Four Species of Montagnulaceae Unrecorded in Korea and Isolated from Plant Litter in Freshwater

  • Freshwater is a diverse and complex environment for fungi and provides several types of habitat including plant litter, sediment, and carcasses. We collected plant litter from the main stream and branch streams of Nakdong River, Sohancheon in Samcheok, and Geumoreum in Jeju. From several samples of plant litter, we isolated 8 fungal strains belonging to 4 Montagnulaceae species unrecorded in Korea: Paraconiothyrium archidendri, Paraphaeosphaeria sporulosa, Paraphaeosphaeria michotii, and Paraphaeosphaeria viridescens. These fungi were identified by phylogenetic analysis using the internal transcribed spacer (ITS) region and examination of morphological characteristics. Montagnulaceae is known as one of the families in Pleosporales and includes coniothyrium-like fungi. In this study, we described phylogenetic analysis and mycological characteristics of these species, and this is the first report of these taxa in Korea.

  • 국내에서 문헌에만 기록되어 있던 장군대사초(Carex poculisquama $K{\ddot{u}}k$.)가 우리나라 석회암지대에 분포하는 것으로 확인되었다. 장군대사초는 과포가 마름모상 타원형이고, 맥 위에만 단모가 있다는 형태적인 특징으로 같은 절내 근연종인 갈사초(Carex ligulata Nees)와 구분된다. 본 연구에서 장군대사초의 실체를 확인하고, 기재, 도해, 화상자료를 통하여 근연종과의 차이점을 검색표로 작성하였다.

  • According to Article 9.3 of the International Code of Nomenclature for algae, fungi, and plants(Shenzhen Code), a lectotype may be selected as the nomenclatural type from the original material, if the name was published without a holotype. While reviewing the genus Scrophularia collected in Northeast Asia, we found that two species, S. alata A. Gray and S. kakudensis Franch., were still untypified. S. alata has three specimens considered as syntypes in two herbaria, Harvard University and the Smithsonian Institution. For S. kakudensis, two specimens considered as syntypes at the Muséum National d'Histoire Naturelle, Paris were classified as normal specimens, not type specimens. Therefore, two species of Scrophularia L. namely, S. alata A. Gray and S. kakudensis Franch., are lectotypified. The lectotypes are kept in the Harvard University and the Muséum National d'Histoire Naturelle, Paris, respectively. Furthermore, some nomenclatural issues related to these names are discussed, and the photographs of the selected lectotypes are provided.

A report of seven unrecorded bacterial species in Korea, isolated from marine sediment

  • In March 2021, marine sediment from East Sea samples were suspended in a 2% NaCl solution, and serial dilution was performed in fresh marine and Reasoner's 2A agar. Isolated bacterial strains were identified based on 16S rRNA gene sequences, and showed at least 98.7% sequence similarity with previously reported bacterial species. Finally, seven bacterial strains which were validly published but not reported in Korea, were obtained. These isolates were allocated to the orders Bacillales and Flavobacteriales. The three Flavobacteriales strains are classified into the family Flavobacteriaceae. The other four Bacillales belong to the families Bacillaceae and Paenibacillaceae. The seven unrecorded bacterial strains in this study are classified into seven different genera, which are assigned to Mesobacillus, Paenibacillus, Gramella, Gillisia, Arenibacter, Fictibacillus, and Brevibacillus. During the investigation, the possibility of excavation of various unrecorded species in domestic marine sediment was confirmed. Gram-staining, cell morphology, physiological and basic biochemical characteristics, and phylogenetic analysis were performed in this study and provided in the description of each strain.

금호분지 내 지리관광자원의 자연지리학적 배경 (Physical Geographical Background of Geotourism Resources in Gumho River Basin)

  • 본 논문에서는 금호분지를 6개의 지형단위(보현산지, 팔공산지, 비슬산지, 영천분지, 경산분지, 대구분지)로 구획한 후, 각 지형단위 내에 분포하는 지리관광자원의 자연지리학적 배경과 분포의 특성을 고찰하였다. 금호분지 내의 지리관광자원은 지형환경(32개, 78%)을 많이 반영하며, 다음으로 지질환경(7개, 17%)을 반영한다. 보현산지 내에는 3개의 지형자원이 분포하며, 팔공산지 내에는 8개의 지형자원이 분포한다. 비슬산지 내에는 5개의 지형자원과 1개의 문화자원이 분포한다. 그리고 영천분지 내에는 2개의 지형자원과 1개의 지질자원, 1개의 생물자원, 그리고 2개의 문화자원이 분포하는데, 생물자원 1개와 문화자원 1개는 지형환경을 반영한다. 정산분지 내에는 1개의 지형자원과 1개의 생물자원, 3개의 지질자원이 분포하며, 생물자원 1개는 지형환경을 반영한다. 대구분지 내에는 8개의 지형자원과 2개의 지질자원, 3개의 생물자원이 분포하는데, 생물자원 2개는 지형환경의 영향을 많이 받는다.

LPS 유도된 HCT116 인간 대장암세포에서 cordycepin의 prostaglandin E2-EP4 receptor 감소 조절을 통한 세포의 이동과 전이 억제 효과 (Cordycepin Inhibits LPS-induced Cell Migration and Invasion in Human Colorectal Carcinoma HCT116 cells through Down-regulation of Prostaglandin E2-EP4 Receptor)

  • Prostaglandin E2(PGE2), a major product of cyclooxygenase-2 (COX-2), plays an important role in the carcinogenesis of many solid tumors, including colorectal cancer. Because PGE2 functions by signaling through PGE2 receptors (Eps), which regulate tumor cell growth, invasion, and migration, there has been a growing amount of interest in the therapeutic potential of targeting Eps. In the present study, we investigated the role of EP4 on the effectiveness of cordycepin in inhibititing the migration and invasion of HCT116 human colorectal carcinoma cells. Our data indicate that cordycepin suppressed lipopolysaccharide (LPS)-enhanced cell migration and invasion through the inactivation of matrix metalloproteinases (MMP)-9 as well as the down-regulation of COX-2 expression and PGE2 production. These events were shown to be associated with the inactivation of EP4 and activation of AMP-activated protein kinase (AMPK). Moreover, the AMPK inhibitor, compound C, as well as AMPK knockdown via siRNA, attenuated the cordycepin-induced inhibition of EP4 expression. Cordycepin treatment also reduced the activation of CREB. These findings indicate that cordycepin suppresses the migration and invasion of HCT116 cells. Through modulating EP4 expression and the AMPK-CREB signaling pathway. Therefore, cordycepin has the potential to serve as a potent anti-cancer agent in therapeutic strategies against colorectal cancer metastasis.

회화나무 열매 추출물에 의한 지방세포 분화 및 지방생성 억제 (Inhibition of Adipocyte Differentiation and Adipogenesis by the Extract from Sophora japonica Fruit)

  • The world-wide rate of obesity is increasing continuously, representing a serious medical threat since it is associated with a variety of diseases including type 2 diabetes, cardiovascular disease, and numerous cancers. Sophora japonicais used as a traditional herb for medicinal purposes in eastern Asia. However, the anti-obesity effects of S. japonicafruit have not been explored. The aim of this study is to investigate the inhibition of adipocyte differentiation and adipogenesis by an ethanol extract of S. japonicafruit (EESF) in 3T3-L1 pre-adipocytes. Our results demonstrate that EESF suppressed the terminal differentiation of 3T3-L1 pre-adipocytes in a dose-dependent manner, as confirmed by a decrease in lipid droplet number and lipid content through Oil Red O staining. EESF significantly reduced the accumulation of cellular triglyceride, which was associated with a significant inhibition of the levels of pro-adipogenic transcription factors, including PPARγ, C/EBPα and C/EBPβ. In addition, EESF potentially down regulated the expression levels of adipocyte-specific proteins, including aP2 and leptin. In particular, EESF treatment effectively enhanced the activation of the AMPK signaling pathway; however, the co-treatment with compound C, an inhibitor of AMPK, significantly restored the EESF-induced inhibition of pro-adipogenic transcription factors and adipocyte-specific genes. These results indicate that EESF may exert an anti-obesity effect by controlling the AMPK signaling pathway, suggesting that the fruit extract of S. japonica may be a potential anti-obesity agent.

Three New Recorded Species of the Physalacriaceae on Ulleung Island, Korea

  • Most known species in the Physalacriaceae are saprotrophs that grow on decaying leaves and wood, and approximately 21 genera in the Physalacriaceae have been reported worldwide. During an ongoing survey of indigenous fungi in Korea, four specimens belonging to the Physalacriaceae were collected on Ulleung Island. These specimens were identified as three species based on morphological characteristics and molecular analysis of rDNA-internal transcribed spacer sequences. Three species in three genera were shown to be new records in Korea: Hymenopellis orientalis, Paraxerula hongoi, and Ponticulomyces orientalis. The latter two are the first records of these genera in Korea. In this study, we provide detailed morphological descriptions of these species and describe their phylogenetic position within the Physalacriaceae.

Vascular plant diversity of the Gogunsan Archipelago in the Korean Peninsula

  • This study was carried out to investigate the flora of six islands belonging to the Gogunsan Archipelago (i.e., Sinsi-do, Seonyu-do, Munyeo-do, Yami-do, Bian-do, and Duri-do) in the Korean Peninsula. As results of five field surveys from March to October of 2016, we have identified 575 total taxa, representing 527 species, five subspecies, 42 varieties, and one hybrid, placed in 358 genera and 118 families. Of these 575 taxa, four are endemic to Korea, six taxa are listed on the Korean Red List of threatened species, 67 are floristic regional indicator plants, and 74 are invasive alien species. In this study, we compared species richness among the islands, and find that the larger the islands, the higher the species richness. In the case of habitat affinity types, forest species were most common, followed by farmland, seacoast, bare ground and wetland species. From similarity analyses based on the composition of vascular plants, each island did not exhibit either local specificity or unique diversity. On the contrary, the proportion of invasive alien and ruderal species may increase by human activities. Investigations and analyses of island flora such as this are important to assess the current status of the flora, predict future vegetation patterns and the spread of the alien species, and establish managment plans of plant diversity.
