Calcium Carbonate Buffer System을 이용한 Bifidobacterium longum의 배양 효율 증대에 관한 연구 (Enhancement of Cultivation Efficiency of Bifidobacterium longum Using Calcium Carbonate Buffer System)

  • 낮은 pH 환경에서 생장이 억제되는 혐기성 유산균인 bifidobacteria의 배양효율 증대를 위한 완충제로서 alginate로 고정화된 평균 지름 2.5 mm의 $CaCO_3$ 비드를 제조하였다. 고정화된 $CaCO_3$ 비드의 완충 효과를 조사하기 위하여 초산, 유산 및 초산과 유산을 3:2 몰농도 비율로 혼합한 산용액을 이용하여 pH 4.0으로 조정된 m-TPY 배지에 첨가되는 $CaCO_3$ 비드의 양을 달리하여 20분간 반응시킨 결과 첨가되는 $CaCO_3$ 비드의 양에 따라 다양한 pH 상승 효과가 나타남을 관찰하였고, 배양액에 첨가되는 $CaCO_3$ 비드의 양을 달리하여 bifidobacteria의 배양 특성을 조사한 결과 배양액에 첨가되는 $CaCO_3$ 비드의 양이 증가할수록 배양액내 완충능력과 균증식도가 크게 증가하였다. 또한, $CaCO_3$ 비드가 30%로 도입된 배양액에 포도당 농도를 0.5%, 1.0%, 1.5%, 2.0%, 2.5% 그리고 5.0%로 각각 첨가하여 당농도에 따른 bifidobacteria의 배양 특성을 조사한 결과 배양액내 당농도가 증가함에 따라 배양액내 균증식도와 대수기의 지속 시간이 크게 증가되었다. 따라서 bifidobacteria의 배양효율 증대를 위하여 완충제로서 고정화된 $CaCO_3$ 비드의 이용은 매우 효과적인 것으로 생각되었다.

Bifidobacteria의 Caco-2 Cell 정착성에 미치는 영향 인자 (Factors Affecting the Adherence of Bifidobacteria to Caco-2 Cell)

  • Adherence of probiotic bacteria to intestinal epithelium is found to be the most principal characteristics among the various physiological functionality. This study was conducted to investigate the effect of bifidobacterial growth properties and condition on the Caco-2 cell adherence and to construct a basic data on adherence-related research. Among 20 strains of bifidobacteris tested, when measured by cell surface hydrophobicity(CSH) and cell agglutination(CA), Bifidobacterium bifidum ATCC29521, Bif. adolescentis K8, and Bif. infantis K9 were selected. Using these strains, variations of Caso-2 cell adherence depending upon experimental condition were analyzed. The results obtained are as follows : Even though Bif. bifidum ATCC29521, Bif. adolescentis K8, and Bif. infantis K9 reached more 85% cell surface hydrophobicity there was no significant difference in cell agglutination, when reached 31.54$\pm$0.54mg/ml. By direct count method for adherence, viable cell count of M3, K1, K2, K8, K9 and K10 reached more 100 counts per 100 Caco-2 cells. When Bif. bifidum ATCC29521, Bif. adolescentistis K8, and Bif. infantis K9 were used to compare the adherence depending upon viable cell counts, reaction time, and growth phase, the more viable cell count, and the more adhered cell counts, the less adherence percentage. In addition, there was no difference in adherence percentage of bifidobacteria when bifidobacteria was incubated from 1 to 8 hrs after Caco-2 cells already formed monolayer. Considering of the effect of growth phase of bifidobacteria on adherence variation, all strains showed the highest adherence during the early stage of stationary phase. In conclusion, adherence of bifidobacteria was affected by strain specificity, viable cell count, and growth activity.

Effect of Skim Milk-Alginate Beads on Survival Rate of Bifidobacteria

  • In this study, an attempt was made to increase the survival rate of bifidobacteria entrapped in alginate in the gastrointestinal tract, and to investigate the potential industrial applications, for example lyophilized capsules and yogurt. First, the protective effect of various food additives on bifidobacterial survivability was determined after exposure to simulated gastric juices and bile salts. The additives used in this study were skim milk (SM), polydextrose (PD), soy fiber (SF), yeast extract (YE), chitosan (CS), $\kappa$-carageenan ($\kappa$-C) and whey, which were added at 0.6% concentration (w/v) to 3% alginate-bifidobacterial solution. In the simulated gastric juices and bile salts, the protective effect of 0.6% skim milk-3% alginate (SM-A) beads on the survival rate of bifidobacteria proved to be higher than the other additives. Second, the hydrogen ion permeation was detected through SM-A vessel without bifidobacterial cells at different SM concentrations (0.2%, 0.4%, 0.6%, 0.8%, and 1.0%). There were no differences in terms of the pH decrease in SM-A vessels at 0.6%, 0.8%, and 1.0% (w/v) SM concentrations. The survival rate of bifidobacteria in SM-A beads would appear to be related to the SM buffering capacity against hydrogen ions and its tendency to reduce the pore size of bead. In this experiment, the survival rate of bifidobacteria entrapped in beads containing 0.6% SM showed the highest viability after exposure to simulated gastric juices for 3h, thereby indicating that 0.6% SM is the optimum concentration fir 3% alginate bead preparation. Third, the effect of SM-A beads on the freeze-drying and yogurt storage for 10 days was investigated. SM-A beads were found to be more efficient for freeze drying and yogurt storage than untrapped cells and the alginate bead. Consequently, the survival rate of bifidobacteria entrapped in SM-A beads was increased in simulated gastric juices, bile salts and probiotic products, such as lyophilized capsules and yogurt, SM-A beads can be expected to produce high value probiotic products.

Effect of Additives on the Viability of Bifidobacteria Loaded in Alginate Poly-l-lysine Microparticles during the Freeze-drying Process

  • Bifidobacteria-loaded alginate poly-l-lysine microparticles (bap microparticles) were prepared using an air atomization method and then freeze-dried. The viability of the bap microparticles was investigated as a function of the amount of the bifidobacteria cultures, and the addition of a yeast extract, cryoprotectants, antioxidants and neutralizer. The size of the bap microparticles with and without the bifidobacteria was $84.8{\pm}28.5\;{\mu}m$ ($mean{\pm}standard$ deviation) and $113.1{\pm}38.5\;{\mu}m$, respectively. The surface morphology was slightly ellipsoid and wrinkled regardless of the incorporating bifidobacteria. The viability gradually decreased with increasing freeze-drying time. Free-flowing powdered bap microparticles were obtained at least 12 h after freeze-drying the wetted slurry of bap microparticles. However, the particles tended to aggregate when either lactose or ascorbic acid was added. The addition of a yeast extract, cryoprotectants (glycerol and lactose), antioxidants ($NaHSO_3$ and ascorbic acid) and neutralizer $(Mg_3(PO_4)_2)$ resulted in a significantly higher viability of the bifidobacteria in the bap microparticles after freeze-drying (0.34-1.84 log) compared with the culture alone.

비피더스균(Bifidobacterium bifidum )에 의해 발효된 우유가 고지방식을 섭취한 흰주와 혈청 지질대사에 미치는 효과에 관한 연구 (The Effect of Fermented Milk by Bifidobacterium bifidum on Serum Lipid Metabolism in Rats Treated High Fat Diet)

  • 본 연구는 건강한 20대 한국 성인의 장에서 내산성과 내담즙성이 강한 비피더스균을 분리하여 비피더스균의 형철 콜레스테롤 저하 효과를 알아보기 위하여 체중 200g 정도 되는 Sprague-Dawley 종 희준 숫컷을 대상으로 우유, 비피더스균을 첨가항 우유, 캡슐화된 비피더스균을 첨가한 우유와 비피더스균에 의해 발혀된 발효유를 투여한 결과 우유, 비피더스 균을 첨가한 우유와 캡슐화된 비피더스균을 첨가항 우유를 투여한 실험군에서 혈청 콜레스테롤 저하 효과가 나타나지 않았으나 비피더스 발효유를 먹인 실험군에서 혈청 콜레스테롤 저하시키는 효과를 볼 수 있었으므로 혈청 콜레스테롤 저하 효과를 나타내는 것은 발효유에 있는 젖산균 그 자체가 아니라 발효유라는 결과를 얻었다.

  • Bifidobacteria의 배양효율을 증대하기 위하여 완충제로써 $CaCO_3$를 Ca-alginate에 고정화한 비드를 제조하여 사용하였다. B. longum KCTC 3218과 한국인 분변에서 분리한 B. longum HLC 3742 균주를 $CaCO_3$ 비드, NaOH, $Na_2CO_3$, $NH_4OH$ 등을 완충제로 각각 사용한 2.5-liter 발효기에서 각각 배양하여 균 증식과 저장에 따른 생존력을 조사한 결과 $CaCO_3$ 비드를 완충제로 사용한 경우가 다른 완충제를 사용한 경우보다 균 증식과 저장 안정성에서 효과적임을 알 수 있었다. 따라서 완충제로써 $CaCO_3$ 비드는 bifidobacteria의 고농도 배양과 생존력 증대에 유용할 것으로 사료되었다.

The Effect of Bifidobacteria and Various Oligosaccharides Consumption on the Risk of Colon Cancer in Rats

  • This study examined the effect of viable bifidobacteria and non-digestible carbohydrates on the cecal pH, colonic neoplastic lesion (aberrant crypt) and proliferating cell nuclear antigen (PCNA) labeling index in carcinogen-treated mts. Animals received s.c. injection of dimethylhydrazine (DMH) (15 mg/kg body weight) twice 3 days apart. Three days after the second carcinogen administration, the treatments were begun. 1he treatments were basal diet (AIN-76) with skim milk (Basal/skim), or the following diets with daily gavage of $10^8$ bifidobacteria: basal (Basal/bifido), $2\%$ fructo-oligosaccharide (FOS/bifido), $2\%$ soybean oligosaccharide (SBO/bifido), $2\%$ wheat bran oligosaccharide (WBO/bifido) and $8.4\%$ wheat bran (WB/bifido). After 4 weeks of treatment, cecal pH was measured using a pH probe. The number of aberrant crypt (AC), aberrant crypt foci (ACF) and crypt multiplicity were enumerated and colonic PCNA labeling index was determined using immunohistochemistry. Cecal pH was significantly reduced in SBO/bifido and FOS/bifido groups compared to control group. However, there were no significant differences in either number of AC or rates of cell proliferation as shown by PCNA labeling index among the groups, although mts fed FOS/bifido reduced the numbers of ACF compared to Basal/skim group. The SBO/bifido group did not reduce the number of ACF or PCNA labeling index. Also, other oligosaccharides did not reduce the risk of colon cancer compared to control group. The concomitant reduction of cecal pH and number of ACF suggest that the combination of bifidobacteria and FOS may reduce the risk of colon cancer.

Physiological Responses of Oxygen-Tolerant Anaerobic Bifidobacterium longum under Oxygen

  • In order to investigate what kind of response anaerobic bifidobacteria has on oxygen stress, five oxygen-tolerant bifidobacteria were isolated from human fecal samples. All were temporarily identified as Bifidobacterium longum through an analysis of carbohydrate utilization patterns and cellular fatty acid profiles. In the presence of oxygen, the lag phase became extended and the cell growth was suppressed. Bifidobacterial cell was able to remove dissolved oxygen in an early stage of growth and to overcome oxygen stress to a certain extent. The cell became long n size and showed a rough surface containing many nodes which were derived from abnormal or incomplete cell division. Cellular fatty acid profiled changed remarkably under a partially aerobic condition, so that the carbon chain of cellular fatty acid became short. All the dimethyl acetals originated from plasmalogen were reduced, any cyclopropane fatty acid, 9, 10-methyleneoctadecanoic acid ($C_{19:0}cyc9,10$), was increased remarkably. Oxygen stress induced a 5.5 kD protein in B. longum JI 1 of the oxygen-teolerant bifidobacteria, that was named Osp protein, and its N-terminal amino acid sequence was as follows: unknown amino acid-Thr-Gly-Val-Arg-Phe-Ser-Asp-Asp-Glu. Therefore, the oxygen-tolerant bifidobacteria seemed to defend against oxygen stress byincreasing the content of short fatty acid and cyclopropane fatty acid, and induction of an oxygen stress protein, but not the plasmalogen.

Inhibitory Effects of Bifidobacterium spp. Isolated from a Healthy Korean on Harmful Enzymes of Human Intestinal Microflora

  • Five hundreds of bifidobacteria were isolated from a healthy Korean and the inhibitory effects of these isloated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli or total human intestinal microflora. In comparison with the results of E. coli or intestinal microflora cultivation, Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525 effectively inhibited harmful enzymes ($\beta$-glucuronidase and tryptophanase) of E. coli and lowered the pH of the culture media. Also they inhibited the harmful enzymes ($\beta$-glucosidase, $\beta$-glucuronidase, tryptophanase and urease) and ammonia production of intestinal microflora, and lowered pH of the culture media by increasing lactic acid bacteria of intestinal microflora. When these isolated bifidobacteria were administered on mice, fecal harmful enzymes were also inhibited. Among tested bifidobacteria, B. breve K-110 had the highest inhibitory effect of fecal harmful enzymes.

Improvement of the electro-transformed cell yield for Bifidobacterium sp. with large DNA

  • In this study, the poor electro-transformant yield of large DNA in bifidobacteria was improved by increasing the DNA concentration, which was amplified by enhancing electroporation conditions: treating the cell wall weakening agent and cell membrane permeabilizing molecule as well as changing the electrical parameter. In the enhanced conditions, the electroporation frequency increased from 15 to 71 times compared to the initial conditions at the same DNA concentration. As the DNA concentration increased, the difference in the electroporation frequency between the two conditions became greater, and the curve of the enhanced conditions seemed to be saturated with a DNA concentration over $4{\mu}g$. The present study provided a clue to the recovery of the electroporation frequency with large DNA and formulated the relationship between the DNA concentration, the DNA size and the electroporation frequency in bifidobacteria. Therefore, this study will contribute to the expansion of molecular genetic studies of bifidobacteria.
