Survivability of Encapsulated Bifidobacterium breve BS2-PB3 during Storage and in Simulated Gastric Juice and Bile Salt

  • Bifidobacterium breve is commonly administered as probiotics in fermented foods or supplements. Accordingly, a sufficient number of live cells must be consumed to experience the benefits of probiotics. Microencapsulation is a technique used to increase the viability and stability of probiotics when exposed to industrial processes, low/high temperature, storage periods, and gastrointestinal conditions. This study aimed to evaluate the effect of microencapsulation with alginate-chitosan and pectin-chitosan matrices on the viability and stability of B. breve BS2-PB3. Microencapsulation was performed using alginate-chitosan (2%, 0.75%) and pectin-chitosan (3%, 0.75%), followed by freeze drying. The encapsulation efficiency of both matrices was 94%, and the survival rate after freeze drying was 96%. After 8 weeks of storage, B. breve BS2-PB3 showed a stable viability, with more than 96% and 83% survival rates at 4℃ and room temperature, respectively. Both encapsulation matrices maintained bacterial viability up to 55% in simulated gastric juice (pH 2) and up to 66% in 0.3% bile salt solution. The administration of microcapsules to mice lowered the fecal pH and increased the defecation frequency. This study demonstrated that microencapsulation using alginate-chitosan and pectin-chitosan could maintain probiotic viability and stability.

Functional Properties of Filamentous Fungi Isolated from the Indonesian Fermented Dried Cassava, with Particular Application on Poultry

  • The study aimed to evaluate the probiotic properties, antioxidant activity and fermentative capacity of Acremonium charticola and Rhizopus oryzae isolated from the Indonesian fermented dried cassava, with particular application on poultry. A. charticola inhibited the growth of Escherichia coli and Aspergillus flavus. A. charticola and R. oryzae grew in potato dextrose agar (PDA) adjusted to pH 3 and 8 or in PDA supplemented with bile salt up to 0.8%. After soaking for 8 hr, the survival rate of A. charticola in the simulated gastric juice (pH 2) and bile solutions (2% bile salt) was lower than that of R. oryzae. A. charticola and R. oryzae exhibited strong antioxidant activities. Compared to unfermented cassava pulp (control), the fibre content of cassava pulp tended to be lower after fermentation with A. charticola for 14 days. The populations of A. charticola and R. oryzae were significantly higher in fermented cassava pulp than in unfermented one. Coliform was higher in cassava pulp fermented with R. oryzae or A. charticola + R. oryzae compared to control after 7 days of fermentation, however, the bacteria were not different between A. charticola-fermented cassava pulp and control. Lactic acid bacteria (LAB) were higher in A. charticola- and R. oryzae-fermented cassava pulp than those in control, however, no difference of LAB was observed between A. charticola + R. oryzae-fermented cassava pulp and control. In conclusion, A. charticola exhibited antibacterial, antifungal and antioxidant activity, gastrointestinal persistence and fermentative capacity that may be beneficial for poultry industry.

Advanced Formulation and Pharmacological Activity of Hydrogel of the Titrated Extract of C. Asiatica

  • Titrated extract of Centella asiatica (TECA) contains three principal ingredients, asiaticoside (AS), asiatic acid (AA), and madecassic acid (MA). These components are known to be clinically effective on systemic scleroderma, abnormal scar formation, and keloids. However, one problem associated with administration of TECA is its low solubility in aqueous as well as oil medium. In this study, various nonionic surfactants and bile salts as anionic surfactant were tested and screened for solubilizing TECA with a view to developing topical hydrogel type of ointment which is stable physicochemically, and has better pharmacological effects. When TECA was incorporated into various nonionic surfactant systems, labrasol had the most potent capacity for solubilizing TECA. In cases of bile salt systems, Na-deoxycholate (Na-DOC) had foremost solubilizing capacity, even more than labrasol. In differential scanning calorimetric study, the peaks of AA, MA, AS and Na-DOC disappeared at the coprecipitate of $1\%$ TECA and $1\%$ Na-DOC, suggesting the optimum condition of Na-DOC for solubilizing TECA. When the physicochemical stability of hydrogel containing this mixture was assessed, it was stable at room temperature for at least one month. Pharmacologically it significantly decreased the size of wound area at the $9^{th}$ day when applied to the wound area of rat dorsal skin. Taken together, solubility of TECA was dramatically improved by using non ionic and anionic surfactant systems, and Na-DOC was found to be the most effective solubilizer of TECA in formulating a TECA-containing hydrogel typed ointment. Moreover this gel was considered to be applicable to clinical use for wound healing effect.

  • Bacillus licheniformis was an useful material for improvement of fecal microflora. The stability of Bacillus licheniformis in low pH (pH 2, 3, 4, 6, 7), artificial bile acid, high concentration of salt (0, 5, 10, 20, $30\%$), and ethanol (0, 4, 8, 16, $32\%$) was investigated in this study. The viability of Bacillus licheniformis was stable for pH 3, 4, 6, 7 Final viability of Bacillus licheniformis was confirmed below $25\%$, in the bile acid. In high concentration of salt, Bacillus licheniformis showed optimal viability in $10\%$ of salt. The viability of Bacillus licheniformis was over $95\%$ in condition of ethanol concentration $4\%,\;8\%,16\%,\;32\%$ contrast with uncontained ethanol at $37^{\circ}C$ for 4h. It was verified that Bacillus licheniformis was stable for pH 4, 6, 7, high concentration of salt and ethanol but was unstable in pH 2 and bile acid.

In Vitro Probiotic Properties of Indigenous Dadih Lactic Acid Bacteria

  • The aim of this research was to identify candidate probiotic lactic bacteria among indigenous dadih lactic isolates. Dadih is an Indonesian traditional fermented milk of West Sumatra which is fermented naturally. Viability of the strain is critical in determining the capacity of lactic bacteria to induce immune stimulation as well as to colonize in the intestinal tract. Therefore, LAB are proposed to exert health promoting or probiotic effects in human, such as inhibition of pathogenic microflora, antimutagenic, and the reduction of cholesterol levels. This manuscript reports in vitro probiotic properties of indigenous dadih lactic bacteria, especially some important colonization factors in GI tract, such as lysozyme, acid and bile tolerance. Bile Salt Hydrolase (BSH) activity, spectrum of bacteriocin, and antimutagenic activity of bacterial cells were also assessed. Twenty dadih lactic isolates were screened further for their tolerance to low pH, at pH 2 and 3 as well as their bile tolerance. There were ten isolates classified as acid and bile acid tolerant, and further screened for lysozyme tolerance, BSH activity. The spectrum of bacteriocin activity of isolates was assayed using cell-free neutralized supernatants by agar spot test against variety of pathogens. Lc. lactis subsp. lactis IS-10285, IS-7386, IS-16183, IS-11857 and IS-29862, L. brevis IS-27560, IS-26958 and IS-23427, Leu.mesen.mesenteroides IS-27526, and L. casei IS-7257 each has good survival rate at low pH values and in the presence of lysozyme, and short lag time in the presence of 0.3 % oxgall. Lc. lactis subsp. lactis IS-11857 and IS-29862 each has high BHS activity, Lc. lactis subsp. lactis IS-10285 and IS-16183 each had a positive spectrum of bacteriocin activity against E. coli 3301 and Lysteria monocytogenes ATCC 19112, while L. brevis IS-26958 has high BHS activity as well as positive spectrum of bacteriocin against E. coli 3301, Lysteria monocytogenes ATCC 19112, and S. aureus IFO 3060. All of the ten dadih lactic strains performed in vitro acid and bile tolerance, indicating a possibility to reach the intestine alive, and display probiotic activities.

Effects of Mustard Leaf(Brassica Juncea) on Cholesterol Metabolism in Rats (갓의 급이가 흰쥐의 Cholesterol 대사에 미치는 영향)

  • To investigate the effects mustard leaf(Brassica Juncea) on Cholesterol metabolism, male Sprague Dawley rate were fed semipurified diets containing 2% or 4% mustard leaf with or without cholesterol for 5 weeks. Plasma cholesterol content decreased significantly by feeding 4% mustard leaf with of without cholesterol for 5 weeks. Plasma cholesterol content decreased significantly by feeding 4% mustard leaf in rats fed 1% cholesterol in the diet. In addition, HDL-cholesterol increased slightly by the feeding of mustard leaf, resulting in a significant increase in the HDL-cholesterol/total cholesterol ratio and a reduction of atherosclerotic index. However, levels of plasma lipids were not influenced by mustared leaf in rats fed cholesterol-free diet. The contents of all classes of lipid in liver increased by dietary cholesterol. Of the liver lipids, triglyceride and cholesterol ester were accumulated most, showing a fatty liver synodrome. Supplementation of mustard leaf to cholesterol-containing diet resulted in a slight decrease in neutral lipid contents of liver. Fecal cholesterol excretion was higher by more than 2.7 and 3.3-fold in rats fed 2 and 4% mustard leaf than in control rats fed cholesterol. Similar trends were found in fecal bile salt excretion; rats fed and 4% mustard leaf excreted more bile salts by more than 1.5 and 2% than those fed control diet containing cholesterol. In summary, mustard leaf may have an antiatherogenci effect of reducing plasma cholesterol level and increasing HDL-cholesterol level. The plasma cholesterol lowering effect of mustard leaf is suggested to be due, at least in part, to increase in fecal excretion of cholesterol and bile acids.

Antibacterial Activity and Probiotic Properties of Lactic Acid Bacteria from Korean Intestine Origin (한국인 장관에서 분리한 유산균의 항균활성 및 프로바이오틱스 특성 연구)

  • The purpose of this study was to investigate the probiotic properties of lactic acid bacterial strains isolated from human feces. The properties were tested on the basis of guideline for probiotic selection protocol such as tolerance for acid or bile salt, autoaggregation, antibiotic resistance, and antimicrobial activity. Total 25 lactic acid bacteria were isolated from human feces, and their antibacterial activity was tested against Staphylococcus aureus, Escherichia coli, E. coli O157:H7, Vibrio parahaemolyticus, V. alginolyticus using an agar diffusion assay. Among them, 4 selected strains were identified by analysis of their 16S rRNA, as Lactobacillus rhamnosus MG316, L. acidophilus MG501, L. reuteri MG505, and L. gasseri MG570. Results show that resistance to low pH and bile salts. Also, the selected strains were resistant to bile acid up to 3% and their autoaggregation rates were as high as 60%. All strains tested were resistance to nalidixic acid and kanamycin.

The Solubilization Behavior of DOPE-Immunoliposomes with Immunoglobulin G(IgG) by Added Bile Salts (Immunoglobulin G(IgG)를 함유한 DOPE 리포솜의 제조와 담즙산염에 의한 용해 특성)

  • The effects of bile salts (BS) on the stability of dioleoylphosphatidylethanolamine (DOPE) liposomes were investigated, observing apparent absorbance of vacant liposomes and calcein release from entrapped liposomes. Unilamellar liposomes were prepared by using a small quantity of palmitoly-immunoglobulin G(IgG) ($2.5{\times}10^{-4}$ mo1/lipid mol) to stabilize the bilayer phase of the unsaturated DOPE which by itself does not form stable liposomes. The destabilization of PE immunoliposomes by papain, clearly demonstrates that the IgG is essential for stabilization of PE bilayer. Approximately 4% of the entrapped calcein was released from the PE liposomes after 1 hr from liposome formation. Calcein release and absorbance of liposomes depended on the BS/lipid ratio because of the solubilization of lipid molecule in bilayer and the formation of mixed micelles. At very low BS concentrations, the incorporation of BS induced BS/lipid aggregates in the outer vesicles monolayer, while high BS concentrations, mixed micelles were formed. Chelate and its conjugates as $3{\alpha},\;7{\alpha},\;12{\alpha}-trihydroxy$ BS induce the concentration of the $3{\alpha}$, $12{\alpha}-dihydroxy$ BS at half-maximal solubilization of immunoliposomes to approximately 2.5-, or 5-fold. Conjugation of BS with glycine or taurine slightly enhanced their capacities to perturb membranes.

Factors Affecting Adhesion of Lactic Acid Bacteria to Caco-2 Cells and Inhibitory Effect on Infection of Salmonella Typhimurium

  • In this study, seven strains isolated from mustard leaf kimchi were screened for their tolerance to simulated gastric and bile juices, the adhesive properties to Caco-2 cells, and the inhibition ability of Salmonella Typhimurium ATCC 29631 adhesion. Lactobacillus acidophilus GK20, Lactobacillus paracasei GK74, and Lactobacillus plantarum GK81, which were resistant to bile as well as gastric juices, possessed high bile-salt hydrolase (BSH) activity towards both sodium glycocholate and sodium taurocholate. The strongest in vitro adherence of $53.96{\pm}4.49%$ was exhibited by L. plantarum GK81 followed by L. acidophilus GK20 with adhesion levels of $40.72{\pm}9.46%$. The adhesion of these strains was significantly (p < 0.05) reduced after exposure to pepsin and heating for 30 min at $80^{\circ}C$. Addition of $Ca^{2+}$ led to a significant (p < 0.05) increase of the adhesion of L. acidophilus GK20, but the adhesion ability of L. plantarum GK81 was not different from the control by the addition of calcium. In the competition and exclusion experiment, the adhesion inhibition of S. Typhimurium by L. plantarum GK81 strain was much higher than the other strains. Moreover, the exclusion inhibition of S. Typhimurium by L. acidophilus GK20 was considerably high, although the inhibition activity of this strain was lower than L. plantarum GK81.

Probiotic Characteristics of Lactobacillus rhamnosus Isolated from Kefir (Kefir로부터 분리한 Lactobacillus rhamnosus의 Probiotic 특성)

  • To search probiotic microorganisms, we isolated Lactobacillus sp. from kefir, The Lactobacillus sp. strain showed $99.5\%$ of identity to species Lactobacillus rhamnosus by API kit. Lactobacillus rhamnosus showed high resistances to acidic environment, which grew well even at pH 2.0 and $1.0\%$ bile salt Enzyme activity of Lactobacillus rhamnosus was higher in amylase ($0.673\;{\mu}mol/min/mg$) than that in xylanase ($0.288\;{\mu}mol/min/mg$), cellulase($0.117\;{\mu}mol/min/mg$) and phytase($0.269\;{\mu}mol/min/mg$). Especially, the Lactobacillus rhamnosus showed high heat stability which remained $1{\times}10^6\;CFU/ml$ at $60^{\circ}C$. The maximum numbers of Lactobacillus rhamnosus on growth owe was reached at 24 h fermentation and pH was decreased to 4.6. The resistances of Lactobacillus rhamnosus to acidic pH and bile salt were better than that of Lactobacillus acidophilus used as control. When Lactobacillus rhamnosus was cultured with E. coli in MRS broth, E. coli was disappeared after 18 h. These result suggest that the isolated Lactobacillus rhamnosus has a useful probiotics properties.
