Ammonia oxidizing bacteria(AOB) oxidize ammonia to nitrite and are important microorganisms which control nitrification. Several environmental factors such as dissolved oxygen(DO), temperature and pH influence the growth of AOB. In this work, to assess the effect of DO concentration on AOB, four aerobic biofilm reactors packed with ceramic media were operated 1, 3, 5 and 7 mgDO/L, respectively. The optimal DO concentration with stable nitrification efficiency in aerobic biofilm reactor was above 5.0 mg/L. To assess the relationship between the DO concentration and the characteristics of AOB in aerobic biofiim reactor, DGGE and cloning based on PCR targeting 16S rRNA and amoA gene were performed. Additionally, INT-DHA activity test was proceeded to estimate the activity of AOB. As the results of DGCE and cloning, the community of AOB and the ratio of Nitrosomonas sp. changed little in spite of different nitrification efficiencies. INT-DHA activity test showed that the activity of AOB decreased as DO concentration decreased. It means that DO concentration does not affect the community of AOB, but the activity of AOB.
The effects of organic supplement (acetate) and dissolved oxygen concentration on the nitrification rate of wastewater were investigated in the 27.7 L pilot-scale airlift biofilm reactor with the granular activated carbon media of 0.613 mm diameter. The ammonium oxidation rate increased stepwise up to 5 kg N/㎥$.$d at the riser air velocity of 0.063 m/s, when the air velocity and the ammonium loading rate were raised alternately. The nitrite build-up was observed during the early stage of the biofilm formation, which disappeared after the reactor operation of 128 days. As increasing the organic loading rate, the organic oxidation rate increased up to 25.0 kg COD/㎥$.$d with the removal efficiency of 94% but the oxidation rates of ammonium and nitrite decreased. The oxidation rates of ammonium and nitrite increased with the dissolved oxygen concentrations. When the pure oxygen was sparged, the ammonium oxidation rate was almost five times higher than that with air at the same velocity.
The objectives of this study were to examine the biodegradation of phenol using the anaerobic fluidized bed reactor(AFBR). Mixed microorganisms were selected from the anaerobic digestion tank, and could be adapted to high concentration of phenol by increasing the phenol concentration 600-3600 mg/l step by step. The results were summarized as follows: 1. The average removal efficiency of phenol was 90%, decreased by increasing concentration of phenol, and then a shock range was 1200~2400 ppm. 2. The production rate of biogas in overall limits was proportional to the concentration of influent phenol. 3. At steady state, compositions of gases were $CH_4$ 55~60%, $C0_2$ 34~43%, respectively. These were similar to that of the theoretical estimates. 4. The production rates of biogas and methane per the molarity of phenol removed were linearly increased, 56.45 l gas/mol-phenol and 29.20 l $CH_4/mol$-phenol. Using this biogas, the recoverable energy was 269.1 kcal/mol phenol. It was 120.2 kcal/g-COD, transforming into the chemical oxygen demand. 5. The bulk of microorganisms existed in suspended section of fluidized bed with type of biofilm and its concentration was 340 mg/g-media. In conclusion, the anaerobic treatment of pure phenol was possible and its removal efficiency, introducing the AFBR, was successful. Also toxic organic compound such as phenol was biodegradable and was recoverable as resource of energy.
The treatment of the washout from small scale incinerator was performed physically, chemically and biologically. The results are as follows. 1. SS, FS removal efficiency of washout wastewater from incinerator was 67.4%, 37.4%, while SS, FS of sewage wastewater was removed 63.2% 35.4% respectively. 2. The optimal conditions for chemical coagulation turned out to be pH 7.5, alum(Al2O3 10%) 30ml/ι and polyelectrolyte(A-601P 0.1%) 4ml/ι. SS 86%, FS 89.5%, BOD 42.5% and CODMn, 63.5% was removed and the removal efficiency of some metals are shown as Pb 93.5%, Zn 86.5% and Fe 80.6%. The concentration of the effluent was SS 9mg/ι, BOD 98.4mg/ι, and CODMn 138.4mg/ι. 3. The removal efficiency in treating washout wastewater of incinerator through HBC-briquet media was getting higher with increasing HRT, and mixed wastewater with 1:1, 1:2 ratio could be met up to the standard limit with higher HRT than 12hr. Under the condition of 1:2 mix ratio and HRT 24 hr, removal efficiency of SS, BOD, CODMn, T-N and T-P was 92.1%, 90%, 87%, 48.2% and 48%, respectively, and the concentration of treated wastewater was SS 2.9 mg/ι, BOD 10.3mg/ι, CODMn 14.1mg/ι, T-N 11.6 mg/ι and T-P 1.3 mg/ι, respectively.
The river water source of Saidabad Surface Water Treatment Plant at Dhaka, Bangladesh, is deteriorated too much to be treated by conventional treatment process due to excessive ammonia pollution. In order to improve the raw water quality before it enters into the main treatment chain, a pilot study was conducted for pre-treatment of the raw water. The objective is to investigate the rate of reduction of ammonia using the Meteor pilot, a biological pretreatment system, which is a laboratory scale Moving Bed Biofilm Reactor with a nominal volume of hundred liters, filled with 50 L of Meteor 660 media. The reduction of ammonia was quite significant on average 73%, while the reduction of COD was in a range from 20 to 60%. The Meteor pilot was able to treat and nitrify the raw water and produce an effluent that respects the guarantee of ammonia < $4.0mg\;NH_3-N/L$ when the raw water ammonia concentration was < $15mg\;NH_3-N/L$. The study identified operating parameters necessary to achieve the desired goal of adequate ammonia removal. The study results would benefit a range of systems across the country by providing guidance on the design and operation of a biological pre-treatment system for ammonia removal.
A novel hydrogen fermentation technique by using polyvinyl alcohol (PVA) gel beads as a biomass carrier was investigated. The hydrogen gas was stably produced throughout the experimental period in a continuous reactor. Even though the hydrogen productivity was suddenly decrease by experimental troubles, the bacteria attached to the PVA gel beads played as an inoculum, it was promptly recovered. The hydrogen yield per glucose was not very high ($1.0-1.2mol-H_2/mol-glucose$), thus the optimization of the experimental conditions such as ORP and HRT should be considered to improve the hydrogen productivity. Bacterial community was stable during experimental period after the PVA gel beads applying, which indicated that applying of biomass carrier was specific to keep not only the biomass but also the bacteria commonly. Clostridium species were phylogenetically detected, which suggested that these bacteria contributed to the hydrogen production in the biofilm attached to the PVA gel beads.
The purpose of this study is to investigate biomass characteristics and organic removal efficiency by changing superficial upflow velocity and organic loading rate in treating alcoholic distillery wastewater. Since the biomass concentration and the thickness of biofilm are very sensitive to superficial upflow velocity, a high concentration of biomass could be achieved by decreasing superficial upflow velocity that lowered the organic removal efficiency. Therefore, superficial upflow velocity should be controlled as to give optimum conditions and removal efficiency. Generally, activated sludge system shows 70% COD removal efficiency at$1.5kgCOD/m^3{\cdot}day$, but the fluidized-bed biofllm reactor shows 80% COD removal efficiency even at 6kgCOD/$m^2{\cdot}day$.
A fixed biofilm reactor system composed of anaerobic, anoxic(1), anoxic(2), aerobic(1) and aerobic(2) reactor was packed with synthetic activated ceramic (SAC) media and adopted to reduce the inhibition effect of low temperature on nitrification activities. The changes of nitrification activity at different wastewater temperature were investigated through the evaluation of temperature coefficient, volatile attached solid (VAS), specific nitrification rate and alkalinity consumption. Operating temperature was varied from 20 to $5^{\circ}C$. In this biofilm system, the specific nitrification rates of $15^{\circ}C$, $10^{\circ}C$ and $5^{\circ}C$ were 0.972, 0.859 and 0.613 when the specific nitrification rate of $20^{\circ}C$ was assumed to 1.00. Moreover the nitrification activity was also observed at $5^{\circ}C$ which is lower temperature than the critical temperature condition for the microorganism of activated sludge system. The specific amount of volatile attached solid (VAS) on media was maintained the range of 13.6-12.5 mg VAS/g media at $20{\sim}10^{\circ}C$. As the temperature was downed to $5^{\circ}C$, VAS was rapidly decreased to 10.9 mg VAS/g media and effluent suspended solids was increased from 3.2 mg/L to 12.0 mg/L due to the detachment of microorganism from SAC media. And alkalinity consumption was lower than theoretical value with 5.23 mg as $CaCO_3$/mg ${NH_4}^+$-N removal at $20^{\circ}C$. Temperature coefficient (${\Theta}$) of nitrification rate ($20^{\circ}C{\sim}5^{\circ}C$) was 1.033. Therefore, this fixed film nitrogen removal process showed superior stability for low temperature condition than conventional suspended growth process.

Space separation method that use independent reactor for nitrification and other reactor for denitrification has been commonly used for biological nitrogen removal process like $A^2O$ process. However, this method needs large space and complicate pipelines and time separation method such as SBR process have a difficulty in continuous treatment. Thus biological nitrogen removal process which is capable of continuous treatment, easy opeation and space saving is urgently required. In this research, submerged moving media was used for a biofilm process and suspended sludge was used for biological nitrogen removal at the same time. In particular DO environment by controlling air flow rate was investigated for simultaneous nitrification/denitrification. Total nitrogen removal in aeration rate more than $67L/min{\cdot}m^3$ showed 51~53% and rose to 65%, 70% and 78% in $50L/min{\cdot}m^3$, $58L/min{\cdot}m^3$ and $25L/min{\cdot}m^3$ respectively. Total phosphorus removal was very low about 10~20% more than $67L/min{\cdot}m^3$ aeration rates. But total phosphorus removal roses when reduces aeration rate by $58L/min{\cdot}m^3$ low and it showed total phosphorus removal of 72% in aeration rate $25L/min{\cdot}m^3$.
A pilot-scale biofilm nitiritation reactor was operated with the reject water from a large wastewater treatment plant. The effects of various operating parameters including pH, temperature, dissolved oxygen, solids and organic concentrations were examined. A stable nitritation was achieved at operating pH range of 7.3 to 8.8 with an alkalinity addition. Higher operating temperature of $35{\pm}0.7^{\circ}C$ achieved more stable nitritation compared to $30{\pm}0.2^{\circ}C$. It has been noticed that nitrite accumulation maintained with DO, solids and organic concentrations range of 0.8 to 3.9 mg/L, 3,400 to 11,000 mg/L, and 86 to 572 mg/L, respectively. It seems that the accumulation of nitrite was caused by both the inhibition of $NO_2{^-}$ oxidizers due to free ammonia and the maintenance of the high operating temperature of $35^{\circ}C$ which promote to accumulate the $NH_4{^+}$ oxidizers in the reactor. According to microbial community analysis of fluorescence in situ hybridization and INT-Dehydrogenase measurements, more nitrifiers were presented in attached form compared to suspended growth.
