Comparative analysis of AGPase proteins and conserved domains in sweetpotato (Ipomoea batatas (L.) Lam.) and its two wild relatives

  • Conserved domains are defined as recurring units in molecular evolution and are commonly used to interpret the molecular function and biochemical structure of proteins. Herein, the ADP-glucose pyrophosphorylase (AGPase) amino acid sequences of three species of the Ipomoea genus [Ipomoea trifida, I. triloba, and I. batatas (L.) Lam. (sweetpotato)] were identified to investigate their physicochemical and biochemical characteristics. The molecular weight, isoelectric point, instability index, and grand average of hyropathy markedly differed among the three species. The aliphatic index values of sweetpotato AGPase proteins were higher in the small subunit than in the large subunit. The AGPase proteins from sweetpotato were found to contain an LbH_G1P_AT_C domain in the C-terminal region and various domains (NTP_transferase, ADP_Glucose_PP, or Glyco_tranf_GTA) in the N-terminal region. Conversely, most of its two relatives (I. trifida and I. triloba) were found to only contain the NTP_transferase domain in the N-terminal region. These findings suggested that these conserved domains were species-specific and related to the subunit types of AGPase proteins. The study may enable research on the AGPase-related specific characteristics of sweetpotatoes that do not exist in the other two species, such as starch metabolism and tuberization mechanism.

Terahertz Spectral Characteristics of Electrolyte Solutions under Different Magnetic Fields

  • Microfluidic chips are new devices that can manipulate liquids at the micrometer level, and terahertz (THz) time-domain spectroscopy has good applicability in biochemical detection. The combination of these two technologies can shorten the distance between sample and THz wave, reduce THz wave absorption by water, and more effectively analyze the kinetics of biochemical reactions in aqueous solutions. This study investigates the effects of different external magnetic field intensities on the THz transmission characteristics of deionized water, CuSO4, CuCl2, (CH3COO)2Cu, Na2SO4, NaCl, and CH3COONa; the THz spectral intensity of the sample solutions decrease with increasing intensity of the applied magnetic field. Analysis shows that the magnetic field leads to a change in the dipole moment of water molecules in water and electrolyte solutions, which enhances not only the hydrogen-bond networking ability of water but also the hydration around ions in electrolyte solutions, increasing the number of hydrogen bonds. Increasing the intensity of this magnetic field further promotes the hydrogen-bond association between water molecules, weakening the THz transmission intensity of the solution.

Monitoring of Water Quality Parameters using Spectroscopic Characteristics of River Water - Ulsan Area (하천 분광특성을 이용한 수질항목 모니터링 연구 - 울산 지역)

  • Spectroscopic characteristics of river water from four major watersheds in the Ulsan area were measured to examine their potential for estimating water quality parameters. The total 176 river samples were collected from 44 sites of small streams within the watersheds during the year 2006. Spectroscopic characteristics investigated included protein-like fluorescence (FLF) intensity, fulvic-like fluorescence (FLF) intensity, terrestrial humic-like fluorescence (TLF) intensity, UV absorbance at 254 nm, and UV absorbance difference at 220 nm and 254 nm. Protein-like fluorescence intensity showed linear relationships with biochemical oxygen demand (BOD), chemical oxygen demand (COD), total phosphorous (TP) concentrations of the samples with the correlation of 0.784, 0.779, and 0.733, respectively. Due to the UV absorption characteristics of nitrate at 220 nm, UV absorbance difference at 220 nm and 254 nm was selected to represent total nitrogen (TN) concentration. Exclusion of some samples with PLF intensity higher than 5.0 improved the correlation between the UV absorbance difference and TN as demonstrated by the increase of the correlation coefficient from 0.392 to 0.784. Instead, for the samples with PLF intensity lower than 5.0, the highest correlation of TN was achieved with UV absorbance at 254 nm. The results suggest that PLF intensity could be used as the estimation index for BOD, COD, and TP concentration of river water, and as the primary screening index for the prediction of TN using UV absorbance difference. Some BOD-based water quality levels among the river water were statistically discriminated by the PLF intensity. Low p-values were obtained from the t-tests on the samples with the first level and the second level (p=0.0003) and the samples with the second and the third levels (p=0.0413). Our combined results demonstrated that the selected spectroscopic characteristics of river water could be utilized as a tool for on-site real-time monitoring and/or the primary estimation of water quality.

The Trophic State Assessment using Biochemical Composition in the Surface Sediments, the Southern Coast of Korea (표층 퇴적물의 생화학적 조성을 이용한 남해연안 영양상태 평가)

  • In order to classify the trophic state and environmental quality of marine coastal system, an approach using the characteristics and biochemical composition in the sediments can be available. This research, including 25 coastal bay, belong to 131 stations, was carried out along the south coasts of Korea in February 2007. Type of sediment, total ogranic carbon, total nitrogen, phytopigments and biochemical composition(proteins, lipids, carbohydrates) were analyzed. Result from Multi-dimensional Scaling(MDS) ordination indicates that four group can be identified. The result of ANOVA with tukey test shows that the concentrations of proteins, carbohydrates and biopolymeric carbon were significantly different to four groups. We propose the trophic state classification for these groups using the biochemical composition of sediment organic matter. I group(Masan, Jinhae, Haengam) has been defined as hypertrophic state, II group(Tongyeong, Goseong;Jaran, Geoje et al.), as eutrophic; III group(Gamak, Deungnyang, Yeoja et al.), as mesotrophic and IV group(Sinan, Jindo, Muan), as oligotrophic. On the basis of results reported in this study, the biochemical composition of sediment organic matter could be considered an useful and sensitive tool for the classification of the trophic state of marine coastal systems.

Purification and Biochemical Properties of Storage Protein SP-2 in Tobacco Budworm (Helicoverpa assulta Guenee) (담배나방 저장단백질 SP-2의 정제 및 생화학적 특성)

  • A storage protein(SP-2) was confirmed in haemolymph during larval-pupal-adult development of tobacco burdworm(Helicoverpa assulta Guenee), and its biochemical characteristics were investigated. The titer of SP-2 showed a peak at mature larva, decreased gradually through the late pupal stage, and became undetectable at adult period. As the results from electrophoretic mnysis, SP-2 was confirmed to be glycolipoprotein(M.W. 332kDa) relatively stable to heat( $\leq$ 68$^{\circ}C$ ). This storage protein was determined to be a tetramer composed of a single subunit with MW of 83kDa, and the isoelectric point was 5.7. The amino acid composition of the SP-2 was characterized. It has relatively high content of methionine and histidine, whereas the contents of tyrosine and phenylalanine were relatively low.

  • Bile salt deconjugation is the most biologically significant reaction among the bacterial alterations of bile acids in the gastrointestinal tract of human and animal. The responsible enzyme, bile salt hydrolase (BSH), catalyzes the hydrolysis of glycineand/or taurine-conjugated bile salts into amino acid residues and deconjugated bile acids. Herein we review current knowledge on the distribution of BSH activity among various microorganisms with respect to their biochemical and molecular characteristics. The proposed physiological impact of BSH activity on the host animal as well as on the BSH-producing bacterial cells is discussed. BSH activity of the probiotic strains is examined on the basis of BSH hypothesis, which was proposed to explain cholesterol-lowering effects of probiotics. Finally, the potential applications of BSH research are briefly discussed.

  • It was confirmed that extracts of some plants were used in folklore medicine as hypoglycemic agents. Of these plants is Azadirachta indica ("neem") which grows in tropical regions. The present study deals with biochemical effects of the "neem" leaf water extract given orally to experimental aminals, especially the hypoglycemic characteristics. Normals as well as alloxan diabetic rats have been used in this work. The results showed that the "neem" leaf extract produced some hypoglycemia in normal rats when given in two doses, while in diabetic rats there was a decrease in blood sugar, but it could not alleviate the diabetic in body weight loss and high percentage mortality, especially with a high dose. It was observed increased with concomitant decrease in the liver fat as compared to normal levels. There was also a drop in liver proteins which was dose-related. The results were compared wiht those obtained with an oral hypoglycemic drug (Glibenclamide).glycemic drug (Glibenclamide).

  • The present study was carried out to investigate the biochemical characteristics, antibiotic susceptibility and toxin(ST, LT, VT1.2 type) production test of 60 Escherichia coli isolated from diseased domestic animals in southern area of Kyungbuk province from April to December 1997. 1. The biochemical and cultural reaction were consistent with the classification criteria of Edwards and Ewing. 2. In antibiotic susceptibility test, 60 E coli showed highly susceptible to CL(96.7%), XNL(86.7%), AN(81.7%), SXT(61.7%), Lin(55%), GM(53.3%), KM(41.7%), N(41.7%), ENR(40%), AM(40%), CF(30%), 5(13.3%) and Te(11.7%), in order. 3. Sixty E coli isolates were multiful resistant to seven or more antibiotics incombination. 4. Three strains for 60 E coli were detected heat-labile enterotoxin(LT) and that's titers were 2, 8 and 16, respectively.

Antibiotic Resistance of Pathogenic Escherichia coli Isolated from Piglets with Diarrhea (설사자돈 유래 대장균의 항생물질 내성에 관하여)

  • The present study was conducted to investigate the biochemical characteristics and anti-biotic resistance of Escherichia coli(E. coli) isolated from piglets with diarrhea in Kyongbuk province during the Period from February to November 1991. 368 E. coli strains were isolated from 382 piglets with diarrhea and the biochemical and cultural reaction were compared with the classification criteria of Edwards and Ewing. Tetracycline and sulfadimethoxine were found to be highly ineffective at in vitro inhibition of the E. coli of piglets origin. The majority of E. coli were susceptible to amikacin, chloramphenicol and gentamicine. 89 (89.0%) of 100 strains of E. coil were resistant to one or more drugs. The organisms resistant to 20 or 3 drugs were 54(60.6%) of 89 strains, whereas 16(17.9%) strains were found to be resistant to one drug. 55(61.8%) out of 89 drug resistance strains carried R factors($R^+$) which were transfer-able to the recipients by conjugation.

  • The microcapsules containing fragrant oil as functional material were prepared by in-situ polymerization with prepolymer that was made from melamine-formaldehyde (MF) as wall material of microcapsules. The effects of polymerization variables, such as the nature and concentration of surfactants, stirring rate, and stirring time, on the size and distribution of the particles were investigated. Fourier transform-infrared spectroscopy (FT-IR), thermal analysis, particle size analysis, scanning electron microscopy (SEM) observation were used to investigate the characteristics of microcapsules. Through the FT-IR and SEM analysis, we found that the prepared microcapsules were containing fragrant oil and the shape of particle was spherical. The nature and concentration of surfactants, stirring rate, and stirring time had profound effects on the particle size and particle size distribution.
