Technology Convergence & Trend Analysis of Biohealth Industry in 5 Countries : Using patent co-classification analysis and text mining (5개국 바이오헬스 산업의 기술융합과 트렌드 분석 : 특허 동시분류분석과 텍스트마이닝을 활용하여)

  • The study aims to identify convergence and trends in technology-based patent data for the biohealth sector in IP5 countries (KR, EP, JP, US, CN) and present the direction of development in that industry. We used patent co-classification analysis-based network analysis and TF-IDF-based text mining as the principal methodology to understand the current state of technology convergence. As a result, the technology convergence cluster in the biohealth industry was derived in three forms: (A) Medical device for treatment, (B) Medical data processing, and (C) Medical device for biometrics. Besides, as a result of trend analysis based on technology convergence results, it is analyzed that Korea is likely to dominate the market with patents with high commercial value in the future as it is derived as a market leader in (B) medical data processing. In particular, the field is expected to require technology convergence activation policies and R&D support strategies for the technology as the possibility of medical data utilization by domestic bio-health companies expands, along with the policy conversion of the "Data 3 Act" passed by the National Assembly in January 2019.

Effect of Capsaicin on the Body fat and Adipocyte in the Diet induced-obese Mice

  • As the obesity has been known to be related with the hyperlipidemia, cardiovascular disease, cerebral apoplexy, fatty liver, and other chronic diseases, recent researches have focused on the functional food materials and their anti-obesity activities. This study was performed to study the effects of vanilloid family capsaicin, major pungent ingredient of hot chillies and peppers, on anti-obesity activities. ICR male mice were fed one of the pellet diet, basal diet, and high fat diet with capsaicin (45 $\mu\textrm{g}$/day) solution for 5 days. Mice in the corresponding control groups were given water for 5 days. In results, capsaicin reduced body weights in any diet groups. Percent weight and cell size of the abdominal white adipose tissue in mice on the high fat diet with capcaicin were significantly lower compared with those in mice on the high fat diet with water. However, percent brown adipose tissue weight per body weight in mice on the high fat diet was not affected by capsaicin. Capsaicin reduced the levels of s-triglyceride and s-total cholesterol in the pellet diet or high fat diet groups. There was no difference in the s-protein levels between the capsaicin group and the control water group. These data indicate that 1) orally administered capsaicin has a reducing effect on the blood triglyceride and total cholesterol levels, and 2) capsaicin has lowering effects on the body weight, percent weight and cell size of the abdominal white adipose tissue.

Effect of Psyllium Seed Husk on the Postprandial Glucose Control and Insulin Secretion Dynamics

  • This study was to investigate the effect of psyllium seed husk (PSYL) on postprandial glucose control and insulin secretion dynamics in Sprague-Dawley rats. In experiment 1, the rise in postprandial serum glucose was monitored during a 240-min period using a maltose loading test In normal rats given 16.6 mg/l00 g B.W./ml of PSYL orally, all the blood glucose levels during the 240-min period did not show statistically significant differences from the corresponding levels in normal rats given water. However, in streptozotocin-induced diabetic rats given the same amount of PSYL, the blood glucose level at 30 min was significantly lower than that in diabetic rats given water, and the peak time of the rise in the postprandial glucose was delayed In experiment 2, the normal (N) and diabetic (Db) rats were given PSYL (25 mg/l00 g B.W./ml/day) orally for 5 days. Blood samples were collected in order to measure the s-glucose and s-insulin levels. The final s-glucose level at day 5 in Db-PSYL was significantly lower than that in the corresponding control rats (Db-CONT) and the final s-insulin level in Db-PSYL was significantly greater than that in Db-CONT. In vitro 40-min pancreas perfusion was performed at day 5 in order to examine the insulin secretion dynamics. Results showed that the amounts of insulin secreted during the first phase (11-20 min) and the second phase (21-40 min) in the Db-PSYL were significantly greater than those in Db-CONT. Therefore, it is concluded that psyllium seed husk could be beneficial for controlling postprandial glucose levels in the stretozotocin-induced diabetic rats, and it may be partially mediated by insulin secretion dynamics.

Effect of Chlorella Diet Supplementation on Blood and Urine Cadmium Levels in Cadmium Poisoned Rats

  • This study was conducted to investigate the influence of chlorella diet supplementation. Blood accumulation and urine excretion levels were measured after the Sprague-Dawley (SD) rats were fed on a chlorella diet supplementation mixed with 40 ppm of CdCl$_2$. Four groups tested for blood accumulation and urine excretion levels. All four groups fed on a basic diet with a cadmium mixture. The diet for the first group contained only basic diet and the cadmium added to the drinking water. The diet for the three other groups contained cadmium to the drinking water, and 1%, 5% and 10% of chlorella added to the basic diet. A concentration of cadmium for the first group showed a 3.2$\pm$0.4 $\mu\textrm{g}$/I blood accumulation level and 41.5$\pm$32.9 $\mu\textrm{g}$/l urinary excretion level, and the second group, which was fed on the basic diet with 1% of chlorella added and cadmium to the drinking water showed a $1.5\pm$0.6 $\mu\textrm{g}$/l blood level and only l4.l$\pm$1.6 $\mu\textrm{g}$/l urinary excretion level. The other two groups, which were fed on 5% and 10% of chlorella concentration and cadmium to the drinking water did not exhibit any notable effects greater than the group fed on 1% concentration of chlorella. The results suggest that the blood accumulation and urinary excretion of Cadmium are influenced by the chlorella diet supplementation from the concentration of 1% of the basic diet.

Extraction Procedures for Free Radical Scavenging Activity from Noni Fruit (Morinda citrifolia) (노니 과실에 함유된 항산화물의 추출 공정)

  • In this study, we investigated maximum yield of radical scavenging activity (RSA) using each of various extraction solvents and procedure from noni friut (Morinda citrifolia). Noni fruits were either sun-dried (FS) or hot-air dried (FO) at $60^{\circ}C$ after steam blanching. For optimum aqueous extraction, noni fruits should be sun dried and autoclave extraction time should not be over 30 min to produce extraction with high RSA with low cost. In case of 50% ethanol extraction, reflux extracts of FS and FO resulted in $IC_{50}$ of 1.92 mg/mL and 3.06 mg/mL at 8 hr. When $IC_{50}$ values were lower than 5 mg/mL, coefficient of correlation was 0.71 indicating that 71% of the phenolic antioxidants in noni fruits were accounted for the activity by scavenging free DPPH. However, coefficient of correlation significantly decreased to 0.63 over $IC_{50}$ values of 5 mg/mL. Autoclave extraction contained chlorogenic acid of $14.69 \;{\mu}g/mL$ and scopoletin of $3.86 \;{\mu}g/mL$. Reflux extraction showed all three compounds, chlorogenic acid ($26.19 \;{\mu}g/mL$), quercetin ($19.59 \;{\mu}g/mL$), and scopoletin ($17.4 \;{\mu}g/mL$). Therefore, the result of this study indicated that the potential antioxidant activities and functional values were obtained significantly with reflux extraction from noni fruit.

Anti-hypertensive Effects of ethanol extract of Phyllostachys Pubescens via Antioxidant Activity (맹종죽의 항산화활성을 통한 항고혈압 효능)

  • Phyllostachys pubescens (Maengiong-Juk), a kind of the bamboo, was reported to have many beneficial pharmacological actions. in this study, of using 70% ethanol extract of Phyllostachys pubescens we investigated its efficacy on angiotensin converting enzyme (ACE) and antioxidant enzyme activities. In addition, vasorelaxant effect was examined in rat aortic rings. The inhibitory effect of ACE activity by Phyllostachys pubescens extract (PPE) was dose-dependently increased by 61.42% at 10mg/ml. PPE relaxed the pre-contracted rat aortic rings with 10$^{-6}$M phenylephrine, showing about 88% at 4.0mg/ml. Sprague Dawley (SD) rats were given different concentrations of PPE mixed in the drinking water for 10 weeks. PPE did not show any difference with control group in blood pressure, body weight (BW) and food intake. However, it revealed the highest total antioxidative effect at dose of 1.0 g/100 g BW in plasma by TEAC assay. Thiobarbituric acid reactive substance (TBARS) and protein carbonyl levels which are markers of tissue peroxidation, were significantly lowed at the same dosage. Furthermore, hepatic antioxidant enzymes such as total superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and catalase activities were also significantly increased by PPE (1.0 g/100 g BW). In conclusion, we suggest that PPE might have antihypertensive effect through increasing antioxidant activities.

Analysis of Pharmacogenetic Information in Korea Drug Labels (국내 허가사항에 반영된 약물 유전정보 분석)

  • Background: Pharmacogenomics is the study of how genetic mutations in patients affect their response to drugs. Pharmacogenomic studies aim to maximize drug effects and minimize adverse drug events. The Food and Drug Administration and the European Medicine Agency published guidelines for pharmacogenetics in 2005 and 2006, respectively; the Korean Ministry of Food and Drug Safety followed suit in 2015. Methods: This study analyzed pharmacogenomic information in the Korean Ministry of Food and Drug Safety's integrated drug information system to evaluate whether domestic pharmaceutical products reflect the current research on pharmacogenomic differences. Results: In June 2020, the Korean pharmacogenomic database contained genomic data on 90 compounds. Of these, 45 compounds were classified as "Antineoplastic and immunomodulating agents." The other 45 non-antineoplastic agents were in the following categories: Anti-infectives, Mental & behavior disorder, Hormone & metabolism related diseases, Cardiovascular system, Skin & subcutaneous tissue disease, Genito-urinary system and sex hormones, Blood and blood forming organs, Nervous system, Alimentary tract and metabolism, Musculo-skeletal system, and Other conditions including the respiratory system. In addition, 30 additives unrelated to the main ingredient were associated with genetic precautions. Conclusion: This study showed that antineoplastic and immunomodulating agents accounted for half the drugs associated with pharmacogenetic information. For antitumor and immunomodulatory drugs, genomic tests were recommended depending on the indication; this was in contrast to genomic testing recommendations for non-antineoplastic medications. Genomic tests were rarely requested or recommended for non-antineoplastic medications because the relationships between genotype and efficacy among those drugs were relatively weak.

Acquisition and Classification of ECG Parameters with Multiple Deep Neural Networks (다중 심층신경망을 이용한 심전도 파라미터의 획득 및 분류)

  • As the proportion of non-contact telemedicine increases and the number of electrocardiogram (ECG) data measured using portable ECG monitors increases, the demand for automatic algorithms that can precisely analyze vast amounts of ECG is increasing. Since the P, QRS, and T waves of the ECG have different shapes depending on the location of electrodes or individual characteristics and often have similar frequency components or amplitudes, it is difficult to distinguish P, QRS and T waves and measure each parameter. In order to measure the widths, intervals and areas of P, QRS, and T waves, a new algorithm that recognizes the start and end points of each wave and automatically measures the time differences and amplitudes between each point is required. In this study, the start and end points of the P, QRS, and T waves were measured using six Deep Neural Networks (DNN) that recognize the start and end points of each wave. Then, by synthesizing the results of all DNNs, 12 parameters for ECG characteristics for each heartbeat were obtained. In the ECG waveform of 10 subjects provided by Physionet, 12 parameters were measured for each of 660 heartbeats, and the 12 parameters measured for each heartbeat well represented the characteristics of the ECG, so it was possible to distinguish them from other subjects' parameters. When the ECG data of 10 subjects were combined into one file and analyzed with the suggested algorithm, 10 types of ECG waveform were observed, and two types of ECG waveform were simultaneously observed in 5 subjects, however, it was not observed that one person had more than two types.

Effect of Green Tea Products on Bone Metabolism Marker in Ovariectomized Rats with High Cholesterol Diet Intake (고콜레스테롤 식이를 섭취한 난소절제 흰쥐에서 녹차가공품이 골 대사 지표에 미치는 효과)

  • This study was designed to evaluate the effect of green tea products (GTP) on bone metabolism marker in ovariectomized (OVX) rats fed high cholesterol diet. Forty Sprague-Dawley female rats, 10 weeks of age ($279{\pm}2g$), were divided into 4 groups and fed on the experimental diets for 6 weeks: sham operated control (Sham-C) and OVX-control (OVX-C) groups treated high cholesterol diet. OVX-GTP 5% (OVX-G5) and OVX-GTP 20% (OVX-G20) groups were treated with high cholesterol diet containing 5% GTP and 20% GTP, respectively. Food efficient ratio was significantly (p<0.05) lower in OVX-G20 than in the other OVX groups. Bone mineral density of femur was not significantly different among the experimental groups in the order of Sham-C>OVX-G5 and OVX-G20>OVX-C. Alkaline phosphatase activities on serum was lower in the GTP supplement groups than in the OVX-C. Estradiol levels of serum were higher in the GTP supplement groups than in the OVX-C. Osteocalcin levels of serum was the lowest in the OVX-G20. Deoxypyridinoline crosslink values of urine, indicator of bone absorption, was the lowest in the OVX-G20 group. The GTP supplemented groups had a lower bone resorption ratio than in the OVX-C group. From the above results, these findings suggest the possibility of using GTP as a functional food materials related to bone metabolism in menopause.
