청과물 저장에 관한 연구(제3보) -환경압력변화가 Tomato 과실의 저장에 미치는 영향에 대하여- (A Study on the Storage of Fresh Fruits and Vegetables(III) -Effects of the Storage of Tomato fruits by controlled atmospheric pressure-)

  • 1. 품종은 노지에서 재배한 복수2호를 사용하였으며 1971년과 1972년 2회에 걸쳐 Tomato 과실의 성숙시기별 호흡량변황 및 저장중 성분변화에 대하여 조사하였다. 2. 환경압력조절에 의한 Tomato 과실의 호흡억제효과는 SAP 구중 660 Torr. 구가 가장 효과적이었다. 3. 성장중 성숙시기별로 채취한 Tomato의 호흡량 변화는 각채취시기별로 차이가 있었으며 Tomato의 일대에 있어서 시기별 숙기에 이른 과실을 경과일수에 따라 호흡량을 조사하면 전체적으로 호흡량의 증감이 sigmoid 형으로 나타났으며 하나의 climacteric pattern을 나타내 주고 있다. 4. 성장중 성숙시기별로 채취한 Tomato 과실의 환경압력변화에 의한 영향은 어느시기에서나 NAP구보다 SAP구의 호흡량이 적었으며 전 기간중 동일한 경향으로 호흡억제 효과가 있었다. 5. 저장중 Tomato 과실의 중량변화는 NAP 구가 SAP 구보다 현저하며 산, 당 및 Vitamine-C의 변화에 있어서는 SAP구가 NAP구 보다 감소율이 적었다.

Viator vitreocola gen. et sp. nov. (Stylonematophyceae), a new red alga on drift glass debris in Oregon and Washington, USA

  • A new encrusting red alga was found growing abundantly on glass debris items that drifted ashore along the coasts of Oregon and Washington. These included discarded fluorescent tubes, incandescent light bulbs, capped liquor bottles, and ball-shaped fishing-net floats. Field collections and unialgal cultures of the alga revealed that it consisted of two morphological phases: a young loosely aggregated turf and a mature consolidated mucilaginous crust. The turf phase consisted of a basal layer of globose cells that produced erect, rarely branched, uniseriate to multiseriate filaments up to $500{\mu}m$ long with closely spaced cells lacking pit-plugs. These filaments expanded in size from their bases to their tips and released single cells as spores. At maturity, a second phase of growth occurred that produced a consolidated crust, up to $370{\mu}m$ thick. It consisted of a basal layer of small, tightly appressed ellipsoidal-to-elongate cells that generated a mucilaginous perithallial matrix containing a second type of filament with irregularly spaced cells often undergoing binary division. At the matrix surface, the original filaments continued to grow and release spores but often also eroded. Individual cells, examined using confocal microscopy and SYBR Green staining, were found to contain a central nucleus, a single highly lobed peripheral chloroplast without a pyrenoid, and numerous chloroplast nucleoids. Morphological data from field and culture isolates and molecular data (rbcL, psbA, and SSU) show that this alga is a new genus and species which we name Viator vitreocola, "a traveller on glass."

Fine Structural Analysis of Secretory Silk Production in the Black Widow Spider, Latrodectus mactans

  • The spinning apparatus and production of secretory silk from silk gland of the black widow spider, Latrodectus mactans were studied with scanning and transmission electron microscopes. The silk glands were located in seven groups on the spinnerets including each pair of major and minor ampullate, 3 pairs of tubuliform, 1 pair of flagelliform, 2 pairs of aggregate, about 50 pairs of pyriform and over 250 pairs of aciniform glands, respect- ively. Each group of silk gland feeds silk into one of the three spinneret pairs. Secretory silk is synthesized from rough endoplasmic reticulum (rER) of glandular epithelial cells. The secretory silk is transported from toe rER into the secretory vacuoles which are grown up by fusion with the surrounding small vesicles including the secretory silk. The secretory vacuoles, which show a gradual increase in electron density with the process of maturity, are formed without involvement of the Golgi complex, suggesting that they do not play an important role in the processing of the secretory silk. The secretory silk products are released by the mechanism of apocrine secretion, losing part of their cytoplasm. Moreover, another type of silk precursor, possibly protein, appears as granular material, and is also discharged to the luminal cavity.

  • In order to improve national competitiveness through technological research and development, the government puts in R&D budget every year and manages to improve the R&D results. Accordingly, various R&D project management methods have applied for successful advancement of technology and product. TRL is a measurement system developed by NASA to assess the maturity of technology since the 1970s. To apply medical device, the characteristics such as regulation, clinical trials are considered as a significant influence. In this study, we would like to derive PMI(performance measures index) for medical device R&D projects by using TRL and stage-gate model. As a result of this study, it is possible to use the PMI for decision making and evaluation in the R&D projects and believed that the objectivity can be ensured by the approval or certification of regulatory authority.

  • Today, a desirable way to manage livestock manure is to activate its utilization as a resource. The production of high quality liquid fertilizer of livestock manure is very important because it increases the use of various liquid fertilizer. However, the result of mature evaluation with a maturity measuring instrument for liquid fertilizer showed that the deviation of concentration between liquid fertilizer did not bring into uniformity. The result is also making sure that quality management for liquid fertilizer is not smoothly made. Quality evaluation for compost and liquid fertilizer includes physical, biological, chemical and microbiological methods, but a chemical method is mainly being implemented due to fairness and field application. Therefore, this study figured out correlation in feces and urine through regression analysis of livestock manure and tried to create a research plan to carry out efficient quality analysis of managing livestock manure.

Gametogenic Cycle by Quantitative Statistical Analysis and the Biological Minimum Size in Protothaca (Notochione) jedoensis (Bivalvia: Veneridae) in Western Korea

  • The gametogenic cycle, the spawning season and the biological minimum sizes in female and male Protothaca (Notochione) jedoensis were investigated by quantitative statistical analysis. In females, monthly changes in the percents of the follicle areas to the ovarian tissue areas and the percents of the oocyte areas to the ovarian tissue areas increased in February and reached the maximum in April, and then gradually decreased from May to July, with the spawning peak between June and July. In males, monthly changes in the percents of the testicular tissue areas to total tissue areas and the percents of the spermatogenic stage areas to the testicular tissue areas increased in February and reached the maximum in April, and then showed a rapid decrease from May to July. From these data, it is apparent that the number of spawning seasons in female and male P. (N.) jedoensis occurred once a year, from May to July. Therefore, P. (N.) jedoensis in both sexes showed a unimodal gametogenic cycle during the year. Compared the gametogenic cycle by quantitative statistical analysis in 2007 with the previous qualitative results of this species, the results of the gametogenic cycle calculated by quantitative statistical analysis showed some differentiations in the spawning seasons evaluated by the gonad index by qualitative histological analysis. The intervals of the beginning of two spawning seasons showed one month between the results of quantitative and qualitative analyses. The biological minimum sizes (considering to 50% of group sexual maturity) in female and male clams by quantitative analysis of this species are 32.01 mm in shell length in females and 30.58 mm in males, respectively. According to the mean shell length fitted to von Bertalanffy's equation, 30.58 and 32.01 mm in shell length were considered to be two years old. Therefore, we assume that both sexes of this population begin reproduction from two years of age.

  • 토마토 상종(商種) 중(中) "강력미수(强力米壽)"를 노지(露地) 재배(栽培)한 것을 mature green stage에서 수확하여 상온(常溫) 감압하(減壓下)에서 처리(處理)하였을 때의 저장 생리(生理)와 관련(關聯)하는 주요성분(主要成分) 및 호흡량(呼吸量), 숙도(熟度), chlorophyll, 지질(脂質)등의 변화(變化)를 조사(調査)한 바, 1. 과실(果實)의 climacteric rise의 발현(發現)은 감압구(減壓區) 보다 10 일(日)이상 지연되었으며, 2, 처리기간(處理期間) 중(中) 숙도(熟度)의 변화(變化)는 감압정도(減壓程度)에 따라 차(差)가 있어, 대체(大體)로 감압도(減壓度)가 높을수록 지연되었다. 이 관계(關係)는 climacteric rise와도 밀접(密接)한 관계가 있었으며 처리구(處理區)에 관계(關係)없이 dark pink stage와 climacteric maximum는 일치(一致)하였다. 3. 과실(果實)의 색상(色相)과 관계(關係)가 깊은 chlorophyll의 감소(減少)도 상압구(常壓區)가 감압구(減壓區)보다 현저하였다. 4. 과실(果實)의 함유지질(含有脂質)은 중성지질(中性脂質), 인지질(燐脂質). 당지질(糖脂質)이었으며 그중(中) 중성지질(中性脂質)이 46%, 인지질(燐脂質)이 32%, 당지질(糖脂質)이 13%이였다. 중성지질(中性脂質)의 구성(構成)은 sterol ester, triglyc ericle, free fatty acid, 1.3 diglyceride, 1.2 diglyceride, sterol, monoglyceride 의 7종(種)이었으며, 이중(中) triglyceride가 49.3%로서 가장 함량(含量)이 많았다. 인지질(燐脂質)은 phosphatidic acid, cardiolipin, phosphatidyl choline, phosphatidyl ethanolamine, phos phatidyl glycerol, phosphatidyl inositol phosphatidyl serine의 7종(種)으로 구성(構成)되며 이중(中) phosphatidyl choline 및 phosphatidyl ethanol amine이 가장 많이 함유되고 있었다. 5. 저장중(貯藏中) 총지질(總脂質)은 시일(時日)이 경과함에 따라 점차(漸次) 감소(減少)하는 경향(傾向)이었는데 상압구(常壓區)에 비하여 감압구(減壓區)의 변화(變化)가 적었다. 중성지질(中性脂質)은 climacteric rise 및 숙도(熟度)와 관련하여 변화(變化)하였으며 climacteric onset기(期)에. 가장 많이 감소(減少)하다가 다시 상승(上昇)하는 경향(傾向)이었다. 중성지질(中性脂質) 중(中) climacteric rise 및 숙도(熟度)와 관련하여 변화(變化)한 것은 diglyceride 및 sterol ester의 2종(種)이었으며 glyceride가 중성지질(中性脂質) 전량(全量)의 변화(變化)와 동일(同一)한 경향(傾向)인데 반(反)하여 sterol ester은 climactric onset까지 증가(增加)하다가 기후(其後) 감소(減少)하였다. 인지질(燐脂質)도 저장기간(貯藏期間) 중(中) 처리구(處理區)에 관계(關係)없이 다같이 감소(減少)되었는데, 그 정도(程度)는 상압구(常壓區)에 비(比)하여 감압구(減壓區)의 변화(變化)가 적었다. 인지질(燐脂質) 중(中) ph. choline과 ph. ethanlamine은 climacteric rise및 숙도(熟度)와 관련하여 변화(變化)하였다. 중성지질(中性脂質) diglyceride 및 sterol ester와 인지질(燐脂質)의 ph. choline 과 ph. ethanol amine은 climacteric rise와 관련하여 변화(變化)하였다.

다래를 이용한 발효주의 제조 및 이화학적 특성 (Manufacturing and Physicochemical Properties of Wine using Hardy Kiwi Fruit (Actinidia arguta))

  • 다양한 기능성을 가지는 다래를 이용하여 발효주의 제조공정을 확립하였다. 다래과실의 수율 향상을 위하여 후숙기간을 5일로 정하고, 효소제 처리는 0.1% cytolase PCL5를 2시간 처리하였을 때 과즙 수율이 53.5%로 무처리구보다 약 2.4배나 수율이 증가하였다. 다래 발효주 제조시 강한 신맛을 감소시키기 위한 감산처리는 0.1%(w/v) $CaCO_3$를 최적농도로 선정하였다. 다래과실을 파쇄한 후, 각각 가수하여 25%(v/w) 처리구(hardy kiwi wine 25)와 50%(v/w) 처리구(hardy kiwi wine 50)를 준비하였다. 각각 처리구에 설탕을 첨가하여 $24^{\circ}brix$ (252 g/l)로 보당하였고 0.1%(w/v) $CaCO_3$, 0.1%(v/v) cytolase PCL5 효소제, 메타중아황산 칼륨(200 ppm)을 첨가한 후, 주모($1.5{\times}10^7$ cell/ml)를 접종하여 $20^{\circ}C$에서 2주 동안 발효하였다. 숙성은 $4^{\circ}C$에서 8주 동안 진행하면서 다래 발효주의 pH와 총산도는 각각 pH 3.4-3.7과 1.12-1.21%로 유지하였다. 알코올 함량과 환원당은 14.3-14.4%와 15-16 g/l로 유지하였다. 다래 발효주의 주요 유기산과 유리당은 citric acid (6.6-6.7 g/l)와 fructose (2.5-2.7 g/l)로 가장 높게 나타내었다. 주요 향기성분으로는 iso-amyl alcohol, iso-butyl alcohol, phenethyl alcohol, ethyl acetate, acetaldehyde 등이 검출되었다. 다래 발효주의 관능평가 결과, 맛과 향, 종합적인 기호도 모두 hardy kiwi wine 50에서 기호도가 가장 우수한 것으로 확인하였다.

수피${\cdot}$돈분 부산물 비료의 부숙단계별 특성 변화 (Changes in Characteristics of Bark and Piggery Manure By-Product Fertilizers During the Composting)

  • 본 연구는 부산물 비료의 부숙도 판정 기준 설정을 위한 기초자료를 구축하기 위하여 우리 나라에서 가장 널리 사용되고 있는 수피 부산물 비료와 돈분 부산물 비료를 대상으로 원재료로부터 완제품 생산까지 부숙단계별 시료의 물리적${\cdot}$화학적${\cdot}$생물학적 특성의 변화를 측정하기 위해 수행되었으며, 그 결과를 요약하면 다음과 같다. 부숙이 완료됨에 따라 수피 부산물 비료의 경우 유색(black)으로, 돈분 부산물의 경우 암갈색으로 변화되었으며, 수피 부산물의 경우는 43일경과 후에, 돈분 부산물의 경우는 40일경과 후에 원 시료의 냄새가 사라지고 퇴비취가 나는 것으로 나타났다. 완숙된 수피 부산물 비료의 pH와 EC는 각각 pH6.5와 $1dS{\cdot}m^{-1}$에서 안정화되었으며, 돈분 부산물 비료의 경우는 pH7.2와 $6dS{\cdot}m^{-1}$에서 안정화되었다. 부숙이 진행됨에 따라 유기물함량은 점차 감소하여 수피 부산물 비료의 경우 120일 경과후 62%, 돈분 부산물 비료의 경우 40일경과 후 59%로 안정화되었다. 부숙 기간 중 총 질소의 함량은 수피 부산물 비료의 경우 $1.1{\sim}1.5%$, 돈분 부산물의 경우 $1.5{\sim}2.2%$를 유지하였다. 두 가지 비료 모두에서 암모니아태 질소는 초기에 증가하다 중기 이후 감소하였고, 질산태 질소는 계속적으로 증가하였으며, 무기태 질소의 전체 함량은 증가하였다. 부숙이 완료된 퇴비의 유기물/질소비는 수피 부산물 비료의 경우 25에서, 돈분 부산물 비료의 경우 27에서 안정화되었다. 부숙이 진행됨에 따라 CEC는 증가하였는데, 완숙단계에서 수피 부산물 비료의 경우 $87cmol(+)\;{\cdot}\;kg^{-1}$으로, 돈분 부산물 비료의 경우$70cmol(+)\;{\cdot}\;kg^{-1}$으로 증가하였다. 조섬유 중 Cellulose와 Hemicellulose는 부숙이 진행됨에 따라 감소하였으나, Lignin의 비율은 점차 증가하였다. 부산물비료의 부숙단계별 시료의 특성변화는 부산물 비료의 부숙도를 평가하는 지표로 활용될 수 있을 것으로 판단된다.

Genetic correlations between behavioural responses and performance traits in laying hens

  • Objective: The aim of the study was to evaluate genetic correlations between the behavioural profile and performance in laying hens as an indirect answer to the question whether the observed behavioural responses are associated with increased levels of stress in these birds. Methods: The assessment of birds' temperament was carried out using the novel objects test. The behavioural test was conducted in two successive generations comprising 9,483 Rhode Island White (RIW) birds (approx. 4,700 individuals per generation) and 4,326 Rhode Island Red (RIR) birds (approx. 2,100 individuals per generation). Based on the recorded responses, the birds were divided into two groups: a fearful profile (1,418 RIW hens and 580 RIR hens) and a brave/curious profile (8,065 RIW hens and 3,746 RIR hens). The birds were subjected to standard assessment of their performance traits, including SM, age at sexual maturity; ST, shell thickness; SG, egg specific gravity; EW, mean egg weight; IP, initial egg production; and HC, number of hatched chicks. The pedigree was three generations deep (including two behaviourrecorded generations). Estimation of the (co)variance components was performed with the Gibbs sampling method, which accounts for the discrete character of the behavioural profile denotation. Results: The analyses revealed negative correlations between the performance traits of the laying hens and the behavioural profile defined as fearful. In the group of fearful RIW birds, delayed sexual maturation (0.22) as well as a decrease in the initial egg production (-0.30), egg weight (-0.54), egg specific gravity (-0.331), shell thickness (-0.11), and the number of hatched chicks (-0.24) could be expected. These correlations were less pronounced in the RIR breed, in which the fearful birds exhibited a decline in hatchability (-0.37), egg specific gravity (-0.11), and the number of hatched chicks (-0.18). There were no correlations in the case of the other traits or they were positive but exhibited a substantial standard error, as for the egg weight. Conclusion: To sum up the results obtained, it can be noted that behavioural responses indicating fearfulness, i.e. escape, avoidance, and approach-avoidance may reflect negative emotions experienced by birds. The negative correlations with performance in the group of fearful hens may indirectly indicate a high level of stress in these birds, especially in the white-feathered birds, where stronger performance-fearfulness correlations were found. Fearful birds should be eliminated from breeding by inclusion of the behavioural profile in the selection criterion in the case of laying hens.
