오미자 박 추출물 및 schizandrin에 의한 암세포 항성장 및 세포사멸 활성 (Anti-proliferative and Pro-apoptotic Activities by Pomace of Schisandra chinensis (Turcz.) Baill. and Schizandrin)

  • 오미자는 다양한 인간 질환을 치료하기 위한 한약재로 사용되어 왔으며, schizandrin과 gomisin A와 같은 다양한 생리활성물질을 함유하고 있는 것으로 알려져 있다. 본 연구에서는 오미자 박으로부터 에탄올 추출물(PSC)을 제조하고, 이들이 대장암 세포인 HCT116의 세포 생존율에 미치는 영향과 ATF3, NAG-1, p21와 같은 pro-apoptotic 유전자의 발현 변화에 미치는 영향을 연구하였다. 오미자 박 에탄올 추출물의 처리는 농도 의존적으로 암세포생존율을 감소시켰으며, 세 가지 pro-apoptotic 유전자의 발현을 모두 증가시켰다. 또한, 오미자 유래의 순수물질인 schizandrin도 세포 생존율을 농도의존적으로 감소시켰으며, ATF3, NAG-1, p21 유전자의 발현을 증가시켰다. 게다가, schizandrin을 처리한 세포에서 PARP cleavage를 확인함으로써 apoptosis가 일어남을 확인하였다. 이러한 PARP cleavage는 NAG-1 siRNA의 transfection에 의해서 회복됨을 확인하였다. 이러한 결과는 schizandrin에 의해 유도되는 apoptosis와 NAG-1의 발현증가가 직접적인 관련이 있음을 나타낸다. 종합적으로, 본 연구결과는 오미자 박 추출물과 schizandrin에 의해 매개되는 항암 활성과 암세포 사멸현상을 이해하는데 도움을 줄 것으로 사료된다.

Evaluation of MiR-34 Family and DNA Methyltransferases 1, 3A, 3B Gene Expression Levels in Hepatocellular Carcinoma Following Treatment with Dendrosomal Nanocurcumin

  • Hepatocellular carcinoma (HCC) is the most common primary malignancy of the liver making up more than 80 percent of cases. It is known to be the sixth most prevalent cancer and the third most frequent cause of cancer related death worldwide. Epigenetic regulation constitutes an important mechanism by which dietary components can selectively activate or inactivate target gene expression. The miR-34 family members including mir-34a, mir-34b and mir-34c are tumor suppressor micro RNAs, which are expressed in the majority of normal tissues. Several studies have indicated silencing of miR-34 expression via DNA methylation in multiple types of cancers. Bioactive nutrients like curcumin (Cur) have excellent anticarcinogenic activity and minimal toxic manifestations in biological systems. This compound has recently been determined to induce epigenetic changes. However, Cur is lipophilic and has a poor systemic bioavailability and poor absorption. Its bioavailability is increased through employing dendrosome nanoparticles. The aim of the current study was to investigate the effect of dendrosomal nanocurcumin (DNC) on expression of mir-34 family members in two HCC cell lines, HepG2 and Huh7. We performed the MTT assay to evaluate DNC and dendrosome effects on cell viability. The ability of DNC to alter expression of the mir-34 family and DNA methyltransferases (DNMT1, DNMT3A and 3B) was evaluated using semi-quantitative and quantitative PCR. We observed the entrance of DNC into HepG2 and Huh7 cells. Gene expression assays indicated that DNC treatment upregulated mir34a, mir34b and mir34c expression (P<0.05) as well as downregulated DNMT1, DNMT3A and DNMT3B expression (P<0.05) in both HepG2 and Huh7 cell lines. DNC also reduced viability of Huh7 and HepG2 cells through restoration of miR-34s expression. We showed that DNC could awaken the epigenetically silenced miR-34 family by downregulation of DNMTs. Our findings suggest that DNC has potential in epigenetic therapy of HCC.

Effects of supplemented culture media from solid-state fermented Isaria cicadae on performance, serum biochemical parameters, serum immune indexes, antioxidant capacity and meat quality of broiler chickens

  • Objective: The objective of this study was to investigate effects of supplementation of culture media from solid-state fermented Isaria cicadae (I. cicadae) on performance, serum biochemical parameters, serum immune indexes, antioxidant capacity and meat quality of broiler chickens. Methods: A total of 648 Arbor Acres male broiler chickens(1 d; average body weight, 42.93±0.47 g) were randomly assigned to 6 treatments, each with six replicates and 18 broiler chickens per replicate. Broiler chickens were fed phase I (d 1 to 21) and phase II (d 22 to 42) diets. The phase I diets were corn and soybean-meal based diets supplemented with 0%, 2%, 4%, 6%, 8%, or 10% culture media from solid-state fermented I. cicadae respectively. The phase II diets were corn and soybean-meal based diets supplemented with 0%, 1.33%, 2.67%, 4.00%, 5.32%, or 6.67% culture media from solid-state fermented I. cicadae respectively. Results: In phase I, the broiler chickens with the supplementation of culture media had increased body weight gain and feed intake (linear and quadratic, p<0.05) with increasing inclusion of culture media. The levels of serum total antioxidant capacity (T-AOC), glutathione (GSH) and superoxide dismutase (SOD) increased linearly (p<0.05). In phase II, levels of serum T-AOC and interleukin-1β increased linearly (p<0.05), and GSH increased (p<0.05). In the kidney, GSH and glutathione peroxidase (GSH-Px) concentrations increased (linear and quadratic, p<0.05) and SOD concentration increased linearly (p<0.05). Compared to the control, shear force and drip loss of breast muscle decreased (linear and quadratic, p<0.05). Drip loss of leg muscle decreased linearly and quadratically (p<0.05). Conclusion: Dietary supplementation of culture media from solid-state fermented I.cicadae which was enriched in both wheat and residual bioactive components of I. cicadae enhanced the growth performance of broiler chickens. It also improved body anti-oxidative status and contributed to improve broiler meat quality.

야생산사(Crataegus pinnatifida BUNGE)씨 생리활성물질의 산화억제 효과 (Effects of Feral Haw (Crataegus pinnatifida BUNGE) Seed Extracts on the Antioxidant Activities)

  • 본 연구에서는 야생산사씨의 항산화 활성을 알아보고자 70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v / v), n-butanol, ethyl acetate (EA) 용매로 추출하여, 시료 농도 0.2 mg/ml, 0.5 mg/ml, 0.8 mg/ml에서 측정 비교 하였다. DPPH, reducing power, FRAP 및 ABTS radical 소거활성은 70% methanol, 70% ethanol, EA, CM, n-butanol 추출물 순으로 높게 측정되어 모두 같은 경향을 나타내었다. 특히, DPPH에서 70% methanol 추출물은 18.22%~65.42%, 70% ethanol 추출물은 15.97%~60.25%의 우수한 radical 소거 활성을 보였다. NO radical 소거 활성은 n-butanol, EA, CM, 70% methanol, 70% ethanol 추출물 순으로 나타났으며, $OH^-$ radical 소거 활성은 n-butanol, 70% methanol, 70% ethanol, CM, EA 추출물 순으로 측정되었다. 모든 용매 추출물은 대조군으로 사용한 표준물질에 비해서는 유의적으로 항산화 활성이 낮은 것으로 나타났다. 산사씨 용매 추출물 중 70% methanol 및 70% ethanol 추출물의 항산화 활성이 대부분 높은 경향이었다. 따라서 산사씨는 우수한 천연 항산화제로 활용이 가능할 것으로 사료된다.

Quali-Quantitative Analysis of Flavonoids for Mulberry Leaf and Fruit of 'Suhyang'

  • BACKGROUND: Globally, mulberry (Morus sp.) is exploited for feeding leaf to silkworms in order to obtain silk fiber or for animal feedstock production. Also, mulberry fruit is known to a by-product that was produced from mulberry tree after harvesting leaves for silkworm rearing, as a yield and consumption of mulberry fruit was increased, it has been fixing to a newincome crop. Mulberry leaves and fruits are used for the health benefits of human beings. Mulberry contains various bioactive components, such as alkaloids and flavonoids. Mulberry flavonoids are an important part of the diet because of their effects on human nutrition. The flavonoids in mulberry leaf and fruit of 'Suhyang'(Morus alba L.) were determined. METHODS AND RESULTS: Flavonoids for mulberry leaf and fruit of 'Suhyang' were analysed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS)technique. An UPLC-DAD-QTOF/MS system was used, and identification of mulberry leaves constituents was carried out on the basis of the complementary information obtained from LC spectra, MS ions, and MS/MS fragments. The mulberry leaf (16 flavonoids) and fruit (9 flavonoids) were isolated and analyzed from Suhyang using UPLC-DAD-QTOF/MS chromatogram. To the best of our knowledge, Quercetin 3-O-(6"-O-malonyl) glucoside and quercetin 3-O-rutinoside (rutin) was detected on the highest content in leaf and fruit, respectively and further research will be devoted to evaluate their biological activity. CONCLUSION: Obtaining information about the concentration of functional materials in mulberry leaves could contribute to the development and promotion of processed, functional products and offer possible industrial use of 'Suhyang', holding promises to enhance the overall profitability of sericulture.

Bacillus subtilis를 이용한 국내산 신이대 잎 발효에 따른 영양성분, 폴리페놀, 항산화능 변화 (Changes of Nutritional Components, Polyphenols, and Antioxidant Activities of Domestic Bamboo Tree (Sasa coreana Nakai) Leaves Fermented with Bacillus subtilis)

  • Bamboo leaf has many nutritional and bioactive compounds such as polyphenols, and it can be used for various foods application. Fermentation is one of appropriate processes that can affect the nutrition, taste and flavor, and antioxidant activities of foods. In this study, a representative domestic bamboo tree (Sasa coreana Nakai) leaves were fermented by Bacillus subtilis KCCM 11315 and the changes of carbohydrates, minerals, amino acids, and phenolic compounds and antioxidant activities were investigated before and after fermentation. During the fermentation period, firstly, the leaf was softened and turned to brown. The constituent carbohydrates were slightly increased from $432.09{\pm}5.38mg/g$ to $458.42{\pm}7.39mg/g$, and free sugars decreased by 95% from $28.12{\pm}2.03mg/g$ to $1.4{\pm}0.14mg/g$. Mineral was $20987.5{\pm}345.1{\mu}g/mL$, which was slightly increased after the fermentation compared to $20804.1{\pm}364.6{\mu}g/mL$ before that. The total amino acids were increased to $73881.94{\pm}137.59mg/100g$ compared to $58464.51{\pm}109.12mg/100g$ before fermentation, and free amino acids decreased by more than 85% from $32782.67{\pm}92.49mg/100g$ to $4827.83{\pm}19.76mg/100g$. Total polyphenols content (TPC) increased from $25.51{\pm}1.04GAE^*mg/g$ to $35.34{\pm}0.91GAE^*mg/g$, and the total flavonoid content (TFC) increased to $80.57{\pm}0.22QE^*mg/g$ compared to $69.64{\pm}0.26QE^*mg/g$. The total catechin content (TCC) of TFC was decreased from $69.64{\pm}0.94mg/L$ to $58.23{\pm}0.76mg/L$. The DPPH radical $IC_{50}$ of bamboo leaves decreased to $2624.85{\pm}37.03{\mu}g/mL$ and the ABTS radical $IC_{50}$ of $187.26{\pm}4.78{\mu}g/mL$ was increased after the fermentation. These results could be used as essential nutritional data before developing processed food products using the bamboo leaf.

재배지에 따른 노니열매 착즙액의 이화학적 특성, 생리활성 성분 및 항산화 활성 (Physicochemical properties, bioactive composition and antioxidant activities of noni fruit juices from different regions of cultivation)

  • 노니의 주요 산지인 사모아, 인도네시아, 중국(하이난), 하와이, 태국, 타히티에서 생산된 노니 열매를 착즙하여 산지별 노니열매 착즙액의 이화학적 특성, 기능성 성분 및 항산화 활성을 비교분석하였다. 산지별 노니열매 착즙액의 pH는 3.63-3.83 수준으로 나타났으며, 색도는 태국산이 다른 산지 노니열매 착즙액보다 L, a, b 값 모두 유의적으로 낮은 값으로 나타났다. 고형분 함량은 $5.97-6.97^{\circ}Brix$ 범위로 하와이산이 $6.97^{\circ}Brix$로 가장 높았으며, 반면 인도네시아산이 $5.97^{\circ}Brix$로 가장 낮은 값을 나타내었다. 태국산이 가장 많은 스코폴레틴과 루틴을 함유하고 있었으며, 인도네시아산이 다른 지역에서 생산된 노니열매 착즙액보다 폴리페놀 함량이 유의적으로 가장 높았다. 항산화 활성 중 DPPH 라디칼 소거능은 인도네시아 > 하와이 > 타히티 > 태국 > 사모아 > 중국 순으로 나타났으며, ABTS 라디칼 소거능은 인도네시아 > 하와이 > 타히티 > 사모아 > 중국 > 태국 순으로 나타나 인도네시아산이 가장 높은 항산화능을 보였다. 이상의 결과, 태국산이 스코폴레틴과 루틴의 함량이 가장 높았으며, 반면 인도네시아산이 가장 우수한 항산화 활성을 가지는 것을 확인하였다.

팽화처리가 홍미삼의 품질특성에 미치는 영향 (Effect of Puffing on Quality Characteristics of Red Ginseng Tail Root)

  • 팽화처리가 홍미삼의 사포닌 성분, 당류, 페놀성분, 미세구조 및 소화율 등에 미치는 영향을 조사하였다. 홍미삼의 추출된 조사포닌 함량은 팽화처리후 약 26.5% 증가하였다. 사포닌 조성에서 팽화홍미삼은 홍미삼에 비해 $Rb_1$, $Rb_2$, Rc, Re, $Rg_1$함량이 다소 감소하였으나 Rd와 $Rg_3$함량은 증가하였고, 특히 $Rg_3$함량은 0.49 mg/g에서 0.72 mg/g로 증가하였다. 가용성 총당은 팽화전후 비슷하였고, 산성다당체 함량은 각각 7.15, 6.44%로 팽화후 다소 감소하였다. 총페놀함량은 7.86%에서 9.94%로 증가하였다. 개별 페놀산의 경우 gentisic acid는 홍미삼에서만, salicylic acid는 팽화후 홍미삼에서만 검출되었고, gallic acid, p-coumaric acid, caffeic acid 및 ferulic acid 등은 팽화로 크게 감소한 반면 syringic acid는 약 6배 증가하였다. 팽화후 홍미삼의 절단면 미세구조는 팽화전보다 훨씬 균일화한 입자모양을 나타내었으며, 팽화전후 펩신소화율은 각각 22.4, 46.2%로 통계적으로 유의한 차이가 있었다 (p<0.05). 본 결과는 팽화처리에 의해 홍미삼의 생리활성성분의 증가와 기호성 및 소화율을 높일 수 있음을 시사하는 것으로 판단되었다.

곤충 유래 항균 펩타이드의 작용 기작 (Mode of Action of Antimicrobial Peptides Identified from Insects)

  • 지구상에 존재하는 전체 동물 중 가장 큰 부분을 차지 하고 있는 곤충은 예로부터 인간의 식품, 농업, 산업, 의약 등의 일상 생활에 이용되어 왔다. 많은 수와 높은 영양학적 가치로 곤충의 생리활성물질이 미개발 생물자원으로 재평가 되고 있다. 곤충은 면역세포, 곤충 혈구세포, 효소들의 연쇄반응 혹은 항균 단백질/펩타이드 같은 방법으로 외부의 감염에 저항성을 가지게 된다. 항균 펩타이드는 곤충의 혈림프의 선천성 면역 시스템 중 주요한 성분중의 하나로 항생제 내성 균주의 출몰이 빈번하게 일어나 해결책으로 새로운 항생제 개발이 시급한 시점에서 유력한 후보물질로 주목 받고 있다. 곤충 유래 항균 펩타이드는 150개가 넘게 분리되었으며 크게 세크로핀, 디펜신, 글라이신/프롤린이 풍부한 펩타이드로 이루어진다. 이 논문에서, 향균 펩타이드를 생산하는 여러 곤충 중에서 벌, 소똥구리, 울도하늘소, 나비 그리고 울도하늘소에서 얻을 수 있는 펩타이드의 종류 그리고 작용 기작에 대해 알아보았다. 이 펩타이드들은 항균효과를 가지고 있으며 멜리틴을 제외하고 적혈구의 용혈 현상이 나타나지 않고 주로 세포막을 붕괴시키거나 세포자살기작을 유도하여 병원성 미생물의 성장을 억제한다. 곤충 유래 펩타이드와 같은 생리활성물질이 그 활용 가능성의 면에서 엄청난 가능성을 가지고 있어 이에 대한 연구는 앞으로 더 주목을 받을 것이다.

Development of New Materials of Ginseng by Nanoparticles

  • For centuries, Panax ginseng Meyer (Korean ginseng) has been widely used as a medicinal herb in Korea, China, and Japan. Ginsenosides are a class of triterpene saponins and recognized as the bioactive components in Korean ginseng. Ginsenosides, which can be classified broadly as protopanaxadiols (PPD), protopanaxatriols (PPT), and oleanolic acids, have been shown to flaunt a vast array of pharmacological activities such as immune-modulatory, anti-inflammatory, anti-tumor, anti-diabetic, and antioxidant effects. In recent years, a number of ginseng and ginsenoside researches have increasingly gained wide attention owing to its unique pharmacological properties. Although good efficacies of ginsenosides have been reported, lack of target specific delivery into tumor sites, low solubility, and low bioavailability due to modifications in gastro-intestinal environments limit their biomedical application in clinical trials. As a result to this major challenge, nanotechnology and drug delivery techniques play a significant role to solve this problematic issue. Thus, we reported the preparation of poly-ethylene glycol (PEG) and glycol chitosan (GC) functionalized to ginsenoside (Compound K and PPD) conjugates via hydrolysable ester bonds with improved aqueous solubility and pH-dependent drug release. In vitro cytotoxicity assays revealed that PEG-CK, and PPD-CK conjugates exhibited lower cytotoxicity compared to bare CK and PPD in HT29 cells. However, GC-CK conjugates exhibited higher and similar cytotoxicity in HT29 and HepG2 cells. Furthermore, GC-CK-treated RAW264.7 cells did not exhibit significant cell death at higher concentration of treatment which supports the biocompatibility of the polymer conjugates. They also inhibited nitric oxide production in lipopolysaccharide (LPS)-induced RAW64.7 cells. In addition to polymer-ginsenoside conjugates, silver (AgNps) and gold nanoparticles (AuNps) have been successfully synthesized by green chemistry using different m. The biosynthesized nanoparticles demonstrated antimicrobial efficacy, anticancer, anti-inflammatory, antioxidant activity, biofilm inhibition, and anticoagulant effect. Special interest on the effective delivery methods of ginsenoside to treatment sites is the focus of metal nanoparticle research.In short, nano-sizing of ginsenoside results in an increased water solubility and bioavailability. The use of nano-sized ginsenoside and P. ginseng mediated metallic nanoparticles is expected to be effective on medical platform against various diseases in the future.
