Taxonomic Study of the Genus Pholiota (Strophariaceae, Basidiomycota) in Korea

  • The genus Pholiota (Strophariaceae, Basidiomycota) is made up of wood-rotting saprotrophic mushrooms characterized by a yellow or brown pileus with scales and/or slimy, and by a brownish smooth spore with a germ pore. However, these features are not enough to distinguish its species, or separate the genus Pholiota from other brown-spored wood-rotting genera such as Hypholoma and Stropharia. Although internal transcribed spacer (ITS) sequencebased identification has improved identification accuracy for species of Pholiota, most Pholiota species in Korea are reported based on morphological features. To evaluate the taxonomy of Pholiota species, we investigated 62 specimens collected from 1999 to 2019 in Korea using ITS sequence analysis and morphological observation. Twelve of the 16 recorded Pholiota species in Korea were identified. While eight species were clearly separated, the ITS analysis did not distinguish three in the Pholiota adiposa complex. Therefore, further investigation is required to distinguish these three species. ITS sequences deposited in GenBank confirm that P. highlandensis exists in Korea. The presence of the other four Pholiota species could not be confirmed through specimens or sequence information in GenBank. A taxonomic key and the ITS sequence data for Korean Pholiota species are included and can be good baselines for further research on Pholiota taxonomy and diversity.

  • Pineal organs of poikilotherm vertebrates transform the environmental light information into a humoral message and a neuronal activity. The former is melatonin, and the latter is modulation of the impulse in ganglion cells. The ganglion cells are physiologically classified into luminosity (achromatic) type and chromatic one, as the neural activity is modulated in two ways. We attempted to classify the pineal ganglion cells with morphological characteristics by means of the three- dimensional reconstruction method. In the pineal ganglion cells of river lamprey, there are two different features, oval and spherical. For comparison of their projection region in the brain, the tracing investigation was also carried out. The application of the neural tracer near mesencephalic tegmentum showed that only oval-shaped ganglion cells were labeled in the pineal organ. These results suggest that the oval-shaped ganglion cell is functionally different from the spherical one.

바이오디젤 생산에 미치는 원료 특성의 영향 (Effects of Properties of Raw Materials on Biodiesel Production)

  • Biodiesel is an alternative and renewable energy source, which is hoped to reduce global dependence on petroleum and environmental problem. Biodiesel produced from a variety of oil sources such as vegetable oil, animal fat and waste oils, and has properties similar to those associated with petro-diesel, including cetane number, volumetric heating value, flash point, viscosity and so on. In this study, we investigate the effect of quality of raw materials on alkali-catalyzed transesterification for producing of biodiesel. The increase of content of free fatty acid and water in oil were caused the sharp decrease of conversion yield. Also, the low purity of methanol in reactant was inhibited the reaction rate. In the case of addition of sodium sulfate as absorbent to prepare catalyst solution, the content of fatty acid methyl ester in product was increased more about 1.6% than that of control. However, the addition of zeolite, sodium chloride and sodium sulfate as absorbent in reactant to remove water generated from reaction did not show any enhancement in the reaction yield. This result may provide useful information with regard to the choice and preparation of raw materials for more economic and efficient biodiesel production.

Status of wetland vascular plant species in Korea

  • We report the status of wetland vascular plant species in Korea including the whole peninsula and its adjacent islands. This analysis was based on database from our previous categorized list. In all, 4,050 taxa have been reported, including 3,769 native and 281 naturalized. Of these, 479 taxa (12%) are considered as wetland vascular indicator species: 240 obligate wetland plants (OBW) and 239 facultative upland plants (FACW). Approximately 31% of those 479 taxa, i.e., 149 taxa, are labelled as aquatic macrophytes. Wetland plants, mostly herbaceous but some woody, inhabit aquatic bodies and wet meadows. Except for two OBW and six FACW taxa, the rest of the plants are summer-green only. The information provided here is valuable for making assessments of wetland ecosystem health, as well as for developing management plans to preserve and restore wetlands and their resident plant species while also creating artificial wetland environments.

치료저항성 우울증의 연구에서 패러다임의 전환 (Paradigm Shift in the Study of Treatment Resistant Depression)

  • Treatment-resistant depression (TRD) is a major public health problem. It is estimated that about 30% of patients with major depressive disorder do not show substantial clinical improvement to somatic or psychosocial treatment. Most of studies for TRD have focused on the subjects already known as TRD. Patients with unipolar depressive episodes that do not respond satisfactorily to numerous sequential treatment regimens were included in the TRD studies. Such post hoc experimental design can be regarded only as consequences of having TRD, rather than as causal risk factors for it. Although informative, data derived from such studies often do not allow a distinction to be made between cause and effect. So, we should shift paradigm toward examining the risk for developing TRD in untreated depressed patients. To deal with this problem, untreated depressed patients should be enrolled in the study to identify biological markers for treatment resistance. The peripheral or central biological markers should be explored before starting treatment. Subsequent systematic administration of treatments with appropriate monitoring in the subjects can determine the risk for developing treatment resistance in untreated individuals. Such information could give a cue to improve the initial diagnosis and provide more effective treatment for TRD.

Perspectives provided by leopard and other cat genomes: how diet determined the evolutionary history of carnivores, omnivores, and herbivores

  • Recent advances in genome sequencing technologies have enabled humans to generate and investigate the genomes of wild species. This includes the big cat family, such as tigers, lions, and leopards. Adding the first high quality leopard genome, we have performed an in-depth comparative analysis to identify the genomic signatures in the evolution of felid to become the top predators on land. Our study focused on how the carnivore genomes, as compared to the omnivore or herbivore genomes, shared evolutionary adaptations in genes associated with nutrient metabolism, muscle strength, agility, and other traits responsible for hunting and meat digestion. We found genetic evidence that genomes represent what animals eat through modifying genes. Highly conserved genetically relevant regions were discovered in genomes at the family level. Also, the Felidae family genomes exhibited low levels of genetic diversity associated with decreased population sizes, presumably because of their strict diet, suggesting their vulnerability and critical conservation status. Our findings can be used for human health enhancement, since we share the same genes as cats with some variation. This is an example how wildlife genomes can be a critical resource for human evolution, providing key genetic marker information for disease treatment.

Development of PCR-Based Screening Methods for Macrolide Type Polyketides in Actinomycetes

  • About two thirds of the naturally occurring antibiotics have been discovered from actinomycetes. Therefore, the probability of discovering further new antibiotics from actinomycetes is declining as many known metabolites are isolated repeatedly. However, various efforts leave been made in order to enhance the probability of discovering novel compounds. In the present study, we have developed new screening strategies based on the antibiotic biosynthetic pathway, and the genetic information, utilizing polymerase chain reaction. We have selected macrolide type polyketides. In order to divide the ansamycin group antibotic of macrolide type polyketides, we have selected 3-amino-5-hydroxybenzoic acid (AHBA) moiety which contains a biosynthetically unique structural element in the group as a target molecules. Oligonucleotide primers were designed to amplify DNA fragments of macrolide type polyketide synthase and AHBA synthase genes from fourteen actinomycetes species. This method was successfully applied to all three of the known macrolide type polyketide produccing actinomycetes tested. In addition, it also identified the presence of potential macrolide type polyketide producing genes from seven actinomycetes that were known to produce none of macrolide type polyketides, and AHBA biosynthetic genes in one actinomycetes. This technique is potentially useful for the screening of new antibiotices and cloning of their biosynthetic genes.

Taxonomic Study of the Genus Abundisporus in Korea

  • The polypore genus Abundisporus Ryvarden is characterized by resupinate to pileate fruitbodies with a purplish brown hymenophore, slightly thick-walled, pale yellowish and non-dextrinoid basidiospores, and causing white rot. A purple color hymenophore, an easily observable and striking character, was considered the main distinctive feature at the generic level within polypores. However, due to highly similar basidiocarp features, species identification within these purple polypores is particularly difficult. Three species of purple colored polypores have been reported in Korea (Abundisporus fuscopurpureus, A. pubertatis, and Fomitopsis rosea). Based on morphological re-examination, ecological information, and sequence analysis of the internal transcribed spacer, we showed that previous classification was incorrect and there is only one species (A. pubertatis) in Korea. We provide a detailed description of A. pubertatis in Korea, as well as a taxonomic key to distinguish wood rot fungi with a purple hymenophore.

  • 그람 음성이며 막대모양의 Sphingobacteriaceae bacterium SH-48은 삼척 소한천에서 분리하였다. SH-48에 대한 유전체 분석을 실시하였으며, G + C 비율이 38.4%인 5,650,162 bp 크기의 염기서열을 얻었다. 유전체 특징은 카로티노이드 생합성 유전자인 crt 유전자 클러스터를 보유하고 있어 균주의 잠재적 중요성을 보여준다. 이러한 유전체 정보는 카로티노이드 생합성 경로에 대한 새로운 정보를 제공한다.

  • 중국 운남의 고유종으로 알려진 꼭두서니과의Ixora auricularis Chun & F. C. How ex W. C. Ko가 베트남에서 발견되었다. 이 종은 Ixora coccinea와 형태적으로 가장 비슷하지만, 잎의 기저는 이저이고, 측맥의 수가 많으며, 탁엽의 길이가 길고, 꽃받침 통의 길이가 짧으며, 화관의 끝이 도피침형, 화관내부의 부속체의 수가 더 많은 점에서 구분된다. 본 종에 대한 기재, 도해, 컬러사진과 함께 근연식물의 검색표를 제시하였다.
