식용대두유에 대한 Chelating agent의 항산화 효과 (Antioxidant Effect of some Chelating Agents on Soybean Oil)

  • 식용대두유의 산화방지를 위하여 항산화제로서 Chelating agents의 항산화효과를 mild한 조건(67ml $O_{2}/min$, $50^{\circ}C$)하에서 A.O.M.법으로 측정하였으며 기기분석법 (UV,IR,MMR)에 의한 측정의 가능여부를 시험한 결과 다음과 같다. 1. Chelating agents의 항산화 효과는 diphenic acid(109.3%), naphthoquinone(111.2%), pyremellitic acid(168.6%), Quinolinic acid(183.6%), naphthalic acid(921.2%) 순으로 증가하였으며, Control에 비하여 각각 9.3, 11.2, 68.6, 83.6, 821.2% 증가하였다. 2. Chelating agents는 항산화 효과를 나타냈으며, 철과 등을 첨가했을 경우에 현저하였다. 또 이들의 항산화 효과는 화합물의 관능기와 분자구조에 따라 다르며 diphenic acid, naphthoquinone 등에서 낮았다. 3. 본 실험과 같이 mild한 조건하에서 NMR 분석법을 이용하여 항산화 효과를 측정할 수 있었으나 UV와 IR은 적당치 않았다.

Effects of Silkworm Hemolymph and Cartilage-specific Extracellular Matrices on Chondrocytes and Periosteum-derived Progenitor Cells

  • In order to enhance the repair of defects in articular cartilage via cell therapy with autologous chondrocytes, as well as with periosteum-derived progenitor cells (PDPCs), silkworm hemolymph (SH) and a variety of cartilage-specific extracellular matrices (ECMs) including type II collagen, proline, chondroitin 4-sulfate, and chondroitin 6-sulfate were assessed with regard to their efficacy as media supplements. SH, a known anti-apoptotic agent, was found to enhance cell growth, as was shown by the results of a 3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) assay. According to the results of reverse transcriptase polymerase chain reaction (RT-PCR) analyses, the cartilage-specific ECMs were found to stimulate the expression of hyaline cartilage-specific genes, most notably type II collagen and Sox9, in monolayer cultures of PDPCs.

  • A novel integrated method for increasing dissolved oxygen concentration in culture media has been developed. It involves adding hydrogen peroxide to the medium, which is then decomposed to oxygen and water by catalase and adding vegetable oil to the medium as antifoam agent and oxygen vector. A new apparatus for automated addition of hydrogen peroxide to the bioreactor to keep the dissolved oxygen concentration constant over the range $10-100%\;{\pm}\;5%$ was tested. A significant increase (over threefold) of cultivation time was obtained while the dissolved oxygen concentration remained stable ($30%\;{\pm}\;5%$). Therefore, use of corn oil mixed with Ca-stearate as oxygen vector and antifoam and hydrogen peroxide as oxygen source to control excessive foam that was generated by microorganism biosurfactant, GB16-BS produced at Bacillus sp. GB16 cultivation was appropriate for stable cultivation.

Sulforaphane Inhibits Osteoclastogenesis by Inhibiting Nuclear Factor-κB

  • We show that sulforaphane inhibits osteoclastogenesis in the presence of macrophage colony-stimulating factor (M-CSF) and receptor for activation of nuclear factor-${\kappa}B$ ligand (RANKL) in osteoclast (OC) precursors. Sulforaphane, an aliphatic isothiocyanate, is a known cancer chemo-preventative agent with anti-oxidative properties. Nuclear factor-${\kappa}B$ (NF-${\kappa}B$) is a critical transcription factor in RANKL-induced osteoclastogenesis, and electrophoretic mobility shift assays (EMSAs) and assay of NF-${\kappa}B$-mediated secreted alkaline phosphatase (SEAP) revealed that sulforaphane selectively inhibited NF-${\kappa}B$ activation induced by RANKL. Inhibition may involve interaction of sulforaphane with thiol groups, since it was prevented by reducing agents.

Urokinase Conjugated with Water-Soluble Dextran

  • Urokinase, a plasminogen activator, was conjugated with dextran by the cyanogen bromide activation-coupling method. The resulting water-soluble conjugate was purified by gel permeation chromatography on Sephadex G-200. The maximal activity was obtained when the ratio of urokinase/dextran was 1/20 for the coupling. The final preparation showed 5 CTA units/mg conjugate, 300 CTA units/mg protein, 8.4 % activity retention, and 47 % protein retention. The urokinase-dextran conjugate had good thermal, pH and storage stabilities. In addition, it showed greater resistance to the inhibitory effect of human plasma than native urokinase. Also in vitro biological half-life of urokinase increased 40 times by this conjugation. In view of activity, excellent stability and increased half-life, the conjugate can be a potential fibrinolytic agent in an injectable form.

닭 우모로부터 추출한 아미노산 생화학제의 고구마뿌리혹선충 증식억제 효과 (Nematicidal Effect of Root-Knot Nematode (Meloidogyne incognita) by Amino Acids Biochemical Agent Extracted from Chicken Feather)

  • 본 연구는 뿌리혹선충의 화학적 방제를 대체할 수 있는 친환경 선충방제제 개발을 목적으로 닭 우모분으로부터 추출한 아미노산 생화학제(feather amino acid; FAA)의 고구마뿌리혹선충에 대한 방제효과를 검정하였다. 다양한 종류의 아미노산을 대상으로 고구마뿌리혹선충 유충 치사효과를 조사한 결과, 5종류(L-asparagine, L-aspartic acid, L-methionine, L-tyrosine, _L-cysteine)의 아미노산이 50 mM 농도에서 50% 이상의 치사율을 나타내었고, L-asparagine 50 mM에서 치사율이 94%로 가장 높았다. 닭 우모분으로부터 추출한 FAA 생화학제의 고구마뿌리혹선충 2령 유충의 치사력과 알 부화억제 효과를 조사한 결과, 1/50 농도에서 80% 이상의 유충 치사력과 74%의 알 부화 억제율을 보였다. 토마토 근권토양에 FAA 생화학제를 처리하고 60일간 재배시험을 통하여 관찰한 결과, 토양 내 유충밀도는 63%, 난낭수는 59%, 뿌리혹 형성 61%의 방제효과를 나타내었다. 이상의 결과로부터 닭 우모분으로부터 추출한 FAA 생화학제는 뿌리혹선충의 친환경 선충방제제로 활용될 수 있을 것으로 사료된다.

세라믹 LDH 나노하이브리드를 이용한 MRI 조영제의 최신 연구동향 (Recent Progress in MRI Contrast Agent with Ceramic LDH Nanohybrids)

  • Ceramic layered double hydroxide (LDH) nanohybrids have attracted considerable interest in biomedical science due to their unique structural feature and characteristics in biological condition. Many studies on LDH nanoparticles have been reported in diagnosis applications including magnetic resonance imaging (MRI) contrast agents in order to not only provide better imaging performance through multimodal imaging strategy, but realize therapeutic function which treat cancers in one platform. This review highlights the recent progress in MRI T1 contrast agent, dual modal imaging system, and MRI-guided drug delivery systems ranging from synthetic method and characterization to evaluation in vitro and in vivo based on the ceramic LDH nanohybrids. Future research directions are also suggested for next-generation bio-imaging contrast agent.

  • Poly (HPMA/NVP) hydrogel was synthesized according to the composition and the con- centration of the crosslinking agent. The swelling degree of the P(HPMA/NVP) hydrogel decreased with increasing of P(HPMA) composition and crosslinking agent. The glass transi- tion and softening temperatures of the P(HPMA/NVP) increased with increasing of P(HPMA) composition. The composition of P(HPMA/NVP) : 70/30 and the concentration of the crosslin- king agent of 0.5 wt % gave the optimum condition in the application to the contact lens. There were also adequate in the mechanical, refractive properties and in vivo test in the P(HPMA/ NVP): 70/30 system.

Chestnut Extracts as new Anti-aging agent

  • Inner nutshell of Castanea crenata has been used as an anti-aging and anti-wrinkle agent from the ancient time in east Asia. In order to develop new anti-aging and anti-wrinkle, ethanolic extract of inner nutshell of Castanea crenata was prepared and various biological activities were evaluated. Cor-285 possessed potent tree radical scavenging activity in vitro compared to gallic acid. Cor-285 showed the preventive effect against UV-induced cytotoxicity of fibroblast at concentration of 25-250mg/ml. When Cor-285 was evaluated for its anti-allergic activity, it effectively inhibited histamine release from mast cells induced by compound 48/80. The inhibitory activity was stronger than that of glycyrrhizinate. Cor-285 also showed on vivo inhibition against delayed hypersensitivity as well as croton-oil induced ear edema in mice when topically applied. These results strongly suggest that Cor-285 may reduce immunoregulatory/inflammatory skin trouble. From the attempts to isolate the scavenger, were isolated. In a clinical trial of twenty healthy volunteers with aged skin, 6 weeks application of Cor-285 decreased wrinkle about 26% and increased moisturizing 20% on the skin. All of these results indicate that Cor-285 may be an effective anti-aging and anti-wrinkle agent.

물달개비의 생물학적 방제인자 물달개비바구미의 생활사 및 기주특이성 (Life Cycle and Host Specificity of Tanysphyrus (Tanysphyroides) major Roelofs (Coleoptera : Curculionidae), a Possible Candidate Agent for the Biological Control of Monochoria vaginalis var. plantaginea)

  • 물달개비는 한국의 논에서 문제되는 난방제 잡초중의 하나이다. 물달개비의 생물학적 방제 인자로 물달개비바구미를 선발하였다. 생물학적 방제에 이용하기 위해 2006년과 2007년 계대사육을 실시하여 형태적, 생태적 특징을 조사하였다. 이 종은 1년에 한세대만 경과하며 성충으로 월동하고, 성충은 6월 하순부터 9월 하순까지 발생한다. 물달개비바구미는 알에서 성충까지 $22{\pm}0.7$일이 소요된다. 주요 농작물과 수생식물 60종을 대상으로 기주특이성을 조사 결과 물달개비의 생물학적 방제 곤충으로서의 가능성을 보였다.
