  • This study focuses on a determination of amount of Pt in the Pt/C for catalysts of polymer electrolyte membrane fuel cells (PEMFC). PEMFC offer low weight and high power density and being considered fur automotive and stationary power applications. The PEMFC behavior is quite complex is influenced by several factors, including catalysts and structure of electrode and membrane type. Catalyst of electrode is important factor for PEMFC. One of the obstacles preventing polymer electrolyte membrane fuel cells from commercialization is the high cost of noble metals to be used as catalyst, such as platinum. To effectively use these metals, they have to be will dispersed to small particles on conductive carbon supports. The optimal amount of Pt in Pt/C was investigated by using polarization curves in single cell with $H_2/O_2$ operation.

돈분을 이용한 열분해공정 바이오오일의 특성 (Characteristics of Bio-oil by Pyrolysis with Pig Feces)

  • 본 연구에서는 돈분을 이용한 열분해공정(pyrolysis)에 의한 바이오오일의 특성을 분석하여 보고하였다. 기본적으로 bio-oil 생산을 위한 pilot auger형 반응기는 $400^{\circ}C{\sim}600^{\circ}C$의 고온을 유지하였다. 바이오오일의 특성은 수질분석, 열량가, 원소분석, GC/MS를 이용한 마이오일의 원소, $^1H$ NMR분광기에 의한 functional group 구명 등을 포함한다. 돈분시료를 이용한 바이오오일 생산량은 pilot auger 반응기의 온도가 $550^{\circ}C$일 때 바이오일 생산율은 질량의 21%로서 최대를 나타내었다. 이 결과는 본 연구에서 연속 auger형 반응기의 이송이 편리하고 bio-oil 생산량이 적지 않아 대체 축분처리기술의 하나로 검토할 수 있음을 보였다. 그러나 auger 반응기의 원료로의 열전도가 유동상 반응조보다 낮아서 향후 이를 개선하기 위한 연구가 성공적으로 수행되면 바이오오일 생산량을 제고시킬 수 있을 것으로 판단된다.

Effect of Lactobacillus mucosae on In vitro Rumen Fermentation Characteristics of Dried Brewers Grain, Methane Production and Bacterial Diversity

  • The effects of Lactobacillus mucosae (L. mucosae), a potential direct fed microbial previously isolated from the rumen of Korean native goat, on the rumen fermentation profile of brewers grain were evaluated. Fermentation was conducted in serum bottles each containing 1% dry matter (DM) of the test substrate and either no L. mucosae (control), 1% 24 h broth culture of L. mucosae (T1), or 1% inoculation with the cell-free culture supernatant (T2). Each serum bottle was filled anaerobically with 100 mL of buffered rumen fluid and sealed prior to incubation for 0, 6, 12, 24, and 48 h from which fermentation parameters were monitored and the microbial diversity was evaluated. The results revealed that T1 had higher total gas production (65.00 mL) than the control (61.33 mL) and T2 (62.00 mL) (p<0.05) at 48 h. Consequently, T1 had significantly lower pH values (p<0.05) than the other groups at 48 h. Ammonia nitrogen ($NH_3$-N), individual and total volatile fatty acids (VFA) concentration and acetate:propionate ratio were higher in T1 and T2 than the control, but T1 and T2 were comparable for these parameters. Total methane ($CH_4$) production and carbon dioxide ($CO_2$) were highest in T1. The percent DM and organic matter digestibilities were comparable between all groups at all times of incubation. The total bacterial population was significantly higher in T1 (p<0.05) at 24 h, but then decreased to levels comparable to the control and T2 at 48 h. The denaturing gradient gel electrophoresis profile of the total bacterial 16s rRNA showed higher similarity between T1 and T2 at 24 h and between the control and T1 at 48 h. Overall, these results suggest that addition of L. mucosae and cell-free supernatant during the in vitro fermentation of dried brewers grain increases the VFA production, but has no effect on digestibility. The addition of L. mucosae can also increase the total bacterial population, but has no significant effect on the total microbial diversity. However, inoculation of the bacterium may increase $CH_4$ and $CO_2$ in vitro.

Screening of Probiotic Activities of Lactobacilli Strains Isolated from Traditional Tibetan Qula, A Raw Yak Milk Cheese

  • In this study, 69 lactobacilli isolated from Tibetan Qula, a raw yak milk cheese, were screened for their potential use as probiotics. The isolates were tested in terms of: Their ability to survive at pH 2.0, pH 3.0, and in the presence of 0.3% bile salts; tolerance of simulated gastric and intestinal juices; antimicrobial activity; sensitivity against 11 specific antibiotics; and their cell surface hydrophobicity. The results show that out of the 69 strains, 29 strains (42%) had survival rates above 90% after 2 h of incubation at pH values of 2.0 or 3.0. Of these 29 strains, 21 strains showed a tolerance for 0.3% bile salt. Incubation of these 21 isolates in simulated gastrointestinal fluid for 3 h revealed survival rates above 90%; the survival rate for 20 of these isolates remained above 90% after 4 h of incubation in simulated intestinal fluid. The viable counts of bacteria after incubation in simulated gastric fluid for 3 h and simulated intestinal fluid for 4 h were both significantly different compared with the counts at 0 h (p<0.001). Further screening performed on the above 20 isolates indicated that all 20 lactobacilli strains exhibited inhibitory activity against Micrococcus luteus ATCC 4698, Bacillus subtilis ATCC 6633, Listeria monocytogenes ATCC 19115, and Salmonella enterica ATCC 43971. Moreover, all of the strains were resistant to vancomycin and streptomycin. Of the 20 strains, three were resistant to all 11 elected antibiotics (ciprofloxacin, erythromycin, tetracycline, penicillin G, ampicillin, streptomycin, polymyxin B, vancomycin, chloramphenicol, rifampicin, and gentamicin) in this study, and five were sensitive to more than half of the antibiotics. Additionally, the cell surface hydrophobicity of seven of the 20 lactobacilli strains was above 70%, including strains Lactobacillus casei 1,133 (92%), Lactobacillus plantarum 1086-1 (82%), Lactobacillus casei 1089 (81%), Lactobacillus casei 1138 (79%), Lactobacillus buchneri 1059 (78%), Lactobacillus plantarum 1141 (75%), and Lactobacillus plantarum 1197 (71%). Together, these results suggest that these seven strains are good probiotic candidates, and that tolerance against bile acid, simulated gastric and intestinal juices, antimicrobial activity, antibiotic resistance, and cell surface hydrophobicity could be adopted for preliminary screening of potentially probiotic lactobacilli.

유기질비료(有機質肥料) Bio-com 시용(施用)이 토양(土壤)의 미생물상(微生物相) 및 화학성(化學性)에 미치는 영향(影響) (Effect of Organic Fertilizer as Bio-com Application on the Changes of Soil Microorganisms, Gas Evolution, and Mineral-N Transformation in Submerged Condition)

  • 상품화(商品化) 되어있는 유기질비료(有機質肥料) Bio-com$^{(R)}$을 토양(土壤)에 시용시(施用時) 토양(土壤)의 pH, Eh, gas 생성(生成), Ammonia 화성율(化成率) 질산화성율(窒酸化成率) 및 미생물(微生物) 변화(變化)에 미치는 영향(影響)을 알기 위하여 일반농가퇴비(一般農家堆肥) 및 양송이 폐상퇴비등(等)을 비교(比較)하여 담수조건(湛水條件)에서 실내시험(室內試驗)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 삼요소구(三要素區)에 비(比)하여 각종(各種) 퇴비시용(堆肥施用)은 토양(土壤) pH를 상승시키는 경향(傾向)이 있었으며 Bio-com$^{(R)}$ 시용(施用)은 타공시유기물(他供試有機物)과 상이(相異)하지 않했다. 2. 시험후(試驗後) 토양(土壤)의 미생물수(微生物數) 변화(變化)는 삼요소(三要素) 혹은 각종(各種) 퇴비(堆肥)를 삼요소(三要素)와 병용(倂用)하므로서 현저(顯著)한 증가(增加)를 보였으며 농가퇴비시용구(農家堆肥施用區)에서 세균수(細菌數)의 증가(增加)가 현저(顯著)하였다. 3. Bio-com$^{(R)}$ 시용(施用)은 항온기간중(恒溫期間中) 질산화성율(窒酸化成率)을 증가(增加)시켰으며 그율(率)은 타처리(他處理)에 비(比)하여 수배정도(數培程度)높아서 밭작물재배지(作物栽培地)에 시용(施用)이 합리적(合理的)이었다. 4. Bio-com$^{(R)}$ 시용(施用)에 의한 gas의 생성(生成)은 일반퇴비(一般堆肥)나 양송이 폐상퇴비(堆肥) 시용(施用)과 비슷한 경향(傾向)을 보였고 $N_2O$의 생성(生成)은 삼요소시용(三要素施用)보다 낮은양(量)을 보였다.

미생물 합성에 의한 poly(3-hydroxybutyrate)의 분자량과 말단관능기 제어 (Control of Molecular Weight and Terminal Groups of Poly(3-hydroxybutyrate) in Bio-synthesis)

  • In the bio-synthesis of poly(3-hydroxybutyrate)(PHB), which is a kind of poly(3-hydroxyalkanoate)(PHA), aimed to control the low molecular weight of PHB and obtain a telechelic PHB. As a result of incubation of R. eutropha at $30^{\circ}C$ with ethylene glycol added as a chain transfer agent, PHB content on the dry cell weight increased up to 24h, however, it decreased after that, and the molecular weight of PHB increased from 9h to 12h, and then, decreased up to 72h. The decrease of the content and the molecular weight of PHB indicates that PHB was decomposed as an energy source in bacterial cells and was incorporated into metabolic pathways. $^1H-NMR$ of the obtained PHB after incubation for 72h was measured to determine the terminal groups of the PHB during incubation. As the results of $^1H-NMR$ measurement, the peaks derived from ethylene glycol in both terminals of PHB were observed. Which indicate that the terminal reaction was caused by the addition of ethylene glycol, and that telechelic PHB having hydroxyl group at the both terminals where molecular weight was controlled was successfully synthesized.

Density Functional Study on the C-H Bond Cleavage of Aldimine by a Rhodium(I) Catalyst

  • We investigated the C-H bond activation mechanism of aldimine by the [RhCl$(PPH_3)_3$] model catalyst using DFT B3LYP//SBKJC/6-31G*/6-31G on GAMESS. Due to their potential utility in organic synthesis, C-H bond activation is one of the most active research fields in organic and organometallic chemistry. C-H bond activation by a transition metal catalyst can be classified into two types of mechanisms: direct C-H bond cleavage by the metal catalyst or a multi-step mechanism via a tetrahedral transition state. There are three structural isomers of [RhCl$(PH_3)_2$] coordinated aldimine that differ in the position of chloride with respect to the molecular plane. By comparing activation energies of the overall reaction pathways that the three isomeric structures follow in each mechanism, we found that the C-H bond activation of aldimine by the [RhCl$(PH_3)_3$] catalyst occurs through the tetrahedral intermediate.

한국인 구치열에서 치간유두 존재와 치아접촉점과 치간골 거리와의 관계 (Relationship between Interdental Papilla Existence & Distance from Interdental Alveolar Crest to Contact Point in the Posterior Dentition of Korean adults)

  • The anatomic structure around interproximal area plays an important role not only in the natural teeth, but also in the implant. The loss of papilla can lead to cosmetic deformity, phonetic problem, food impaction on the anterior dentition, and masticatory problem, food impaction and proximal caries on the posterior dentition. The purpose of this study was to evaluate the relationship between interdental papilla existence and distance from contact point to alveolar crest in Korean posteior dentition. 45 Korean adult patients(31males, 14 females) participated in this study. Measurements were carreid out total 126 interproximal areas, 18 first premolar, 31 second premolar, 40 first molar, and 37 second molar areas. Papilla index was recorded as suggested by Jemt. Distance between contact point and alveolar crest measrued by Florida $probe^{R}$, after flap elevation. Each distance was measured 10 times by every 0.1mm unit. The results showed that the mean Papilla index 1.37 and mean distance between contact point and alveolar crest was 7.44mm. The correlation between the Papilla index and distance was high negative correlation(Pearson correlation=-0.47), and it was statistically significant(P=0.000) When the distance between contact point and alveolar crest was 5mm, the loss of papilla was appeared almost in half cases. When the distance was 6mm, the papilla loss was present 95%, when 7mm, the papilla loss was 100%.
