The Physico-Chemical Properties and Cooking Qualities of Barley Isogenic Lines (보리 Isogenic Lines의 이화학적 품질과 취반특성)

  • This study is presented to investigate the physico-chemical properties and qualities for the starch isogenic lines bred in barley(Hordeum vulgare L.).The pearling yield showed higher value in Franubet and Wafranubet than others, but the whiteness of pearled barley varied with the cultivars. The milling rate showed excellence in Franubet, fractured granular lines, whereas that of the waxy and round endosperm isogenic lines is poor. Franubet, the lines having fractures starch granule, was highest in starch content. The highest value in swelling power and water-binding capacity of the barley was proved by the lines having waxy and fractured starch endosperms. The amylogram patterns indicated that the waxy lines such as Wanubet and Wafranubet have lower initial gelatinization temperature and higher maximum viscosity than those of the normal lines. ${\beta}-Glucan\;viscosity$ was generally higher in waxy barley but changed greatly with the cultivars. The water absorption, soluble solid and expansibility in cooking property showed the highest value in waxy and fractured starch granular lines, and the whiteness of cooked barley did not differ with cultivars.

  • Starch granules of mungbean observed by microscope and scanning microscope were oval or round, $8{\sim}13{\mu}$ wide, and $18{\sim}30{\mu}m$ long. X-ray diffraction of the starch granules resulted weak crystallinity at $2{\theta}\;16.9^{\circ}$ The blue value of the starch was 0.36, amylose content 22.7%, alkali number 8.52, ferricyanide number 1.06, and water binding capacity of 81.6%. Swelling of the starch was negligible until $50^{\circ}C$, thereafter it increased rapidly. Optical transmittance of 0.3% starch suspension was increased rapidly from $65^{\circ}C$ and the gelatinization at $65{\sim}90^{\circ}C$ was of single stage. Amylogram patterns of the 6.7 and 8% starch solution were similar with no peak viscosity. The time constant for retrogradation of 40% starch gel stored at $21^{\circ}C$ was 1.99 days.

Iron and vitamin D status in breastfed infants and their mothers

  • Purpose: We assessed the relationships between iron and vitamin D statuses in breastfed infants and their mothers and evaluated the determinants of iron and vitamin D deficiencies in breastfed infants. Methods: Seventy breastfed infants aged 4-24 months and their mothers participated in this study from February 2012 to May 2013. Complete blood counts, total iron binding capacity, and levels of C-reactive protein, iron, ferritin, calcium, phosphate, alkaline phosphatase, and 25-hydroxyvitamin D (25(OH)D) in infants and their mothers were measured. Results: A history of maternal prepregnancy anemia was associated with lower ferritin and 25(OH)D levels in both infants and their mothers. The 25(OH)D level of infants correlated with maternal 25(OH) D levels. The independent risk factors for iron deficiency in breastfed infants were the duration of breastfeeding (odds ratio [OR], 6.54; 95% confidence interval [CI], 1.09-39.2; P=0.04) and infant body weight (OR, 2.65; 95% CI, 1.07-6.56; P=0.04). The determinants for vitamin D deficiency were the infant's age (OR, 0.15; 95% CI, 0.02-0.97; P=0.046) and maternal 25(OH)D level (OR, 0.74; 95% CI, 0.59-0.92; P=0.01). Conclusion: A maternal history of prepregnancy anemia requiring iron therapy was associated with lower current ferritin and 25(OH)D levels in both infants and their mothers. Therefore, physicians should monitor not only iron but also vitamin D levels in infants who are breastfed by mothers who had prepregnancy anemia.

Effect of Expression of Genes in the Sphingolipid Synthesis Pathway on the Biosynthesis of Ceramide in Saccharomyces cerevisiae

  • Ceramide is important not only for the maintenance of the barrier function of the skin but also for the water-binding capacity of the stratum corneum. Although the exact role of ceramide in the human skin is not fully understood, ceramide has become a widely used ingredient in cosmetic and pharmaceutical industries. Compared with other microorganisms, yeast is more suitable for the production of ceramide because yeast grows fast and is non-toxic. However, production of ceramide from yeast has not been widely studied and most work in this area has been carried out using Saccharomyces cerevisiae. Regulating the genes that are involved in sphingolipid synthesis is necessary to increase ceramide production. In this study, we investigated the effect of the genes involved in the synthesis of ceramide, lcb1, lcb2, tsc10, lac1, lag1, and sur2, on ceramide production levels. The genes were cloned into pYES2 high copy number vectors. S. cerevisiae was cultivated on YPDG medium at $30^{\circ}C$. Ceramide was purified from the cell extracts by solvent extraction and the ceramide content was analyzed by HPLC using ELSD. The maximum production of ceramide (9.8 mg ceramide/g cell) was obtained when the tsc10 gene was amplified by the pYES2 vector. Real-time RT-PCR analysis showed that the increase in ceramide content was proportional to the increase in the tsc10 gene expression level, which was 4.56 times higher than that of the control strain.

Binding of Zeolites to Inorganic Fiber using Covalent Linkers (공유결합을 이용한 무기질 섬유와 제올라이트의 결합)

  • Zeolites with excellent adsorption capacity of volatile organic compounds were attached onto inorganic fibers which were the raw materials of honeycomb-type adsorbers. The amounts of zeolite particles attached onto the fibers considerably increased by treating them with hydrochloric acid, sulfuric acid, or hydrofluoric acid. Various functional groups such as chloropropyl, aminopropyl and epoxy groups of silane compounds, and amine groups of polyethylenimine were employed as covalent linkage materials between the fibers and zeolite particles. The state of the fibers coated with zeolite particles was examined by scanning electron microscopy, and the amounts of zeolite particles bound to the fibers were estimated from their BET surface areas. The largest amount was obtained when polyethylenimine was employed as a linkage material. Polyethylenimine was the most effective for attaching zeolite particles onto the inorganic fibers among various linkers employed.

Quality Characteristics of Naengmyon Added with Persimmon (Diospyos kaki L. folium) Leaf Powder (감잎분말을 첨가한 냉면의 품질 특성)

  • This study was conducted to develop naengmyon added with persimmon leaf powder. To find out the proper mixing ratio, physicochemical properties were investigated. There was a little difference in water binding capacity between control and naengmyon added with persimmon leaf powder. Amylogram showed that initial pasting temperature and peak point value were increased as increment of adding amount of persimmon leaf powder. After cooking, the weight of a naengmyon was increased but cooking loss and turbidity were decreased with increment of persimmon leaf powder. At a glance of color difference values, there was a distinctive boundary near at 6%. But similar at over 6% as increasing adding amount of persimon leaf powder texture characteristics such as hardness, adhesiveness, cohesiveness, springness and gumminess were declined. From overall experimental result, the proper mixing ratio of persimmon leaf powder for naengmyon was 4%.

Physicochemical Properties of Hydroxypropylated Chestnut Starch (Hydroxypropyl화 밤 전분의 이화학적 특성)

  • Physicochemical properties of the native chestnut starch(NCS) and hydroxypropylated chestnut starch(HCS) with propylene oxide were investigated. The water binding capacity and blue value of chestnut starch were increased by hydroxypropylation. With increasing temperature, the swelling power and solubility were increased and those of HCS were higher. The light transmittance of HCS was higher than it of NCS. Endotherm characteristics were studied with DSC showed that temperatures of initial gelatinization of NCS and HCS were $64.44^{\circ}C$ and $62.80^{\circ}C$, respectively. The temperature of initial gelatinization and enthalpies gelatinization in chestnut starch were decreased by hydroxypropylation. And the viscosity of HCS was higher than that of NCS.

  • Strontium selective chromatographic material $(Sr-Spec^{TM})$ was investigated for separation of radiostrontium from environmental soil and water sample. This chromatographic material has great capacity of binding of strontium ion in nitric acid media, and has selectivity to permit the separation of stontium from bulk amount of calcium. But the extraction of strontium was reduced by the other interfering ions such as K and Ba. So, in order to apply this material to the soil sample, prior removal treatment of K and Ba was needed. But the Sr-Spec material could provides simple and effective methods for the separation and removal of radiostrontium from liquid sample.

Quality Characteristics of Wet Noodles with Allium hookeri Powder (삼채(Allium hookeri) 분말을 첨가한 생면의 품질 특성)

  • This study investigated the quality of wet noodles added with Allium hookeri powder. Wet noodles were prepared by addition of 0, 2, 6, and 10% powder to flour of the basic formulation. The water binding capacity of Allium hookeri powder was higher than that of flour. Swelling power and solubility increased with increased temperature. The weight, water absorption, volume, and turbidity values of cooked noodles showed no significant differences. The lightness value decreased with an increase in Allium hookeri powder content. The redness and yellowness values increased with an increase in Allium hookeri powder content. The textural properties of cooked noodles decreased with an increase in Allium hookeri powder content. Antioxidative activity as measured by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity of wet noodles increased as the concentration of Allium hookeri powder increased. The highest quality noodles were obtained with 2% Allium hookeri powder in the wet noodle formula.

Though the effectiveness of ceramide is not understood fully, ceramide has become a widely used ingredient in cosmetic and pharmaceutical industries. However, ceramide production from Saccharomyces cerevisiae has not been widely studied and the quantity are very low. Gene deletion in the cell is used frequently to investigate the function of gene and verification research of drug target. Specially, deletion mutant library is useful for a large amount functional analysis of gene. In this study, deletion mutants of genes on the metabolic pathway of ceramide synthesis in S. cerevisiae were grown in a batch culture and the cellular content of ceramide was measured. The ceramide content was highest in ${\triangle}$ydc1 mutant and 6 mg ceramide/g cell was obtained.
