Brief Introduction of Research Progresses in Control and Biocontrol of Clubroot Disease in China

  • Clubroot disease of crucifers has occurred since 1957. It has spread to the whole China, especially in the southwest and nourtheast where it causes 30-80% loss in some fields. The disease has being expanded in the recent years as seeds are imported and the floating seedling system practices. For its effective control, the Ministry of Agriculture of China set up a program in 2010 and a research team led by Dr. Yueqiu HE, Yunnan Agricultural University. The team includes 20 main reseachers of 11 universities and 5 institutions. After 5 years, the team has made a lot of progresses in disease occurrence regulation, resources collection, resistance identification and breeding, biological agent exploration, formulation, chemicals evaluation, and control strategy. About 1200 collections of local and commercial crucifers were identified in the field and by artificiall inoculation in the laboratories, 10 resistant cultivars were breeded including 7 Chinese cabbages and 3 cabbages. More than 800 antagostic strains were isolated including bacteria, stretomyces and fungi. Around 100 chemicals were evaluated in the field and greenhouse based on its control effect, among them, 6 showed high control effect, especially fluazinam and cyazofamid could control about 80% the disease. However, fluzinam has negative effect on soil microbes. Clubroot disease could not be controlled by bioagents and chemicals once when the pathogen Plasmodiophora brassicae infected its hosts and set up the parasitic relationship. We found the earlier the pathogent infected its host, the severer the disease was. Therefore, early control was the most effective. For Chinese cabbage, all controlling measures should be taken in the early 30 days because the new infection could not cause severe symptom after 30 days of seeding. For example, a biocontrol agent, Bacillus subtilis Strain XF-1 could control the disease 70%-85% averagely when it mixed with seedling substrate and was drenching 3 times after transplanting, i.e. immediately, 7 days, 14 days. XF-1 has been deeply researched in control mechanisms, its genome, and development and application of biocontrol formulate. It could produce antagonistic protein, enzyme, antibiotics and IAA, which promoted rhizogenesis and growth. Its The genome was sequenced by Illumina/Solexa Genome Analyzer to assembled into 20 scaffolds then the gaps between scaffolds were filled by long fragment PCR amplification to obtain complet genmone with 4,061,186 bp in size. The whole genome was found to have 43.8% GC, 108 tandem repeats with an average of 2.65 copies and 84 transposons. The CDSs were predicted as 3,853 in which 112 CDSs were predicted to secondary metabolite biosynthesis, transport and catabolism. Among those, five NRPS/PKS giant gene clusters being responsible for the biosynthesis of polyketide (pksABCDEFHJLMNRS in size 72.9 kb), surfactin(srfABCD, 26.148 kb, bacilysin(bacABCDE 5.903 kb), bacillibactin(dhbABCEF, 11.774 kb) and fengycin(ppsABCDE, 37.799 kb) have high homolgous to fuction confirmed biosynthesis gene in other strain. Moreover, there are many of key regulatory genes for secondary metabolites from XF-1, such as comABPQKX Z, degQ, sfp, yczE, degU, ycxABCD and ywfG. were also predicted. Therefore, XF-1 has potential of biosynthesis for secondary metabolites surfactin, fengycin, bacillibactin, bacilysin and Bacillaene. Thirty two compounds were detected from cell extracts of XF-1 by MALDI-TOF-MS, including one Macrolactin (m/z 441.06), two fusaricidin (m/z 850.493 and 968.515), one circulocin (m/z 852.509), nine surfactin (m/z 1044.656~1102.652), five iturin (m/z 1096.631~1150.57) and forty fengycin (m/z 1449.79~1543.805). The top three compositions types (contening 56.67% of total extract) are surfactin, iturin and fengycin, in which the most abundant is the surfactin type composition 30.37% of total extract and in second place is the fengycin with 23.28% content with rich diversity of chemical structure, and the smallest one is the iturin with 3.02% content. Moreover, the same main compositions were detected in Bacillus sp.355 which is also a good effects biocontol bacterial for controlling the clubroot of crucifer. Wherefore those compounds surfactin, iturin and fengycin maybe the main active compositions of XF-1 against P. brassicae. Twenty one fengycin type compounds were evaluate by LC-ESI-MS/MS with antifungal activities, including fengycin A $C_{16{\sim}C19}$, fengycin B $C_{14{\sim}C17}$, fengycin C $C_{15{\sim}C18}$, fengycin D $C_{15{\sim}C18}$ and fengycin S $C_{15{\sim}C18}$. Furthermore, one novel compound was identified as Dehydroxyfengycin $C_{17}$ according its MS, 1D and 2D NMR spectral data, which molecular weight is 1488.8480 Da and formula $C_{75}H_{116}N_{12}O_{19}$. The fengycin type compounds (FTCPs $250{\mu}g/mL$) were used to treat the resting spores of P. brassicae ($10^7/mL$) by detecting leakage of the cytoplasm components and cell destruction. After 12 h treatment, the absorbencies at 260 nm (A260) and at 280 nm (A280) increased gradually to approaching the maximum of absorbance, accompanying the collapse of P. brassicae resting spores, and nearly no complete cells were observed at 24 h treatment. The results suggested that the cells could be lyzed by the FTCPs of XF-1, and the diversity of FTCPs was mainly attributed to a mechanism of clubroot disease biocontrol. In the five selected medium MOLP, PSA, LB, Landy and LD, the most suitable for growth of strain medium is MOLP, and the least for strains longevity is the Landy sucrose medium. However, the lipopeptide highest yield is in Landy sucrose medium. The lipopeptides in five medium were analyzed with HPLC, and the results showed that lipopeptides component were same, while their contents from B. subtilis XF-1 fermented in five medium were different. We found that it is the lipopeptides content but ingredients of XF-1 could be impacted by medium and lacking of nutrition seems promoting lipopeptides secretion from XF-1. The volatile components with inhibition fungal Cylindrocarpon spp. activity which were collect in sealed vesel were detected with metheds of HS-SPME-GC-MS in eight biocontrol Bacillus species and four positive mutant strains of XF-1 mutagenized with chemical mutagens, respectively. They have same main volatile components including pyrazine, aldehydes, oxazolidinone and sulfide which are composed of 91.62% in XF-1, in which, the most abundant is the pyrazine type composition with 47.03%, and in second place is the aldehydes with 23.84%, and the third place is oxazolidinone with 15.68%, and the smallest ones is the sulfide with 5.07%.

A Synergistic Effect of Chitosan and Lactic Acid Bacteria on the Control of Cruciferous Vegetable Diseases

  • Two lactic acid bacteria (LAB) designated J02 and J13 were recovered from fermented vegetables based on their ability to suppress soft rot disease caused by Pectobacterium carotovorum subsp. carotovorum (Pcc) on radish. J02 and J13 were identified as Lactobacillus pentosus and Leuconostoc fallax, respectively. The ability of J02 and J13 to suppress plant diseases is highly dependent on chitosan. LAB alone has no effect and chitosan alone has only a moderate effect on disease reduction. However, J02 or J13 broth cultures plus chitosan display a strong inhibitory effect against plant pathogens and significantly reduces disease severity. LAB strains after being cultured in fish surimi (agricultural waste) and glycerol or sucrose-containing medium and mixed with chitosan, reduce three cruciferous vegetable diseases, including cabbage black spot caused by Alternaria brassicicola, black rot caused by Xanthomonas campestris pv. campestris, and soft rot caused by Pcc. Experimental trials reveal that multiple applications are more effective than a single application. In-vitro assays also reveal the J02/chitosan mixture is antagonistic against Colletotrichum higginsianum, Sclerotium rolfsii, and Fusarium oxysporum f. sp. rapae, indicating a broad-spectrum activity of LAB/chitosan. Overall, our results indicate that a synergistic combination of LAB and chitosan offers a promising approach to biocontrol.

Enhancement of Biocontrol Activity of Serratia plymuthica A21 -4 Toward Phytophthora Blight of Pepper by Amendment of Nutritional Condition

  • Serratia plymuthim A21-4 strongly inhibits the mycelial growth, zoospore formation, and cystospore germination of Phytophthor spp and Pythium species. The bacterial isolate produced antifungal substance and chitinase. The bacteria also enhanced to plant growth remarkably in low nutritional condition. The application of cell suspension of A21-4 to pepper seedlings in greenhouse experiments and soil drenching in farmer's field was proved successfully to control the phythophthora blight of pepper. For the effective control, however, relatively high density of cell number(10$\^$9/cfu/$m\ell$) is required. Density effect was similar in plant growth promoting activity of A21-4. Though this investigation we improved the problem with changes of culture condition of bacteria and some nutritional amendment.

Bacillus sp. BS061 Suppresses Powdery Mildew and Gray Mold

  • The use of a microorganism, or its secretions, to prevent plant disease offers an attractive alternative or supplement to synthetic fungicides for the management of plant disease without the negative effects of chemical control mechanisms. During a screening for microorganisms with the potential to be used as microbial fungicides, Bacillus sp. BS061 was isolated from a plant leaf. The strain BS061 potently inhibited the mycelial growth of Botrytis cinerea, and significantly reduced disease incidence of powdery mildew in cucumber and strawberry. We also found that the culture filtrate of BS061 inhibited the mycelial growth of various plant pathogens.

Isolation, Purification, and Structural Identification of an Antifungal Compound from a Trichoderma Strain

  • Trichoderma strain T-33 has been demonstrated to have inhibitory effect on the fungus species Cytospora chrysosperma. Here, an active antifungal compound was obtained from Trichoderma strain T-33 extract via combined separation technologies, including organic solvent extraction, liquid chromatography, and thin-layer chromatography. The purified compound was further characterized by advanced analytical technologies to elucidate its chemical structure. Results indicated that the active antifungal compound in Trichoderma strain T-33 extract is 2,5-cyclohexadiene-1,4-dione-2,6-bis (1,1-dimethylethyl).

Bacterial endophytes from ginseng and their biotechnological application

  • Ginseng has been well-known as a medicinal plant for thousands of years. Bacterial endophytes ubiquitously colonize the inside tissues of ginseng without any disease symptoms. The identification of bacterial endophytes is conducted through either the internal transcribed spacer region combined with ribosomal sequences or metagenomics. Bacterial endophyte communities differ in their diversity and composition profile, depending on the geographical location, cultivation condition, and tissue, age, and species of ginseng. Bacterial endophytes have a significant effect on the growth of ginseng through indole-3-acetic acid (IAA) and siderophore production, phosphate solubilization, and nitrogen fixation. Moreover, bacterial endophytes can protect ginseng by acting as biocontrol agents. Interestingly, bacterial endophytes isolated from Panax species have the potential to produce ginsenosides and bioactive metabolites, which can be used in the production of food and medicine. The ability of bacterial endophytes to transform major ginsenosides into minor ginsenosides using β-glucosidase is gaining increasing attention as a promising biotechnology. Recently, metabolic engineering has accelerated the possibilities for potential applications of bacterial endophytes in producing beneficial secondary metabolites.

토끼풀(Trifolium repens L.) 방제용 생물제제 Penicillium oxalicum (PENOX)의 발아, 생장, 포자생성 및 탄소원이용에 미치는 수분활성 및 온도의 영향 (Effect of Water Activity and Temperature on Growth, Germination, Sporulation, and Utilization of Carbon Source of Penicillium oxalicum (PENOX) as a Biocontrol Agent(BCA) for control of Clover(Trifolium repens L.))

  • Penicillium oxalicum (PENOX)은 잔디밭 토끼풀(Trifolium repens L.)의 방제를 위한 생물제제(biocontrol agent, BCA)로서의 잠재성을 보여왔다. 미생물제초균류에 의한 제초활성은 다양한 환경조건하에서 이들의 발아율, 생장력 및 기질이용성 등에 크게 영향을 받는다. 따라서 본 연구는 PENOX의 다양한 수분확성($0.94{\sim}0.995\;a_w$) 및 온도조건($18{\sim}30^{\circ}C$)하에서의 균사생장, 포자발아율 및 포자생성량을 glycerol로 조정된 MEA(malt extract agar) 배지를 사용하여 in vitro상에서 조사하였다. PENOX의 균사생장은 높은 수분활성조건($0.995\;a_w$)에서 가장 좋았으며 $30^{\circ}C$에서 가장 양호하였다. 또한 $0.995\;a_w$에서는 저온($20^{\circ}C$)에서도 균사생장이 대체로 양호하여 수분활성이 약간 낮은 조건($0.97\;a_w$)에서의 최대 균사생장율과 비슷한 균사생장율을 나타냈다. $0.97\;a_w$에서의 균사생장율은 $25^{\circ}C$$30^{\circ}C$조건간에 대체로 비슷한 결과를 나타냈으며 저온($20^{\circ}C$)에서는 매우 감소하였다. PENOX의 포자발아율도 $0.995\;a_w/30^{\circ}C$에서 가장 높았으며 수분활성이 감소할수록 낮아지는 경향을 나타냈다. 저온($18^{\circ}C$)에서의 발아전 지체시간(lag phase time)은 전 $a_w$ 조건하에서 6시간 이상 그리고 낮은 수분활성조건($0.94\;a_w$)하에서는 18과 $25^{\circ}C$에서 각 18, 12시간 이상으로 나타났다. 그러나 포자생성은 $0.995\;a_w$의 높은 수분조건에 비해 약간 낮은 $0.97\;a_w$ 조건에서, 그리고 고온($30^{\circ}C$)에 비해 저온조건($20^{\circ}C$) 하에서 훨씬 양호하였다. 한편, GN MicroPlate를 이용해 PENOX의 기질로서의 95개 탄소원에 대한 이용성을 조사한 결과 $0.995\;a_w/25^{\circ}C$에서의 생태적 생식역값(niche size)은 86으로 가장 높았으며 수분스트레스조건($0.955\;a_w$)에서는 그 이용성이 낮아져 그 값은 65를 나타냈다. 그리고 고온($30^{\circ}C$)에서는 $0.995\;a_w$$0.955\;a_w$에서는 각각 84와 50을 나타내 $0.995\;a_w$에서 보다 $0.955\;a_w$ 조건에서 탄소원 이용시 온도스트레스에 더 민감하게 반응하는 것으로 나타났다. 이러한 균류생태생리학적인 정보는 효과적인 BCA의 개발을 위한 기초자료로서 유용하게 활용될 수 있을 것이다.

Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

이유자돈에서 Salmonella Typhimurium 감염에 대한 박테리오파지의 방어 효능 (Protective effect of bacteriophages against Salmonella Typhimurium infection in weaned piglets)

  • Salmonellosis has caused heavy losses in swine industry and implications for public health. Recently, the urgent problem of antibiotic resistance due to multidrug-resistant Salmonella spp. has been on the rise. The use of host-specific bateriophages as a biocontrol is one possible alternative. In this study, clinical signs, growth performance, quantification and detection of antigen, histopathological changes of gastrointestinal tracts were analyzed comparatively in weaned piglets according to administration of bacteriophages and challenge with Salmonella (S.) Typhimurium. Piglets challenged with S. Typhimurium after administered with bacteriophages showed reduced clinical signs, higher growth performance, lower bacterial shedding, lower quantificational value of antigens in intestines, higher V/C ratio and higher the number of goblet cells in intestines than piglets administered without bacteriophage and challenged with S. Typhimurium. These results indicate that feeding contained with bacteriophages has effect to prevent infection of S. Typhimurium in weaned piglets and suggest that a use of bacteriophage can be considered a valid antibiotic alternative.

선충 기생 전적 진균의 접종원 농도와 온도조건에 따른 성충감염 및 집단 감소효과 (Decrease of Nematode Population by Introduction of Nematophagous Fungi into The Soil as Affected by Inoculum Concentration and Temperature in Vitro)

  • Five nematophagous fungi were evaluated for their nematicidal effect in vitro on Rhabditis sp. and Meloidogyne hapla in soil. Inocula of Arthrobotrys arthrobotryoides, A. conoides, A. oligospora, Dactylella lobata, and Fusarium oxyaporum were grown in moistened corn-sandy soil and chopped potato-sandy soil media, and incubated at 26$^{\circ}C$ for one week. The prepared inocula were incorporated in autoclaved sandy soil, mixing thoroughly at rates equ-invalent to 1:50, 1:100, 1:200, and 1:400, repectively, before 80g of the mixture carrying 100 Rhabditis sp. was put into petri plates. Nematophagous fungi effectively teduced the popuation of Rhabditis sp. in soil in a week or two following treatment of the incula at concentration of 1:50 and 1:100. The optimum was at $25^{\circ}C$ for nematicidial effect as high as 80-100%. The at the rate of 1:100 prepared incula were incorporated in auto-claved soil, where 100 Juveniles M. hapla were introduced per 80% soil. All fungi infected the M. hapla effectively in soil, caysing more than 90% mortality within one week. This result indicated the potential value of these fungi as promising biocontrol agents.
