Preparation of New Binaphthol Derived Chiral Stationary Phases

  • To develop more advanced binaphthol derived CSPs, two new 1,1-bi-2-naphthol derived chiral stationary phases (CSPs) (CSP 1 and CSP 2) were prepared by connecting with silica gel at a hydroxy (OH) group of binaphthol. Previously reported two 1,1'-binaphthyl-2,2'-diamine derived CSPs (CSP 3 and CSP 4) were also prepared to compare the role of their functional groups (-OH and -$NH_2$) with CSP 1 and CSP 2. Enantioseparation of randomly selected, 11 chiral compounds on these four CSPs was performed and the results compared to each other. 3,5-Dinitrobenzoyl (3,5-DNB) derivatives of each CSP showed better results than the others and binaphthyldiamine derived CSP showed slightly better than binaphthol derived one.

  • Binaphthol-derived chiral ketones 1a-c were synthesized and were shown to serve as active catalysts for asymmetric epoxidation of olefins using Oxone, although their enantioselectivities were not high. However, very interestingly, the stereochemical outcome of the resulting epoxides implicates that in the epoxidation using 1a-c, the planar transition state may be more favorable than the spiro transition state.

  • An easy and convenient synthetic route to (S)-2-hydroxy-2'-(3-phenyluryl-benzyl)-1,1'-binaphthyl-3-carboxaldehyde (1), capable of recognizing tryptophan by fluorescence has been developed. The binol carboxaldehyde 1 exhibited a high selectivity to L-tryptophan over other examined L-${\alpha}$-amino acids such as alanine, phenylalanine, glutamine, arginine, lysine, serine, threonine, aspartat, valine, histidine and cysteine, with a fluorescence "turn-on" signal. In addition, 1 displayed chiral discrimination with good enantioselectivity toward L-tryptophan over D-tryptophan through different fluorescence enhancement factors.

Synthesis of Novel H8-Binaphthol-based Chiral Receptors and Their Applications in Enantioselective Recognition of 1,2-Amino alcohols and Chirality Conversion of L-Amino acids to D-Amino acids

  • Novel $H_8$-binaphthol-based chiral receptors appended with an uryl moiety (2a) and a guanidinium moiety (2b) have been designed and synthesized for the enantioselective recognition of 1,2-amino alcohols via reversible imine formation. The selectivities ($K_R/K_S$ = 9.8 ~ 19.4) of 2b in imine formation with 1,2-amino alcohols are higher than those of 2a ($K_R/K_S$ = 1.8 ~ 4.5). Similar efficiency trend have been observed in the conversion of L-amino acids to D-amino acids, i.e., the efficiency of the receptor 2b (D/L ratio: 4.3 ~ 10.1) is superior to 2a (D/L ratio: 4.0 ~ 8.7).

  • A new thiourea based receptor (1) was synthesized and applied to fluoride ion recognition in acetonitrile solution. Receptor 1 displayed dual changes in absorption and fluorescence emission intensities selectively for fluoride ions. The interaction of 1 with fluoride undergoes a deprotonation process that is confirmed by $^1H$ NMR titration.
