High Alloying Degree of Carbon Supported Pt-Ru Alloy Nanoparticles Applying Anhydrous Ethanol as a Solvent

  • Alloying degree is an important structural factor of PtRu catalysts for direct methanol fuel cells (DMFC). In this work, carbon supported PtRu catalysts were synthesized by reduction method using anhydrous ethanol as a solvent and $NaBH_4$ as a reducing agent. Using anhydrous ethanol as a solvent resulted in high alloying degree and good dispersion. The morphological structure and crystallanity of synthesized catalysts were characterized by X-ray diffraction (XRD), high resolution transmission electron microscope (HR-TEM). CO stripping and methanol oxidation reaction were measured. Due to high alloying degree catalyst prepared in anhydrous ethanol, exhibited low onset potential for methanol oxidation and negative peak shift of CO oxidation than commercial sample. Consequently, samples, applying ethanol as a solvent, exhibited not only enhanced CO oxidation, but also increased methanol oxidation reaction (MOR) activity compared with commercial PtRu/C (40 wt%, E-tek) and 40 wt% PtRu/C prepared in water solution.

A New Analytical Method to Determine the Purity of Synthetic Fluorophores using Single Molecule Detection Technique

  • A new assay technique to distinguish between pure compounds and the isomeric mixtures has been suggested using single molecule (SM) fluorescence detection technique. Since the number of emission spots in a fluorophorespread film prepared from a genuine dye solution was determined by experimental condition, the deviation of spot numbers from the expected values could be considered to be an indication of lower purity of the sample solution. The lower limit of sample concentration for this assay was determined to be $5{\times}10^{-10}$ M to show uniform number of expected spots within 10% uncertainties in our experimental condition. An individual fluorescence intensity distribution for a mixture of isomers having doubly different emissivities was simulated by adding distributions obtained from Cy3 and nile red (NR) independently. The result indicated that the mixture could be identified from the pure compounds through the difference in the number of Gaussian functions to fit the distribution. This new assay technique can be applied to the purity test for synthetic biofluorophores which are usually prepared in small quantities not enough for classical ensemble assays.

  • Many methods have been developed for the remediation of contaminated soil and groundwater. Among those technologies, in-situ bioremediation is most likely to be cost-effective method for petroleum hydrocarbon contamination. But the in-situ bioremediation can require more time to remediate hydrocarbon-contaminated soil and groundwater than other methods. Therefore we intended to save time of in-situ bioremediation using a biological additive to activate indigenous microbes in soil. The additive, 'Inipol EAP 22' stimulates the growth of specific flora, significantly accelerating the speed at which hydrocarbons are biodegraded. And it hans been tested in accordance with protocol approved by the USEPA and is registered on the National Contingency Plan Product Schedule List. In the experiment, three soil samples contaminated with fuel oil were prepared in the same concentration. Inipol EAP 22 was not added to one sample and was added to the other two samples with 5% and 10% of hydrocarbon by weight respectively. And $CO_2$gas derived from bacterial respiration was analyzed in each samples for 15 days. As a result, 145% and 153% of $CO_2$ evolution (microbial respiration) against the sample without 'Inipol EAP 22' occurred in samples with 'Inipol EAP 22' addition of 5% and 10%, respectively

On-line Monitoring of a Glucose Concentration on a Fermentation Process of Wine for an Automatic Control of a Fermentation Process (발효공정 자동제어를 위한 포도주 발효 중 포도당 농도 온라인 측정)

  • A flow injection analysis method (FIA), which analyzes sample conditions after injecting a sample and reagents into a continuous stream, are recognized as the most adequate analyzing method according to the increase of sampling frequency, the decrease of measuring time and the diversity of measuring targets. Specially, the FIA is considered to be used effectively for the control of a fermentation process to produce fermentation food and useful microbial production by activation of a fermentation industry for development of biological materials. In this study, a flow injection analysis sensor unit was developed for on-line monitoring of the fermentation process. The performance was verified by on-line measuring the concentration of glucose of the fermentation process of wine. The glucose concentrations of the samples were measured every 12 hours during the whole fermentation process and compared with those by a HPLC. The concentration relative errors of glucose on the fermentation process of wine showed below 30% within 72 hours and over 50% after the 72 hours. The sensor unit had potential to on-line monitoring of the fermentation process but some problems to overcome for an commercial application.

  • We have developed and been using laboratory order communication system which is a computerized laboratory request and reception system wi th bar code between inpatient or outpatient and the clinical laboratory in Chungbuk National Unversity Hospital. Work flows are as follows: Tests are requested by the physicians through hospital information system without issuing request forms. Bar code stickers containing demographics of patient and other informations such as sample number, slip code and specimen code are printed and attached to smaple tubes. At the department of clinical pathology, smaples are received through the bar code reader. Area numbers are automatically created and laboratory work numbers are determined. Worklists can be issued by each section of laboratory when needed. Our order communication system alleviates the human labor such as specimen labelling and making worklist and reduces clerical errors that occur from sample collection to laboratory analysis.

Improvement of Histopathological Sample Preparation by Employing Microwave Heating Method on Frozen Section Specimens

  • Biological samples can be fixed either by chemical method by using chemical solution or physical methods by using heat treatment. The problem in traditional heat fixation is unsatisfactory quality due to uneven heat conduction in specimen and loss of inner cell contents. Chemical fixation method also bears several intrinsic problems like the limit in specimen size, time consumption in fixative impregnation, and loss of low molecular weight cell components. These factors deteriorate the quality of fixed specimen, thus limit the magnification and contrast of tissue pictures. Microwave heat has been reported to be a good alternative to current chemical methods to overcome these problem. In this study, we tried to introduce the microwave energy method to routine fixation work in hospital. We replaced chemical fixative with saline to provide moderate reaction condition, and used frozen section to reduce time for sample preparation. Temperature was measured at each experiment. The fixation of rat kidney tissue with 2.45 GHz electromagnetic wave and saline showed similar result to the control group fixed with traditional chemical method. Human tumor tissue fixed with 2.45 GHz electromagnetic in frozen section was improved in terms of histochemistry of PAS and immunohistochemistry of tumor marker like cytokeratin. Total turnaround time was reduced from $24\sim38$ h to to $2\sim4$ h. In conclusion, the quality of samples prepared by microwave heating method was at least as good as that of traditional method. If the condition for the fixation of different specimens is standardized, this new method could be applied to routine work in hospital, and could save working time as well.

Estimating the Abundance and Fishing Mortality of Pacific Cod Gadus macrocephalus during the Spawning Season in Jinhae Bay, Korea, Using a Mark-Recapture Method (표지방류 조사를 통한 거제 외포 주변해역 대구(Gadus macrocephalus) 자원량과 어획사망률 추정)

  • We estimated the population size and fishing mortality of Pacific cod Gadus macrocephalus during the spawning season in waters off Woipo, Geoje Island, Korea, using a mark-recapture method. We marked and released 51 cod>50 cm in total length; six were recaptured by local fishermen during the period from December 15 to 31, 2009. The estimated population size was ca. 180,000 and the fishing mortality of the exploitable cod was 26%. Although we could assume a closed population due to the short survey period, we evaluated the uncertainty in the estimates by applying bootstrap resampling because the sample size was small. The estimated 95% confidence interval was 94,000-568,000 for the population size and 8-49% for fishing mortality. Our study demonstrated that the application of mark-recapture methods and bootstrap resampling can be useful in stock assessment for fisheries management in Korea, but requires a larger sample size, spatially extensive coverage, and sophisticated mark-recapture models based on a refined sampling design for reliable stock assessment and biological reference points in sustainable cod management.

Development of an Auto Dilution Unit of Substrate Solutionfor a Flow Injection Type Biosensor (흐름주입식 바이오센서용 기질용액 자동희석 장치 개발)

  • For development of an on-line monitoring unit of fermentation process, an auto dilution unit based on traditional chemical and biological analytical method was developed and the performance was evaluated. The dilution unit was constructed with two syringe pumps and flow direction change valves and fully automated. Total delivery volume of two pumps using distilled water was measured to confirm the operating stability And diluted concentrations of three substrate solutions (glucose, lactic acid, ethanol) were compared with a standard method with a high performance liquid chromatograph (glucose, lactic acid) and gas chromatograph (ethanol). Relative error values of total delivery volume of the pumps were below 3% and standard deviation values were 0.003 (n=5). Relative error values of diluted concentration of the dilution unit measurements were below 2% with 1/10 of dilution ratio and 70, $80{\mu}{\ell}$ of sample volume conditions for glucose and lactic acid, 1/30 of dilution ratio and $70{\mu}{\ell}$ of sample volume conditions for ethanol, respectively. In case of the ethanol, cause of the evaporative characteristics, the relative error values showed over 5% whole experimental conditions.

  • A two-dimensional pulsed doppler system for ultrasonic blood velocity doppler signals is studied and implemented. The second-order sampling method and serial data processing procedures are utillized in the sys- tem, which eliminates the untuning problems at phase channels in the quadrature detection method as well as in the channels of parallel data processing. rho digital signal processor used in this system allows a hardware savings and flexible design options. The efficiency of the various mean frequency estimators in the second-order sampling system is examined by computer simulation as a function of the intersequence sample delay time. The temporal delay for the quadrature component is changed from $1/(4f_o){\;}to{\;}3/(4f_o){\;}and{\;}5/(4f_o)$ where to is the center frequency of the transducer, It is found that autocorrelator is the optimum frequency estimator for the second-order sampling: with !he intersequence sample delay of $1/(4f_o){\;}to{\;}3/(4f_o){\;}and{\;}5/(4f_o)$. The qualitative variation and information proportional to blood velocity in the vessel system are obtained in the VIVO experiments.

  • Optical coherence tomography(OCT) is a noninvasive optical imaging tool for biomedical applications. OCT can provide depth resolved two/three dimensional morphological images on biological samples. In this paper, we integrated an OCT system that was composed of an SLED(Superluminescent Light Emitting Diode, ${\lambda}_0$=1305 nm bandwith= 141 nm), a reference arm adopting a rapid scanning optical delay line(RSOD) to get high speed imaging, and a sample arm that used a micro electro mechanical systems(MEMS) scanning mirror. The sample arm contained a compact probe for imaging dental structures. The performance of the system was evaluated by imaging in-vivo human teeth with dental calculus, and the results indicated distinct appearance of dental calculus from enamel, gum or decayed teeth. The developed probe and system could successfully confirm the presence of dental calculus with a very high spatial resolution($6{\mu}m$).
