Effect of Silk in Silk/PLGA Hybrid Films on Attachment and Proliferation of Human Aortic Endothelial Cells (실크/PLGA 하이브리드 필름에서 실크가 인간 대동맥 내피세포의 부착과 증식에 미치는 효과)

  • The vascular endothelial cells are the inner layers of blood vessels. It regulates the function of blood vessels and proliferation of vascular smooth muscle cells. Poly(lactide-co-glycolic acid) (PLGA) is a biodegradable synthetic polymer with a well-controlled degradation rate and an acceptable mechanical strength. It can be easily fabricated into many shapes. Silk consists of 18 amino acids. It found important for attaching cells cultured in vitro, and maintaining cell functions. In this study, we fabricated silk/PLGA biomaterial hybrid films of 0, 10, 20, 40 and 80 wt% silk. We performed MTT, SEM, ELISA, and immunocytochemistry analyses. We confirmed the adhesion and the proliferation of HAECs on silk/PLGA according to the content of silk, and 40 wt% silk/PLGA hybrid films have superior adhesion and proliferation properties. These results demonstrate that silk/PLGA hybrid films provide suitable surfaces for HAECs, and there is the effect of silk on cell growth and proliferation.

  • We studied on preparation of nanoparticles modified surface using biodegradable polymer, poly(DL-lactide-co-glycolide) (PLGA). Two kinds of PLGA nanoparticles were prepared by a spontaneous emulsification solvent diffusion (SESD) method using cetyltrimethylammonium chloride (CTAC) and tetradecyltrimethylammonium bromide (TTAB) as a cationic surfactant and polyethylene glycol-block-polypropylene glycol copolymer (Lutrol F68) as a nonionic surfactant. Model protein was coated on the surface of nanoparticles by the ionic complexation. The model protein was that influenza vaccine ($H_3N_2,\;H_1N_1$, B strain) labeled with NHS-fluorescein. The sizes of cationic nanoparticles were 140-160 nm and the surface charges were 50-60 mV. The sizes of nonionic nanoprticles were 80-90 nm and the surface charge was -10 mV. After coating vaccine on the surface of nanoparticles, the sizes of cationic nanoparticles were increased to 380-400 nm and the size of nonionic nanoparticles was not increased. The amount of coated vaccine on the cationic nanoparticles was 22.73 ${\mu}g$/mg.

Biotreatment Technologies for Air Pollution Control (생물학적 처리기술을 이용한 대기오염 제어)

  • Biological treatment is a relatively recent air pollution control technology in which off-gases containing biodegradable odors and volatile organic compounds(VOCs) are vented through microbes. It is a promising alternative to conventional air pollution control methods. Bioreactors for air pollution control have found most of their success in the treatment of dilute and high flow waste air streams containing VOCs and odor compounds. They offer several advantages over traditional technologies such as incineration or adsorption. These include lower treatment costs, absence of formation of secondary pollutants, no spent chemicals, low energy demand and low temperature treatment. The three most widely used technologies are described, namely biofiltration, biotrickling filtration, bioscrubbing. The most widely used bioreactor for air pollution control is biofilter, but it has several limitations. In the past years major progress has been accomplished in the development of vapor phase bioreaction systems, for solving problems of biofilter. Biotrickling filters are more complex than biofilters, but are usually more effective, especially for the treatment of compounds which are difficult to degrade or compounds that generate acidic by-products. This, paper reviews fundamental and theoretical/practical aspect of air pollution control in biofilter, biotrickling filter and bioscrubber, focusing more extensively on biotrickling filtration. Special emphasis is given to the operating parameters and the factors influencing performance for air pollution control, and cost estimation in biotreatment technologies.

Isolation of Microorganisms and Development of Microbial Augmentation for Treatment of Paper Mill Wastewater (제지폐수 처리용 미생물의 분리 및 복합 미생물제제의 개발)

  • This study was performed to investigate the effects of microbial augmentation on the biological treatment of paper mill wastewater. Three bacteria (KN11, KN13, KN27) capable of degrading aromatic compounds and a bacterial strain (GT21) producing an extracellular cellulase were isolated from soil and wastewater by selective enrichment culture. Through morphological, physiological, and biochemical taxonomies, isolated strains of KN11, KN13, KN27, and GT21 were identified as Acinetobacter sp., Neisseria sp., Bacillus sp., and Pseudomonas sp. and named Acinetobacter sp. KN11, Neisseria sp. KN13, Bacillus sp. KN27, and Pseudomonas sp. GT21, respectively. For analysis of non-biodegradable and chemical oxygen demand (COD)-increasing matter in a paper mill wastewater, we utilized GC/MS to detect aromatic compounds and their derivatives containing several substituted functional groups. The microbial augmentation, J30 formulated with the mixture of bacteria including Acinetobacter sp. KN11, Neisseria sp. KN13, Bacillus sp. KN27, and Pseudomonas sp. GT21, was used for the treatment of paper mill wastewater. The optimum temperature and pH for COD removal of the microbial augmentation, J30, were $30^{\circ}C$ and 7.5, respectively. For evaluation of the industrial applicability of the microbial augmentation, J30 in the pilot test, treatment efficiency was examined using paper mill wastewater. The microbial augmentation, J30, showed a COD removal rate of 87%. On the basis of the above results, we designed the wastewater treatment process of the activated sludge system.

Biodegradation Characteristics of Poly-3-hydroxybutyrate, $Sky-Green^R$ and $Mater-Bi^R$ by Soil Bacteria (토양세균의 Poly-3-hydroxybutyrate,$Sky-Green^R$$Mater-Bi^R$분해 특성)

  • Degradation behavior of the three commercial biodegradable polymers, namely poly(3-hydroxybutyrate) (PHB) Sky-Green/sup R/ (SG) and Mater-Bi/sup R/ (MB) was investigated using bacteria isolated from activated sludge and farm soil. Three PHB degrading bacteria, three SG degrading bacteria and one MB degrading bacteria were isolated. The PHB degrading bacteria were identified to be Flavimonas oryzihabitans, Corynebacterium pseudodiphtheriticum and Micrococcus diversus, while Pseudomonas vesicuraris, Pasteurlla multocida and Flavobacterium odoratum were identified as SG degrading bacteria. As for MB, Pseudomonas vesicuraris was isolated. The shake flask test for 28 days indicated that the rate of biodegradation of PHB, SG and MB in terms of weight loss were about 44∼69% 25∼32% and 29% respectively. The surface morphology of PHB, SG andMB films before and after degradation by microorganisms in an activated sludge soil was observed under SEM, demonstrating that the film surface had a very porous structure, and that microorganisms colonized heavily on the film surface. TOC and pH variation as a result of abiotic hydrolysis, or microbial growth in the absence of the polymers were compared to those due to degradation by F. oryzihabitans. Abiotic hydrolysis of PHB was three times as fast as that of SG and MB. Addition of yeast extract to the basal liquid medium accelerated the biodegradation of the polymers. Biodegradation of PHB was always faster than that of SG and MB irrespectively of the presence of yeast extract in the basal liquid medium.

Refractory Textile Wastewater Treatment Using Cell-Immobilized Polyethylene glycol Media (PEG 포괄고정화담체를 이용한 난분해성 염색폐수 처리)

  • This study investigated the removal of recalcitrant organics in dyeing wastewater using a fluidized bed reactor(FBR) that contained cell-immobilized pellets. The pellets were manufactured and condensing the gel phase by mixing PEG-polymer and cells to form micro-porous PEG-polymer pellets whose size were ${\Phi}\;4mm{\times}H\;4mm$ on average. An industrial activated sludge without any pre-adaptation was used for the cell immobilization because it gave an equivalent removal efficiency to a pre-adapted sludges. The feed was obtained from an effluent of a biological treatment plant, which contained $SCOD_{Cr}$ of 330 mg/L and $SBOD_5$ of 20 mg/L. The $SCOD_{Cr}$ removal efficiency was over 45% and the effluent $COD_{Mn}$ concentration was less than 100 mg/L at HRTs from 6 to 24 hrs. The optimum HRT in the FBR was determined as 12 hrs considering the removal efficiency and cost. When a raw wastewater containing 768 mg/L of $COD_{Cr}$ was fed to the FBR, the effluent $COD_{Cr}$ concentration increased only slightly, giving a 70% of $COD_{Cr}$ removal or a 97% of $BCOD_5$ removal. This indicated that the FBR had an excellent capability of biodegradable organics removal also. In conclusion, the FBR could be applied to textile wastewater treatment in place of an activated sludge process.

Combined Treatment of Livestock Wastewater with Sewage Using Phanerochaete chrysosporium PSBL-1 (Phanerochaete chrysosporium PSBL-1을 이용한 축산폐수와 하수의 연계처리)

  • We studied possibility of mixing treatment of livestock wastewater and sewage using Phanerochaete chrysosporium PSBL-1. Our study showed that 97.6% of SS and 95% of T-P removal efficiency was achieved when 2 mL BF02(a coagulant) and 100 mL C-210EL(a cationic polymer) were added to the mixture(2:1, v/v) of livestock wastewater and sewage. We studied treatment characteristic of Phanerochaete chrysosporium PSBL-1, after were mixed pretreated wastewater and sewage by dillution ten times about livestock wastewater. The removal efficiency of NBDCOD(non-biodegradable COD), $NH_3-N$ and T-N was increased according to increase of pH. That is, T-N concentration of effluent was satisfied 60 mg/L by drain water waterqulity standard of livestock wastewater public treatment facilities with 35 mg/L from a lapse of five days at pH 6.7, 51 mg/L from a lapse of three days at pH 8 and 33 mg/L from a lapse of one day at pH 10. Moreover $COD_{Mn}$ concentration of effluent was satisfied 40 mg/L by drain water waterqulity standard of livestock wastewater public treatment facilities after a laps of one day at all pH. Organics and nitrogen concentrations of effluent were higher case with addition of V.A.(veratryl alcohol) than case without addition of V.A.(veratryl alcohol). $COD_{Mn}$ concentration of effluent satisfied drain water qulity standard of livestock wastewater public treatment facilities from a lapse of one day, when C/N rate(3:1) of influent was not controled. T-N satisfied that from a lapse of two days, when C/N rate was controled with $4{\sim}6$.

Development and Full-scale Application of the Alternative Carbon Source Based on the Substrate Compatibility (미생물 순응 호환성에 기반한 대체탄소원 개발 및 실용화 사례)

  • J sewage treatment plant (WWTP) in Busan has used methanol as an external carbon source for the biological denitrification process. Methanol is widely used. but rather expensive and very dangerous in handling. Therefore, it has been required that the economic alternative carbon source must be developed. By-product from a fine chemical industry can be Purified by removing high molecular weight substances using the ultrafilter membrane separation process and RBDCOD fraction becomes $98{\sim}99%$ of COD substances in the purified by-product. The purified by-product containing three types of alcohols, methanol, prophylenglycol and methoxypropanol; showed similar chemical characteristics to the methanol, a main external carbon source, in biodegradation pathway. Shown above, the compatibility between main and alternative carbon sources has been achieved. Also very short or no adaptation period is necessary in the case of exchanging these carbon sources. The compatibility between external carbon sources is an essential element for stabilizing WWTP operations. During the full-scale application test of the by-product, the alternative carbon source line got on par with the treatment efficiency of the methanol line. With the test result, J-WWTP changed methanol to a fine chemical by-product, in two out of three J-WWTP lines. Moreover, it is expected that 55.4% of the external carbon source cost reduction can be achieved in the alternative carbon source applied lines.

Structural and Kinetic Characteristics of 1,4-Dioxane-Degrading Bacterial Consortia Containing the Phylum TM7

  • 1,4-Dioxane-degrading bacterial consortia were enriched from forest soil (FS) and activated sludge (AS) using a defined medium containing 1,4-dioxane as the sole carbon source. These two enrichments cultures appeared to have inducible tetrahydrofuran/dioxane and propane degradation enzymes. According to qPCR results on the 16S rRNA and soluble di-iron monooxygenase genes, the relative abundances of 1,4-dioxane-degrading bacteria to total bacteria in FS and AS were 29.4% and 57.8%, respectively. For FS, the cell growth yields (Y), maximum specific degradation rate ($V_{max}$), and half-saturation concentration ($K_m$) were 0.58 mg-protein/mg-dioxane, $0.037mg-dioxane/mg-protein{\cdot}h$, and 93.9 mg/l, respectively. For AS, Y, $V_{max}$, and $K_m$ were 0.34 mg-protein/mg-dioxane, $0.078mg-dioxane/mg-protein{\cdot}h$, and 181.3 mg/l, respectively. These kinetics data of FS and AS were similar to previously reported values. Based on bacterial community analysis on 16S rRNA gene sequences of the two enrichment cultures, the FS consortium was identified to contain 38.3% of Mycobacterium and 10.6% of Afipia, similar to previously reported literature. Meanwhile, 49.5% of the AS consortium belonged to the candidate division TM7, which has never been reported to be involved in 1,4-dioxane biodegradation. However, recent studies suggested that TM7 bacteria were associated with degradation of non-biodegradable and hazardous materials. Therefore, our results showed that previously unknown 1,4-dioxane-degrading bacteria might play an important role in enriched AS. Although the metabolic capability and ecophysiological significance of the predominant TM7 bacteria in AS enrichment culture remain unclear, our data reveal hidden characteristics of the TM7 phylum and provide a perspective for studying this previously uncultured phylotype.

Clinical Study on Therapeutic Effects of Biodegradable membrane $Biomesh^{(R)}$ and autogenous bone grafts in infrabony defects (골내치주낭에 $Biomesh^{(R)}$ 차단막과 자가골이식의 치료효과에 대한 연구)

  • The ultimate goal of periodontal disease therapy is to promote the regeneration of lost periodontal tissue, there has been many attempts to develop a method to achieve this goal, but none of them was completely successful. This study was designed to compare the effects of treatment using resorbable barrier membrane($Biomesh^{?}$) in combination with autogenous bone graft material with control treated by only modified Widman flap. 22 infrabony defecs from 10 patients with chronic periodontitis were used for this study, 10 sites of them were treated with resorbable barrier membrane and autogenous bone graft material as experimental group and 12 site were treated by only modified Widman flap as control group. Clinical parameters including probing depth, gingival recession, bone probing depth and loss of attachment were recorded at 6-8 months later, and the significance of the changes was statistically analyzed. The results are as follows : 1. Probing depth of the two group was reduced with statistically significance(P<0.05), but this changes were not different between the two experiment, control group with statistically significance. 2. Gingival recession showed statistically significant increase in control group(P<0.05), but not in experimental group, and initial values of the two group were in statistically significant difference(P<0.05). 3. Bone probing depth showed statistically significant decrease in experimental group(P<0.05), but not in control group, and this changes were different between the two experiment, control group with statistically significance(P<0.05). 4. Loss of attachment showed statistically significant decrease in experimental group(P<0.05), but not in control group, and this changes were different between the two experiment, control group with statistically significance(P<0.05) On the basis of these results, treatment using resorbable barrier membrane in combination with autogenous bone graft material improve the probing depth, bone probing depth and loss of attachment in infrabony defects.
