The Association of Blood Pressures and Blood Biochemical Properties with BMI in Health Checkup Examinees (종합건강검진 수검자들의 비만수준과 혈압, 혈액학적 및 혈액생화학적 지표와의 관련성)

  • The purpose of this study was to investigate the association of blood pressures, hematological and blood biochemical properties according to the degree of obesity based on differential combinations of BMI in health checkup examinees. Study subjects were 3,731 adults of 20 years and over (2,312 males, 1,419 females), not recognized as taking medicines for or having cardiovascular diseases, who underwent health package check-up at the Korea Association of Health Promotion, Daejeon Branch from Jan. 2007 to Dec. 2009. As a results, the mean values of SBP, DBP, TG, TC, LDL-C, ALT, AST, GGT, ALP were significantly increased with higher level of BMI, but the HDL-C was significantly decreased with higher level of BMI. On correlation of BMI with various factors, the BMI was positively correlated with SBP, DBP, TG, TC, LDL-C and GGT, while the HDL-C was negatively correlated. Based on the result of ordinal logistic regression analysis, the risk ratio to the abnormal level of SBP, DBP, TG, ALT and GGT were significantly increased as BMI increased.

A Study on the Riboflavin Nutritional Status by Biochemical Tests in Healthy Female College Students in Korea (생화학적(生化學的) 측정방법(測定方法)에 의한 우리나라 여대생(女大生)들의 리보플라빈 영양상태(營養狀態)에 관한 연구(硏究))

  • A dietary survey by 3-day food record, clinical examination, urinary riboflavin excretion, and erythrocyte glutathione reductase activity coefficient ( EGRAC ) were measured on forty -eight female college students residing in Seoul. Thirty -one students were on normal diet and seventeen were lacto - ovo vegetarians. The results are as following : (1) Students had lower intakes of energy, similar intakes of protein and higher intake of riboflavin compared to Korean RDA for their age group. Vegetarians were lower in energy and protein intakes but were similar in RF compared to omnivores. (2) Mean EGRAC values were $1.24{\pm}0.03$ for all subjects, $1.19{\pm}0.04$ for omnivores, and $1.32{\pm}0.06$ for vegetarians. Percentages of subjects with EGRAC values above 1.2 were 65% in all subjects, 55% in omnivores, and 82% in vegetarians. Therefore, it was concluded that vegetarians were more severe in biochemical lesions than omnivores despite the fact that their RF intakes were similar to omnivores. (3) Eighteen students who had EGRAC values above 1.2 were given daily supplementation of 5mg RF for one week and EGRAC determination was repeated. They all returned to normal range. (4) Correlation analyses showed that there was no significant correlation between the EGRAC values and dietary intakes of nutrients. Possible explanations for this were given. From these results, it was concluded that even though dietary RF intakes of the subjects were higher than Korean RDA levels, biochemical deficiency among the subjects was remarkably high if the criteria of EGRAC > 1.2 were used. More investigations are necessary to establish a criteria for biochemical deficiency and RDA levels of riboflavin for Koreans.

Experimenting biochemical oxygen demand decay rates of Malaysian river water in a laboratory flume

  • Lack of information on the Biochemical Oxygen Demand (BOD) decay rates of river water under the tropical environment has triggered this study with an aim to fill the gap. Raw sewage, treated sewage, river water and tap water were mixed in different proportions to represent river water receiving varying amounts and types of wastewater and fed in a laboratory flume in batch mode. Water samples were recirculated in the flume for 30 h and BOD and Carbonaceous BOD (CBOD) concentrations were measured at least six times. Decay rates were obtained by fitting the measured data in the first order kinetic equation. After conducting 12 experiments, the range of BOD and CBOD decay rates were found to be 0.191 to 0.92 per day and 0.107 to 0.875 per day, respectively. Median decay rates were 0.344 and 0.258 per day for BOD and CBOD, respectively, which are slightly higher than the reported values in literatures. A relationship between CBOD decay rate and BOD decay rate is proposed as $k_{CBOD}=0.8642_{k_{BOD}}-0.0349$ where, $k_{CBOD}$ is CBOD decay rate and $k_{BOD}$ is BOD decay rate. The equation can be useful to extrapolate either of the decay rates when any of the rates is unknown.

  • In this study, we investigated the optimal medium composition for bacterial cellulose (BC) production by Gluconacetobacter sp. RKY5. Among the various kinds of carbon sources, glycerol was the most efficient as a sole carbon source and its optimal concentration for BC production was 15 g/L. The optimal concentration of yeast extract as a nitrogen source for BC production was found to be 8 g/L. $K_{2}HPO_{4}$ and acetic acid were selected respectively as a phosphate source and a secondary substrate, and both optimal concentrations were 3 g/L. The amount of produced BC was 4.59 g/L in a static culture and 6.5 g/L in a shaking culture condition with 150 rpm. These values were 2.1 and 2.7 times higher than those in a static (2.16 g/L) and a shaking (2.41 g/L) cultures using HS medium generally used for BC production.

  • Wheat sprout (Triticum aestivum) shows excellent nutritional and health effects due to the contents in amino acids, minerals, and other nutrients rich in chlorophyll and vitamins. In this study, spent coffee grounds were used to cultivate the wheat sprout for 12 day. An amount of 0%, 20%, 40%, 60%, 80%, and 100% (w/w, based on commercial sterile soil media) spent coffee grounds were used under the same conditions. Total length and weight of wheat sprout, aboveand below-ground length and weight, and the chloropyll contents were compared. Soil media were analyzed before and after wheat cultivation, showing that 40% and 60% (w/w) coffee media promoted wheat growth in view of length and weight. Chlorophyll contents in each group showed almost constant values, while 100% (w/w) coffee media led to a slight decrease. In conclusion, spent coffee grounds stimulated wheat growth, showing nearly stable contents of chlorophyll.

  • The carcinogenic potential of steviol, a metabolite of/ stevioside (a compound that is used as a sweetener for food and drink), was examined in hamsters of both sexes. Groups of 55 male and 55 female hamsters were given diets containing steviol at 0, 100 and 500 mg/kg diet for 22 months in males and 18 months in females. After 6, 12 and 22 months in males and 18 months in females. hamsters from each group were sacrificed for hematological and biochemical tests. Growth food utilization and consumption, general appearance and mortality were similar in treated and control groups. The mean life span of hamsters given steviol was not significantly different from that of the controls. No treatment-related changes were observed in hematological, urinary and biochemical values at any stage of the study. There was no significantly altered development of neoplastic or non-neoplastic lesions attributable to steviol treatment in any organ or tissue. The highest level oj steviol in the diet which still causes no effects in hamsters was 500 mg/kg diet, under the experimental conditions used.

Relationship between Lead Levels in Blood and Urine and Laboratory Test Values among Lead Exposed Workers (저농도 연 폭로근로자들의 혈중 및 요중 연량과 생화학적 검사치들 간의 관련성)

  • The study subjects included the workers who were estimated to have been exposed in a low density of lead-laden glaze from pottery manufacturers. They were evaluated in terms of PbB and PbU as indices of lead concentration in the body, and Hb, Hct, ALAD, and FEP as indices of biochemical lead poisoning. Consequently, the study results revealed the relationship between each of indices and its effectiveness to indicate the degree of occupational lead exposure. 1. The mean of PbB was $22.85{\;}{\mu}g/d{\ell}$(range; $4.2~44.7{\;}{\mu}g/d{\ell}$) and PbU $45.69{\mu}g/d{\ell}$(range; $6.7~87.5{\mu}g/d{\ell}$), to indicate the level of lead exposure in the subjects. 2. While there was a high, linear correlation between PbB, PbU and FEP (r=0.835, r=0.855, respectively), a low correlation existed between PbB and ALAD(r=-0.240). 3. While there was a high, linear correlation between PbU and FEP(r=0.825), a low correlation existed between PbU and ALAD(r=-0.352). The above results suggest that FEP can be used as a highly useful biochemical index of lead poisoning, based on its high correlation with PbB and PbU indicating lead concentration in the body.

  • To evaluate iron nutritional status and to compare biochemical indices used for evaluation of iron nutriture, fasting blood sample was taken from 96 Korean female college student and hemoglobin(Hb) concentration, hematocrit(Hct), serum iron, total iron binding capacity(TIBC) and serum ferritin concentration were measured and transferrin(TF) saturation was calculated. Prevalence of iron deficiency varied from 4.2% when judged from Hb concentration and Hct to 38.5% and 40.6% when TIBC and serum ferritin concentration were used. TIBC was negatively correlated with serum ferritin(r=0.4561, p<0.001), while other indices showed positive but less significant correlations(p<0.05). Regarding iron status judged by serum ferritin was regarded as true iron status, the present cutoff points of Hb, Hct, serum iron, TF saturation were very insensitive in identifying the subjects with iron deficiency. Only TIBC was suitable for detecting iron deficiency in the study subjects. It is concluded from the study results that iron deficiency in young Korean adult women would be significantly underestimated with presently used biochemical measurements except TIBC and new cutoff points of these indices were proposed based on sensitivity, specificity measured prevalence, and positive and negative predictive values.

Docking and QSAR studies of PARP-1 Inhibitors (PARP-1 억제제의 Docking 및 QSAR 연구)

  • Poly(ADP-ribose)polymerase-1 (PARP-1) is a nuclear enzyme involved in various physical functions related to genomic repair, and PARP inhibitors have therapeutic application in a variety of neurological diseases. Docking and the QSAR (quantitative structure-activity relationships) studies for 52 PARP-1 inhibitors were conducted using FlexX algorithm, comparative molecular field analysis (CoMFA), and hologram quantitative structure-activity relationship analysis (HQSAR). The resultant FlexX model showed a reasonable correlation (r$^{2}$ = 0.701) between predicted activity and observed activity. Partial least squares analysis produced statistically significant models with q$^{2}$ values of 0.795 (SDEP=0.690, r$^{2}$=0.940, s=0.367) and 0.796 (SDEP=0.678, r$^{2}$ = 0.919, s=0.427) for CoMFA and HQSAR, respectively. The models for the entire inhibitor set were validated by prediction test and scrambling in both QSAR methods. In this work, combination of docking, CoMFA with 3D descriptors and HQSAR based on molecular fragments provided an improved understanding in the interaction between the inhibitors and the PARP. This can be utilized for virtual screening to design novel PARP-1 inhibitors.

Effects of the Velocity Waveform of the Physiological Flow on the Hemodynamics in the Bifurcated Tube

  • The periodicity of the physiological flow has been the major interest of analytic research in this field up to now Among the mechanical forces stimulating the biochemical reaction of endothelial cells on the wall, the wall shear stresses show the strongest effect to the biochemical product. The objective of present study is to find the effects of velocity waveform on the wall shear stresses and pressure distribution along the artery and to present some correlation of the velocity waveform with the clinical observations. In order to investigate the complex flow phenomena in the bifurcated tube, constitutive equations, which are suitable to describe the rheological properties of the non-Newtonian fluids, are determined, and pulsatile momemtum equations are solved by the finite volume prediction. The results show that pressure and wall shear stresses are related to the velocity waveform of the physiological flow and the blood viscosity. And the variational tendency of the wall shear stresses along the flow direction is very similar to the applied sinusoidal and physiological velocity waveforms, but the stress values are quite different depending on the local region. Under the sinusoidal velocity waveform, a Newtonian fluid and blood show big differences in velocity. pressure, and wall shear stress as a function of time, but the differences under the physiological velocity waveform are negligibly small.
