연골 재생에 대한 실크 단백질의 가능성 (The Possibility of Silk Protein to the Chondrogenesis)

  • 최근 다양한 생체재료를 이용하여 연골재생과 관련한 많은 연구가 진행되고 있다. 실크단백질은 생체적합성이 뛰어나며, 우수한 기계적 강도를 가지고 있는 천연 고분자 물질로서 최근 생체재료로 사용하기위한 연구가 세계적으로 많이 이루어지고 있다. 본 연구는 실크단백질이 연골재생에 효과가 있는지를 확인하기위하여 수행되었다. 우리는 연골세포를 코뼈로부터 분리하고, 3종류의 배지 (DMEM, DMEM/F12, RPMI)와 서로 다른 농도의 ascorbic acid를 사용하여 최적 배양조건을 확립하였다. 그 결과 우리가 분리한 연골세포는 10% FBS와 $100{\mu}M$ ascorbic acid가 함유된 DMEM배지에서 가장 잘 생장하였다. 연골에 대한 실크의 영향을 관찰하기위해서, 실크 피브로인 용액을 제작하고 이를 멸균한것과 멸균하지 않은 것으로 구분하여 연골세포 배양 시 첨가하여 연골분화에 대한 마커인자인 제2형 콜라겐의 발현량을 측정하였다. 멸균하지 않은 실크 피브로인 첨가시 제2형 콜라겐의 발현량이 2.7배 증가하였으나, 멸균된 실크 피브로인의 첨가는 제2형 콜라겐의 발현량을 오히려 감소시켰다. 또한 실크 피브로인은 제10형 콜라겐의 발현을 증가시키는 것을 확인하였다. 이 효과는 특히 연골세포를 3차원 배양할 때 더 컸다. 본 연구결과를 통하여 우리는 연골을 재생하는데 있어서 실크 단백질을 가능성을 보았으며, 향후 연구에서 연골재생과 실크의 관계를 좀 더 정밀하게 파악하고자 한다.

혈액대용물질의 제조 및 특성 평가 (Estimate of Characteristics and Manufacture of Blood Substitutes)

  • 본 연구는 헤모글로빈을 계란의 인지질로 microencapsulation한 hemosome과 PFC 유화액을 제조하여 산소전달 특성을 향상시키고자 하였다. 실험결과 fluosol-DA를 sonication하여 평균입자크기가 437 nm의 안정성 있는 유화액을 제조할 수 있었으며, glycerol의 농도를 증가시킴으로써 유화시간을 줄이고 안정도를 높일 수 있었다. 소 혈액으로부터 추출한 헤모글로빈을 달걀노른자로부터 추출한 인지질로 microencapsulation하여 직경 $0.8{\mu}m$의 hemosome을 얻을 수 있었다. Hemosome의 산소포화도 곡선은 정상혈액의 적혈구와 같이 S자형을 보였으며 $P_{50}$은 24 mmHg이었다. Hemosome과 혈액을 1:4(V/V%)의 혼합한 혼합용액에서의 산소 포화도는 정상혈액과 유사한 결과를 보였으며, PFC 유화액과 혈액을 1:4(V/V%)로 혼합한 혼합용액에도 정상혈액과 유사한 결과를 보였다.

다공성 폴리우레탄을 이용한 동정맥 누관의 개발 (Development of U-shaped Arterialvenous Shunt Using Porous Polyurethane)

  • 다공성 인공 혈관을 제조하기 위한 새로운 기술에 대하여 연구하였다. 제조방법을 달리하여 만든 다공성의 시편을 준비하여 여러 가지 물성을 비교하였다. 용매/비용매 교환법에 의한 다공성 부여 방법과 고분자 용액의 냉각에 따른 상전이 현상을 이용하여 고분자 용액의 농도와 냉각속도를 달리하여 인공 혈관 시편을 제작하였다. 고분자 용액의 농도가 감소할수록 그리고 냉각속도가 작을수록 인공 혈관 재료로서 유리한 물성을 지닐 수 있음을 알 수 있었다. 위의 방법으로 다공성을 조절함으로써 재료의 기계적 물겅 또한 조절할 수 있음을 알 수 있었다. 냉각 속도를 조절하여 다공성을 부여하는 방법을 이용하여, 고안된 몰드를 사용하여 기존에는 만들기 어려웠던 균일한 두게의 U자형 소구경 인공 혈관을 만들 수 있었다. 비교적 간단한 도구로 균일한 물성을 지니며 원하는 물성의 인공 혈관을 제조할 수 있었다. 온도를 조절하여 고분자 박막에 다공성을 부여하는 기술은 다양한 기능의 의료용 고분자에 접목하여 필요로 하는 기능성을 부여할 수 있는 중요한 기술로 이용할 수 있을 것이다.

센서 응답의 Time-Profile 을 이용한 전자 후각 (E-Nose) 시스템의 Vapor 인식 성능 향상 (Improved Vapor Recognition in Electronic Nose (E-Nose) System by Using the Time-Profile of Sensor Array Response)

  • 전자후각(I-nose)시스템은, 전통적으로 음식물이나 플라스틱 제재의 생산에서 자동화된 품질관리 시스템에 사용되어 왔으나, 최근 호흡가스 등을 통해 당뇨, 호흡 및 소화기 질환과 감염 등을 검사하는 진단영역으로 그 응용분야를 확대하고 있다 이러한 질병과 연관이 없는 휘발성 유기화합물(volatile organic compound, YOC) 에 대하여 I-nose 의 센서어래이는 센서 물질과 휘발성 화합물 사이의 반응으로 인해 고유한 반응을 보이며, 신호의 profile 에 그 흔적을 남긴다. 본 연구에서는 센서어래이의 반응 신호를 profile 형태로 유지 및 분석함으로써 I-nose 의 가스시료 인식 성능을 보다 향상시킬 수 있는 방법을 제안하였다. 신호의 profile 에는 패턴인식을 위한 기존의 개별적인 특성(feature) 보다 많은 정보가 들어 있으며, 이를 가스의 구분에 효과적으로 이용하기 위해 디지털 영상처리에서 사용되는 패턴 매칭을 응용한time-profile방법을 새롭게 제안하였다. 제안된 방법의 검증을 위해, 반도체 공정에 의해 제작된 16채널의 초소형 가스센서 어래이를 사용해 측정된 8종류의 각기 다른 가스시료들에 대하여, 동종 및 이종 가스간의 매칭의 정도를 산출하였다. 기존의 방법과 비교한 결과 동종과 이종 가스간의 뚜렷한 구별이 가능하여 이를 패턴인식에 사용하면 E-nose의 가스 인식 성능을 향상시킬 것으로 전망된다.

세포 배양 가능한 커버슬립형 초음파 변환자 (Cell-cultivable ultrasonic transducer integrated on glass-coverslip)

  • 초음파 뇌 자극술을 통하여 뇌 심부의 국소 지역에 있는 뇌 세포의 활성화를 유도할 수 있으며, 이를 통하여 저하된 뇌 기능을 치료하는데 효과가 있음이 보고되어 왔다. 반면, 초음파 자극의 종류에 따라 신경 변조의 효율과 방향이 달라질 수 있음이 알려져 있어, 적절한 초음파 자극의 종류를 확립하는 연구가 중요하다. 따라서, 본 논문에서는 이를 효과적으로 최적화 하기 위해 세포 배양시 사용되는 커버슬립 기반의 초음파 변환자를 제안하고자 한다. 균일한 초음파 자극을 전도하기 위해서 폴리머 압전소자(Poly-vinylidene fluoride-trifluorethylene, PVDF-TrFE)를 스핀 코팅하고 패를린 절연층을 상단에 적층시켜 음압 출력을 극대화 시켰다. 개발된 초음파 변환자 융합 커버슬립은 초음파자극기 표면에 배양된 수십개의 신경세포에 균일하고 정확한 초음파 자극을 전달 할 수 있고, 자극에 따른 세포의 반응을 형광 현미경으로 실시간 관찰 가능하다. 따라서, 동일한 초음파 자극에 대한 세포의 반응 신호를 최대 수십개 세포로부터 동시에 획득 가능하므로, 반응 신호를 평균 한다면 낮은 강도의 초음파 자극에 따른 뇌 세포의 미세한 반응을 검출할 수 있을 뿐만 아니라, 초음파 변환자와 물의 표면 등에서 발생하는 정현파에 의한 자극의 왜곡 현상을 줄일 수 있어서 사용자가 원하는 초음파 자극을 정확하게 세포로 전달 가능하다. 이렇게 개발된 초음파 변환자를 통해 변환자 표면에 배양된 별세포에서 6 MHz, 0.2 MPa의 저강도 초음파 자극에 의해 유도된 칼슘 반응을 성공적으로 관찰할 수 있었다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The frame and leaflet of the polymer valve were made from polyurethane, To evaluate the hemodynamic performance of the polymer valve a comparative study of flow dynamics past a polymer valve and a St. Jude Medical prosthetic valve under physiological pulsatile flow conditions in vitro was made. Comparisons between the valves were made on the transvalvular pressure drop, regurgitation volume and maximum valve opening area. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate. The results showed that the functional characteristics of the polymer valve compared favorably with those of the mechanical valve at higher heart rate. 30621 T00401030621 ^x Explosive evaporative removal process of biological tissue by absorption of a CW laser has been simulated by using gelatin and a multimode Nd:YAG laser. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. One of the major problems in EIT has been the inaccessibility of internal voltage or current data in finding the internal impedance values. We propose a new image reconstruction method using internal current density data measured by NMR. We obtained a two-dimensional current density distribution within a phantom by processing the real and imaginary MR images from a 4.77 NMR machine. We implemented a resistivity mage reconstruction algorithm using the finite element method and sensitivity matrix. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.   • 본 연구에서는 관상의 Poly(lactic-co-glycolic acid) (PLGA) 담체에 대한 인간의 초자연골과 탄성연골의 형성정도를 살펴보았다. 담체는 PLGA의 분자량에 따라서는 110,000 g/mol과 220,000 g/mol을 비교하였고 내경 유지를 위하여 내경측에는 220,000 g/mol, 외경측에는 110,000 g/mol 의 복합체를 만들거나, 비분해성 고분자 폴리에틸렌 튜브와 110,000 g/mol PLGA의 담체와의 결합도 시도하였다. PLGA 담체들은 주사전자현미경으로 단면 구조를 관찰하였다. 각각의 담체에 20세 미만의 환자들의 비중격에서 채취된 초자연골과 귀에서 채취된 탄성연골에서 분리한 연골세포를 심었다. 분리된 연골세포는 두 번의 계대배양을 거쳐 각각의 PLGA 담체에 심었고 일주일동안 생체 외 환경에서 배양하였다. 각각의 세포와 담체의 복합체를 nude mouse의 배부 좌, 우로 피하조직에 이식하고 8주 뒤 H&E 염색으로 조직 검사를 시행하였다. 110,000 g/mol의 PLGA담체의 연골조직은 잘 형성되어 있었지만 그 내경은 유지되지 못하였다. 반면 220,000 g/mol의 PLGA담체의 연골조직은 내경은 유지하였으나 연골조직이 부분적으로 형성되어 있고 성숙한 연골조직의 양이 많지 않았다. 초자연골 세포에 비교하여 탄성연골 세포가 같은 조건하에서 연골조직을 더 많이 형성한 것으로 나타났다. 관상의 유지를 위하여 220,000 9/mol PLGA 담체를 내경측에 110,000 g/mol PLGA 담체를 외경측으로 한 담체에서는 연골조직 형성이 잘 되지 않았으나 내경측에 폴리에틸렌 튜브를 끼운 110,000 g/mol PLGA 담체에서는 조직 형성과 내경유지가 잘 되었고 원래의 담체와 거의 유사한 형태로 유지되었다. 분화된 연골세포도 조직 소견으로 확인할 수 있었다. 이 1mm 내경의 관상 연골조직은 인공 기관지나 식도 등을 위한 동물 실험과 인공 합성 튜브의 대체 등 앞으로 많은 응용분야가 기대된다.
