ZEN-NOH Central Research Institute for Feed and Livestock) and a cloned calf from three-successive-cloning (1997, Tsunoda et al.). According to the survey of MAFF of Japan, over 500 calves were born until this year and a half of them were already brought to the market for beef. 4. After the report of "Dolly", in February 1997, the first somatic cell clone female calves were born in July 1998 as the fruits of the joint research organized by Dr. Tsunoda in Kinki University (Kato et al, 2000). The male calves were born in August and September 1998 by the collaboration with NIAI and Kagoshima Prefecture. Then 244 calves, four pigs and a kid of goat were now born in 36 institutes of Japan. 5. Somatic cell cloning in farm animal production will bring us an effective reproductive method of elite-dairy- cows, super-cows and excellent bulls. The effect of making copy farm animal is also related to the reservation of genetic resources and re-creation of a male bull from a castrated steer of excellent marbling beef. Cloning of genetically modified animals is most promising to making pig organs transplant to people and providing protein drugs in milk of pig, goat and cattle.

Hormone Analysis during Artificial Estrus Induction in Korean Black Goats (흑염소의 인공수정 발정동기화 처리방법에 따른 호르몬 변화분석)

  • The purpose of the study is to investigate the change in the blood progesterone (P4) and estrogen (E2) levels when applying different estrus induction protocols to Korean black goats, and this was done to gain understanding about their reproduction physiology. For the experiment, we performed three estrus induction protocols that are commonly used in bovine: controlled internal drug release (CIDR) + prostaglandin $F2{\alpha}$ ($PGF2{\alpha}$), $PGF2{\alpha}$ + gonadotropin-releasing hormone (GnRH) + $PGF2{\alpha}$, and CIDR + $PGF2{\alpha}$ + PMSG. The P4 and E2 concentrations showed different patterns until the last treatment of the three protocols. However, similar concentration patterns were shown after the last treatment in all the protocols. In conclusion, we monitored the blood P4 and E2 levels in Korean black goats following three different estrus induction protocols. Our findings may be used in other breeding programs of Korean black goats, such as artificial insemination and embryo transfer.

Prolificacy and Its Relationship with Age, Body Weight, Parity, Previous Litter Size and Body Linear Type Traits in Meat-type Goats

  • Data on age and body weight at breeding, parity, previous litter size, days open and some descriptive body linear traits from 389 meat-type, prolific Black Bengal goats in Tripura State of India, were collected for 3 and 1/2 years (2007 to 2010) and analyzed using logistic regression model. The objectives of the study were i) to evaluate the effect of age and body weight at breeding, parity, previous litter size and days open on litter size of does; and ii) to investigate if body linear type traits influenced litter size in meat-type, prolific goats. The incidence of 68.39% multiple births with a prolificacy rate of 175.07% was recorded. Higher age (>2.69 year), higher parity order (>2.31), more body weight at breeding (>20.5 kg) and larger previous litter size (>1.65) showed an increase likelihood of multiple litter size when compared to single litter size. There was a strong, positive relationship between litter size and various body linear type traits like neck length (>22.78 cm), body length (>54.86 cm), withers height (>48.85 cm), croup height (>50.67 cm), distance between tuber coxae bones (>11.38 cm) and distance between tuber ischii bones (>4.56 cm) for discriminating the goats bearing multiple fetuses from those bearing a single fetus.

A Comparative Study on Fresh and Frozen Embryo Transfer after Superovulation in Black Bengal Goats(Capra-hircus)

  • The experiment was divided into two phases. In phase-I fresh embryos were transferred and in Phase-II frozen embryos were transferred. Embryos were collected by using Dulbecco's phosphate buffered saline. In phase-I total of 65 ova were collected out of 107 ovulation in 18 goats. Recovery of ova was 60.74%, of which 51 (78.46%) was fertilized. Sixteen embryos were transferred to 10 recipient goats and kidding was observed in 6 goats, that produced 10 kids. Thus, 62.50% embryo survival and 60% kidding were achieved in phase-I. In phase-II of the experiment, 17 regular cyclic Black Bengal goats were used. The main purpose was to study the viability of caprine embryos after cryopreservation. In this phase the embryos were collected and frozen using Bio-cool freezers. A two step addition of cryoprotectants (5% glycerol and 10% glycerol) and three-step dilution of cryoprotectants with 1mole (M) sucrose was used. Embryos were preserved for 10 to 45 days. Out of 27 embryos preserved, 18 were recovered after freezing and thawing (37$^{\circ}C$ water bath) with 33.33% embryonic loss. Seventeen frozen and thawed embryos were transferred in 9 recipient goats, out of which kidding was observed in 6 goats and 7 kids were produced, giving a 66.66% kidding and embryo survival of 41.17%. The technique utilized for fresh and frozen embryo transfer can be successfully utilized to produce goats of superior genetic merits. The protocol used for addition of cryoprotectant, freezing, thawing and dilution was found suitable for caprine embryo freezing.

A Study of Ku on the costume of the variant races dynasty in China-based on Khitai, Mongols and Manchus costume- (중국 민왕조 복식에서 에 관한 연구)

  • The purpose of this study is to investigate the Ku which was worn by nomadic tribes which was Khitai, Mongols and Manchus. The results of this study can be summarized as follows. 1. The fur clothes were very important clothes that nomadic tribes put on in order to protect them against the cold. The Ku was an outfit for protection against the cold. It was made of ani-mal fur and its style was tight sleeve and long dress. 2. The Ku was worn in all social classes including emperor, nobles and the masses in ancient China and Asia dynasty. After the nomadic tribes estabilished dynasty in China the style of Ku was changed. All kinds of animal fur was used to make the Ku but the volume of fur, kinds and colors were classified severely by their social position. In Khitai dynasty the Ku was worn in same style. But the emperor wore the Ku of silver marten. The noble class wore the Ku of purple, black and blue marten and silver squirrel skin. The masses wore the Ku of the other colors marten, sheep, squirrel and dessert fox skin. In Mongol dynasty, the emperor wore the Ku of purple marten, silver squirrel, silver fox and black fox skin. The nobles wore the Ku of the other colors marten and squirrel. The masses wore the Ku of degraded animal fur. Before the Manchus estabilished the varient races dynasty in China, the rich man preferred the Ku of marten, squirrel, fox, goat furs. But the poorman preferred the Ku of cow, horses, pig, sheep, cat, dog, snake and deer skin. After the manchus conquered the China, the manners of dress were changed. Generally the fur did not appear in right side of garment, but high quality or noble fur appeared outside. Sometimes the inside or outside of Ku was made of silk fabrics and sleeves, neck-band was made of the high quality fur.

Follicular Stimulation and Laparoscopic Ovum Pick-up (LOPU) in Repeatedly Superovulated Korean Black Goats (반복적인 과배란 처치 경험이 있는 한국흑염소에서 난포 자극 및 복강경을 이용한 난포란 채취(LOPU))

  • Laparoscopic ovum pick-up (LOPU) is a convenient method for collecting oocytes in small ruminants. LOPU has the advantage of being a less invasive means of oocyte collection, thereby allowing for a repeated usage of the oocyte donor animals. A total of 25 Korean black goats were used in the winter season (December to February) and LOPU was applied to the goats which had been treated for superovulation more than two times during the last twelve months. Estrus was synchronized with an intravaginal insert containing 0.3 g progesterone for 10 to 12 days. Ovaries were hyperstimulated with eCG 1,000 IU oneshot, FSH with eCG (50 mg / 1,000 IU; 70 mg / 500 IU; 70 mg / 1,000 IU) oneshot or FSH multiple-shot with eCG oneshot ($20mg{\times}6/300IU$) given intramuscularly 72 h prior to LOPU. For these groups, the number of follicles (mean ${\pm}$ SEM) observed which developed to larger than 2 mm in diameter were $1.6{\pm}2.5$, $4.3{\pm}3.1$, $5.5{\pm}4.2$, $6.6{\pm}2.1$ and $8.8{\pm}7.8$, respectively. Oocytes were aspirated by using OPU needles and a vacuum pump. The overall oocyte retrieval rates were 41.4%. Oocytes were matured in TCM-199 supplemented with 10% (w/v) bovine serum albumin + $10{\mu}g/ml$ FSH + $1{\mu}g/ml$ $17{\beta}$-estradiol for 27 h at $39^{\circ}C$ in 5% $CO_2$ in air. Oocytes were parthenogenetically activated by ionomycin combined with 6-diethylaminopurine (6-DMAP). Total oocyte maturation and cleavage rate were 67.3% and 78.8%, respectively. In summary, LOPU is a useful oocyte collection method in Korean black goats that can provide immature oocytes for transgenesis or nuclear transfer.

COMPARATIVE STUDY ON THE DETECTION OF PORPHYROMONAS ENDODONTALIS BY ANAEROBIC CULTURE, IIF AND DNA PROBE METHOD IN INFECTED ROOT CANALS (감염 근관에서 혐기성 배양법과 간접 면역 형광법 및 DNA 프로브법에 의한 Porphyromonas endodontalis의 검출에 관한 비교 연구)

  • There are many advantages when using IIF and DNA probe methods over anaerobic culture method in that they are time-and effort-saving, more precise and more sensitive. Furthermore, in IIF and DNA probe methods, the detection is possible only with small amount of bacteria, the quantitative analysis is possible, and the cell viability is not necessary. The purpose of this study is to observe the incidence of P.endodontalis by carrying out anaerobic culture, IIF and colony lift using DNA probe method respectively, and to compare these 3 methods in terms of effectiveness and sensitivity in order to identify the most effective detection method. 30 teeth with at least one clinical symptoms, with single canal, and with pulp necrosis were sampled. For sampling bacteria, access cavity was prepared after disinfecting tooth and its surroundings. Then the paper point was inserted up to the periapical area, leave there for a while, and finally it was placed into PRAS Ringer's sol. and PBS sol. In anaerobic culture method, P.endodontalis was identified by biochemical tests after subculturing black and brown colonies which were produced after 7 days of incubation on BAP and Brucella BAP in anaerobic chamber. To identify P.endodontalis in IIF method, species-specific polyclonal rabbit-antisera of P.endodontalis(ATCC 35406) was reacted with sampled PBS sol. dispensed onto glass slide, and then P.endodontalis was examined by phase contrast microscopy after incubating with Goat anti-rabbit lgG conjugated to Fluorescein isothiocyanate. For colony lift using DNA probe method, membranes were laid over colonies on the surface of BAP and were hybridized with cloned DNA probe of P.endodontalis. The existence of P.endodontalis was then identified by the methods of chemiluminescent detection and color metric detection. Black colony was found in 11 teeth out of 30 teeth and P.endodontalis was detected in 6 teeth (20 %) by anaerobic culture method, 16 teeth (53 %) by IIF method, and 7 teeth (23 %) by DNA probe method. IIF method is significantly better in detecting P.endodontalis than DNA probe method and anaerobic culture method. There was no significant differences between DNA probe method and anaerobic culture method. There was significant correlation between the formation of black colony and the existence of P.endodontalis. The probability of detecting P.endodontalis when black colony being present is 2.89 times higher than when not being present. There was significant relationship between the foul odor of clinical symptoms and P.endodontalis. The sensitivity of existing P.endodontalis when foul odor being present was 93.75 %, while the specificity of not existing P.endodontalis when foul odor not being present was 28.57 %. These results suggested that the probes of P.endodontalis will be used to decide the method and prognosis in endodontic treatments.

Effects of Crude Protein Levels in Total Mixed Rations on Growth Performance and Meat Quality in Growing Korean Black Goats

  • This study was carried out to investigate effects of crude protein (CP) levels in total mixed rations (TMR) on growth performance and meat quality in growing Korean black goats. Thirty-six 6-month-old growing black male goats (initial body weight =17.95${\pm}$0.13 kg) were used. The goats were randomly divided into four treatments and 9 animals were allotted to each treatment. In terms of treatments, CP levels in TMR were 14, 16, 18, and 20% of dry matter (DM) and all diets were isocaloric (2.4 Mcal ME/kg DM). The experiment lasted for 20 weeks with a 15-day adaptation period. After the end of the trial, five goats from each treatment were slaughtered to measure carcass characteristics and meat quality. Average daily gain (ADG) significantly (p<0.05) increased as dietary CP levels increased, but the highest ADG was found on the 18% CP treatment. The higher CP levels in TMR significantly (p<0.05) decreased feed conversion ratio, but the lowest ratio was on the 18% CP treatment, suggesting a higher bioavailability of nutrients by balanced energy and protein levels on this treatment. There was no significant difference in dressing percentage among treatments, but there was a tendency for levels more than 16% CP in TMR to have a comparatively higher dressing percentage than the 14% treatment. The percentages of meat, fat and bone were not significantly influenced by dietary CP levels. Dietary CP levels did not influence chemical composition of goat meat. Furthermore, there was no significant difference among treatments in any of the sensory parameters except for tenderness. Tenderness was similar for 18 and 20% treatments, which were significantly (p<0.05) higher than the 14% treatment. The results from this study suggested that an adequate CP level in TMR for achieving optimal growth performance and meat quality of growing Korean black goats might be 18% of DM, and that dietary CP level above 18% seemed not to further increase growth performance and meat quality.

Effects of Feeding Total Mixed Rations Containing Different Winter Forage Crop Silages on Feed Intake, Nutrient Digestibility and Blood Characteristics in Korean Black Goats (동계사료작물 사일리지를 첨가한 TMR 급여가 흑염소의 사료섭취량, 영양소 소화율 및 혈액성상에 미치는 영향)

  • This study was conducted to determine the feed value of total mixed rations (TMR) containing different winter forage crop silages on feed intakes, daily gain, nutrient digestibility, and nitrogen retention in Korean black goats. A total of 12 male goats were used in this study and each goat was housed in individual metabolism crates. The treatments were four diets, consisting of whole crop barley silage (T1), rape silage (T2), rye silage (T3), and Italian ryegrass silage (T4). The experiment was designed as a $4{\times}4$ Latin square arrangement in three replicates. Daily intakes for dietary dry matter (DM), organic matter (OM), crude protein (CP) per head were highest in rape silage treatment, and lowest in whole barley silage (p<0.05). Digestible intakes for CP and acid detergent fiber were highest in rape silage treatment. The daily gain of rape, rye, and Italian ryegrass silages were significantly (p<0.05) higher than that of whole crop barley silage. Digestibility of DM. CP, fiber were highest in whole crop barley silage, and lowest in rape silage (p<0.05). Nitrogen (N) intake and N retention were significantly highest in rape silage and lowest in whole crop barley silage (p<0.05). The results showed that, among different winter forage crop silages, rape silage incorporation into TMR had the best performances indicating increases of feed intakes and N retention in Korean black goats.
