Synergistic Antimicrobial Action of Thymol and Sodium Bisulfate against Burkholderia cepacia and Xanthomonas maltophilia Isolated from the Space Shuttle Water System

  • A combination of thymol and sodium bisulfate was found to be an effective biocidal agent against strains of Burkholderia cepacia and of Xanthomonas maltophilia that were found in the space shuttle water system. Potassium iodide (KI), the biocidal agent used in the past, had a minimum inhibitory concentration (MIC) of 50,000 ppm against the two B. cepacia (541 STS-81 and 1119 STS-91) strains, whereas that of thymol and sodium bisulfate was 2,400 and 950 ppm, which was 21 and 53 times lower than that of KI for B. cepacia, respectively. The MIC value for the combination of thymol and sodium bisulfate was 4 times lower than that for thymol or sodium bisulfate alone against B. cepacia (541 STS-81, 1119 STS-91) or Pseudomonas cepacia (ATCC 31941). The fractional inhibitory concentration (FIC) of the combination of thymol and sodium bisulfate for all organisms tested was less than 0.5, indicating a strong synergistic effect.

Inhibitory Effects of Grapefuit Seed Extract on Growth and Aflatoxin Production of Aspergillus parasiticus (Grapefruit 종자추출물을 이용한 Aspergillus parasiticus의 생육 및 Aflatoxin 생성억제 효과)

  • This study was conducted to determine the potential of grapefruit seed extract to support Aspergillus parasilicus growth and aflatoxin production. The grapefruit seed extract inhibited the growth and aflatoxin production of the fungi in the level of more than 4,000 ppm and 3,000 ppm in the medium, respectively. Grapefruit seed extract appears to block the conversion of acetate, averufin and versiconal acetate into aflatoxin in vitro experiments. The addition of grapefruit seed extract to the feeding experiment systems did not inhibit the incorporation of 14C-labeled versicolorin A, versicolorin A hemiacetal and sterigmatocystin into aflatoxin. In the electron microscopic examination the biocidal action of grapefruit seed extract was related to the disturbance of cell menbrane funtion, inhibiting cellular respiration.

Effect of Polyphosphates on the Growth of Penicillium griseofulvum and the Production of Patulin (Penicillium griseofulvum 성장과 Patulin 생성에 미치는 인산염의 효과)

  • To extend the shelf lives of rice and corn products, the effects of the polyphosphates[$Na(PO_3)n$, n=11] on the growth of Penicillium griseofulvum and patulin production were investigated. The growth was completely inhibited in the potatoes dextrose agar medium treated with 2% polyphosphate. Moisture content had a considerable influence on the production of patulin. At 30% moisture content, the amounts of patulin produced in rice and corn were $61.40 \mu g/ml$ and $40.74 \mu g/ml$, respectively, but the level of the toxin was significantly decreased to 93~95% by addition of 1% polyphosphates. No patulin was detected in both rice and corn medium added 2% polyphosphate when the incubation time prolonged. The result of scanning electron microscopy was supposed that the biocidal action of polyphosphate on fungi was related to the collapse of cell wall structure.

  • To prevent food-poisoning outbreaks by Vibrio vulnificus the antimicrobial efficacy of grapefruit seed extract (GFSE) was examined. Minimal inhibitory concentration (in vitro) for the microorganism was found to be 50∼100 ppm. Transmission electron micrographs of Vibrio vulnificus showed the biocidal action of this natural antimicrobial agent would be related to specific respiratory effect coupled with the destruction of permeable function of microbial cell membrane. After Anguilla japonica GFSE-injected to the body was incubated in the seawater contaminated by Vibrio vulnifiucs the fish meats were taken up, mixed with control diet and used as a diet in the feeding experiment. In this experiment the effect of GFSE treated with fish muscle on body weight, protein efficiency ratio, serum enzymes and serum blood components of broiler chicks was investigated. It is proved from this study that there is neither Vibriosis nor toxicity associated with GFSE itself an fish meats treated with it when it is injected to the fish body at a level of 250 ppm or less.

Anthocyanins from Clitoria ternatea Attenuate Food-Borne Penicillium expansum and its Potential Application as Food Biopreservative

  • Clitoria ternatea or Commonly known blue pea, is a perennial climber crop native to Asian countries. The current study was aimed to evaluate the antimicrobial activity C. ternatea extract on food borne microorganisms and its antifungal effect on Penicillium expansum. The extract showed significant antimicrobial activity against 3 Gram positive bacteria, 2 Gram negative bacteria and 1 filamentous fungus on disc diffusion assay. The extract also showed good biocidal effect on all Gram positive bacteria tested and P. expansum. However, the kill curve analysis revealed that the fungicidal activity of the extract against P. expansum conidia was depend on the concentration of the extract and the time of exposure of the conidia to the extract. The scanning electron micrograph of the extract treated P. expansum culture showed alterations in the morphology of fungal hyphae. The germination of P. expansum conidia was completely inhibited and conidial development was totally suppressed by the extract, suggesting the possible mode of action of anthocyanin. Besides, the extract also exhibited 5.0-log suppression of microbial growth relative to control in the rice model. The results indicate the potential use of the C. ternatea anthocyanin as food biopreservative.
