Isolation and Characterization of Bacteriophages Against Pseudomonas syringae pv. actinidiae Causing Bacterial Canker Disease in Kiwifruit

  • Pseudomonas syringae pv. actinidiae causes bacterial canker disease in kiwifruit. Owing to the prohibition of agricultural antibiotic use in major kiwifruit-cultivating countries, alternative methods need to be developed to manage this disease. Bacteriophages are viruses that specifically infect target bacteria and have recently been reconsidered as potential biological control agents for bacterial pathogens owing to their specificity in terms of host range. In this study, we isolated bacteriophages against P. syringae pv. actinidiae from soils collected from kiwifruit orchards in Korea and selected seven bacteriophages for further characterization based on restriction enzyme digestion patterns of genomic DNA. Among the studied bacteriophages, two belong to the Myoviridae family and three belong to the Podoviridae family, based on morphology observed by transmission electron microscopy. The host range of the selected bacteriophages was confirmed using 18 strains of P. syringae pv. actinidiae, including the Psa2 and Psa3 groups, and some were also effective against other P. syringae pathovars. Lytic activity of the selected bacteriophages was sustained in vitro until 80 h, and their activity remained stable up to 50℃, at pH 11, and under UV-B light. These results indicate that the isolated bacteriophages are specific to P. syringae species and are resistant to various environmental factors, implying their potential use in control of bacterial canker disease in kiwifruits.

기내 검정법을 이용한 국내 과수 화상병 방제제 선발 (In Vitro Screening of Antibacterial Agents for Suppression of Fire Blight Disease in Korea)

  • 국내 과수 화상병은 2015년 사과 및 배에서 발생하여 큰 피해를 주고 있으나, 이에 대한 적절한 방제제는 등록되어 있지 않다. 다만 농촌진흥청은 무름병과 세균점무늬병 등 다른 식물세균병해에 등록되어 있는 살세균제들을 직권등록 시켜놓은 상태이다. 그러나 직권등록이란, 약효는 보증하지 않으면서 약해만을 보증하는 제도로, 국내에서 과수 화상병이 지속적으로 발생되고 있는 상황에서는 적절한 방제제를 선발하는 것이 요구되어지기에 본 연구를 수행하였다. 직권등록 국내 과수 화상병 방제제 중, 각각 4종의 농용항생제와 구리 합성 화학물 그리고 생물학적 방제제 2종에 대한 기내 검정법을 통하여 국내 과수 화상병원세균 억제 및 병징확산 억제효과를 검증하였다. Kasugamycin, Oxytetracycline 및 streptomycin의 항생제는 효과가 우수하였으며, oxolinic acid는 꽃에서의 억제효과는 우수하였지만 신초 및 미성숙과 기내검정에서는 다소 효과가 감소하였다. 4종의 구리 합성 화학물은 항생제만큼의 억제효과는 나타내지 않았지만, 사용가능한 수준으로 조사되었다. 또한 2종의 생물적 방제제는 처리시기 등을고려할 경우, 친환경 재배지에서는 사용 가능할 것으로 판단되었다. 한편 국내 과수 화상병원세균은 선발 방제제 중 항생제 저항성 유전자들을 가지고 있지 않고,화분매개충에 대한 영향도 없는 것으로 조사되었다.

Comparative Proteome Analysis of Two Antagonist Bacillus subtilis Strains

  • Natural wild-type strains of Bacillus subtilis are extensively used in agriculture as biocontrol agents for plants. This study examined two antagonist B. subtilis strains, KB-1111 and KB-1122, and the results illustrated that KB-1122 was a more potent inhibitor of the indicator pathogen than KB-1111. Thus, to investigate the intrinsic differences between the two antagonist strains under normal culture conditions, samples of KB-1111 and KB-1122 were analyzed using MALDI-TOF-MS. The main differences were related to 20 abundant intracellular and 17 extracellular proteins. When searching the NCBI database, a number of the differentially expressed proteins were identified, including 11 cellular proteins and 10 secretory proteins. Among these proteins, class III stress-response-related ATPase, aconitate hydratase, alpha-amylase precursor, and a secretory protein, endo-l, 4-beta-glucanase, were differentially expressed by the two strains. These results are useful to comprehend the intrinsic differences between the antagonism of KB-1111 and KB-1122.

상추균핵병의 생물적 방제를 위한 Bacillus amyloliquefaciens M27 선발 (Selection of Bacillus amyloliquefaciens M27 for Biocontrol on Lettuce Sclerotinia Rot)

  • 시설재배작물 상추의 Sclerotinia sclerotiorum와 Sclerotinia minor에 의한 균핵병을 생물적으로 방제하기 위하여 분리한 수원을 비롯한 6개 지역에서 토양 등에서 29균주의 세균을 분리하였다. 분리세균에 대하여 균사생육저지원법과 생물검정법을 통하여 세균 M27와 RM43균주가 선발되었다. 이 선발균주에서 Bacillus amyloliquefaciens M27균주가 균핵병균을 가장 효과적으로 억제하였다.

생물방제균 Bacillus subtilis YB-70이 생산하는 항진균성 항생물질의 분리 및 구조결정

  • A potential biocontrol bacterium, YB-70 was isolated from a rhizosphere in suppressive soil and identified as a strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on Fusarium solani with the culture filterates from B. subtilis YB-70, we found that antifungal mechanism of B. subtilis YB-70 was mediated by antibiotic substances produced from the bacterium. These antifungal substances were appeared to be hear-resistant, micromolecular, and ethy alcohol soluble. Antifungal agents produced by B. subtilis YB-70 showed strong inhibified against root-rotting fungi F. solani in in vivo pot test. An antifungal substance. YBS-1s, was purified from the culture broth of B. subtilis YB-70 by isoelectronic precipitation, silica gel column chromatography and Sephadex LH-20 column chromatography analysis by Fab-MASS, $^{1}$H-NMR, $^{13}$C-NMR, DEPT, and amino acid analyzer revealed that the YBS-1A was a peptide antibiotics of iturin class containing seven amino acids from five different groups, and the other(YBS-1B) was an analogue of iturin group composed of 11 amino acids with larher molecular weight of about 1, 500 dalton, which was lager than that of iturin A.

Screening rhizobacteria for biological control of root rot and Phytophthora blight on glnseng.

  • Ginseng (Panax ginseng) is one of the most widely cultivated medicinal herbs in Korea. During 3 or 5 years cultivation of ginseng, yield losses can reach as high as 30-60% due to numerous diseases in Korea. Among 106 Bacillus strains isolated from various plant internal roots, we selected three promising biocontrol agents by screening against root rot caused by Cylindrocarpon destructan in a greenhouse. Preinoculation of selected isolates to seed or one-year-old root resulted in stimulation of shoot and/or root growth of seedlings, and control of root rot in infested soils with Cylindronrpon destructans (P=0.05). Furthermore, drenching of selected isolates on seedling-growing pots reduced the incidence of Phytophthora blight when the seedlings were challenged with zoospores of Phytophthora cactorum (P=0.05). However, isolates B1141 and B1142 did not show any antifungal activity against various soilborne pathogens while B1146 did in vitro. Our results provide an insight that rhizobacteria can induce resistance against various plant diseases on ginseng even if any resistant breeds have been unknown on ginseng yet.

Survey of Egg- and Cyst-parasitic Fungi of Potato Cyst Nematode in Indonesia

  • Twelve fungal isolates out of 123 isolates obtained from cysts and soils of potato cyst nematode (PCN)-infested fields in Central Java, Indonesia had parasitic abilities of over 50% on PCN eggs or females (cysts) in vitro pathogenicity tests. Cultural and morphological characters and DNA sequences of ribosomal genes in ITS region revealed that they were four isolates of Gliocladium (Trichoderma) virens, three isolates of Fusarium oxysporum, one of F. lateritium, one of Penicillium tritinum and two of Taralomyces spp. A hundred percent infections occurred in eggs or cysts by three fungal isolates G. virens, F. oxysporum and P. oxalicum, suggesting that these fungi may have a good potential for the PCN biocontrol. Especially, G. virens isolates, which occurred most frequently in the PCN-infested potato fields and are known to be highly adaptable to varying habitats, may be developed as reliable agents for controlling PCN with both egg- and cyst-parasitic capabilities and with high ecological adaptabilities.

  • Biotechnological researches in Central Asian Research Institute of Sericulture were developed since 1981 when laboratory of artificial diets for mulberry silk­worm was created. The researches were directed not only to creation of accessible artificial diets (AD) for off-season mass rearing of economical beneficial insect and for industrialization of sericulture but also to obtaining of easy reproducible source of animal proteins, carbohydrates, lipids, enzymes, enzymes inhibitors and other metabolites - bioresource for manufacture of oriental medicine preparations. Space experiments with silkworms became possible with use of AD. These experiments used mulberry silkworm as high effective board test organism with outstanding developmental, reproductive and transgenic properties having impor­tant significance in future of cosmonautics. Introduction to practice of bioprotective and ecologically pure method of drying and sterilizing of green cocoons and by-products of silk manufacture allows to increase efficiency of cocoons reeling technology as well as to preserve nativity of biological active substances in mulberry silkworm pupae that is unachievable by traditional methods of raw material processing. New spheres of application of two important silk proteins - fibroin and sericin in medicine, cosmetics, for bio- and nanotechnology are opening with use of biotechnological approaches. At present the achievements and experience of R & D in these fields are used in creation of modern pests biocontrol agents. Identification and Characterization of the Antifungal Substances of a Novel Streptomyces cavourensis NA4

  • A new actinomycete strain NA4 was isolated from a deep-sea sediment collected from the South China Sea and showed promising antifungal activities against soilborne fungal pathogens. It was identified as Streptomyces cavourensis by morphological, physiological, and phylogenetic analyses based on its 16S rRNA gene sequence. The main antifungal components were isolated and identified from the fermentation culture as bafilomycins B1 and C1. These compounds exhibited significant antifungal activities and a broad antifungal spectrum. The results suggest that the Streptomyces cavourensis NA4 and bafilomycins B1 and C1 could be used as potential biocontrol agents for soilborne fungal diseases of plants.

Biological Control of Apple Anthracnose by Paenibacillus polymyxa APEC128, an Antagonistic Rhizobacterium

  • The present study investigated the suppression of the disease development of anthracnose caused by Colletotrichum gloeosporioides and C. acutatum in harvested apples using an antagonistic rhizobacterium Paenibacillus polymyxa APEC128 (APEC128). Out of 30 bacterial isolates from apple rhizosphere screened for antagonistic activity, the most effective strain was APEC128 as inferred from the size of the inhibition zone. This strain showed a greater growth in brain-heart infusion (BHI) broth compared to other growth media. There was a reduction in anthracnose symptoms caused by the two fungal pathogens in harvested apples after their treatment with APEC128 in comparison with non-treated control. This effect is explained by the increased production of protease and amylase by APEC128, which might have inhibited mycelial growth. In apples treated with different APEC128 suspensions, the disease caused by C. gloeosporioides and C. acutatum was greatly suppressed (by 83.6% and 79%, respectively) in treatments with the concentration of $1{\times}10^8$ colony forming units (cfu)/ml compared to other lower dosages, suggesting that the suppression of anthracnose development on harvested apples is dose-dependent. These results indicated that APEC128 is one of the promising agents in the biocontrol of apple anthracnose, which might help to increase the shelf-life of apple fruit during the post-harvest period.
