Oenococcus oeni의 접종 시기가 국내 MBA 포도의 와인 품질에 미치는 영향 (Influence of Oenococcus oeni Inoculation Time on the Quality of MBA Wine)

  • 본 연구는 Oenococcus oeni의 동시접종(co-incculation)이 국내 MBA(Muscat Baily A) 와인의 품질에 미치는 영향을 연구하였다. 이번 실험을 위해 시료와인이 5가지로 구분되었다. 실험결과는 S+O.Co 와인처럼 Oeococcus oeni의 동시접종이 와인생산 시간의 단축과 고품질의 와인생산 가능성을 보여주었다. 다른 시료 와인들과 비교해 봤을 때, S+O.Co 와인은 가장 많은 Oeococcus oeni의 생균수를 보유했으며, 사과산 함량은 빠른 속도로 상당량이 감소되었으나, 젖산은 빠른 속도로 매우 많은 양이 증가되었다. Oeococcus oeni와 효모의 동시접종은 알코올 발효와 젖산 발효를 14일에서 16일이면 동시에 끝낼 수 있다. 따라서 이번 연구는 Oeococcus oeni가 동시 접종된 와인이 품질 향상, 와인양조 과정의 시간적 단축, 생산성 증대 효과를 보여주었다.

Evaluation of Boar Sperm Viability by MTT Reduction Assay in Beltsville Thawing Solution Extender

  • MTT (3-(4, 5-dimethyl thiazol-2-yl)-2, 5-diphenyl tetrazolium bromide) reduction assay is a method that validates the viability of an active cell. Dehydrogenase in mitochondria converts yellow colored insoluble tetrazolium salt to purple colored water-soluble formazan. Sperm also have mitochondria in the midpiece, therefore sperm viability could be evaluated by MTT reduction assay. Several studies have already demonstrated the capability of application of the MTT reduction assay to sperm of several species in Hepes-BSA buffer. Because most liquid semen was diluted in extender like BTS (Beltsville Thawing Solution), Modena or Androhep when it is used or transferred, semen needed another dilution in Hepes-BSA buffer to assess sperm viability. In this study, we evaluated boar sperm viability especially in BTS extended semen and compared the efficiency of this test with eosin-nigrosin staining. We used the fresh BTS extended semen from a local A.I center. Semen sample was diluted to $3.0{\times}10^7$ sperms/ml in BTS. The rates of formazan production were measured in 96-well microtiter plates immediately and 1h after incubation at $17^{\circ}C$ using a spectrophotometer at wave length 560 nm. Simultaneously, split samples of the same semen were tested, using eosin-nigrosin staining to compare the efficiency of the MTT assay of sperm viability in BTS. The correlation between the results of these tests was calculated using Student-t test and ANOVA. The results revealed a strong correlation between the results of MTT reduction rate and the results that were simultaneously determined by eosin-nigrosin staining at 1 h. In conclusion, the MTT reduction test was an effective and simple method to validate sperm viability and it could be used as a simple tool to evaluate sperm viability in the local A.I center and laboratory.

Rapid Diagnosis of CMT1A Duplications and HNPP Deletions by Multiplex Microsatellite PCR

  • Charcot-Marie-Tooth (CMT) disease and hereditary neuropathy with liability to pressure palsies (HNPP) are frequent forms of genetically heterogeneous peripheral neuropathies. Reciprocal unequal crossover between flanking CMT1A-REPs on chromosome 17p11.2-p12 is a major cause of CMT type 1A (CMT1A) and HNPP. The importance of a sensitive and rapid method for identifying the CMT1A duplication and HNPP deletion is being emphasized. In the present study, we established a molecular diagnostic method for the CMT1A duplication and HNPP deletion based on hexaplex PCR of 6 microsatellite markers (D17S921, D17S9B, D17S9A, D17S918, D17S4A and D17S2230). The method is highly time-, cost- and sample-saving because the six markers are amplified by a single PCR reaction and resolved with a single capillary in 3 h. Several statistical and forensic estimates indicated that most of these markers are likely to be useful for diagnosing the peripheral neuropathies. Reproducibility, as determined by concordance between independent tests, was estimated to be 100%. The likelihood that genotypes of all six markers are homozygous in randomly selected individuals was calculated to be $1.6{\times}10^{-4}$, which indicates that the statistical error rate for this diagnosis of HNPP deletion is only 0.016%.

Sphingopyxis panaciterrae sp. nov., Isolated from Soil of Ginseng Field

  • A Gram-negative, strictly aerobic, motile bacterial strain, designated Gsoil $124^T$, was isolated from a soil sample taken from a ginseng field in Pocheon Province (South Korea). The isolate contained Q-10 as the predominant lipoquinone, plus $C_{18:1}\;{\omega}7c$ and summed feature 4 ($C_{16:1}\;{\omega}6c$ and/or iso-$C_{15:0}$ 2-OH) as the major fatty acids. The G+C content of the genomic DNA was 68.1 mol%, and the major polar lipids consisted of sphingoglycolipid, phosphatidylglycerol, phosphatidylcholine, and phosphatidylethanolamine. A comparative 16S rRNA gene sequence analysis showed that strain Gsoil $124^T$ was most closely related to Sphingopyxis chilensis (98.7%), Sphingopyxis alaskensis (98.2%), Sphingopyxis witflariensis (98.2%), Sphingopyxis taejonensis (98.0%), and Sphingopyxis macrogoltabida (97.6%). However, the DNA-DNA relatedness between strain Gsoil $124^T$ and its phylogenetically closest neighbors was less than 22%. Thus, on the basis of its phenotypic properties and phylogenetic distinctiveness, strain Gsoil $124^T$ should be classified as representing a novel species in the genus Sphingopyxis, for which the name Sphingopyxis panaciterrae sp. nov. is proposed. The type strain is Gsoil $124^T$ (=KCTC $12580^T$=LMG $24003^T$).

  • The magnetism of$Fe_3O_4$ nanoparticles was applied to magnetic bio-panning process for finding specific sequences against $Fe_3O_4$ crystal phase. Vibrating sample magnetometer (VSM) measurement showed that the coercivity of 30 Oe and the saturation magnetization of 55 emu/g were sufficient in controlling particle movement and magnetizing particles in the media, respectively. This ferrimagnetism of nanoparticles practically enhanced panning efficiency by exaggerating centrifuge step and preventing particle loss. Sequencing results showed that histidine which was commonly found in peptide sequences played an important role in the binding onto $Fe_3O_4$ nanoparticle surface. However, various possible motifs were also observed from several neighboring amino acids of histidine.

Lack of association between the VEGFA gene polymorphisms and preterm birth in Korean women

  • Preterm birth (PTB), a pregnancy-related disease, is defined as a birth before 37 weeks of gestation. It is a major cause of maternal mortality and morbidity worldwide, and its incidence rate is steadily increasing. Various genetic factors can contribute to the etiology of PTB. Vascular endothelial growth factor A (VEGFA) gene is an important angiogenic gene and its polymorphisms have been reported to be associated with PTB development. Therefore, we conducted a case-control study to evaluate the association between VEGFA rs699947, rs2010963, and rs3025039 polymorphisms and PTB in Korean women. A total of 271 subjects (116 patients with PTB and 155 women at ≥38 weeks of gestation) were analyzed in this study. The genotyping of VEGFA gene polymorphisms was performed using polymerase chain reaction- restriction fragment length polymorphism. No significant association between the patients with PTB and the control groups was confirmed. In the combination analysis, we found a significant association between PTB and VEGFA rs699947 CC-rs2010963 GG-rs3025039 CC combination (odds ratio, 3.77; 95% confidence interval, 1.091 to 13.032; p = 0.031). The VEGFA rs699947, rs2010963, and rs3025039 polymorphisms might have no genetic association with the pathogenesis of PTB in Korean women. However, the combination analysis indicates the possibility that VEGFA acts in PTB pathophysiology. Therefore, larger sample sets and replication studies are required to further elucidate our findings.

  • Polarization-sensitive optical coherence tomography (PS-OCT) was developed to image highly scattering tissues with accounting for polarization effects in the sample. These polarization-sensitive images can provide additional information on the structure of the tissue because of a polarization state of the light is changed at its interaction with biological tissues.   • The development of detection techniques is needed, in order for regulating authorities to determine whether or not a particular food sample has been irradiated, and label it accordingly so that a consumer's free choice can be exercised. The chemical and physical changes brought about in foods by practical doses of irradiation are very small, and therefore very sensitive methods are required. A number of promising approaches have been developed and evaluated. These include chemical, physical and biological methods ranging from the very simple to highly sophisticated techniques.
