  • Endothelin (ET) is a 21 amino acid peptide with multifunctional effects on the vasculature as well as a variety of other cell types such as respiratory, gastrointestinal, urogenital, endocrine, central nervous systems, and others. Endothelin has emerged as a modulator by autocrine and paracrine actions for many cellular activities, including vasoconstriction, cell proliferation, hormone production, neurotransmitter and/or neuromodulator. The endothelin family consists of three closely related peptides, ET-1, ET-2, and ET-3 derived from separate genes, such as chromosome 6, 1, and 20, respectively. ET-1 is the predominant isoform produced in the cardiovascular system and about which most is known. Endothelin receptors are seven-transmembrane GTP-binding protein-coupled receptors, which are classified into endothelin-A (ETA) and endothelin-B (ETB) receptors. Interestingly, recent evidence is accumulating to suggest that ET -1 may contribute to a variety of pain states such as allodynia and hyperalgesia in animals and humans. Therefore, in this review the biological characteristics and contraction-related mechanism of endothelin-1 in mammalian cells will be summarized. Especially, we focus on multifunctional roles for ET-1 in noxious stimulation-induced pain for the study of pain specialized physical therapy.

염건 조피볼락(Sebastes schlegeli) 제조 중 ADH 및 ALDH의 활성변화 (Changes in Alcohol Dehydrogenase (ADH) and Acetaldehyde Dehydrogenase (ALDH) Activity during the Processing of Salt-Dried Rockfish Sebastes schlegeli)

  • The objective of this study was to determine the processing conditions for salt dried rockfish Sebastes schlegeli by sun drying and cold-air drying, as measured by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. We processed salt dried rockfish samples. The salinity of rockfish samples was within 1% following salting with 25% salt brine for 3 h. The moisture content of salt dried rockfish was found to reduce linearly from 70.12 to 39.5 g/100 g over the same time interval. The water activities of salt dried rockfish by sun and cold-air drying were 0.94 and 0.87, respectively, after three days of drying. Acid values (AV) were 10.71 and 5.96 mg KOH/g, respectively, after the three day drying period. The ADH activity in a water extract from salt dried rockfish following sun and cold-air drying for 24 h was 228.5% and 226.1% at 13.3 mg/mL, respectively, and was higher than that when drying lasted for 48 and 72 h. The ALDH activity was not affected but both ADH and ALDH activity tended to decrease as the drying time increased from 24 to 72 h. The conditions of processing for the best quality of salt dried rockfish were determined to be drying with a cold-air system for 24 h. These results indicated that water extracts from salt dried rockfish have valuable biological attributes owing to the metabolizing of alcohol and can provide useful information for the design of drying systems for salt dried rockfish.

인공부도의 생물학적 처리 기술 비교 연구: 인공부도의 조류의 저감 효과 개선을 위한 여재와 물벼룩 적용 (Comparative Study on Biological Technology in Artificial Floating Island: Application of Media and Daphnia to Algal Biomass Control)

  • 호소의 경관성을 향상 시킬 수 있은 자연정화법인 인공식물섬을 설계하고 수질개선 효율을 향상시키기 위하여 여재, 식생과 물벼룩을 혼용하는 방법을 개발하였다. 따라서 조류 제거 효율을 알아보기 위한 각 혼합시스템은 G-BD-여재와 물벼룩, G-OB-식물과 여재, G-BOD-여재 식물과 물벼룩, G-C-대조군으로 실험 및 평가하였다. 각 혼합시스템 현장적용성을 평가하기 위하여 chl-a, TN과 TP의 제거효율을 조사 하였고, 그 결과 G-BD의 평균제거 효율은 69.24%, 16.61%, -0.61%; G-OB는 평균 68.39%, 14.11%, 10.52%; G-BOD는 평균 78.30%, 6.69%, 25.09%; G-C는 평균 35.42%, -3.47%, -25.18%로 조사되었다. 결과에 따라 제안하면 여재, 식생과 동물플랑크톤을 혼용한 시스템이 조류제어방면에서 효율적으로 나타났다. 하지만 영양염류 제어관리에서는 식생과 여재를 혼합한 시스템이 가장 효율적이다.

대청호 저토의 N, P및 Si 영양염 함량과 조류생장잠재력 (The Contents of Nitrogen, Phosphorus, Silicon Nutrient and Algal Growth Potential (AGP) in the Sediment of Taechong Reservoir)

  • 대청호내 3개 지점에서 저토의 영양염을 조사하여 수층과 식물플랑크톤에 대한 생장 잠재력에 미치는 영향을 파악하였다. 저토-수층의 경계면에서 DO는 0.5 mg $O_{2}$/l 이하로서 무산소 상태였다. 공극수의 전기전도도는 저토보다 1.9${\sim}$2.6배 더 높았고 수심이 얕을수록 변동폭이 컸다. Eh는 -12${\sim}$-148mV 범위였고 가비중은 1.17${\sim}$1.30g/cm$^{3}$ 범위로서 지역간에 차이가 크지 않았다. 함수율과 유기물 함량은 각각 58%${\sim}$72%, 8${\sim}$13% 범위로서 댐 부근에서 높았고 옥천천에서 낮았다. 저토의 입도 조성은 사질이 97%를 차지하였다. 저토내 총세균수와 규조류의 세포밀도는 수심이 깊은 하류에서 높았다. 공극수와 치환성의 질소와 인 함량에서 입자성이 용존성보다 풍부하였고, 무기 질소로는 NH$_{4}$가 대부분이었다. 또한 유기와 무기 질소 형태로 볼 때, 공극수내 질소함량은 무기 질소가 많았고, 치환성 질소는 유기 질소가 많았다. 공극수 인 함량에서 용존 유기 인이 많았고 치환성은 무기 인이 많았다. 저토내 규소는 치환성이 최대 63%로서 공극수보다 훨씬 많았다. 대청호의 저토는 유기물이 풍부한 부영양 상태로 평가되었다.

자동배양기를 이용한 미생물 검출 (Detection of Microbial Growth in an Automated Culture System)

  • 미생물 자동배양기를 이용한 감염성 물질의 검출은 시험적 오차의 경감은 물론 분리율 향상과 시간 단축을 가능하게 하였다. BacT/ALERT 3D 자동배양기는 미생물성장 시 발생되는 이산화탄소를 비색법으로 검출하는 장비로서 임상시료를 이용한 미생물 배양과 검출에 이용되어왔다. 자동배양기의 효율성을 검증하고 의약품의 무균 시험에 적용가능한지를 평가하기 위하여 6종의 세균을 이용하여 중식 및 검출특성을 분석하였다. 3종의 호기성세균 Pseudomonas aeruginosa, Micrococcus Iuteus, Bacillus subtilis와 Staphylococcus aureus는 1 CFU가 31.44 시간 내에 검출되었고, 혐기성균인 Clostridium sporogenes는 15.96시간에 균의 중식이 감지되었다. 저성장 혐기성세균인 Propionibacterium acnes는 $10^4$ CFU의 균수에서 검출에 129.36시간이 소요되었다. 이 같은 결과는전통적 직접 배양법보다 검출감도에 있어서 $10\sim10^5$배 높고 검출시간을 $2\sim10$10시간 단축한 것으로서 자동배양법이 미생물 증식과 검출에 효율적임을 말해준다. 따라서 본 시험에서 이용한 자동배양법은 임상시료에서의 감염원 진단뿐 아니라 생물의약품의 무균시험에 이용 가능하다고 판단된다.

  • Bacterial biofilms are analogous to multi-cellular organisms or to clonal communities of higher organisms. In this respect, it can be demonstrated that biofilms display the type of genetic variation associated with macroorganisms. The formation of genetic variants from biofilms is the result of internally produced and regulated signals and the appearance of these variants coincides with dispersal from the biofilm. Moreover, the generation of such variation, has similar outcomes for the bacterial community, where diversification of phenotypic traits ensures that the bacterial community optimizes its chances of success when dispersing or surviving when challenged with environmental stress. These observations increase the complexity with which we view bacteria and also suggest that microbial systems can serve as models for the testing of eukaryotic ecological theories.

국내 논필지 모니터링 자료를 이용한 APEX-Paddy 모델 적용성 평가 (Evaluating the Performance of APEX-Paddy Model using the Monitoring Data of Paddy Fields in Iksan, South Korea)

  • APEX 모형은 다양한 영농 활동의 토양과 물환경에 대한 영향을 필지 및 유역 규모로 평가하기 위해 개발된 모형이다. 최근 APEX의 주요 기작을 바탕으로 논에서의 수도작 운영에 따른 물수지, 양분 유출에 대한 모의가 가능하도록 한 APEX-Paddy가 고안된 바 있다. 본 연구에서는 익산 지역의 논 시험포 모니터링 자료를 이용하여 APEX-Paddy 모형의 적용성을 평가하고자 하였다. 2013년과 2014년의 논 유출량과 부하량 자료를 수집하고 자동보정 툴 APEX-CUTE 4.1과 추가적 수동보정을 통해 모형의 모의성능을 검토하고 한계점을 고찰하였다. 연구결과, 논의 물수지와 질소 배출부하량은 대체로 합리적인 수준의 모의성능을 보이는 한편 유사량과 인 배출부하량 모의에 있어 논의 담수상태 유사배출 기작에 대한 고려가 미흡하여 모의성능에 한계가 있는 것으로 분석되었으며 원인에 대해 고찰하였다. 더불어 자동보정 툴의 적용에 있어 매개변수 민감도를 바탕으로 한 수동보정 결과보다 정확도가 다소 떨어지는 경향을 보여 그 활용에 유의가 필요한 것으로 판단되었다.

Consequences of Water Induced Disasters to Livelihood Activities in Nepal

  • BACKGROUND: The changes in the climatic conditions have brought potentially significant new challenges, most critical are likely to be its impact on local livelihoods, agriculture, biodiversity and environments. Water induced disasters such as landslides, floods, erratic rain etc., are very common in developing countries which lead to changes in biological, geophysical and socioeconomic elements. The extent of damages caused by natural disasters is more sever in least developing countries. However, disasters affect women and men differently. In most of the cases women have to carry more burden as compared to their male counterpart during the period of disasters. METHODS AND RESULTS: This study examines the impact of disasters on the local livelihood especially agriculture and income generating activities of women in three districts of Nepal. The study uses the primary data collected following an exploratory approach, based on an intensive field study. The general findings of the study revealed that women had to experience hard time as compared to their male counterpart both during and after the disaster happen. Women are responsible for caring their children, collecting firewood, fetching water, collecting grass for livestock and performing household chores. Whereas, men are mainly involved in out-migration and remained out-side home most of the time. After the disaster occurred, most of the women had to struggle to support their lives as well as had to work longer hours than men during reconstruction period. Nepal follows patriarchal system and men can afford more leisure time as compared to women. During the disaster period, some of the households lost their agricultural lands, livestock and other properties. These losses created some additional workload to women respondent, however at the same time; they learn to build confidence, self-respect, self-esteem, and self-dependency.Although Nepal is predominantly agriculture, majority of the farmers are at subsistence level. In addition, men and women have different roles which differ with the variation in agro-production systems. Moreover women are extensively involved in agricultural activities though their importances were not recognized. Denial of land ownership and denial of access to resources as well as migration of male counterparts are some of the major reasons for affecting the agricultural environments for women in Nepal. CONCLUSION: The shelter reconstruction program has definitely brought positive change in women's access to decision making. The gradual increase in number of women respondent in access to decision making in different areas is a positive change and this has also provided them with a unique opportunity to change their gendered status in society.Furthermore, the exodus out-flow of male counterparts accelerated the additional burden and workload on women.

생태계보전협력금 제도 활성화를 위한 부과금 개선 방안 연구 (A study on Improvement and Invigoration of Cooperation Charge on Conservation Ecosystem Fund)

  • Korea introduced the cooperation charge on conservation of ecosystem for minimizing damage of ecosystem due to development projects and their effects and for preparing resources for natural environment conservation projects. Advanced countries have made efforts by expanding investment in natural environment conservation and restoring projects to promote prevention of global warming and improvement of biological diversity, are establishing nationwide strategies and plans. To examine the reality of projects by returns of the cooperation charge on conservation of ecosystem, microsite projects in schools and public facilities take the largest share while their project budgets are only about 100~300 KRW, relatively small, which might be attributable to budget restrictions in accordance with the calculating method of levying cooperation charge on conservation of ecosystem and problems of project proceeding in the system of returning fund for projects in general. The conclusion which this study suggests on invigoration of cooperation charge on conservation of ecosystem and its operation are as followings. First, although the cooperation charge on conservation of ecosystem has been introduced in 2001, the amount of imposition per unit area remains unchanged. It is desirable to increase the amount into $1,400KRW/m^2$ as of August, 2011 as the price index has been continuously rising for the past 10 years and the upward adjustment of imposition per unit area should be notified by the decree of the Ministry of Environment every January. Second, the ceiling amount of the cooperation charge on conservation of ecosystem should be abolished. Now the ceiling amount is defined as 1 billion KRW but it was found that there was not any ceiling amount specified according to the comparative analysis of similar systems with the Korean environmental improvement fund. The ceiling should be abolished so that medium level businesses are carried out and ecosystem recovering projects in the true sense of the word can be made smoothly. Third, weight should be introduced in calculating amounts in accordance with ecologic and economic values. Harmony between development and environment can be achieved by applying differentiated weights of constant regional coefficient by use zone and ecologic and economic values. Continuous efforts of improving cooperation charge on conservation of ecosystem should be made more than anything else so that projects by returns of cooperation charge on conservation of ecosystem get effectiveness.

단상 고온 및 중온 혐기성 하수 슬러지의 소화 공정 비교 (Comparison of Single-stage Thermophilic and Mesophilic Anaerobic Sewage Sludge Digestion)

  • 본 연구에서는 하수 슬러지를 대상으로 단상 고온($55^{\circ}C$)과 중온($35^{\circ}C$) 혐기성 소화조의 안정성, 고형물 제거 및 메탄생성을 관찰하였다. 실험 결과 별도의 pH 조절 없이 고온($R_{TAD}$, reactor of thermophilic anaerobic digestion) 및 중온 ($R_{MAD}$, reactor of mesophilic anaerobic digestion) 소화조 내 pH의 경우 혐기 소화 시 안정적인 미생물 활성을 나타낸다고 알려진 6.5-8.0 사이의 값을 소화 기간 내내 유지하는 것을 관찰할 수 있었다. 또한, total alkalinity (TA)의 경우 $R_{TAD}$$R_{MAD}$ 모두 3-4 g $CaCO_3/L$의 높은 값을 안정적으로 유지하였다. 고형물 제거의 경우 순응 기간 후 $R_{TAD}$에서 43.3%, $R_{MAD}$에서 33.6%의 VS 제거율을 보이며 $R_{TAD}$에서 약 10% 높은 VS 제거율은 제거율을 보였다. 유기산의 경우 $R_{TAD}$$R_{MAD}$에서 순응 기간 직후 검출되지 않았다. $R_{TAD}$$R_{MAD}$ 모두 순응 기간 후 안정적인 메탄 생성을 보였으며, $R_{TAD}$에서 $R_{MAD}$에 비해 31.4% 향상된 메탄 생성률이 관찰되었다($R_{TAD}$; 243 mL $CH_4/L/d$; $R_{MAD}$ : 185 mL $CH_4/L/d$). 반면, 두소화조 $R_{TAD}$$R_{MAD}$에서의 메탄 수율은 유사한 값을 보였다.
